Development of a monoclonal antibody against the CD3ε of olive flounder (Paralichthys olivaceus) and its application in evaluating immune response related to CD3ε.

The T cell receptor (TCR) is the binding site of antigen and is responsible for specifically activating the adaptive immune response. CD3, an essential component of the CD3-TCR complex, is known to be composed of γδ and ε chains in teleost. However, there are few monoclonal antibodies (mAb) available to identify these molecules on T cells, so we aimed to produce a mAb against CD3ε to improve our understanding of T cell immune response in olive flounder (Paralichthys olivaceus). CD3ε recombinant protein was expressed in yeast, the expression of which was confirmed by SDS-PAGE, MALDI-TOF/TOF MS

Outbreak of hirame rhabdovirus infection in cultured spotted sea bass Lateolabrax maculatus on the western coast of Korea.

In this study, we determined the cause of a disease outbreak in spotted sea bass, Lateolabrax maculatus reared in culture cages on the western coast of Korea in 2013. The major signs in the diseased fish exhibited were haemorrhaging on the membranes of the abdomen, gastrointestinal organs and opercular gills, as well as an enlarged spleen. No external morphological signs of infection were visible, except for a darkening in colour. No parasites or pathological bacteria were isolated from the diseased fish; however, epithelioma papulosum cyprini (EPC) cells inoculated with tissue homogenates fro

Autophagy-inducing peptides from mammalian VSV and fish VHSV rhabdoviral G glycoproteins (G) as models for the development of new therapeutic molecules.

It has not been elucidated whether or not autophagy is induced by rhabdoviral G glycoproteins (G) in vertebrate organisms for which rhabdovirus infection is lethal. Our work provides the first evidence that both mammalian (vesicular stomatitis virus, VSV) and fish (viral hemorrhagic septicemia virus, VHSV, and spring viremia carp virus, SVCV) rhabdoviral Gs induce an autophagic antiviral program in vertebrate cell lines. The transcriptomic profiles obtained from zebrafish genetically immunized with either Gsvcv or Gvhsv suggest that autophagy is induced shortly after immunization and therefore

Infectious hematopoietic necrosis virus (IHNV) persistence in Sockeye Salmon: influence on brain transcriptome and subsequent response to the viral mimic poly(I:C).

BACKGROUND: Sockeye Salmon are an iconic species widely distributed throughout the North Pacific. A devastating pathogen of Sockeye Salmon is infectious hematopoietic necrosis virus (IHNV, genus Novirhabdovirus, family Rhabdoviridae). It has been postulated that IHNV is maintained in salmon populations by persisting over the life of its host and/or by residing in natural reservoirs other than its susceptible hosts. Herein we demonstrate the presence of IHNV in the brain of Sockeye Salmon that survived an experimentally-induced outbreak, suggesting the presence of viral persistence in this susc

Temperature modulates the protein dynamics of the antigen presentation pathway in VHSV IVb-infected rainbow trout (Oncorhynchus mykiss) cell lines.

Rainbow trout face challenges due to climate change, increasing their susceptibility to pathogens like viruses. The endogenous antigen presentation pathway is essential for antiviral immunity by presenting intracellular antigens to cytotoxic lymphocytes. While previous studies show impaired expression of antigen presentation molecules at suboptimal temperatures, little is known about how temperature affects the protein function of the antigen presentation pathway during viral infections. This study aims to understand how temperature modulates the antigen presentation pathway in rainbow trout i

Comparative study on antigen persistence and immunoprotective efficacy of intramuscular and intraperitoneal injections of squalene - aluminium hydroxide (Sq + Al) adjuvanted viral hemorrhagic septicaemia virus vaccine in olive flounder (Paralichthys olivaceus).

The profitability of the olive flounder (Paralichthys olivaceus) aquaculture industry in Korea depends on high production and maintenance of flesh quality, as consumers prefer to eat raw flounders from aquaria and relish the raw muscles as 'sashimi'. For sustaining high production, easy-to-deliver and efficient vaccination strategies against serious pathogens, such as viral hemorrhagic septicemia virus (VHSV), is very important as it cause considerable losses to the industry. Whereas, a safe and non-invasive vaccine formulation that is free from unacceptable side-effects and does not devalue t

Development of sandwich ELISA for IL-10 quantification in olive flounder (Paralichthys olivaceus) and its application to protein-level analysis during viral hemorrhagic septicemia virus (VHSV) infection.

Interleukin-10 (IL-10) is an anti-inflammatory cytokine that plays a key role in maintaining immune homeostasis. Although studies in teleosts have examined IL-10 gene cloning, mRNA expression, and the functions of recombinant IL-10, analyses at the endogenous protein level remain limited. Considering IL-10 is biologically active only when secreted as a protein, direct detection and quantification of endogenous IL-10 protein are essential to accurately elucidate its immunomodulatory function. In this study, we developed mouse monoclonal and rabbit polyclonal anti-IL-10 antibodies that specifica

Designing and Cloning of the Gene Vaccine Carrying the Viral Haemorrhagic Septicaemia Multi-Epitope Gene in the pNZ8121 Secretion Vector.

The viral haemorrhagic septicaemia virus (VHSV) is considered a very significant disease affecting fish. The objective of this project is to develop a multi-epitope vaccine targeting viral hemorrhagic fever using the reverse vaccinology approach. Epitope prediction was conducted using the immunogenic components of VHSVV. A multi-epitope vaccine was developed by combining the immunogenic proteins' most potent B- and T-cell epitopes with the adjuvant. Following that, a comprehensive evaluation was carried out on many facets of the formulated vaccine, including its physicochemical properties, ant

Identification and expression analysis of long noncoding RNAs in response to infection with VHSV in rainbow trout.

Viral infections in the aquaculture industry present a significant challenge that requires further investigation. Emerging evidence suggests that long noncoding RNAs (lncRNAs) play a crucial role in regulating various biological processes, including immune responses. However, most studies have focused on model organisms, leaving a gap in knowledge about non-coding RNAs in response to viral hemorrhagic septicemia (VHS) in rainbow trout. In this study, we investigated the profile of non-coding RNAs (ncRNAs) and their targeting-related subjects in the spleen of rainbow trout challenged with the V

Use of Poly(I:C) Stabilized with Chitosan As a Vaccine-Adjuvant Against Viral Hemorrhagic Septicemia Virus Infection in Zebrafish.

There is an urgent need for more efficient viral vaccines in finfish aquaculture worldwide. Here, we report the use of poly(I:C) stabilized with chitosan as an adjuvant for development of better finfish vaccines. The adjuvant was co-injected with inactivated viral hemorrhagic septicemia virus (VHSV) (CSpIC+iV vaccine) in adult zebrafish and its efficiency in protection against VHSV infection was compared to a live, attenuated VHS virus vaccine (aV). Both free and stabilized poly(I:C) were strong inducers of an antiviral state, measured by transcriptional activation of the genes of viral sensor

Establishment and identification of spleen cell line from tiger puffer fish (Takifugu rubripes) and immune transcriptome response to IHNV.

The tiger puffer fish (Takifugu rubripes) is highly valued both economically and for its culinary appeal. However, it currently faces a significant risk of disease. This study aimed to establish a spleen cell line to aid in disease prevention and control for this species. We established a new stable cell line (TRSC) from the spleen tissue of the tiger puffer fish. The TRSC cells exhibited stable growth over more than 90 generations in L-15 medium supplemented with 4 ng/mL recombinant human basic fibroblast growth factor (bFGF) and 2 ng/mL recombinant human leukemia inhibitory factor (LIF) at 2

Differences between DNA vaccine and single-cycle viral vaccine in the ability of cross-protection against viral hemorrhagic septicemia virus (VHSV) and infectious hematopoietic necrosis virus (IHNV).

Vaccination procedures can be stressful for fish and can bring severe side effects. Therefore, vaccines that can minimize the number of administrations and maximize cross-protection against multiple serotypes, genotypes, or even different species would be highly advantageous. In the present study, we investigated the cross-protective ability of two types of vaccines - viral hemorrhagic septicemia virus (VHSV) G protein-expressing DNA vaccine and G gene-deleted single-cycle VHSV genotype IVa (rVHSV-ΔG) vaccine - against both VHSV genotype Ia and infectious hematopoietic necrosis virus (IHNV) in

DNA vaccine dual-expressing viral hemorrhagic septicemia virus glycoprotein and C-C motif chemokine ligand 19 induces the expression of immune-related genes in zebrafish (Danio rerio).

Glycoprotein (G protein)-based DNA vaccines are effective in protecting aquaculture fish from rhabdoviruses but the degree of immune response they elicit depends on plasmid concentration and antigen cassette. Here, we developed a DNA vaccine using the viral hemorrhagic septicemia virus G (VG) gene and chemokine (C-C motif) ligand 19 (CCL19)a.2 regulated by the CMV promoter as the molecular adjuvant. After transfection of the prepared plasmid (pVG + CCL19) into epithelioma papulosum cyprini cells, mRNA expression was confirmed through quantitative real-time polymerase chain reaction. The vaccin

Effect of CXCL12-expressing viral hemorrhagic septicemia virus replicon particles on leukocytes migration and vaccine efficacy in olive flounder (Paralichthys olivaceus).

Viral replicon particles are single-cycle viruses defective for function(s) needed for viral replication, which allow them to be recognized as a safer form for the vaccination of animals compared to attenuated live viruses. However, deletion of genes that are critical for the induction of protective immunity can diminish the vaccine potential of viral replicon particles. Therefore, the manipulation of viral replicon particles to produce a molecular adjuvant can be a way to increase immunogenicity of vaccines based on viral replicon particles. Chemokines are a class of chemotactic cytokines tha

Development of the reverse genetics system for viral hemorrhagic septicemia virus genotype IVa and its application in antiviral compound screening.

Viral hemorrhagic septicemia virus (VHSV) is a major pathogen affecting freshwater and marine fish species, posing a significant threat to global aquaculture. Reverse genetics systems are essential for studying viral replication, and host interactions, as well as developing vaccines and therapeutics. In this study, we developed a reverse genetics platform for VHSVLB2018 strain, a genetically distinct VHSV genotype IVa strain which exhibits low genomic identity with other Asian isolates, using a dual RNA polymerase I/II transcription vector. We successfully rescued recombinant VHSV in mammalian

Origin of Public Memory B Cell Clones in Fish After Antiviral Vaccination.

Vaccination induces "public" antibody clonotypes common to all individuals of a species, that may mediate universal protection against pathogens. Only few studies tried to trace back the origin of these public B-cell clones. Here we used Illumina sequencing and computational modeling to unveil the mechanisms shaping the structure of the fish memory antibody response against an attenuated Viral Hemorrhagic Septicemia rhabdovirus. After vaccination, a persistent memory response with a public VH5JH5 IgM component was composed of dominant antibodies shared among all individuals. The rearrangement

Antibody-dependent immune response of olive flounder (Paralichthys olivaceus) induced by inactivated viral hemorrhagic septicemia virus (VHSV) vaccine.

In teleosts, vaccine-induced protection is commonly assessed using serum antibody titers; however, these measurements often fail to fully explain protective outcomes due to limited understanding of B cell differentiation and antibody effector functions. In olive flounder (Paralichthys olivaceus), the cellular basis of antibody-mediated immunity remains poorly characterized. In this study, we investigated B cell differentiation, antibody production, and antibody-dependent immune responses post-immunization with an inactivated viral hemorrhagic septicemia virus (VHSV) vaccine. Nanoencapsulation of inactivated-viral vaccine using chitosan nanoparticles: Evaluation of its protective efficacy and immune modulatory effects in olive flounder (Paralichthys olivaceus) against viral haemorrhagic septicaemia virus (VHSV) infection.

Viral haemorrhagic septicaemia virus (VHSV), a (-) ssRNA virus belonging to the genus Novirhabdovirus of rhabdoviridae family, is the aetiological agent of viral haemorrhagic septicaemia (VHS) disease which causes huge economic losses in farmed olive flounder (Paralichthys olivaceus) and significant mortalities among several other marine fish species in Korea, Japan, and China. Previously, we developed an inactivated vaccine viz., formalin-inactivated VHSV mixed with squalene as adjuvant which was effective in conferring protective immunity (58-76% relative percentage survival) against VHSV bu
