Spontaneous nitroblue tetrazolium (NBT) reduction was evaluated in neutrophils from patients with the different types and forms of leprosy, and compared with reduction obtained form cells from normal controls. Leucocytes from the same subjects were stimulated in vitro by endotoxin, and the rise in percentage of cells reducing NBT was determined. Patients of all groups, with the exception of those with reactional lepromatous leprosy (RLL) had an essentially normal proportion of reducing cells. Neutrophils were normally activated by endotoxin. A luminol enhanced chemiluminescence micromethod has been adapted for use in prenatal diagnosis of chronic granulomatous disease (CGD). After validation of the assay in normal adults, newborns, fetuses, CGD carriers, and CGD patients, the fetuses of two pregnant CGD carriers were tested after fetoscopic aspiration of fetal blood. W. M. Willingham and J. R. J. Sorenson have recently reported [1988) Biochem. Biophys. Res. Commun. 150, 252-258) that the Cu(II)EDTA complex catalyzes the dismutation of O-2 and thereby call into question several earlier reports. These authors concluded that the widespread use of EDTA to suppress dismutation of O-2 by adventitious Cu(II) and other metals, during assays for superoxide dismutase, is unsound. We have undertaken a clarification of this issue by examining the ability of Cu(II)EDTA to catalyze the dismutation of O-2 and have found this complex to be devoid of this activity at pH 6.

Leukocyte function was studied in a child with elevated IgE and many infections. At age 7 months, chemotaxis was decreased, but improved first toward formylated peptides and finally toward complement. A patient with chronic meningococcemia with the unusual features of subungual splinter hemorrhages, a pericardial friction rub, and a culture positive skin lesion is presented. The preparation and properties of the Cu(II) complex Cu(SAS)2.H2O are reported for the antiinflammatory drug Salsalate (SAS). The diffuse reflectance spectra and magnetic moments are consistent with a dinuclear structure as found for [Cu(aspirinate)2(H2O)]2. The Cu(II) complex exhibits an increased superoxide dismutase activity compared with the parent drug molecule in the nitroblue tetrazolium assay.

Fifty-six healthy neonates and children as well as 33 patients with juvenile rheumatoid arthritis and other diseases of the locomotor system were examined using a modification of the NBT test. Unusual inclusions with some of the features of "reducing bodies" were encountered in the skeletal muscle biopsy of a 2.5-year-old boy with childhood-onset acid maltase deficiency. The biopsy revealed a vacuolar myopathy with lysosomal storage of glycogen and eosinophilic refractile inclusions in myofibers, which appeared dark blue with the menadione-nitroblue tetrazolium reaction. The significance of the association of inclusions with reducing properties in the setting of acid maltase deficiency is discussed.

The superoxide dismutase-mimetic activity and the electrochemical behavior of the binuclear complex [Cu(TSA)2py]2 is reported. The complex exhibits a marked SOD activity in the nitroblue tetrazolium assay. Its redox response is in agrement with two copper atoms partially coupled through the carboxylate moieties. The electrode reactions lie in the range of the superoxide/peroxide and oxygen/superoxide couples. This fact is indicative that this complex can act as catalyst for the SOD reaction.
