Neurons as will and representation.

Memory recollections and voluntary actions are often perceived as spontaneously generated irrespective of external stimuli. Although products of our neurons, they are only rarely accessible in humans at the neuronal level. Here I review insights gleaned from unique neurosurgical opportunities to record and stimulate single-neuron activity in people who can declare their thoughts, memories and wishes. I discuss evidence that the subjective experience of human recollection and that of voluntary action arise from the activity of two internal neuronal generators, the former from medial temporal lo

Potassium channels in omnipause neurons.

Potassium (K+) channels are major contributors to fast and precise action potential generation. The aim of this study was to establish the immunoreactivity profile of several potassium channels in omnipause neurons (OPNs), which play a central role in premotor saccadic circuitry. To accomplish this, we histochemically examined monkey and human brainstem sections using antibodies against the voltage gated K+-channels KV1.1, KV3.1b and K+-Cl- cotransporter (KCC2). We found that OPNs of both species were positive for all three K+-antibodies and that the staining patterns were similar for both spe

Neurons Are the Inflammatory Problem.

The discovery that reflex neuronal circuits control immunity provided a new mechanistic understanding of infection and injury. Pinho-Ribeiro et al. (2018) report that lethal tissue destruction from streptococcal necrotizing fasciitis ("flesh-eating disease") occurs because sensory neurons reflexively inhibit frontline immune responses.

Gene length matters in neurons.

A recent study by Gabel et al. (2015) found that Mecp2, the gene mutated in Rett syndrome, represses long (> 100 kb) genes associated with neuronal physiology and connectivity by binding to methylated CA sites in DNA. This study adds to a growing body of literature implicating gene length and transcriptional mechanisms in neurodevelopmental and neurodegenerative disorders.

Performance limitations of relay neurons.

Relay cells are prevalent throughout sensory systems and receive two types of inputs: driving and modulating. The driving input contains receptive field properties that must be transmitted while the modulating input alters the specifics of transmission. For example, the visual thalamus contains relay neurons that receive driving inputs from the retina that encode a visual image, and modulating inputs from reticular activating system and layer 6 of visual cortex that control what aspects of the image will be relayed back to visual cortex for perception. What gets relayed depends on several fact

DNA instability in postmitotic neurons.

Huntington's disease (HD) is caused by a CAG repeat expansion that is unstable upon germ-line transmission and exhibits mosaicism in somatic tissues. We show that region-specific CAG repeat mosaicism profiles are conserved between several mouse models of HD and therefore develop in a predetermined manner. Furthermore, we demonstrate that these synchronous, radical changes in CAG repeat size occur in terminally differentiated neurons. In HD this ongoing mutation of the repeat continuously generates genetically distinct neuronal populations in the adult brain of mouse models and HD patients. The

Gain modulation of respiratory neurons.

A possible mechanism underlying adaptive control of the respiratory system is gain modulation of the discharge frequency (F(n)) patterns of medullary respiratory neurons mediated by GABA(A) receptors. Antagonism of GABA(A) receptors with bicuculline results in an F(n) pattern that is an amplified replica of the underlying control pattern. The contours of F(n) patterns remain proportional to one another. Studies suggest that a tonic GABA(A)ergic input constrains the control- and reflexly-induced activities of these neurons to about 35-50% of the discharge rate without this inhibitory input. The

Extracellular pH and stimulated neurons.

The use of ion-sensitive microelectrodes enabled us to follow the dynamic changes in extracellular pH (pHe) together with those in the extracellular concentration of some biologically important ions, particularly K+ and Ca2+. Activity-related changes in pHe were studied in isolated spinal cords of frogs and in spinal cords of rats in vivo. Repetitive electrical stimulation of an afferent input led either to triphasic alkaline-acid-acid changes (90% of frogs) or to triphasic alkaline-acid-alkaline changes (10% of frogs and rats) with the greatest changes in the lower dorsal horns. The transient

Functional interactions between anteroventral periventricular kisspeptin neurons and gonadotropin-releasing hormone neurons in female mice.

Kisspeptin neurons in the rostral hypothalamus are hypothesized to initiate preovulatory gonadotropin-releasing hormone (GnRH) surges by causing estradiol-dependent activation of GnRH neuron action potential firing and subsequent GnRH release. To determine if estradiol or ovarian cycle stage modulate functional connectivity in this circuit, anteroventral-periventricular (AVPV) area kisspeptin neurons were photostimulated while recording firing and/or evoked synaptic currents from preoptic area GnRH neurons in acutely-prepared mouse brain slices. Slices were prepared 2-days post ovariectomy (OV

RVLM C1 Neurons Innervate Sacral as well as Thoracolumbar Autonomic Preganglionic Neurons in the Rat.

We examined the distribution of axons throughout the spinal cord of the rat that were either immunoreactive for the adrenaline-synthesizing enzyme, phenylethanolamine N-methyltransferase (PNMT), or derived from medullary C1 neurons, one of the three groups of neurons in the brain that synthesize PNMT. We observed that PMNT-immunoreactive axons, as well as C1 axons labelled with GFP from viral transduction, innervate most, but not all, sympathetic preganglionic neurons in the thoracolumbar spinal cord. GFP-positive C1 axons provided innervation to sympathetic preganglionic neurons that expresse

Parafacial neurons in the human brainstem express specific markers for neurons of the retrotrapezoid nucleus.

The retrotrapezoid nucleus (RTN) is a hub for respiratory chemoregulation in the mammal brainstem that integrates chemosensory information from peripheral sites and central relays. Chemosensitive neurons of the RTN express specific genetic and molecular determinants, which have been used to identify RTN precise location within the brainstem of rodents and nonhuman primates. Based on a comparative approach, we hypothesized that among mammals, neurons exhibiting the same specific molecular and genetic signature would have the same function. The co-expression of preprogalanin (PPGAL) and SLC17A6

Current controversies in Niemann-Pick C1 disease: steroids or gangliosides; neurons or neurons and glia.

There has been a recent explosion in research on Niemann-Pick type C disease. Much of the work has used mouse models or cells in culture to elucidate the pathophysiological mechanisms resulting in the phenotype of the disease. This work has generated several contrasting views on the mechanism, which are labeled 'controversies' here. In this review, two of these controversies are explored. The first concerns which stored materials are causative in the disease: cholesterol, gangliosides and sphingolipids, or something else? The second concerns which cells in the body require Npc1 in order to fun

Metabolites of cerebellar neurons and hippocampal neurons play opposite roles in pathogenesis of Alzheimer's disease.

Metabolites of neural cells, is known to have a significant effect on the normal physiology and function of neurons in brain. However, whether they play a role in pathogenesis of neurodegenerative diseases is unknown. Here, we show that metabolites of neurons play essential role in the pathogenesis of Alzheimer's disease (AD). Firstly, in vivo and in vitro metabolites of cerebellar neurons both significantly induced the expression of Abeta-degrading enzymes in the hippocampus and cerebral cortex and promoted Abeta clearance. Moreover, metabolites of cerebellar neurons significantly reduced bra

Dopamine transporter expression distinguishes dopaminergic neurons from other catecholaminergic neurons in the developing zebrafish embryo.

To characterize the formation of the dopaminergic system in the developing zebrafish CNS, we cloned cDNAs encoding tyrosine hydroxylase (th), an enzyme in dopamine synthesis, and the dopamine transporter (dat), a membrane transport protein which terminates dopamine action by re-uptake. Dopaminergic neurons are first detected between 18 and 19 h post-fertilization in a cluster of cells in the ventral diencephalon. Subsequently, th and dat detection identifies dopaminergic neurons in the olfactory bulb, the pretectum, the retina and the locus coeruleus. Neurons expressing th but not dat are adre

Regulation of Neuronal Activity in Hypothalamic Vasopressin Neurons.

Vasopressin is a peptide hormone secreted from the posterior pituitary gland in response to various physiological and/or pathological stimuli, including changes in body fluid volume and osmolality and stress exposure. Vasopressin secretion is controlled by the electrical activity of the vasopressinergic magnocellular neurosecretory cells located in the hypothalamic supraoptic nucleus and paraventricular nucleus. Vasopressin release can occur somatodendritically in the hypothalamus or at the level of pituitary axon terminals. The electrical activity of the vasopressin neurons assumes specific p

PVNCRF Neurons Regulate Migraine-Like Allodynia by Activating CRFR2 on Spinal Trigeminal Caudalis Glutamatergic Neurons.

Migraine is a prevalent and debilitating neurological disorder with poorly understood neural mechanisms. Here, we characterize a hypothalamic-trigeminal pathway involved in the regulation of migraine-like allodynia. Using a combination of monosynaptic circuit, fiber photometry-based calcium imaging, and behavioral assays in a nitroglycerin (NTG)-induced murine model, we observed hyperactivity in corticotropin-releasing factor (CRF)-expressing neurons within the hypothalamic paraventricular nucleus (PVN). Activation or inhibition of PVNCRF neurons mimicked or blocked migraine-like allodynia, re

Most Early-Born Subplate Neurons Persist as Layer 6b Neurons in the Adult Mouse Neocortex.

Subplate neurons (SpNs) are early-born neurons in the mammalian neocortex and play a pivotal role in cortical development. It is widely believed that SpNs are predominantly lost due to postnatal cell death, with only a few remnants contributing to layer 6b (L6b) neurons in the adult neocortex, but the extent to which SpNs persist as L6b neurons remains largely unknown. To address this, we conducted a comprehensive birthdate analysis using multiple ethynyl deoxyuridine (EdU) injections over 24 h, thereby precisely tracking the survival and distribution of SpNs in the adult mouse neocortex. We f

Entorhinal Cortex Wolframin-1-expressing neurons propagate tau to CA1 neurons and impair hippocampal memory.

Tau pathology emerges early in Alzheimer's disease within entorhinal cortex layer II (ECII) and reaches hippocampal CA1, but how this circuit-level spread translates into sex-dependent vulnerability remains unclear. Using a circuit-defined model in which P301L human tau is expressed selectively in Wolframin-1 (Wfs1 + ) ECII neurons and propagates to CA1, we found that the extent and proximal-distal distribution of tau-positive CA1 neurons were comparable in males and females. Despite similar propagation, females exhibited broad hippocampal-dependent cognitive impairment (working memory, object
