A quantitative analysis of genomic instability in lymphoid and plasma cell neoplasms based on the PIG-A gene.

It has been proposed that hypermutability is necessary to account for the high frequency of mutations in cancer. However, historically, the mutation rate (mu) has been difficult to measure directly, and increased cell turnover or selection could provide an alternative explanation. We recently developed an assay for mu using PIG-A as a sentinel gene and estimated that its average value is 10.6 x 10(-7) mutations per cell division in B-lymphoblastoid cell lines (BLCLs) from normal donors. Here we have measured mu in human malignancies and found that it was elevated in cell lines derived from T c

Low Protein Z levels in patients with plasma cell neoplasms are inversely correlated with IL-6 levels.

UNLABELLED: Patients with multiple myeloma (MM) have an increased thrombotic risk, but pathogenesis remains uncertain. Low levels of Protein Z (PZ), a vitamin K-dependent plasma protein, are associated with venous as well as arterial thrombosis. The purpose of this study was to analyze PZ levels in patients with plasma cell neoplasms. PATIENTS AND METHODS: The study consisted of 64 plasma cells neoplasm patients and 42 healthy individuals. Clinical investigations included measurement of plasma PZ and IL-6 levels. RESULTS: PZ levels in patients with plasma cell neoplasms were significantly lo

[Concomitant myeloid sarcoma and plasma cell myeloma--case report of a hitherto undescribed association of two neoplasms].

A 65-year-old male patient initially presented with a penile tumour, identified as myeloid sarcoma. Subsequent bone marrow investigation revealed the presence of a synchronous plasma cell myeloma, without any evidence of a bone marrow-based myeloproliferative disorder. Although the course of both neoplasms was progressive, with increasing bone marrow involvement by the plasma cell myeloma and cutaneous dissemination of the myeloid sarcoma, both neoplasms remained confined to their original tissue compartments. Different biology of both tumours, including markedly divergent response to therapie

Andrographolide Induces Ferroptosis-associated Cytotoxicity and PUFA/MUFA Phospholipid Remodeling in Plasma Cell Neoplasm Cell Lines.

BACKGROUND/AIM: Andrographolide (Andro), a diterpene lactone from Andrographis paniculata, induces reactive oxygen species (ROS)-dependent apoptosis in hematologic malignancy cells. This study investigated whether Andro also induces ferroptosis-associated cytotoxicity in plasma cell neoplasm cell lines. MATERIALS AND METHODS: H929 and ARH77 cells were treated with Andro in the presence or absence of Ferrostatin-1 (Fer-1). Cell viability, ROS generation, Annexin V positivity, intracellular iron, lipid peroxidation, FACL4/GPX4 expression, nuclear factor kappa B (NF-κB)/cyclooxygenase-2 (COX-2)

Evaluating murine double minute 2 status as a stratification tool for risk-adapted management in plasma cell neoplasms.

BACKGROUND: The E3 ubiquitin ligase murine double minute 2 (MDM2) is a key negative regulator of the tumor suppressor protein p53 and has been implicated in the development of various cancers, including hematologic malignancies. In multiple myeloma (MM), increased MDM2 expression has been reported and may play a role in disease progression and resistance to therapy. Despite this, the prognostic implications of MDM2 detected through immunohistochemistry (IHC) remain insufficiently defined. AIM: To evaluate the clinical, pathological, and prognostic significance of MDM2 expression in plasma cel

Primary exploration of cell-free DNA in the plasma of patients with parathyroid neoplasms using next-generation sequencing.

BACKGROUND AND AIMS: Plasma cell-free DNA (cfDNA) has been used to monitor gene mutations and diagnose tumors. Discriminating parathyroid carcinoma (PC) from parathyroid adenoma (PA) before surgery is difficult because of the overlap in clinical features between parathyroid neoplasms. We aimed to detect cfDNA mutations in plasma samples from PC and PA patients before surgery to predict the CDC73 status in tumor tissue and help in the differential diagnosis of parathyroid neoplasms. MATERIALS AND METHODS: Eighteen PC patients and 13 PA patients were enrolled. Plasma cfDNA was detected using ne

Gastrointestinal bleeding as initial presentation of extramedullary plasma cell neoplasms: A case report and review of the literature.

BackgroundPlasma-cell neoplasms rarely involve the gastrointestinal tract and manifest as gastrointestinal bleeding. Plasmablastic myeloma is an aggressive plasma cell neoplasm associated with poor outcomes. A small number of cases with gastrointestinal involvement is reported in the literature and therefore high index of suspicion is essential for avoiding delays in diagnosis and treatment.Case summaryOur aim is to present our experience of a 70-year-old patient with a secondary presentation of plasmablastic myeloma manifesting as unstable upper gastrointestinal bleeding and to review the lit

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) induces peripheral blood abnormalities and plasma cell neoplasms resembling multiple myeloma in mice.

Although epidemiologic studies have suggested a possible association between occupational exposures to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and the risk of development of multiple myeloma, definitive evidence in support of this association is lacking. In the present study, we employed the Vk*Myc mouse model of multiple myeloma to assess the impact of TCDD exposure on multiple myeloma pathogenesis. TCDD induced splenomegaly and multiple peripheral blood abnormalities, including anemia and high serum IgG levels. In addition, TCDD triggered bone lytic lesions, as well as renal tubular casts

An unusual case of a plasma cell neoplasm with an IgG3lambda myeloma and a gamma3 heavy chain disease protein.

A unique case of gamma3 heavy chain disease with two related serum proteins is reported. One molecule appears to be an IgG3lambda myeloma protein. The second molecule is a dimer of a shortened gamma3 heavy chain that has an unblocked amino terminus and lacks the VH and CH1 domains. Its probable origin as a synthetic product is discussed. The clinical and pathologic features of this patient resemble those of other patients with gamma heavy chain disease more than those of patients with multiple myeloma. It seems likely that the heavy chain disease protein is the result of a mutational event in

Localized Tonsillar Amyloidosis Associated With a Plasma Cell Neoplasm Presenting as an Oropharyngeal Mass: A Diagnostic Challenge.

Amyloidosis is a rare and heterogeneous group of disorders characterized by extracellular deposition of misfolded proteins, which may present as either localized or systemic disease. In the head and neck region, the presentation of amyloidosis is uncommon and may clinically mimic neoplastic processes. We report the case of a 58-year-old man with a five-year history of progressive oropharyngeal symptoms, initially perceived as a foreign body sensation and intermittent odynophagia, later evolving to dysphagia to solids. Physical examination revealed marked asymmetry of the palatine tonsils due t

Case Report: Concomitant Diagnosis of Plasma Cell Leukemia in Patient With JAK2 Positive Myeloproliferative Neoplasm.

Plasma cell dyscrasias and myeloproliferative neoplasms (MPN) are hematologic malignancies arising from two distinct hematopoietic cell lineages. They rarely occur concomitantly. Here, we report a case of a patient with a recent diagnosis of a JAK2 V617F positive MPN who presented with a new diagnosis of plasma cell leukemia. The patient had presented to the hospital with a leukocytosis predominantly comprised of plasma cells, followed by work-up involving peripheral blood flow cytometry, FISH analysis, and bone-marrow biopsy. FISH analysis was suggestive of a common progenitor cell for these

Risk stratification of plasma cell neoplasm: insights from plasma cell-specific cytoplasmic immunoglobulin fluorescence in situ hybridization (cIg FISH) vs. conventional FISH.

UNLABELLED: We directly compared the results of routine fluorescence in situ hybridization (FISH) and plasma cell-specific cytoplasmic immunoglobulin (cIg) FISH from 75 paired samples for myeloma risk stratification. CIg FISH improves test specificity and sensitivity and tends to eliminate borderline results. It proves that most plasma cells (PCs) consistently carry the abnormality in myelomas with an IGH translocation, whereas routine FISH detects these cells only at variably low levels. BACKGROUND: Routine cytogenetic analysis of plasma cell neoplasms (PCNs) has a low sensitivity. Conventio

Low cholesterol levels are associated with increasing risk of plasma cell neoplasm: A UK biobank cohort study.

BACKGROUND: Plasma cell neoplasms are a group of hematologic neoplasms that often develop in the elderly population. The relationship between cholesterol levels and hematologic malignancy has been identified in population studies. However, it is still unclear if there is a relationship between cholesterol levels and plasma cell neoplasm in European ancestry. METHODS: Prospective cohorts included 502,507 individuals from the UK Biobank who were followed up to 2019 and assessed total cholesterol(TC) levels, low-density lipoprotein (LDL) levels, high-density lipoprotein (HDL) levels, apolipoprot
