ENETS standardized (synoptic) reporting for neuroendocrine tumour pathology.

In recent years the WHO classification of neuroendocrine neoplasms (NEN) has evolved. Nomenclature as well as thresholds for grading have changed leading to potential confusion and lack of comparability of tumour reports. Therefore, the European Neuroendocrine Tumour Society (ENETS) has set-up an interdisciplinary working group to develop templates for a pathology data set for standardised reporting of NEN. Experts of various disciplines, members of the ENETS Advisory Board, formed a taskforce that discussed and decided on the structure, content and the number of templates needed for reporting

Leptin in humans: lessons from translational research.

Leptin has emerged over the past decade as a key hormone in not only the regulation of food intake and energy expenditure but also in the regulation of neuroendocrine and immune function as well as the modulation of glucose and fat metabolism as shown by numerous observational and interventional studies in humans with (complete) congenital or relative leptin deficiency. These results have led to proof-of-concept studies that have investigated the effect of leptin administration in subjects with complete (congenital) leptin deficiency caused by mutations in the leptin gene as well as in humans

Integration of endocrine signals that regulate insect ecdysis.

The extremely large number of insects and members of allied groups alive today suggests that molting--shedding of an old cuticle--may be one of the most commonly performed behaviors on our planet. Removal of an old cuticle in insects is associated with stereotyped, species-specific patterns of behavior referred to as ecdysis. It has been recognized for decades that the initiation of ecdysis is under hormonal control, but until recently many of the key peptides that regulate ecdysis were unknown. The report in 1996 of a new ecdysis-triggering hormone (ETH) sparked an era of significant advances

Immunofluorescent localization of insulin-like material in the median neurosecretory cells of the blowfly, Calliphora vomitoria (Diptera).

Immunocytochemical staining has shown that the median neurosecretory cells (MNC) of the brain of the blowfly, Calliphora vomitoria, contain an insulin-like material which cross reacts with antibodies to bovine insulin. There are 24-26 paraldehyde fuchsin-positive MNC of which only 6-8 show the specific insulin-like immunoreactivity.

Corazonin mediates photoperiodically induced diapause in the bean bug Riptortus pedestris.

Insects in mid- to high latitudes predict unfavorable seasons through changing photoperiod, and undergo diapause to survive harsh conditions. The circadian clock and brain neurosecretory systems are key components in regulating photoperiodic diapause. However, the neuronal pathways linking these systems, and the neuropeptides involved in photoperiodic diapause, remain poorly understood. We investigated the role of corazonin (CRZ), a conserved neuropeptide regulating metabolic stress and reproduction, in the bean bug Riptortus pedestris, a species highly sensitive to short days for diapause ind

Specific immunoelectronmicroscopic localization of vasopressin and oxytocin in the neurohypophysis of the rat.

An immunoelectronmicroscopic method for the specific localization of neurohypophyseal hormones was developed in neurohypophyses of Wistar and Brattleboro rats, the latter strain being homozygous for diabetes insipidus. If the proper precautions were omitted, a marked cross reactivity between anti-vasopressin and anti-oxytocin preparations was found. Cross reaction of an anti-vasopressin plasma with oxytocin, at a dilution of less than 1:1600, resulted in electron density of all granules within neurosecretory fibres of the Brattleboro and Wistar neurohypophyses. However, this cross reactivity c

Adrenergic receptor activation hyperpolarizes the caudal neurosecretory cells of the flounder, Platichthys flesus.

The physiological factors that govern activity of the caudal neurosecretory system in teleost fish are poorly understood. Immunocytochemical evidence indicates that the neurosecretory Dahlgren cells are innervated by descending monoaminergic fibres. Using intracellular recording techniques in an isolated preparation of the posterior spinal cord of the flounder (Platichthys flesus) we have demonstrated that superfusion of adrenaline or noradrenaline (10(-7) - 10(-3) M) causes hyperpolarization of Dahlgren cells (up to -30 mV). This hyperpolarization is likely to reflect an inhibitory effect of

Photo-neuro-immuno-endocrinology: How the ultraviolet radiation regulates the body, brain, and immune system.

Ultraviolet radiation (UVR) is primarily recognized for its detrimental effects such as cancerogenesis, skin aging, eye damage, and autoimmune disorders. With exception of ultraviolet B (UVB) requirement in the production of vitamin D3, the positive role of UVR in modulation of homeostasis is underappreciated. Skin exposure to UVR triggers local responses secondary to the induction of chemical, hormonal, immune, and neural signals that are defined by the chromophores and extent of UVR penetration into skin compartments. These responses are not random and are coordinated by the cutaneous neuro-

Vth International Symposium on Sjögren's syndrome. Clinical aspects and therapy.

There is increasing awareness that sicca syndrome can lead to ocular, oral and systemic complications. However, a wide variety of pathogenetic processes may lead to the same end result of diminished glandular secretion. Recognition of these different etiologies will lead to improved therapies. On the one hand, an active inflammatory/autoimmune process may respond to immunomodulatory drugs. On the other hands, processes that predominantly involve "neuroendocrine" dysfunction (unrelated to immune activity) of the gland will require a different approach.

[Noise: neuro-endocrine aspects].

Experimental study of neuro-endocrinological effects of noise stress shows neuro-mediator modifications. Exposure to stress induces a marked stimulation of the adrenergic structures which increases plasma concentrations of ACTH and glucosteroids. The increased secretion of the cortico-releasing factor (CRT) from the hypothalamo-hypophyseal system mediates a complex cascade in which other neuro-modulators also participate. Within the CNS, CRF activates the sympathetic nervous system, raising plasma concentrations of epinephrine and nor-epinephrine. These neuro-mediator and endocrine variations

Neuroendocrinological studies on depression with special reference to research at the Max-Planck-Institute of Psychiatry.

Hypercortisolism due to Cushing's syndrome or glucocorticoid therapy induces disturbances in several other endocrine systems and may also cause mental changes, predominantly depression of various degrees. On the other hand, it has repeatedly been shown that endogenous depression is often accompanied by hypercortisolemia, usually of a modest degree, and/or by changes in other hormonal systems similar to those observed in Cushing's syndrome and during treatment with glucocorticoids. Research performed at the MPIP on 327 psychiatric patients and 103 healthy subjects has demonstrated that, in cont

The Contribution of Cell Imaging to the Study of Anterior Pituitary Function and Its Regulation.

Advances in the knowledge of the neuroendocrine system are closely related to the development of cellular imaging and labeling techniques. This synergy ranges from the staining techniques that allowed the first characterizations of the anterior pituitary gland, its relationship with the hypothalamus, and the birth of neuroendocrinology; through the development of fluorescence microscopy applications, specific labeling strategies, transgenic systems, and intracellular calcium sensors that enabled the study of processes and dynamics at the cellular and tissue level; until the advent of super-res

Which patient requires neuroendocrine assessment following traumatic brain injury, when and how?

Traumatic brain injury (TBI) is an important public health problem, particularly among young adults in industrialized countries. Hypopituitarism is a common occurrence among survivors of TBI and may contribute to the associated morbidity seen in the acute and chronic phases following injury. The available data suggest that survivors of moderate to severe TBI should undergo screening for hypopituitarism particularly in the first year after injury. This requires a close liaison between endocrinologists, neurosurgeons, neuropsychologists, intensive care and rehabilitation physicians. Patients who

Mitral prolapse. A heart anomaly in a clinical neuroendocrine context.

Mitral valve prolapse was identified as a separate nosological entity by Barlow in 1963. A characteristic of this cardiac anomaly is blood reflux into the left atrium during the systole owing to the lack of adhesion between valve flaps. The presence of symptoms linked to neuroendocrine dysfunctions or to the autonomic nervous system lead to the onset of the pathology known as mitral valve prolapse syndrome (MVPs). It is usually diagnosed by chance in asymptomatic patients during routine tests. MVPs includes complex alterations to the neurovegetative system and a high clinical incidence of neur

Immunocytochemical identification of alpha-endorphin-like material in neurones of the brain and corpus cardiacum of the blowfly, Calliphora vomitoria (Diptera).

A group of the 24-26 paraldehyde fuchsin-positive median neurosecretory cells (MNC) in the pars intercerebralis of the brain of the blowfly, Calliphora vomitoria, has shown immunoreactivity towards three different antibodies to alpha-endorphin, a peptide that corresponds to the amino acid sequence present between residues 61 and 76 of the precursor molecule, beta-lipotropin (beta-LPH). The immunoreactive material could be followed in axons within the median bundle, the tract through which neurosecretory material from the MNC is passed down to the corpus cardiacum (CC). The alpha-endorphin-immu

[Relationships between the brain and the immune system].

The concept that the brain can modulate activity the immune system stems from the theory of stress. Recent advances in the study of the inter-relationships between the central nervous system and the immune system have demonstrated a vast network of communication pathways between the two systems. Lymphoid organs are innervated by branches of the autonomic nervous system. Accessory immune cells and lymphocytes have membrane receptors for most neurotransmitters and neuropeptides. These receptors are functional, and their activation leads to changes in immune functions, including cell proliferatio

Low number of neurosecretory vesicles in neuroblastoma impairs massive catecholamine release and prevents hypertension.

INTRODUCTION: Neuroblastoma (NB) is a pediatric cancer of the developing sympathetic nervous system. It produces and releases metanephrines, which are used as biomarkers for diagnosis in plasma and urine. However, plasma catecholamine concentrations remain generally normal in children with NB. Thus, unlike pheochromocytoma and paraganglioma (PHEO/PGL), two other non-epithelial neuroendocrine tumors, hypertension is not part of the usual clinical picture of patients with NB. This suggests that the mode of production and secretion of catecholamines and metanephrines in NB is different from that
