The Co-Infection of ISKNV-II and RGNNV Resulting in Mass Mortality of Juvenile Asian Seabass (Lates calcarifer), Zhuhai, Southern China.

Infectious spleen and kidney necrosis virus (ISKNV) and nervous necrosis virus (NNV) are two common and important causative agents in marine-cultured fish. However, high viral loads of both ISKNV and NNV in the same clinical case is unusual. In this study, a mass mortality event of Asian seabass Lates calcarifer juveniles occurred in Zhuhai, the main Asian seabass cultured area in mainland China. The fish samples were pooled for pathogen identification and both high viral loads of ISKNV and NNV were detected by real-time microfluidic quantitative PCR and conventional PCR. Immunohistochemistry

Understanding the interaction between Betanodavirus and its host for the development of prophylactic measures for viral encephalopathy and retinopathy.

Over the last three decades, the causative agent of viral encephalopathy and retinopathy (VER) disease has become a serious problem of marine finfish aquaculture, and more recently the disease has also been associated with farmed freshwater fish. The virus has been classified as a Betanodavirus within the family Nodaviridae, and the fact that Betanodaviruses are known to affect more than 120 different farmed and wild fish and invertebrate species, highlights the risk that Betanodaviruses pose to global aquaculture production. Betanodaviruses have been clustered into four genotypes, based on th

A formalin-inactivated immunogen against viral encephalopathy and retinopathy (VER) disease in European sea bass (Dicentrarchus labrax): immunological and protection effects.

The European sea bass (Dicentrarchus labrax) is an important farmed fish species in the Mediterranean area, very sensitive to the infection by encephalopathy and retinopathy virus (VERv), or Betanodavirus, which causes massive mortalities. Effective vaccines to fight the pathology are not yet available and in this work we describe a promising intraperitoneal immunization route against VERv of sea bass juveniles. We performed intraperitoneal and immersion immunization trials with a VERv (isolate 283.2009 RGNNV) inactivated by formalin, β-propiolactone and heat treatment. Interestingly, the intr

Nodavirus protein A's interdomain elbow controls RNA replication organelle formation and function.

Positive-strand [(+)RNA] viruses replicate their RNA genomes in poorly understood membrane-associated replication organelles (ROs). Cryo-electron microscopy of nodaviral ROs revealed that viral RNA replication protein A, with polymerase and RNA capping domains, forms a "crown" of two stacked 12-mer rings at the RO's opening to the cytosol, providing powerful foundations for analyzing RO formation and function. The lower proto-crown is a ring of 12 polymerases with RNA capping domains clustered to form a central floor. The upper crown mirrors the polymerase ring but has RNA capping domains proj

ZNFX1 functions as a compensatory dsRNA recognition receptor to exert antiviral effect in orange-spotted grouper.

Zinc finger NFX1-type containing 1 (ZNFX1) has been established as a critical mediator of the antiviral response in mammals, functioning through dsRNA recognition and priority activation of the retinoic acid-inducible gene I (RIG-I)-like receptor (RLR) signaling pathway. However, the role of its fish ortholog, particularly in relation to aquatic virus interactions, remains elusive. The absence of the RIG-I homolog, a key pattern recognition receptor, in multiple Actinopterygii may compromise their innate antiviral immune responses. Here, ZNFX1 in Epinephelus coioides (EcZNFX1) is identified as

Rationally designed self-assembled peptide nanofibers provoke robust humoral immunity against nervous necrosis virus.

Nervous necrosis virus (NNV) poses devastating mortality risks to almost all marine fish, causing significant economic losses to the marine fish aquaculture industry, particularly in juvenile groupers. However, no effective NNV vaccine is available. To counter this, we report a smart strategy of a rationally designed modular peptide vaccine platform (CP64-83-Q11) that recapitulates the antigen peptide CP64-83 and a Q11 self-assembling domain. An anti-NNV phage antibody library was constructed and used for screening anti-NNV antibody. Bioinformatics and surface-plasmon resonance were utilized t

M1-type polarized macrophage contributes to brain damage through CXCR3.2/CXCL11 pathways after RGNNV infection in grouper.

The vertebrate central nervous system (CNS) is the most complex system of the body. The CNS, especially the brain, is generally regarded as immune-privileged. However, the specialized immune strategies in the brain and how immune cells, specifically macrophages in the brain, respond to virus invasion remain poorly understood. Therefore, this study aimed to examine the potential immune response of macrophages in the brain of orange-spotted groupers (Epinephelus coioides) following red-spotted grouper nervous necrosis virus (RGNNV) infection. We observed that RGNNV induced macrophages to produce

Surveillance for nervous necrosis virus-specific antibodies in barramundi Lates calcarifer in Australian hatcheries.

We conducted single point-in-time and repeated cross-sectional studies of the prevalence of antibodies against nervous necrosis virus (NNV) in populations of adult barramundi Lates calcarifer in Australia. Serum samples collected between 2002 and 2012 were analyzed with indirect ELISA (n = 468). Most of the samples were sourced from broodstock with unknown exposure history, and these were compared with reference populations with confirmed history of exposure to NNV. Non-lethally collected gonad fluid samples from economically valuable barramundi broodstock (n = 164) were tested for the presenc

Drosophila C virus systemic infection leads to intestinal obstruction.

UNLABELLED: Drosophila C virus (DCV) is a positive-sense RNA virus belonging to the Dicistroviridae family. This natural pathogen of the model organism Drosophila melanogaster is commonly used to investigate antiviral host defense in flies, which involves both RNA interference and inducible responses. Although lethality is used routinely as a readout for the efficiency of the antiviral immune response in these studies, virus-induced pathologies in flies still are poorly understood. Here, we characterize the pathogenesis associated with systemic DCV infection. Comparison of the transcriptome of

Viral nerve necrosis in hatchery-produced fry of Asian seabass Lates calcarifer: sequential microscopic analysis of histopathology.

We studied the natural progression of viral nerve necrosis (VNN) in larvae of Asian seabass Lates calcarifer Bloch from 0 to 40 days post-hatch (dph). The hatchlings were reared in the vicinity of a confirmed nodavirus-affected older batch. Using light and electron microscopy (EM), we made a sequential analysis of histopathological manifestations in nerve tissue and other organs. There were no changes from the day of hatching until 4 dph. Larvae at 4 dph had viral particles in the intramuscular spaces underlying the skin, but the nerve cells of the brain were normal. The first signs of necrosi

Potential involvement of persistently infected cells in protective immunity of grouper fish convalesced from nervous necrosis virus infection.

Nervous necrosis virus (NNV) is a simple RNA virus with high pathogenicity across over 170 fish species. Two experimental models have been developed to efficiently induce convalescent fish using live viruses: (1) pre-administering polyinosinic:polycytidylic acid (poly(I:C)) to induce a non-specific antiviral state, and (2) maintaining fish at suboptimal temperatures for disease onset. Notably, these convalescent fish exhibit robust long-term protective immunity despite having undetectable levels of neutralizing antibodies. This study synthesizes established in vivo and in vitro data on the kin

Immunity to nervous necrosis virus infections of orange-spotted grouper (Epinephelus coioides) by vaccination with virus-like particles.

Nervous necrosis virus (NNV) is a kind of the betanodaviruses, which can cause viral nervous necrosis (VNN) and massive mortality in larval and juvenile stages of orange-spotted grouper (Epinephelus coioides). Due to the lack of viral genomes, virus-like particles (VLPs) are considered as one of the most promising candidates in vaccine study to control this disease. In this study, a type of VLPs, which was engineered on the basis of orange-spotted grouper nervous necrosis virus (OGNNV), was produced from prokaryotes. They possessed the similar structure and size to the native NNV. In addition,

Development of a Recombinant Protein Vaccine Based on Cell-Free Protein Synthesis for Sevenband Grouper Epinephelus septemfasciatus Against Viral Nervous Necrosis.

Sevenband grouper, Epinephelus septemfasciatus, is becoming an important aquaculture species in Korea. However, viral nervous necrosis disease is a large problem causing mass mortality in sevenband grouper aquaculture. Recombinant protein vaccines are one of the best methods to reduce these economic losses. However, the cell-based expression method mainly produces inclusion bodies and requires additional procedures. In this study, we expressed a recombinant viral coat protein of sevenband grouper nervous necrosis virus (NNV) using a cell-free protein synthesis system. The purified recombinant

Oral vaccination via virus-like particles encapsulated in Lactococcus lactis.

Nervous necrosis virus (NNV) causes severe disease in marine fish, leading to high mortality and significant economic losses in aquaculture. Effective oral vaccines are urgently needed for large-scale, labour-efficient immunization of farmed fish. In this study, NNV capsid protein was successfully expressed in Lactococcus lactis (L. lactis) and evaluated as a vaccine candidate in Asian seabass (Lates calcarifer) fingerlings. The capsid protein assembled into virus-like particles (VLPs), structurally similar to those produced in E. coli. Intraperitoneal injection of purified VLPs elicited stron

Transcriptome analysis reveals the DNA replication genes response to Vibrio anguillarum and NNV infection in Jinhu grouper (Epinephelus fuscoguttatus♀ × Epinephelus tukulal♂).

Vibrio anguillarum acts as an infectious agent in the aquaculture industry that causes a fatal hemolytic septicaemic disease in fish and shellfish. Viral nervous necrosis (VNN) disease seriously impacts the healthy development of the aquaculture industry. While the detrimental effects of V. anguillarum and NNV have been widely researched on freshwater and marine fish, whether they affect DNA replication in fish is unclear. In this study, we used Jinhu grouper as a model to investigate the influence of V. anguillarum and NNV on their DNA replication. By RNA-seq analysis in conjunction with gene

Plant Expression of Trans-Encapsidated Chimeric Viral Vaccines with Animal RNA Replicons: An Update.

In this protocol, we outline how to produce a chimeric viral vaccine in a biosafety level 1 (BSL1) environment. An animal viral vector RNA encapsidated with tobacco mosaic virus (TMV) coat protein can be fully assembled in planta. Agrobacterium cultures containing each component are inoculated together into tobacco leaves and the self-assembled hybrid chimeric viral vaccine is harvested 4 days later and purified with a simple PEG precipitation. The viral RNA delivery vector is derived from the BSL1 insect virus, Flock House virus (FHV), and replicates in human and animal cells but does not spr

Establishment and characterization of a novel cell line from spleen of humpback grouper (Cromileptes altivelis) and its application in pathogens susceptibility.

The humpback grouper (Cromileptes altivelis) is a marine fish of significant commercial value and has been identified as a potential candidate for marine aquaculture. In this research, a cell line named as CAS cells was developed from the spleen tissue of the humpback grouper. This cell line has been successfully passaged for over 100 passages, with most cells exhibiting a fibroblast-like morphology. After being cryopreserved in liquid nitrogen for three months, the survival rate of the cells was over 80 %. The sequencing of the COI and 18S rRNA genes has verified that the cell line is sourced

An improved oral vaccine with molecular adjuvant β-defensin protects grouper against nervous necrosis virus infection.

Nervous necrosis virus (NNV) is one of the most important fish viral pathogens infecting more than 120 fish species worldwide. Due to the mass mortality rates often seen among larvae and juveniles, few effective vaccines against NNV were developed up to now. Here, the protective effect of recombinant coat protein (CP) from red-spotted grouper nervous necrosis virus (RGNNV) fused with grouper β-defensin (DEFB) as an oral vaccine was evaluated using Artemia as a biocarrier delivery system in pearl gentian grouper (Epinephelus lanceolatus♂ × Epinephelus fuscoguttatus♀). Feeding with Artemia encap

Tuning intracellular immunity by Nodamura virus B2 protein enhances self-amplifying RNA activity.

Self-amplifying RNA (saRNA) offers a powerful approach for vaccine development, as its replicative nature within cells provides prolonged antigen expression together with self-adjuvanticity. However, its potency is often limited by innate immune responses, particularly those triggered by intracellular double-stranded RNA (dsRNA), highlighting the need for strategies that can boost saRNA efficacy without compromising its immunostimulatory properties. Here we show that the cis-expression of the Nodamura virus (NoV) B2 protein markedly enhances saRNA-driven transgene expression by alleviating key

EcCXCR4b influences RGNNV proliferation by interacting with the RGNNV capsid protein.

UNLABELLED: Red-spotted grouper nervous necrosis virus (RGNNV) is an intensely virulent pathogen affecting cultured fish, capable of infecting various reared marine and freshwater species worldwide and causing severe financial losses in the aquaculture business. The capsid protein (CP) of RGNNV, as the viral surface's sole structural protein, is essential for mediating RGNNV binding to susceptible host cells. However, the cellular receptors involved in RGNNV entry are still mostly unknown. In this study, immunoprecipitation-mass spectrometry (IP-MS) was employed to screen for candidate protein
