Expression of CARD6, an NF-kappaB activator, in gastric, colorectal and oesophageal cancers.

AIMS: Activation of nuclear factor-kappa B (NF-kappaB) signalling is considered a crucial mechanism in the development of cancers. Caspase-associated recruitment domain 6 (CARD6) is a protein that activates NF-kappaB signalling evoked by RIP1, RIP2, Bcl-10 and MEKK. In this study, we analysed tissue expression of CARD6 protein in oesophageal squamous cell carcinoma (ESCC), gastric adenocarcinomas (GC) and colorectal adenocarcinomas (CRC). METHODS: We analysed the expression of CARD6 protein in 58 ESCC, 100 GC and 103 CRC patients' tissues by immunohistochemistry using a tissue microarray (TMA

A segment of the Mecp2 promoter is sufficient to drive expression in neurons.

Rett syndrome (RTT) is caused by mutations in the gene encoding methyl CpG-binding protein 2 (MeCP2). Although MeCP2 shows widespread expression in both neuronal and non-neuronal tissues, the symptoms of RTT are largely neurological. Herein, we have identified the regulatory region of the mouse Mecp2 gene that is sufficient for its restricted expression in neurons. A segment of the Mecp2 gene (-677/+56) exhibited strong promoter activity in neuronal cell lines and cortical neurons, but was inactive in non-neuronal cells and glia. The region necessary for neuronal-specific promoter activity was

Cistanche attenuates acute kidney injury by inhibiting ferroptosis through the disruption of Keap1-Nrf2 interaction.

ETHNOPHARMACOLOGICAL RELEVANCE: Ferroptosis, an iron-dependent regulated cell death, critically contributes to acute kidney injury (AKI) progression. Emerging evidence suggests ferroptosis inhibition as a therapeutic strategy against AKI. Cistanche (Cis), a kidney-tonifying herb in traditional Chinese medicine, exhibits antioxidant activity via nuclear factor erythroid 2-related factor 2 (Nrf2) activation. However, the precise molecular mechanism underlying cistanche-mediated Nrf2 activation remains undefined, and its nephroprotective potential in AKI is incompletely characterized. AIM OF THE

Peroxisome proliferator-activated receptor-delta induces cell proliferation by a cyclin E1-dependent mechanism and is up-regulated in thyroid tumors.

Peroxisome proliferator-activated receptors (PPARs) are lipid-sensing nuclear receptors that have been implicated in multiple physiologic processes including cancer. Here, we determine that PPARdelta induces cell proliferation through a novel cyclin E1-dependent mechanism and is up-regulated in many human thyroid tumors. The expression of PPARdelta was induced coordinately with proliferation in primary human thyroid cells by the activation of serum, thyroid-stimulating hormone/cyclic AMP, or epidermal growth factor/mitogen-activated protein kinase mitogenic signaling pathways. Low molecular weight fucoidan fraction ameliorates inflammation and deterioration of skin barrier in fine-dust stimulated keratinocytes.

Recently evidence linking the effects of fine-dust (FD) on skin inflammation is exaggerating. Fucoidan derived from brown algae has great potential for ameliorating oxidative stress and inflammation. Herein, a fucoidan fraction (SHC4-6) was purified from an enzymatic (Celluclast) extract of an invasive seaweed, Sargassum horneri following gradient ethanol precipitation and anion exchange chromatography. Effectiveness of SHC4-6 in ameliorating FD (from Beijing, China)-induced inflammatory responses in HaCaT keratinocytes and recovery of skin barrier dysfunction was evaluated. SHC4-6 was compris

Prognostic Value of Ki67 Percentage, WT-1 Expression and p16/CDKN2A Deletion in Diffuse Malignant Peritoneal Mesothelioma: A Single-Centre Cohort Study.

Diffuse malignant peritoneal mesothelioma (DMPM) is a rare malignant neoplasm with a poor survival. Although some advances in knowledge have been obtained for the pleural form, much less is known about DMPM. Advantages in terms of prognosis are still limited and strong efforts need to be made. The aim of our study was to correlate several histological and molecular factors with survival in a large cohort of 45 DMPMs. We evaluated histotype, nuclear grade, mitotic count, necrosis, inflammation, desmoplastic reaction, Ki67 percentage, WT-1 expression, p16 protein by immunohistochemistry and CDKN

Ameliorative Effect and Mechanism of Huoxiang-Huanglian on TNBS-Induced Inflammatory Bowel Disease in Zebrafish.

The Huoxiang-Huanglian (HH) drug pair has demonstrated efficacy in treating enteritis, yet its underlying mechanisms remain unclear. We integrated UPLC-MS, network pharmacology, and metabolomics to identify HH components and predict targets for inflammatory bowel disease (IBD). Molecular docking assessed interactions between bioactive compounds and core targets. A 2,4,6-trinitrobenzenesulfonic acid (TNBS)-induced zebrafish enteritis model was treated with HH (100, 200, and 400 μg/mL) or dexamethasone. Colon histopathology was evaluated via H&E and AB-PAS staining. Inflammatory cytokines, oxida

Extramammary Paget's disease presenting as alopecia neoplastica.

Extramammary Paget's disease (EMPD) is a rare malignant neoplasm with a predilection for the apocrine-rich anogenital skin and less commonly for the axilla. The tumor rarely occurs in non-apocrine bearing regions where it is referred to as ectopic EMPD. Here, we report a case of a patient that presented with a poorly circumscribed, erythematous plaque with patchy alopecia on the scalp. Histology showed pagetoid infiltration of the epidermis by atypical single and nested cells with abundant amphophilic cytoplasm with nuclear hyperchromasia, extending along the adnexae. Caloric restriction attenuates NLRP3 inflammasome activation and oxidative stress in leukocytes of patients with obesity.

Obesity promotes chronic low-grade inflammation through metabolic stress and activation of the NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome. The effects of caloric restriction on this pathway in humans are poorly understood; therefore, we evaluated the impact of a 6-month hypocaloric dietary intervention in 45 adults with obesity by assessing the metabolic, inflammatory, oxidative stress, and inflammasome-related markers in peripheral blood mononuclear cells. Weight loss improved metabolic parameters, reduced oxidative stress, and decreased the expression of nuclear

Protective potential of bempedoic acid as an AMPK activator in tamoxifen-induced steatohepatitis in rats.

Despite the beneficial therapeutic effects of tamoxifen (TAMX) against breast cancer, long-term treatment with TAMX enhances the development of metabolic dysfunction-associated steatotic liver disease (MASLD), including steatohepatitis. Bempedoic acid (BA) is a recently approved lipid-lowering drug for dyslipidemia. Our study aimed to examine the potential impact of BA against TAMX-induced steatohepatitis in rats and underline the possible molecular pathways involved. Twenty-four adult female rats were allocated into 4 groups (n = 6): Control, TAMX (45 mg/kg), BA (15 mg/kg), and BA (30 mg/kg)

Repetitively hypoxic preconditioning attenuates ischemia/reperfusion-induced liver dysfunction through upregulation of hypoxia-induced factor-1 alpha-dependent mitochondrial Bcl-xl in rat.

Repetitive hypoxic preconditioning (HP) enforces protective effects to subsequently severe hypoxic/ischemic stress. We hypothesized that HP may provide protection against ischemia/reperfusion (I/R) injury in rat livers via hypoxia-induced factor-1 alpha (HIF-1α)/reactive oxygen species (ROS)-dependent defensive mechanisms. Female Wistar rats were exposed to hypoxia (15 h/day) in a hypobaric hypoxic chamber (5500 m) for HP induction, whereas the others were kept in sea level. These rats were subjected to 45 min of hepatic ischemia by portal vein occlusion followed by 6 h of reperfusion. We eval

Response of mouse skin to tattooing: use of SKH-1 mice as a surrogate model for human tattooing.

Tattooing is a popular cosmetic practice involving more than 45 million US citizens. Since the toxicology of tattoo inks and pigments used to formulate tattoo inks has not been reported, we studied the immunological impact of tattooing and determined recovery time from this trauma. SKH-1 hairless mice were tattooed using commercial tattoo inks or suspensions of titanium dioxide, cadmium sulfide, or iron oxide, and sacrificed at 0.5, 1, 3, 4, 7, or 14 days post-tattooing. Histological evaluation revealed dermal hemorrhage at 0.5 and 1 day. Acute inflammation and epidermal necrosis were initiate

NF-kappaB mediated glucocorticoid response in the inner ear after acoustic trauma.

The inner ear of humans and experimental animals demonstrate an abundance of glucocorticoid receptors (GR). Glucocorticoids (GC) are widely used to treat different hearing disorders; yet the mechanisms of GC action on the inner ear are unknown. We demonstrate how GR can directly modulate hearing sensitivity in response to a moderate acoustic trauma that results in a hearing loss (10-30 dB). The GC agonist (dexamethasone) and the drugs (metyrapone + RU 486) showed opposing effects on hearing threshold shifts. GC agonist (dexamethasone) decreased the hearing threshold whereas pre-treatment with

Pterostilbene: a potential therapeutic agent for fibrotic diseases.

Fibrosis is a prevailing pathology in chronic diseases and accounts for 45% of deaths in developed countries. This condition is primarily identified by the transformation of fibroblasts into myofibroblasts and the overproduction of extracellular matrix (ECM) by myofibroblasts. Pterostilbene (PTS) is a natural analogue of resveratrol and is most commonly found in blueberries. Research has shown that PTS exerts a wide range of pharmacological effects, such as antioxidant, anti-inflammatory, and anticancer effects. As a result, PTS has the potential to prevent and cure numerous diseases. Emerging
