Optogenetic approaches for investigating neural pathways implicated in schizophrenia and related disorders.

Optogenetic approaches have been rapidly adopted by neuroscientists in order to control the activity of neurons with high temporal, spatial and genetic specificity. By expressing light-sensitive microbial opsins within a genetically-specified population of neurons, flashes of light can be used to activate these opsins and thereby modulate the targeted cells in a spatially and temporally defined manner. Thus, optogenetics can be used to activate very specific sets of neurons or projections at particular times, either within freely behaving animals, or in reduced preparations such as brain slice

Studies of the central neural pathways to the stomach and Zusanli (ST36).

The purpose of this morphological study was to investigate the relation between the meridian, meridian points and viscera using neuroanatomical tracers. The common locations of the spinal cord and brain projecting to the stomach and Zusanli were observed following injection of CTB (cholera toxin B subunit) and pseudorabies viruses (PRV-Ba, Bartha strain and PRV-Ba-Gal, galactosidase insertion) into the stomach and Zusanli (ST36). After 4-5 days of survival following injection into twelve rats, they were perfused, and their spinal cords and brains were frozen sectioned (30 microm). These sectio

Poliovirus spreads from muscle to the central nervous system by neural pathways.

A transgenic mouse model was used to address an unsolved question in the pathogenesis of poliomyelitis: how poliovirus invades the central nervous system (CNS). LD50 values for intramuscular and intracerebral inoculation of poliovirus in transgenic mice expressing poliovirus receptors (TgPVR mice) were similar. After intramuscular inoculation with poliovirus, paralysis was observed first in the inoculated limb. In contrast, localization of initial paralysis to the inoculated limb was not observed in normal mice inoculated intramuscularly with the mouse-adapted P2/Lansing poliovirus strain. Aft

NTS Neural Subtypes and Satiety Pathways: taVNS as a Novel Neuromodulatory Intervention.

The global obesity epidemic has become one of the most severe public health challenges of the 21st century, and existing treatment strategies have limitations such as poor long-term adherence, significant side effects, or high invasiveness. Dysregulation of feeding behavior is the core driver of obesity, and its precise regulation depends on the complete transmission of gut-brain axis signals. The nucleus of the solitary tract (NTS), as the first central relay station for visceral afferent signals, is a key hub for the regulation of feeding homeostasis. This article reviews the peripheral sign

Gut instincts: Multi-level control and neural pathways involved in gastrointestinal interoception.

Interoception is the process by which the nervous system senses and regulates the body's internal state to maintain homeostasis. Interoceptive processes in the gastrointestinal tract are carried out by multiple levels of sensory-motor control that are organized into hierarchical neural circuits, with different levels of command being engaged under various circumstances. The enteric nervous system provides local intrinsic control, initiating and sustaining functions like peristalsis and secretory reflexes. To coordinate motility and secretions across distant regions of the gut, the ENS recruits

Regional heterogeneity of brain insulin resistance: From molecular pathways to neural circuits.

Brain insulin resistance (BIR) has emerged as a critical link between metabolic dysfunction and cognitive and behavioral impairment. As the twin epidemics of metabolic and brain disorders accelerate globally, understanding the precise mechanisms of BIR has become a clinical and scientific priority. However, growing evidence indicates that BIR does not manifest uniformly across the central nervous system. Instead, insulin signaling dysfunction exhibit marked regional and circuit-specific heterogeneity across the hippocampus, prefrontal cortex, hypothalamus, amygdala, and mesolimbic reward pathw

Impact of emotional trauma on brainstem: Unlocking the effects on neural pathways.

The brainstem, far from being a simple relay center, emerges as a sophisticated processor of emotional trauma, orchestrating complex neural responses that reshape our understanding of trauma biology. This chapter explores the convoluted relationship between emotional trauma and brainstem function, revealing how traumatic experiences trigger precise quantum-level changes in neural circuits. Through examining chronobiological rhythms and neuroimmune interactions, we uncover the dynamic nature of trauma-induced brainstem adaptations. Our exploration extends to the fascinating brainstem-gut axis,

CHD8 regulates neurodevelopmental pathways associated with autism spectrum disorder in neural progenitors.

Truncating mutations of chromodomain helicase DNA-binding protein 8 (CHD8), and of many other genes with diverse functions, are strong-effect risk factors for autism spectrum disorder (ASD), suggesting multiple mechanisms of pathogenesis. We explored the transcriptional networks that CHD8 regulates in neural progenitor cells (NPCs) by reducing its expression and then integrating transcriptome sequencing (RNA sequencing) with genome-wide CHD8 binding (ChIP sequencing). Suppressing CHD8 to levels comparable with the loss of a single allele caused altered expression of 1,756 genes, 64.9% of which

Neural tube development requires the cooperation of p53- and Gadd45a-associated pathways.

BACKGROUND: Numerous genetically engineered mouse models for neural tube defects (NTDs) exist, and some of the implicated proteins are functionally related. For example, the growth arrest and DNA damage-inducible protein Gadd45a and tumor suppressor p53 are functionally similar, and both are involved in neural tube development (Gadd45a- and Trp53-null embryos show low levels of exencephaly). To assess their roles in neural tube development, we generated double-null mice from Gadd45a- and Trp53-null mice, as well as from cyclin-dependent kinase inhibitor (Cdkn1a) (p21)-null and xeroderma pigmen

Neural pathways regulating Brunner's gland secretion in guinea pig duodenum in vitro.

This study examined the neural pathways innervating Brunner's glands using a novel in vitro model of acinar secretion from Brunner's glands in submucosal preparations from the guinea pig duodenum. Neural pathways were activated by focal electrical stimulation and excitatory agonists, and videomicroscopy was used to monitor dilation of acinar lumen. Electrical stimulation of perivascular nerves evoked large dilations that were blocked by TTX (1 microM) or the muscarinic receptor antagonist 4-diphenylacetoxy-N-(2-chloroethyl)-piperidine hydrochloride (1 microM). The nicotinic agonist 1,1-dimethy

Selective Neural Pathway Targeting by Using Lentiviral Vectors.

Animal behaviors are controlled through a complex neural network in the brain. To explore the nature of brain functions, it is critical to investigate the behavioral and physiological roles of specific neuronal types that comprise this network. Immunotoxin (ITX)-mediated cell targeting is a technology designed to eliminate specific cell types that are genetically engineered to express a foreign receptor, utilizing the cytotoxic activity of a recombinant ITX. Lentiviral vectors with highly efficient retrograde gene transfer (HiRet) and neuron-specific retrograde gene transfer (NeuRet) propertie

Human assembloid model of the ascending neural sensory pathway.

Somatosensory pathways convey crucial information about pain, touch, itch and body part movement from peripheral organs to the central nervous system1,2. Despite substantial needs to understand how these pathways assemble and to develop pain therapeutics, clinical translation remains challenging. This is probably related to species-specific features and the lack of in vitro models of the polysynaptic pathway. Here we established a human ascending somatosensory assembloid (hASA), a four-part assembloid generated from human pluripotent stem cells that integrates somatosensory, spinal, thalamic a

Neural Correlates of the Dual-Pathway Model for ADHD in Adolescents.

OBJECTIVE: The dual-pathway model has been proposed to explain the heterogeneity in symptoms of attention deficit hyperactivity disorder (ADHD) by two independent psychological pathways based on distinct brain circuits. The authors sought to test whether the hypothesized cognitive and motivational pathways have separable neural correlates. METHODS: In a longitudinal community-based cohort of 1,963 adolescents, the neuroanatomical correlates of ADHD were identified by a voxel-wise association analysis and then validated using an independent clinical sample (99 never-medicated patients with ADH

Neural inhibition of inflammation: the cholinergic anti-inflammatory pathway.

The innate immune system is activated by infection and injury to release pro-inflammatory cytokines, which activate macrophages and neutrophils and modulate specific cellular responses. The magnitude of the cytokine response is critical, because a deficient response may result in secondary infections, while an excessive response may be more injurious than the original insult. We recently described a neural pathway, termed the "cholinergic anti-inflammatory pathway", that reflexively monitors and adjusts the inflammatory response by inhibiting pro-inflammatory cytokine synthesis. Inhibitory neural pathway regulating gastric emptying in rats.

The relaxation of the pylorus is one of the most important factors for promoting gastric emptying. However, the role of inhibitory neurotransmitters in the regulation of pyloric relaxation and gastric emptying remains unclear. In this study, we investigated the effects of NO biosynthesis inhibitor, N(G)-nitro-L-arginine methyl ester (L-NAME), and calcium dependent potassium channel blocker, apamin, on vagal stimulation-induced pyloric relaxation and gastric emptying in rats. Olfactory neural pathway in mouse hepatitis virus nasoencephalitis.

The mechanism of brain infection with mouse hepatitis virus-JHM was studied in BALB/cByJ mice following intranasal inoculation, and found to be a consequence of direct viral spread along olfactory nerves into olfactory bulbs of the brain. Infection was followed sequentially from nose to brain, using microscopy, immunohistochemistry and virus quantification. Lesions, antigen and virus were observed in the olfactory bulb and anterior brain as early as 2 days and posterior brain by 4 days after inoculation. Viral antigen extended through nasal mucosa into submucosa, then coursed along the olfacto

Inhibitory control-related neural pathways of early adversity to internalizing problems in preadolescence.

OBJECTIVE: Exposure to early life adversity has long-reaching effects on the psychopathology of adolescents. However, some individuals exposed to early life stressors, such as threat and deprivation, do not later present with symptoms. Previous research suggests that neural mechanisms may play a role in differentiating pathways from early life threat or deprivation to internalizing symptoms. METHOD: This study utilizes neuroimaging to examine the relationship between early adversity experiences and internalizing symptoms later on. Data from the ABCD® study were used to investigate whether inh

Unveiling the Neural Pathways of Empathic Pain: Implications for Modulation Strategies and Beyond.

Empathic pain, a core manifestation of empathy, encompasses an individual's perceptual sensitivity, evaluative judgment, and emotional responsiveness to the suffering of others. This capacity is vital for healthy social interaction, as it fosters prosocial behavior, inhibits aggression, and upholds moral norms critical to social development. This article provides a comprehensive review of current research on empathic pain, examining its conceptual foundations and the underlying neural mechanisms. In particular, it explores the activation of key brain regions, including the anterior cingulate c

Effect of Stress-Related Neural Pathways on the Cardiovascular Benefit of Physical Activity.

BACKGROUND: The mechanisms underlying the psychological and cardiovascular disease (CVD) benefits of physical activity (PA) are not fully understood. OBJECTIVES: This study tested whether PA: 1) attenuates stress-related neural activity, which is known to potentiate CVD and for its role in anxiety/depression; 2) decreases CVD in part through this neural effect; and 3) has a greater impact on CVD risk among individuals with depression. METHODS: Participants from the Mass General Brigham Biobank who completed a PA survey were studied. A subset underwent 18F-fluorodeoxyglucose positron emission
