Obesity is associated with greater variability of reward signals in the nucleus accumbens.

Binge eating disorder (BED) is characterized by repeated episodes of binge eating accompanied by a loss of control. Although the neurobiological underpinnings of binge eating (BE) episodes are not fully understood, there are indications that variability in nucleus accumbens (NAcc) responses could lead to increased variability in food intake. Here, we assessed whether BED is associated with higher intra-individual variability in behavioral and neuroimaging indices of reward responses. To this end, patients with BED (n = 35, MBMI = 33.2 kg/m2 ± 6.8), participants with subsyndromal BED (n = 21, M

Synaptic plasticity in cocaine-seeking ensembles of the nucleus accumbens core.

Cue-induced seeking engages neuronal ensembles within the nucleus accumbens core (NAcore), with neuronal ensembles defined here as neurons coactivated during specific behavioral experiences that have been implicated in cued-reinstatement. Although transient synaptic plasticity has been widely observed in unidentified ensemble and non-ensemble neuronal populations in the NAcore during reinstatement, its expression within behaviorally relevant ensembles remains unclear. Here, we used c-Fos-TRAP2-based tagging to characterize structural and functional synaptic plasticity within ensembles during c

Nucleus accumbens dynamics in food reward seeking and regulation by sleep.

Reward-seeking behavior is essential for survival and is greatly influenced by experience, internal states, and physiological factors such as sleep. The nucleus accumbens (NAc) is reward processing hub that integrates external and internal signals to regulate reward-seeking behaviors. However, it is not well understood how NAc activities during reward seeking may be shaped by learning experience, and to what extent that it may be subject to physiological regulations such as sleep. Here, we used in vivo fiber photometry to monitor calcium (Ca2+) activities in the NAc of male and female mice und

nNOS/NO in nucleus accumbens medicates NKCC1 in neonatal repetitive pain.

Exposure to repetitive pain procedures in the neonatal intensive care unit (NICU) leads to long-term effects on pain response in adults. However, the underlying mechanisms in the central nervous system (CNS) are unclear. This study aimed to invesitgate the role of neuronal nitric oxide synthase (nNOS) and nitric oxide (NO) in the nucleus accumbens (NAc), including increased protein expression, cell number, and specific enzyme activity, which helps accelerate pain. We found that repetitive pain induced the upregulation of stargazin, but there was no significant difference in N-ethylmaleimide se

Investigation of Poststroke Depression Following a Nucleus Accumbens Infarct in Mice.

BACKGROUND: Poststroke depression (PSD) affects ≈33% of individuals 1 year after a stroke. Blood-brain barrier (BBB) dysfunction in the nucleus accumbens (NAc), a hub for emotional processing, reward, and mood regulation, has been linked to stress-induced depressive-like behaviors in male mice. Neurovascular alterations were also observed in postmortem tissue samples from men with a diagnosis of major depression. Thus, we aimed to investigate if BBB changes in the NAc could contribute to PSD pathophysiology. METHODS: Stereotaxic injection of ET-1 (endothelin-1), a potent vasoconstrictor, was

Functional Connectivity of the Nucleus Accumbens and Changes in Appetite in Patients With Depression.

IMPORTANCE: Major depressive disorder (MDD) is characterized by a substantial burden on health, including changes in appetite and body weight. Heterogeneity of depressive symptoms has hampered the identification of biomarkers that robustly generalize to most patients, thus calling for symptom-based mapping. OBJECTIVE: To define the functional architecture of the reward circuit subserving increases vs decreases in appetite and body weight in patients with MDD by specifying their contributions and influence on disease biomarkers using resting-state functional connectivity (FC). DESIGN, SETTING

Selective filtering of excitatory inputs to nucleus accumbens by dopamine and serotonin.

The detailed mechanisms by which dopamine (DA) and serotonin (5-HT) act in the nucleus accumbens (NAc) to influence motivated behaviors in distinct ways remain largely unknown. Here, we examined whether DA and 5-HT selectively modulate excitatory synaptic transmission in NAc medium spiny neurons in an input-specific manner. DA reduced excitatory postsynaptic currents (EPSCs) generated by paraventricular thalamus (PVT) inputs but not by ventral hippocampus (vHip), basolateral amygdala (BLA), or medial prefrontal cortex (mPFC) inputs. In contrast, 5-HT reduced EPSCs generated by inputs from all

Impaired glutamate homeostasis in the nucleus accumbens in human cocaine addiction.

Cocaine addiction is characterized by overwhelming craving for the substance, which drives its escalating use despite adverse consequences. Animal models suggest a disrupted glutamate homeostasis in the nucleus accumbens to underlie addiction-like behavior. After chronic administration of cocaine, rodents show decreased levels of accumbal glutamate, whereas drug-seeking reinstatement is associated with enhanced glutamatergic transmission. However, due to technical obstacles, the role of disturbed glutamate homeostasis for cocaine addiction in humans remains only partially understood, and accor

Methamphetamine leads to the alterations of microRNA profiles in the nucleus accumbens of rats.

CONTEXT: MicroRNA (miRNA) is an important regulator of gene expression. Methamphetamine (METH) induces a variety of alterations in different systems by affecting gene expression, but the effects of METH on miRNA profiles need to be elucidated. OBJECTIVES: This study develops a rat model of METH addiction, and analyzes the expression profile alterations of miRNA in nucleus accumbens (NAc) of the METH-addicted rats. MATERIALS AND METHODS: Sprague-Dawley rats were administered 10 mg/kg METH or vehicle twice a day for 4 weeks. The addictive behaviour of rats was estimated by CPP test. The pathol

mTOR signaling in the nucleus accumbens mediates behavioral sensitization to methamphetamine.

Chronic psychostimulant treatment in rodents readily produces behavioral sensitization, which reflects altered brain function in response to repeated drug exposure. Numerous morphological and biochemical investigations implicate altered neural plasticity in striatal medium spiny neurons (MSNs) as an essential component in behavioral sensitization. The mammalian target of the rapamycin (mTOR) signaling pathway, a key regulator of synaptic neuroplasticity, in the ventral striatum of methamphetamine (METH) -sensitized mice was investigated to determine if a link exists with the development of MET

Regulation of BAZ1A and nucleosome positioning in the nucleus accumbens in response to cocaine.

Chromatin regulation, in particular ATP-dependent chromatin remodelers, have previously been shown to be important in the regulation of reward-related behaviors in animal models of mental illnesses. Here we demonstrate that BAZ1A, an accessory subunit of the ISWI family of chromatin remodeling complexes, is downregulated in the nucleus accumbens (NAc) of mice exposed repeatedly to cocaine and of cocaine-addicted humans. Viral-mediated overexpression of BAZ1A in mouse NAc reduces cocaine reward as assessed by conditioned place preference (CPP), but increases cocaine-induced locomotor activation

Exploring the neural mechanisms of finasteride: a proteomic analysis in the nucleus accumbens.

The enzyme 5α-reductase (5αR) catalyzes the conversion of progesterone and testosterone into neuroactive steroids implicated in a wide array of behavioral functions. The prototypical 5αR inhibitor, finasteride (FIN), is clinically approved for the treatment of androgenic alopecia and benign prostatic hyperplasia. Recent evidence has shown that FIN, albeit generally well tolerated, can induce untoward psychological effects in a subset of patients; furthermore, this drug may have therapeutic efficacy for a number of different neuropsychiatric conditions, ranging from Tourette syndrome to schizop

AUTS2 in the nucleus accumbens is essential for heroin-induced behavioral sensitization.

Autism susceptibility candidate 2 (AUTS2) is a gene associated with autism and mental retardation. Recent studies have suggested an association of the AUTS2 gene with heroin dependence, and reduced AUTS2 gene expression may confer increased susceptibility to heroin dependence. However, the functional role of the AUTS2 protein in regulating enduring neuroadaptations in response to heroin exposure has not been established. Here, we investigated the effects of acute and chronic heroin exposure on AUTS2 mRNA and protein expression in the nucleus accumbens (NAc) and caudate-putamen (CPu) to determi

Delay of gratification in childhood linked to cortical interactions with the nucleus accumbens.

Delay of gratification (DG) is the ability to forego immediate temptations in the service of obtaining larger, delayed rewards. An extensive body of behavioral research has revealed that DG ability in childhood is associated with a host of important outcomes throughout development, and that attentional focus away from temptations underlies this ability. In this study, we conducted a functional magnetic resonance imaging study to identify the neural underpinnings of individual differences in DG among children. We observed a relationship between behavior during the classic DG task, a well-studie

Invigoration of reward seeking by cue and proximity encoding in the nucleus accumbens.

A key function of the nucleus accumbens is to promote vigorous reward seeking, but the corresponding neural mechanism has not been identified despite many years of research. Here, we study cued flexible approach behavior, a form of reward seeking that strongly depends on the accumbens, and we describe a robust, single-cell neural correlate of behavioral vigor in the excitatory response of accumbens neurons to reward-predictive cues. Well before locomotion begins, this cue-evoked excitation predicts both the movement initiation latency and the speed of subsequent flexible approach responses, bu

Biological substrates of reward and aversion: a nucleus accumbens activity hypothesis.

The nucleus accumbens (NAc) is a critical element of the mesocorticolimbic system, a brain circuit implicated in reward and motivation. This basal forebrain structure receives dopamine (DA) input from the ventral tegmental area (VTA) and glutamate (GLU) input from regions including the prefrontal cortex (PFC), amygdala (AMG), and hippocampus (HIP). As such, it integrates inputs from limbic and cortical regions, linking motivation with action. The NAc has a well-established role in mediating the rewarding effects of drugs of abuse and natural rewards such as food and sexual behavior. However, a

Nucleus accumbens long-term depression and the expression of behavioral sensitization.

Drug-dependent neural plasticity related to drug addiction and schizophrenia can be modeled in animals as behavioral sensitization, which is induced by repeated noncontingent or self-administration of many drugs of abuse. Molecular mechanisms that are critical for behavioral sensitization have yet to be specified. Long-term depression (LTD) of alpha-amino-3-hydroxy-5-methyl-isoxazole-4-propionic acid receptor (AMPAR)-mediated synaptic transmission in the brain has been proposed as a cellular substrate for learning and memory. The expression of LTD in the nucleus accumbens (NAc) required clathr

Hashimoto's encephalopathy with selective involvement of the nucleus accumbens: a case report.

Hashimoto's encephalopathy (HE) is an acute or subacute relapsing disorder usually affecting euthyroid patients with evidence of autoimmune thyroiditis. The neurological manifestations are non-specific, with subacute cognitive impairment, movement disorders, generalized seizures, focal neurological symptoms such as stroke-like episodes, or psychiatric disturbances. Autoimmune phenomena are likely to play an etiological role. Magnetic resonance imaging (MRI) findings are usually normal or show non-specific changes. We report the case of an 11-year-old girl with autoimmune thyroiditis who presen

Thought disorder and nucleus accumbens in childhood: a structural MRI study.

Thought disorder has been described as a hallmark feature in both adult and childhood-onset schizophrenia. The nucleus accumbens (NAc) has been repeatedly proposed as a critical station for modulating gating of information flow and processing of information within the thalamocortical circuitry. The aim of the present study was to investigate the relationship of thought disorder measures, which were administered to 12 children with schizophrenia and 15 healthy age-matched controls, and NAc volumes obtained from high-resolution volumetric magnetic resonance imaging analyses.
