DNA-Modified Nanowire Thermometer Uncovers Thermal Responses of Neuronal Soma and Neurites to H2O2 Treatment.

Determining the temperature changes of the soma and the neurites with high spatial resolution would be of significance for investigating physiological activities and abnormal protein aggregation or misfolding, which can further offer insights into the underlying mechanisms of neurodegenerative diseases. In this work, we utilized a DNA hairpin structure with tetramethylrhodamine at its 3' end to modify the surface of silicon nanowires (SiNWs) and obtained a SiNW-based thermometer. The thermometer exhibited high spatial resolution, excellent temperature response, favorable anti-interference capa

Selective PrP-like protein, doppel immunoreactivity in dystrophic neurites of senile plaques in Alzheimer's disease.

Doppel (Dpl) is a prion-like protein encoded by the gene PRND, which has been found downstream of the prion gene PRNP in several species. The present study examines by immunohistochemistry Dpl expression in brain samples from 10 patients with Alzheimer's disease (AD), three patients with Pick's disease, four patients with Parkinson's disease, eight patients with diffuse Lewy body disease (DLBD), six patients with sporadic Creutzfeldt-Jakob disease (CJD) methionine/methionine at the codon 129, two patients with sporadic CJD methionine/valine at the codon 129 and numerous kuru plaques in the cer

Xenopus temporal retinal neurites collapse on contact with glial cells from caudal tectum in vitro.

Nasal and temporal retinal neurites were confronted in culture with glial cells from the rostral and caudal parts of the optic tectum and with glial cells from the diencephalon. Twenty of each of the six classes of encounter between individual growth cones and isolated glial cells were analysed by time-lapse videorecording. The results show that growth cones from the temporal retina collapse when they contact glial cells from the caudal tectum, but do not collapse when they contact glia from other areas. Growth cones of nasal retinal fibres do not collapse on contact with any of the glial type

Ferroptosis and pathogenesis of neuritic plaques in Alzheimer disease.

Neuritic plaques are pathognomonic and terminal lesions of Alzheimer disease (AD). They embody AD pathogenesis because they harbor in one space critical pathologic features of the disease: amyloid deposits, neurofibrillary degeneration, neuroinflammation, and iron accumulation. Neuritic plaques are thought to arise from the conversion of diffuse extracellular deposits of amyloid-β protein (Aβ), and it is believed that during conversion, amyloid toxicity creates the dystrophic neurites of neuritic plaques, as well as neurofibrillary tangles However, recent evidence from human postmortem studies

Neuritic Plaques - Gateways to Understanding Alzheimer's Disease.

Extracellular deposits of amyloid-β (Aβ) in the form of plaques are one of the main pathological hallmarks of Alzheimer's disease (AD). Over the years, many different Aβ plaque morphologies such as neuritic plaques, dense cored plaques, cotton wool plaques, coarse-grain plaques, and diffuse plaques have been described in AD postmortem brain tissues, but correlation of a given plaque type with AD progression or AD symptoms is not clear. Furthermore, the exact trigger causing the development of one Aβ plaque morphological subtype over the other is still unknown. Here, we review the current knowl

Neurite outgrowth in retinal ganglion cell culture.

Retinal ganglion cells (RGC) are the projection neurons of the eye. The RGC is the primary cell type injured in a variety of diseases of the optic nerve, including glaucoma and optic neuritis. The most well-established extrinsic signal of RGC survival and axonal outgrowth is the neurotrophin brain-derived neurotrophic factor. An immunopurification system has been adapted in order to filter large enough quantities of RGCs from the mixed population of retinal neurons in order to perform high-throughput screening in a 96-well format. Using this assay, the screening of a combinatorial chemical lib

Is silica involved in neuritic (senile) plaque formation?

The agent responsible for inducing neuritic (senile) plaque formation in senile dementia of the Alzheimer's type and in the ageing non-demented brain is unknown. Other workers have detected a high concentration of silicon in the rims and cores of senile neuritic plaques. We have therefore looked at whether the reaction of brain tissue to silica particles resembles a neuritic plaque. In this study both fine (10 nm) and coarse (less than 5 microns) particles of silica have been introduced into the brains of rats and mice using a wide range of doses and several methods of administration. The reac

Sp1 in Astrocyte Is Important for Neurite Outgrowth and Synaptogenesis.

In this study, we found that Sp1 was highly expressed in astrocytes, implying that Sp1 might be important for the function of astrocytes. Sp1/GFAP-Cre-ERT2 conditional knockout mice were constructed to study the role of Sp1 in astrocytes. Knockout of Sp1 in astrocytes altered astrocytic morphology and decreased GFAP expression in the cortex and hippocampus but did not affect cell viability. Loss of Sp1 in astrocytes decreased the number of neurons in the cortex and hippocampus. Conditioned medium from primary astrocytes with Sp1 knockout disrupted neuronal dendritic outgrowth and synapse forma

Fasciculation and defasciculation of neurite bundles on micropatterned substrates.

We describe experiments of fasciculation, i.e., bundling, of chick sensory neurites on 2D striped substrates. By Fourier decomposition, we separate left-going and right-going neurite components from in vitro images, and we find first that neurite bundles orient toward preferred angles with respect to the stripe direction, and second that in vitro bundles travel in leftward and rightward directions nearly uninterrupted by crossings of bundles traveling in the opposing direction. We explore mechanisms that lead to these behaviors, and summarize implications for future models for neurite outgrowt

A conditioning lesion enhances sympathetic neurite outgrowth.

Axonal regeneration can be influenced by a conditioning lesion (an axonal injury made prior to a second test lesion). Previously, sympathetic neurons in vivo were shown to respond to a conditioning lesion with decreased neurite outgrowth, in contrast to the enhanced outgrowth observed in all other peripheral neurons examined. The present experiments tested the effects of a conditioning lesion on neurite outgrowth in vitro from the superior cervical ganglion (SCG) and the impact of several factors on that response. Ganglia axotomized 1 week earlier and then explanted in Matrigel or collagen gel

Primary neuritic leprosy in a black South African.

A case of primary neuritic leprosy in a black South African is described, in which the multiple peripheral nerves were affected. The clinical picture and electrophysiological studies are in keeping with a picture of mononeuritis multiplex. Selective involvement of the facial nerve branches with normal blink reflex latencies was observed. The biopsy of the sural nerve disclosed features most consistent with borderline leprosy.

Immunohistological analysis of nerve granulomas in neuritic leprosy.

Immunohistological analysis of infiltrates of nerves in patients with neuritic leprosy was carried out using monoclonal antibodies defining T cell subsets, Langerhans cells, HLA DR antigens, and indirect immunofluorescence. In all, eight nerves were analyzed. 2 of the 8 nerves showed epithelioid cell granulomas surrounded by large numbers of lymphocytes. The predominant lymphocytes in these granulomas were activated T cells expressing CD3 and HLA DR antigens. The proportion of CD3+ and CD4+ cells was higher than that of CD8+ cells. The ratio of CD4+/CD8+ cells in these two biopsy specimens was

Somatostatin immunoreactivity in neuritic plaques of Alzheimer's patients.

Senile dementia of the Alzheimer's type can be diagnosed with certainty only by examining neurofibrillary tangles and neuritic plaques under the microscope. Recently, it has been suggested that the condition is linked to specific neurotransmitter systems, with a decline of cortical acetylcholine, choline acetyltransferase, cholinergic neurones projecting to the cortex, cortical noradrenaline content, locus coeruleus neurones and cortical somatostatic content. Using immunocytochemical methods, we here report that somatostatin-immunoreactive processes are present in neuritic plaques in human Alz

Infectious etiology of neuritic (senile) plaques in mice.

Brains of inbred female VM mice infected with scrapie agent were studied with the use of the Bodian silver impregnation method and by electron microscopy. In brains affected with scrapie, after an incubation period of between 587 and 655 days, numerous primitive, classical, and amyloid plaques were found. No plaques of any type were seen in the control mice.

Inhibition of dynein but not kinesin induces aberrant focal accumulation of neurofilaments within axonal neurites.

Studies from several laboratories indicate that the microtubule motors kinesin and dynein respectively participate in anterograde and retrograde axonal transport of neurofilaments. Inhibition of dynein function by transfection with a construct expressing dynamitin or intracellular delivery of anti-dynein antibodies accelerates anterograde transport, which has been interpreted to indicate that the opposing action of both motors mediates the normal distribution of neurofilaments along axons. Herein, we demonstrate that, while expression of relatively low levels of exogenous dynamitin indeed acce

Cholinergic differentiation of rat sympathetic neurons in culture: effects of factors applied to distal neurites.

Cholinergic properties are induced in sympathetic neurons by several factors applied to entire neurons in culture. Evidence from work with the rat sweat gland model indicates that factors located in target tissues can induce cholinergic differentiation in vivo. We now report that when leukemia inhibitory factor (LIF), heart cell-conditioned medium (HCCM), or dermal fibroblast-conditioned medium (DFCM) is applied to only distal neurites in compartmented cultures of rat sympathetic neurons, the neurons exhibit an increase in specific choline acetyltransferase activity and a concomitant decrease
