Necator americanus infection: a longitudinal study of an urban area in Nigeria.

A longitudinal study was carried out in Owerri, capital of Imo State, Nigeria, to assess the pattern of Necator americanus infection during a 27-month period. The mean annual prevalence fell from 59.0% in 1978, to 55.5% in 1979 and 51.1% in 1980. The egg count (apparent intensity of infection) fell in a similar pattern. Maximum and minimum conversion rates were 55.2 and 13.7%, respectively, and conversion rates fell by approximately 20% between 1979 and 1980. The highest reversion rate was 55.0% between 1979 and 1980. The population density of available third-stage larvae fluctuated seasonally

[Necator americanus uncinariasis in an infant born and residing in Mexico City].

A six-month-old infant is reported with the unquestionable diagnosis diagnosis of Necator americanus uncinariasis. The mother was also infected with the same hookworm. The patient was born and has always dwelt in Mexico City, where helminthiasis is considered extremely rare. The epidemiologic studies determined that the dwelling conditions were inadequate for the transmittal of the agent through the soil. The authors emphasize the importance of the presumptive diagnosis in those cases suggesting the disease and to carry out studies to confirm the existence of other infectious means

Antigen expression during development of the human hookworm, Necator americanus (Nematoda).

The accumulated and de novo synthesized antigens expressed by L3, L4 and adult Necator americanus, recognized by both the natural host, man, and the experimental host, the hamster, were identified by immunoblotting and immunoprecipitation analysis. Following infection of neonatal hamsters serum samples were taken on days 17, 35 and 117. Only serum taken 117 days after infection showed significant reactivity in immunoblotting experiments, recognizing adult epitopes of 30,000, 33,000, 48,000 and 69,000 mol. wt thereby suggesting that few accumulated antigens are shared between developmental stag

Necator americanus: optimization of the golden hamster model for testing anthelmintic drugs.

Laboratory golden hamsters (Mesocricetus auratus) were infected with Necator americanus under several different parasite and host conditions to optimize the model for testing anthelminthic drugs. The results confirmed that male hamsters were more susceptible to infection than females. Host age in the range of 5-15 weeks was not a factor that impacted on adult worm burden, and similar worm burdens were achieved using doses of 150, 250 or 500 N. americanus L3 (NaL3). The largest numbers of adult hookworms were recovered on days 21-28 post-infection, with a significant decrease at days 40-50 post

Construction and analysis of cDNA library of Necator americanus third stage larvae.

OBJECTIVE: To obtain the genetic information on Necator americanus and to search for the purpose genes. METHODS: mRNA was isolated from the third stage larvae of Necator americanus maintained in hamsters. Double strand cDNA was synthesized and ligated to lambda ZAPII vector to construct the cDNA library. Expressed sequence tages (ESTs) were obtained by single pass sequencing of randomly isolated cDNA clones from the established library. RESULTS: A cDNA library of N. americanus was successfully constructed with high recombinant efficiency. The titer of unamplified library was 1 x 10(7). The i

Density-dependent regulation of the growth of the hookworms Necator americanus and Ancylostoma ceylanicum.

Laboratory bred DSN hamsters were exposed to varying doses of infective larvae of Ancylostoma ceylanicum (orally) or Necator americanus (percutaneously) and were autopsied at times which corresponded to a period immediately before cessation of growth of worms or soon afterwards. A total of 829 (404 male and 425 female) A. ceylanicum and 1582 (781 male and 801 female) N. americanus were measured. At worm burdens of fewer than 100, the length of A. ceylanicum appeared to increase with infection intensity and no evidence was found that growth was retarded under crowded conditions. Effect of single-dose albendazole and single-dose mebendazole on Necator americanus.

One hundred two children (43 males and 59 females) aged 6 to 14 years with positive stool examination by Kato-Katz and/or Harada-Mori culture techniques for N. americanus, were randomly divided into two groups. Group I with 48 children were treated with a single dose albendazole, 400 mg. Group II, 54 children, received a single dose mebendazole, 600 mg. After treatment, repeated stool examination was performed on Day 14, Day 21 and Day 28. The children were considered cured when stool examination was negative on all three occasions by both methods. The cure rate was 64% in Group I and 11% in G

Antigen-induced lymphocyte blastogenesis in a hookworm (Necator americanus) infection in man.

A lymphocyte blastogenic technique was employed to examine cell-mediated immunity in a Necator americanus infection. Infective third-stage larvae of N. americanus were jsed for antigen preparation, and lymphocyte cultures were prepared from peripheral blood taken on days 3, 17, and 51 after infection with 250 third-stage larvae of N. americanus. At 3 days post-infection lymphocyte transformation was not induced; at 17 and 51 days after infection significant transformation was obtained.

The effect of novel aromatic heterocycle substituted aminamidine derivatives on Necator americanus.

BACKGROUND: The efficacy of current drugs against hookworms at a single dose is highly variable across regions, age groups and infection intensity. Extensive and repeated use of these drugs also leads to potential drug resistance. Therefore, novel drugs are required for sustained disease control. OBJECTIVES: Novel aromatic heterocycle substituted aminamidine derivatives (AADs) were synthesized based on tribendimine (TBD), and their in vivo potency against Necator americanus was tested. METHODS: The efficacy of the AADs was tested in male hamsters. Oral and IV pharmacokinetic parameters were

Metabolic alterations in the hamster co-infected with Schistosoma japonicum and Necator americanus.

Co-infection with hookworm and schistosomes is a common phenomenon in sub-Saharan Africa, as well as in parts of South America and southeast Asia. As a first step towards understanding the metabolic response of a hookworm-schistosome co-infection in humans, we investigated the metabolic consequences of co-infection in an animal model, using a nuclear magnetic resonance (NMR)-based metabolic profiling technique, combined with multivariate statistical analysis. Urine and serum samples were obtained from hamsters experimentally infected with 250 Necator americanus infective L(3) and 100 Schistoso

Dose-ranging study for trials of therapeutic infection with Necator americanus in humans.

Epidemiological studies suggest that a hookworm infection producing 50 eggs/gram of feces may protect against asthma. We conducted a dose-ranging study to identify the dose of hookworm larvae necessary to achieve 50 eggs/gram of feces for therapeutic trials of asthma. Ten healthy subjects without asthma or airway hyperresponsiveness to inhaled methacholine received 10, 25, 50, or 100 Necator americanus larvae administered double blind to an area of skin on the arm. Subjects were seen weekly for 12 weeks and were then treated with mebendazole. Skin itching at the entry site and gastrointestinal

Necator americanus in inbred mice: a re-evaluation of primary infection kinetics.

The course of a primary Necator americanus infection was studied in the lungs and small intestines of syngeneic mice. Following percutaneous infection no difference in initial larval establishment in the lungs was found between male BALB/c, NIH or B10.G mice. However, significant differences in the subsequent kinetics of infection were demonstrated between the BALB/c and NIH strains. Lung worm burdens declined more slowly in NIH mice than in BALB/c strain. Surprisingly, however, a greater proportion of larvae remaining in the lungs of BALB/c mice, 9 days p.i., were trapped than in NIH mice. Ne

The epidemiology and population biology of Necator americanus infection in a rural community in Zimbabwe.

Baseline data from an epidemiological study of hookworm infection in a rural community in Zimbabwe are presented. The infection status of an age-stratified sample of the community was assessed using anthelmintic expulsion techniques. Necator americanus was the only helminth parasite found to be present. The age-prevalence and intensity profiles rose asymptotically to an adult prevalence of about 80% and adult mean burden of 7.7 worms per host. The overall mean burden was 4.8 worms per host. The frequency distribution of N. americanus was overdispersed and well described by the negative binomia

Adherence of human eosinophils to infective filariform larvae of Necator americanus in vitro.

Eosinophilia is common in hookworm infection but the interaction between eosinophils and the larval stage of the parasite is poorly understood. The present study was conducted to test the ability of the eosinophils to adhere to infective filariform larvae of Necator americanus in vitro. Adherence of eosinophils to the larvae was found to be serum dependent. Antibody facilitated eosinophil adherence but this was maximal in the presence of complement. The adherence was greatly diminished by EGTA treated normal human serum (NHS) and was completely abolished when NHS was treated with either EDTA o

Necator americanus and Ancylostoma ceylanicum: development of protocols for dual infection in hamsters.

Two-day-old baby hamsters were infected initially with the infective larvae of hamster-adapted human hookworm, Necator americanus (NaL3). After a specified period they were again infected orally with infective larvae of Ancylostoma ceylanicum (AcL3). Three weeks after the second infection they were killed and the establishment of N. americanus and A. ceylanicum was assessed. The effect of different infection levels and exposure period of N. americanus on the concurrent establishment of A. ceylanicum was also examined. An infection with 50 NaL3 percutaneously, and 3 weeks later, a second infect
