Adsorption mechanism of an antimicrobial peptide on carbonaceous surfaces: A molecular dynamics study.

Peptides are versatile molecules with applications spanning from biotechnology to nanomedicine. They exhibit a good capability to unbundle carbon nanotubes (CNT) by improving their solubility in water. Furthermore, they are a powerful drug delivery system since they can easily be uptaken by living cells, and their high surface-to-volume ratio facilitates the adsorption of molecules of different natures. Therefore, understanding the interaction mechanism between peptides and CNT is important for designing novel therapeutical agents. In this paper, the mechanisms of the adsorption of antimicrobi

The Use of Osteo-Inductive 3D-Printed Scaffolds Covered with a Pleiotrophin Peptide for Bone Defects: An In Vivo Experimental Study.

The present study investigated the effect of a 3D-printed nanocomposite scaffold on bone healing in vivo. The scaffolds used were made from the bioresorbable thermoplastic polycaprolactone polymer, blended with Multi-Walled Carbon Nanotubes functionalized with chitosan, and manufactured with a rectilinear infill pattern and interconnected pores of 500 μm in size. The study included three groups of 10 Wistar rats, in which a 2 mm bone defect was created in the middle of the right femur. In the scaffold/peptide group, the gap was filled with the scaffold loaded with a peptide corresponding to hu

Co-assembly and self-sorting are not mutually exclusive in enantiomeric peptide systems.

Co-assembly and self-sorting in multicomponent systems are typically treated as mutually exclusive outcomes. Here, we show that this distinction is incomplete. Using enantiomeric peptide nanotubes, we demonstrate that a single co-assembled structure forms only at an equimolar composition, yet remains internally self-sorted into compositionally distinct domains. Contrast-matched neutron scattering directly reveals this segregation and shows that the co-assembled structures adopt layered architectures rather than simple molecular-level mixing. These results establish that co-assembly and self-so

Nontoxic mRNA vehicles based on electrically neutral self-assembling peptides.

The cationic molecules and low-molecular-weight amphiphiles commonly used in nonviral gene delivery vehicles often induce adverse nonspecific effects, notably cytotoxicity and detergent-like unfavourable membrane activity. To address these limitations, here we report the development of a nontoxic mRNA-condensing agent based on electrically neutral self-assembling peptides (ENSPs). Driven by cooperative hydrogen bonding and hydrophobic interactions, ENSPs coassemble with mRNA into discrete nanotube-shaped complexes. ENSP-based mRNA vehicles are almost nontoxic to mammalian cells and exhibit neg

One-Pot Fabrication of Pentapeptide Bio-Aerogel via Direct Alcogelation.

Bio-aerogels are a class of ultra-lightweight, extremely porous, solid materials that are engineered from biopolymers, e.g., polysaccharides, proteins, or peptides. Such biomaterials are of interest for medical and environmental applications, such as molecular filtration, wound healing, or tissue engineering. In this context, ultrashort peptides, defined as sequences of up to ten amino acids, are attractive self-assembling building blocks for bio-aerogels, due to their low-cost synthesis, chemical tunability, and potential biocompatibility. Building on our previous report demonstrating the int

Self-Assembling Gemini-Type Amphiphiles From Helix-Loop-Helix Peptides.

Amphiphilic peptides can be designed to self-assemble into various nanostructures, such as nanorods, nanotubes, and vesicles, depending on their sequence and secondary structure. We have designed a self-assembling amphiphilic peptide that folds into a hairpin-like helix-loop-helix (HLH) motif. The α-helical segments are rich in hydrophobic leucine residues, while the loop segment is positively charged. The morphology of the peptide self-assemblies can be controlled by the preparation method. When prepared from an ethanolic solution at room temperature, these amphiphilic HLHPs self-assemble int

Ultrasensitive diagnosis for an anthrax-protective antigen based on a polyvalent directed peptide polymer coupled to zinc oxide nanorods.

A flexible poly-D-lysine polymer conjugated with different target-binding peptides is demonstrated with an ultralow concentration detection limit compared to those of other conventional detection systems. This polyvalent directed peptide polymer (PDPP) exhibits increased binding affinity and detects anthrax protective antigen at low levels using a well-known zinc oxide nanorod detection system.

Innovative Self-Assembly of 15-Mer Chimeric α-Peptide-Oligourea Foldamers toward Cl--Selective Ion Channels.

Constructing artificial ion channels is a challenging task. Herein, the de novo design of transmembrane ion channels made up of amphiphilic peptide-oligourea chimeric helices is described. They consist of an oligourea segment (7-mer) attached to the C-terminus of a short peptide (8-mer). Mass spectrometry (MS) and transmission electron microscopy (TEM) analyses show that in an aqueous solution, two of these chimeras (HPU-E and HPU-N) independently form defined oligomeric structures. TEM also shows that they form fiber bundles. The third related chimera HPU-F does not oligomerize (MS) but forms

Unique aggregation of anthrax (Bacillus anthracis) spores by sugar-coated single-walled carbon nanotubes.

There has been significant interest in the binding of anthrax spores by molecular species, but with only limited success. Proteins and more recently peptides were used. However, despite the known presence of carbohydrates on the spore surface, carbohydrate-carbohydrate interactions have hardly been explored likely because of the lack of required specific platform for synthetic carbohydrates. We report the successful use of single-walled carbon nanotubes as a truly unique scaffold for displaying multivalent monosaccharide ligands that bind effectively to anthrax spores with divalent cation medi

Tumour sensitization via the extended intratumoural release of a STING agonist and camptothecin from a self-assembled hydrogel.

Tumours with an immunosuppressive microenvironment respond poorly to therapy. Activation of the stimulator of interferon genes (STING) pathway can enhance intratumoural immune activation, but STING agonists are associated with high toxicity and degrade prematurely, which limits their effectiveness. Here, we show that the extended intratumoural release of the STING agonist cyclic di-AMP transforms the tumour microenvironment from immunosuppressive to immunostimulatory, increasing the efficacy of antitumour therapies. The STING agonist was electrostatically complexed with nanotubes comprising a

Interpreting the function of cell penetrating peptide (RGD) in drug transport to the cell membrane: a computational approach.

Carbon nanotubes (CNT) have unique properties that make them an excellent option for use as drug carriers. However, to make them safe for the human body, their walls are typically coated with a layer of peptide, which also helps to neutralize their toxicity. Additionally, a specific peptide sequence can be used to deliver therapeutic agents exclusively to cancer cells. In recent years, considerable progress has been made in the development of drug delivery systems (DDS) for drug delivery by computer-assisted. The present study inquires about the loading of ketoprofen (Ket) and naproxen (Nap) c

Biomolecular interactions of carbon nanotubes with amyloid-β proteins.

Carbon nanotubes (CNTs) have attracted considerable attention due to their prominent advantages of adsorption, tensile strength, electronic conductivity, and surface functionalization using proteins or functional groups. Recent studies suggested that carbon quantum dots, graphene, and hybrid materials doped with CNTs can effectively inhibit the fibrillization of amyloid-β proteins (Aβ), which represents a significant signal of early Alzheimer's disease (AD). The investigations on the CNT-Aβ interaction can elucidate the reaction mechanism and reveal the basic reaction phenomena of CNTs in AD p

Morphology Transition of Tripeptides from Porous Spherical to Rod-like Nanostructures for Small-Molecule Adsorption.

Peptides possess a remarkable propensity to adopt distinct morphologies, ranging from simple aggregates to complex structures such as fibrils and nanotubes. Morphology transformation in peptides is intricately linked to the physicochemical properties of peptides and external factors such as pH, temperature, and solvent conditions. Thus, it is more complex. Herein, we present four tripeptides, Boc-LVL-OMe (LVL), Boc-IVI-OMe (IVI), Boc-LVI-OMe (LVI), and Boc-IVL-OMe (IVL), which undergo direct morphological transformation (except for Boc-IVL-OMe) with time. The morphological transformation from

Incorporation of single-wall carbon nanotubes into an organic polymer monolithic stationary phase for mu-HPLC and capillary electrochromatography.

Single-wall carbon nanotubes (SWNT) were incorporated into an organic polymer monolith containing vinylbenzyl chloride (VBC) and ethylene dimethacrylate (EDMA) to form a novel monolithic stationary phase for high-performance liquid chromatography (HPLC) and capillary electrochromatography (CEC). The retention behavior of neutral compounds on this poly(VBC-EDMA-SWNT) monolith was examined by separating a mixture of small organic molecules using micro-HPLC. The result indicated that incorporation of SWNT enhanced chromatographic retention of small neutral molecules in reversed-phase HPLC presuma

Exploring the influence of carbon nanoparticles on the formation of β-sheet-rich oligomers of IAPP₂₂₋₂₈ peptide by molecular dynamics simulation.

Recent advances in nanotechnologies have led to wide use of nanomaterials in biomedical field. However, nanoparticles are found to interfere with protein misfolding and aggregation associated with many human diseases. It is still a controversial issue whether nanoparticles inhibit or promote protein aggregation. In this study, we used molecular dynamics simulations to explore the effects of three kinds of carbon nanomaterials including graphene, carbon nanotube and C₆₀ on the aggregation behavior of islet amyloid polypeptide fragment 22-28 (IAPP₂₂₋₂₈). The diverse behaviors of IAPP₂₂₋₂₈ peptid

Theoretical design of the cyclic lipopeptide nanotube as a molecular channel in the lipid bilayer, molecular dynamics and quantum mechanics approach.

In this article, cyclic peptides (CP) with lipid substituents were theoretically designed. The dynamical behavior of the CP dimers and the cyclic peptide nanotube (CPNT) without lipid substituents in the solution (water and chloroform) during the 50 ns molecular dynamic (MD) simulations has been investigated. As a result, the CP dimers and CPNT in a non-polar solvent are more stable than in a polar solvent and CPNT is a good container for non-polar small molecules such as chloroform. The effect of the lipid substituents on the CP dimers and CPNT has been investigated in the next stage of our s

Optimized titanium dioxide nanotubes for dental implants: Estimation of mechanical properties and effects on the biological behaviors of human gingival fibroblasts and oral bacteria.

The long-term successes of implant restorations rely on both appropriate osseointegration and robust soft tissue integration (STI). Numerous studies have reported that titanium dioxide nanotube (TNT) arrays formed by electrochemical anodization (EA) can promote early osteogenesis, but the mechanical stability of such modifications is often ignored and remains underexplored. In addition, relatively little research has been done on their effects on soft tissues integration. In this study, we developed mechanically robust TNT arrays using an optimized EA system. Subsequently, we immobilized a pep

Transfer of polyglutamine aggregates in neuronal cells occurs in tunneling nanotubes.

Huntington's disease (HD) is a dominantly inherited neurodegenerative disease caused by CAG expansion in the huntingtin gene, which adds a homopolymeric tract of polyglutamine (polyQ) to the encoded protein leading to the formation of toxic aggregates. Despite rapidly accumulating evidences supporting a role for intercellular transmission of protein aggregates, little is known about whether and how huntingtin (Htt) misfolding progresses through the brain. It has been recently reported that synthetic polyQ peptides and recombinant fragments of mutant Htt are readily internalized in cell culture

Atomistic organization and characterization of tube-like assemblies comprising peptide-polymer conjugates: computer simulation studies.

The structure and stability of the nanotube obtained by assembling peptide-polymer conjugates consisting of two poly(n-butyl acrylate) blocks coupled to the cyclic (D-alt-L)-octapeptide cyc[(L-Gln-D-Ala-L-Lys-D-Ala)2], have been investigated at the molecular level using atomistic molecular dynamics simulations. The effect of the wrapping polymer shells in the tube-like core, which consists of stacked beta-sheet cyclopeptides, has been examined by simulating assemblies of both unsubstituted cyclopeptides, and conjugates in chloroform and N,N-dimethylformamide solutions. Furthermore, the influen
