New molecular tools in Neospora caninum for studying apicomplexan parasite proteins.

Источник пометил публикацию как отозванную

The development of molecular genetics has greatly enhanced the study of the biology and pathology associated with parasites of the phylum Apicomplexa. We have established a system specifically designed for Neospora caninum, and used this system as a heterologous platform for the expression of foreign genes. Plasmid constructs containing fluorescent proteins or targeted genes of Toxoplasma gondii, driven by N. caninum promoters, have yielded robust expression and correct trafficking of target gene products as assessed by immunofluorescence assays and Western blot analyses. Using this approach,

Neospora caninum and Toxoplasma gondii serodiagnosis in human immunodeficiency virus carriers.

INTRODUCTION: Neospora caninum and Toxoplasma gondii belong to the Sarcocystidae family, and both have one definitive and various intermediary hosts. Owing to their weak immune systems, immunocompromised persons might be prone to opportunistic infections. The aim of this study was to investigate the presence of anti- N. caninum and anti- T. gondii antibodies in immunocompromised individuals. METHODS: This cross-sectional study investigated the rates of N. caninum and T. gondii , as assessed using immunofluorescent antibody reaction (IFAT) with 1:50 and 1:16 dilution, respectively, in patients

The Distribution of Neospora caninum Secretory Proteins in Mouse and Calf Brains.

Neospora caninum, as well as Toxoplasma gondii, secrete proteins that facilitate the invasion of host cells and the regulation of host immune response and metabolism. However, the localization of the secretory proteins in infected animal brains has not been studied in detail. Here, we investigate the brain and intracellular distribution of the secretory proteins in experimentally infected mice and naturally infected calves through histopathology and immunohistochemistry (IHC) to detect surface antigen 1 (NcSAG1), cyclophilin (NcCYP), profilin (NcPF), dense granule protein 6 (NcGRA6), and NcGRA

Molecular investigation of Neospora caninum in cattle in the Khomas region of Namibia.

Neosporosis is a leading cause of bovine abortions worldwide, with a substantial economic impact because of reproductive losses, costing the cattle industry billions of dollars annually. Despite the global significance of Neospora caninum, molecular studies on this protozoan parasite have yet to be conducted in cattle in Southern Africa, leaving a critical knowledge gap in countries like Namibia, where beef production is vital to the economy. This study aimed to detect latent N. caninum infections in beef cattle brain and heart muscle samples collected from abattoirs and blood samples from Neo

Seroprevalence of Toxoplasma gondii and Neospora caninum infections in cattle in Mongolia.

Toxoplasma gondii and Neospora caninum are protozoan parasites that cause huge economic losses in animal industries worldwide. N. caninum can cause abortion storms and high culling rates in cattle, whereas T. gondii infection is a significant concern in both human and animals because it can induce abortion and clinical symptoms in immunocompromised hosts. The aim of this study was to determine the seroprevalence of T. gondii and N. caninum in cattle in Mongolia. Specific antibodies to T. gondii and N. caninum were detected by using an indirect enzyme-linked immunosorbent assay (iELISA) based o

Identification and characterization of GRA6/GRA7 of Neospora caninum in MDBK cells.

Neospora caninum, an apicomplexan parasite, is recognized as a major bovine abortifacient. Dense granule antigens (GRAs) play important roles in the formation and modification of parasitophorous vacuoles (PVs) in Toxoplasma gondii. However, a few studies investigating GRAs have been reported in N. caninum. The aim of the present study was to characterize the dense GRA6/GRA7 of N. caninum in PVs using MDBK cells as a host cell model. Neospora caninum was inoculated into MDBK cells, and changes were observed using a transmission electron microscope (TEM). Neospora caninum GRA6/GRA7 were identifi

Neospora caninum infection as a cause of reproductive failure in a sheep flock.

Neospora caninum has been detected only sporadically in cases of ovine abortion, and it has therefore traditionally been considered as an unimportant parasite in small ruminants. This study was carried out with the aim of identifying the pathogen causing serious reproductive problems on a commercial sheep farm. Sera from all rams and ewes tested negative for antibodies against Border disease virus, Schmallenberg virus and Coxiella burnetii, and infections by these agents were therefore ruled out. Nevertheless, seropositivity to N. caninum and/or Toxoplasma gondii was detected, although the ser

Genetic susceptibility to Neospora caninum infection in Holstein cattle in Ontario.

Neospora caninum has been reported to be an important cause of endemic fetal loss and occasional abortion epidemics in cattle around the world. This study examined 12,016 sera collected from 9723 Holstein cows on 125 herds across Ontario in 1998, 1999, and 2000. An enzyme-linked immunosorbent assay was used to examine the sera for antibodies to N. caninum. The overall prevalence of N. caninum antibodies in the cattle was 11.2% and the prevalence in individual herds varied from 0 to 70.4%. Among 3109 daughter-dam pairs, 619 positive dams had 252 positive daughters, giving a detected vertical tr

Seroprevalence of Neospora caninum Infection in Shelter Dogs from Hanoi, Vietnam.

Neosporosis, caused by N. caninum, is an emerging protozoan disease responsible for significant economic losses in the global dairy and meat industries, primarily due to abortion in cattle. Dogs serve as both definitive and intermediate hosts and play a key role in the parasite transmission cycle. Currently, effective control strategies remain limited, partly due to insufficient information on infection status. In Vietnam, data on N. caninum infection are scarce and mainly limited to cattle and buffalo. In this study, an indirect enzyme-linked immunosorbent assay (iELISA) based on recombinant
