Development of a LAMP assay for the rapid visual detection of the emerging tick-borne Songling virus.

BACKGROUND: Songling virus (SGLV) within the genus Orthonairovirus, family Nairoviridae, is an emerging tick-borne virus associated with human febrile illness. However, no rapid detection method for SGLV has been established. METHODS: In this study, four primer sets targeting the nucleocapsid protein gene of SGLV were designed for use in the LAMP assay and evaluated to identify the optimal primer set. Recombinant plasmids were constructed and utilized for assessing the sensitivity of the assay. Tacheng tick virus 1 (TcTV-1)-, Beiji nairovirus (BJNV)-, Yezo virus (YEZV)-, severe fever with thr

The crystal structure of the Hazara virus nucleocapsid protein.

BACKGROUND: Hazara virus (HAZV) is a member of the Bunyaviridae family of segmented negative stranded RNA viruses, and shares the same serogroup as Crimean-Congo haemorrhagic fever virus (CCHFV). CCHFV is responsible for fatal human disease with a mortality rate approaching 30 %, which has an increased recent incidence within southern Europe. There are no preventative or therapeutic treatments for CCHFV-mediated disease, and thus CCHFV is classified as a hazard group 4 pathogen. In contrast HAZV is not associated with serious human disease, although infection of interferon receptor knockout mi

Seroprevalence of five arboviruses in Zebu cattle in the Central African Republic.

Sequential blood samples taken from 237 adult and 147 young Zebu cattle in the Central African Republic (CAR) were examined by enzyme-linked immunosorbent assay for the presence of antibodies to Rift Valley Fever (RVF), Dugbe (DUG), Crimean-Congo haemorrhagic fever (CCHF), Chikungunya (CHIK) and Wesselsbron (WSL) viruses. Antibodies to DUG and CCHF viruses were detected in 70% and 61% respectively, of the sera from adult cattle, probably due to common antigens. This hypothesis is supported by the fact that CCHF virus was isolated only 3 times from ticks, whereas DUG virus was isolated from alm

Genomic and phylogenetic profiling of RNA of tick-borne arboviruses in Hainan Island, China.

Ticks act as vectors and hosts of numerous arboviruses. Examples of medically important arboviruses include the tick-borne encephalitis virus, Crimean Congo hemorrhagic fever, and severe fever with thrombocytopenia syndrome. Recently, some novel arboviruses have been identified in blood specimens of patients with unexplained fever and a history of tick bites in Inner Mongolia. Consequently, tick-borne viruses are a major focus of infectious disease research. However, the spectrum of tick-borne viruses in subtropical areas of China has yet to be sufficiently characterized. In this study, we col

Detection of tick-borne bacteria and babesia with zoonotic potential in Argas (Carios) vespertilionis (Latreille, 1802) ticks from British bats.

Ticks host a wide range of zoonotic pathogens and are a significant source of diseases that affect humans and livestock. However, little is known about the pathogens associated with bat ticks. We have collected ectoparasites from bat carcasses over a seven year period. Nucleic acids (DNA and RNA) were extracted from 296 ticks removed from bats and the species designation was confirmed in all ticks as Argas (Carios) vespertilionis. A subset of these samples (n = 120) were tested for the presence of zoonotic pathogens by molecular methods. Babesia species, Rickettsia spp., within the spotted fev

Hemophagocytosis in a case with Crimean-Congo hemorrhagic fever and an overview of possible pathogenesis with current evidence.

Hemophagocytic lymphohistiocytosis (HLH) is a clinicopathologic condition characterized by high fever, hepatosplenomegaly, cytopenia, hyperferritinemia, and increased hemophagocytic macrophage proliferation and activation in the reticuloendothelial system. Primary HLH is familial and is a fatal disease that begins during early childhood. Secondary HLH may be acquired after intense activation of the immune system due to infection. Clinical and biologic symptoms result from cytokines secreted by T-lymphocytes and macrophages. Subtypes of primary HLH are caused by genetic defects in several cell

Development of an entirely cloned cDNA-based reverse genetics system for Tofla virus of orthonairovirus.

The genus Orthonairovirus includes highly pathogenic tick-borne viruses such as the Crimean-Congo hemorrhagic fever orthonairovirus (CCHFV). A reverse genetics system is an indispensable tool for determining the viral factors related to pathogenicity. Tofla orthonairovirus (TFLV) is a recently identified virus isolated from ticks in Japan and our research has suggested that TFLV is a useful model for studying pathogenic orthonairoviruses. In this study, we successfully established a reverse genetics system for TFLV using T7 RNA polymerase. Recombinant TFLV was generated by transfecting cloned

Crimean-Congo haemorrhagic fever.

Crimean-Congo haemorrhagic fever (CCHF) is one of the most widespread arboviroses in the world. It is present in Africa, south-east Europe, the Middle East and Asia. It is caused by a nairovirus (Bunyaviridae family) transmitted by several species of ticks. The geographical distribution of the disease coincides with the distribution of Hyalomma ticks. While infected livestock do not show signs of illness, humans are severely affected, with a high mortality rate. The most common symptoms are high fever, dizziness, headache, vomiting and haemorrhages. Pathogenesis studies in interferon-receptor-

An indirect enzyme-linked immunoassay (I-ELISA) based on the recombinant nucleocapsid protein (rNp) of Rift Valley fever virus was validated for the detection of specific IgG antibody in human sera. Validation data sets derived from testing sera collected in Africa (n=2967) were categorized according to the results of a virus neutralisation test. The assay had high intra- and inter-plate repeatability in routine runs. No detectable cross-reactions between IgG antibodies generated from mice experimentally infected with viruses representing genus Phlebovirus, Nairovirus, Orthobunyavirus and Bhan

Lethal C57BL/6 mouse model for Yezo virus infection and antiviral therapeutic evaluation.

Yezo virus (YEZV) is an emerging tick-borne orthonairovirus that causes febrile illness and hepatic injury in humans. However, its pathogenesis and effective antiviral interventions remain poorly characterized. In this study, we generated a lethal, mouse-adapted YEZV strain through 30 serial passages in C57BL/6 mice. The adapted strain successfully recapitulated key clinical and pathological features of human YEZV infection, including leukopenia, thrombocytopenia, and hepatitis, as evidenced by elevated serum transaminases, high hepatic viral loads, and histopathological necroinflammatory lesi

Crimean-Congo haemorrhagic fever virus in Kazakhstan (1948-2013).

Crimean-Congo haemorrhagic fever (CCHF) is a pathogenic and often fatal arboviral disease with a distribution spanning large areas of Africa, Europe and Asia. The causative agent is a negative-sense single-stranded RNA virus classified within the Nairovirus genus of the Bunyaviridae family. Cases of CCHF have been officially recorded in Kazakhstan since the disease was first officially reported in modern medicine. Serological surveillance of human and animal populations provide evidence that the virus was perpetually circulating in a local enzoonotic cycle involving mammals, ticks and humans i
