Evidence for different classes of calcitonin binding sites in rat CNS: an autoradiographic study with carbo-calcitonin.

The distribution of binding sites in the rat CNS for a synthetic analogue of eel calcitonin, [Asu1-7]eel calcitonin (carboCT) was investigated. This distribution was compared to that for the natural peptide to see whether a modified molecule would also reveal different classes of binding sites for CT. The regional distribution of 125I-carboCT binding in coronal sections of rat CNS was examined by an in vitro autoradiographic technique. Non-specific binding was assessed after addition of excess cold carboCT or eel CT and the results showed that carboCT binding is specific and that it is displac

Higher Prevalence of Diabetes in Pontine Infarction than in Other Posterior Circulation Strokes.

BACKGROUND: Pontine infarction is the major subtype of posterior circulation stroke, and diabetes is more common in pontine infarction patients than in anterior circulation stroke patients. Whether the prevalence of diabetes remains homogenous within the posterior circulation stroke population is unclear. The present study is aimed at investigating the prevalence of diabetes in pontine infarction and comparing it to other subtypes of posterior circulation stroke. METHODS: We conducted a multicenter case-control study. Patients with posterior circulation stroke were screened. The subjects were

Relationship between cardiovascular neurones and descending antinociceptive pathways in the rostral ventrolateral medulla of the cat.

It has been previously reported that injection of neuroexcitatory compounds into the rostral ventrolateral medulla (RVLM) can produce an inhibition of nociceptive reflexes, often associated with a rise in arterial blood pressure. The aim of this study was to determine whether the subretrofacial (SRF) nucleus, which is a highly circumscribed group of cells within the RVLM known to play a major role in cardiovascular regulation also has an antinociceptive function. In barbiturate-anaesthetised and paralysed cats, unilateral microinjections of the neuroexcitatory compound sodium glutamate (8-20 n

Anatomy and motor pathways of the electric organ of skates.

The electric organ of skates is a paired structure within the tail consisting of two longitudinal columns of electrocytes contained within the lateral musculature on each side of the vertebral column. The electrocytes develop from hypaxial skeletal muscle fibers, and, depending upon the species, are generally classified as either cup-shaped or disc-shaped. The disc-shaped electrocytes are considered to be the more derived type. Regardless of the morphology of the electrocyte, the electric organ discharge of all skates is characterized as a weak asynchronous and long-lasting signal. Although re

Dual action of substance P in nociception and pain modulation at spinal level.

In the present study, the activation of the nociceptive dorsal horn neurons in rats by iontophoretic substance P (SP) has been observed. Thus, the role of SP in spinal nociception is further identified. In addition, the inhibitory effects of iontophoretic SP on nociceptive response (C response) of dorsal horn neurons can also be observed even in rats that have undergone dorsal half transection of spinal cord. These inhibitory effects can be partially blocked by pretreatment with iontophoretic bicuculline but not by naloxone. It indicates that the SP-induced inhibitory effects on the nociceptiv

Dextromethorphan suppresses both formalin-induced nociceptive behavior and the formalin-induced increase in spinal cord c-fos mRNA.

The injection of dilute formalin results in a stereotyped nociceptive behavioral response. Administration of dextromethorphan (s.c.) but not saline, 30 min prior to intraplantar formalin injection prevents this nociceptive response in a dose-dependent manner. In addition, intraplantar formalin reliably induces c-fos mRNA in the ipsilateral spinal dorsal horn as assessed with quantitative solution hybridization at 30 min postinjection. No change in c-fos mRNA was detected in the contralateral spinal dorsal horn, nucleus raphe magnus, periaqueductal grey, medial thalamus, or sensorimotor cortex.

Extracellular signal-regulated kinase activation in the chronic constriction injury model of neuropathic pain in anaesthetized rats.

BACKGROUND: The role of extracellular signal-regulated kinases (ERKs) in nociception has been explored in the last years. While in spinal cord their activation is frequently correlated with pain or acute noxious stimuli, supraspinally, this association is not so evident and remains unclear. This study aims to evaluate ERK1/2 activation in the spinal cord and brainstem nuclei upon neuropathy and/or an additional mechanical stimulus. METHODS: Acute noxious mechanical stimulation was applied in the left hindpaw of anaesthetized SHAM-operated and chronic constriction injured (CCI, neuropathic pai

A potential role for two brainstem nuclei in craniovascular nociception and the triggering of migraine headache.

AIM: To use an animal model of migraine to test whether migraine headache might arise from a brainstem-trigeminal nucleus pathway. METHODS: We measured evoked and spontaneous activity of second-order trigeminovascular neurons in rats to test whether the activity of these neurons increased following the induction of cortical spreading depression or the imposition of light flash - two potential migraine triggers, or headache provokers. We then tested whether drugs that could activate, or inactivate, neurons of the nucleus raphe magnus or the periaqueductal gray matter, would affect any such inc

Neuromodulation of Cardiac Ischemic Pain: Role of the Autonomic Nervous System and Vasopressin.

Cardiac pain is an index of cardiac ischemia that helps the detection of cardiac hypoxia and adjustment of activity in the sufferer. Drivers and thresholds of cardiac pain markedly differ in different subjects and can oscillate in the same individual, showing a distinct circadian rhythmicity and clinical picture. In patients with syndrome X or silent ischemia, cardiac pain intensity may cause neurogenic stress that potentiates the cardiac work and intensifies the cardiac hypoxia and discomfort of the patient. The reasons for individual differences in cardiac pain sensation are not fully unders

Morphine microinjected into the nucleus tractus solitarius and rostral ventrolateral medullary nucleus enhances somatosympathetic A- and C- reflexes in anesthetized rats.

The modulatory effects of morphine microinjected into localized areas of the brainstem on somatosympathetic A- and C-reflexes were examined in urethane-anesthetized rats. Somatosympathetic A- and C-reflexes were elicited in a branch of the inferior cardiac nerve by electrical stimulation of myelinated (A) and unmyelinated (C) afferent fibers in the tibial nerve. Morphine (0.002-0.2 microgram/50 nl) was microinjected into the rostral, intermediate and caudal parts of the nucleus tractus solitarius (NTS), the rostral ventrolateral medullary nucleus (RVLM), the caudal ventrolateral medullary nucl

[Is dorsal vagal complex the key nucleus of acupuncture regulation of gastric function?].

Acupuncture has remarkable effects on treating functional gastrointestinal diseases, but its central mechanism is not clear. At present, the research has mainly focused on several central nuclei, such as the dorsal vagus complex (DVC), nucleus raphe magnus (NRM), locus coeruleus (LC), subnucleus reticularis dorsalis (SRD), hypothalamic paraventricular nucleus (PVN), cerebellar fastigial nucleus (FN), central amygdala (CeA), etc. It is not clear whether the nuclei are involved in acupuncture regulation of gastric function through certain interrelation. Effects of somatostatin, a somatostatin agonist, and an antagonist, on a putative migraine trigger pathway.

OBJECTIVE: To determine whether somatostatin (SST) could be a cortico-brainstem neurotransmitter involved in producing the headache of migraine. BACKGROUND: There is evidence to support the idea that a cortico-brainstem-trigeminal nucleus neuraxis might be responsible for producing migraine headache; we have suggested that SST may be one of the neurotransmitters involved. METHODS: Rats were anesthetised and prepared for recording neurons in either the periaqueductal gray matter (PAG) or nucleus raphe magnus (NRM), as well as the trigeminal nucleus caudalis (TNC). Monoaminergic descending pathways contribute to modulation of neuropathic pain by increasing-intensity treadmill exercise after peripheral nerve injury.

This study characterizes the impact of increasing-intensity treadmill exercise (iTR) on noradrenergic (NE) and serotonergic (5HT) modulation of neuropathic pain. Following sciatic nerve transection and repair (SNTR) rats developed significant mechanical and thermal hyperalgesia that was partially prevented by iTR performed during the first 2weeks after injury. Marked decrease in the expression of 5HT2A and α1A and β-, but not α2A adrenergic receptors in the spinal cord dorsal horn was associated to SNTR and recovered by iTR, particularly in lamina II. iTR significantly increased 5HT2A in peria

Role of substantia Nigra dopaminergic neurons in respiratory modulation and limitations of levodopa in Parkinson's disease.

The substantia nigra pars compacta (SNpc), a midbrain region enriched with dopaminergic neurons projecting to the dorsal striatum, is essential for motor control and has been implicated in respiratory modulation. In Parkinson's disease (PD) models, the loss of SNpc dopaminergic neurons correlates with baseline respiratory deficits, suggesting a potential link between dopaminergic dysfunction and respiratory impairments. To explore this, we used adult transgenic mice (Vglut2cre Ai6 and Vgatcre Ai6) to map neurotransmitter phenotypes, as well as DATcre mice for pharmacogenetic modulation of SNpc

Relationship of presympathetic-premotor neurons to the serotonergic transmitter system in the rat brainstem.

Numerous physiological conditions and emotionally motivated behaviors require concomitant activation of somatomotor and sympathetic efferents. Using a virally mediated retrograde transsynaptic tract-tracing approach, we have previously determined locations of presympathetic-premotor neurons (PSPMNs) in the rat brainstem. These putative dual-function neurons send projections to somatomotor and sympathetic targets and likely participate in sympatho-somatomotor integration. A significant portion of these neurons is found within brainstem areas known to contain serotonergic neurons. Thus, we hypot

Central antiaversive and antinociceptive effects of anterior pretectal nucleus stimulation: attenuation of autonomic and aversive effects of medial hypothalamic stimulation.

Previous studies have shown that stimulation of the rat anterior pretectal nucleus (APtN) strongly depresses a spinal reflex to noxious heat without causing significant aversion or depression of other motor responses. It is not known if APtN stimulation can similarly reduce the aversiveness of electrical stimulation of the brain, nor is it known if APtN stimulation is itself rewarding or aversive. This study used a simple switch-off paradigm to examine the rewarding properties of APtN stimulation at different sites throughout the nucleus and also used the tail-flick test to determine if the st

Physiological and anatomic evidence for functional subclasses of serotonergic raphe magnus cells.

Serotonergic cells in the medullary nucleus raphe magnus (RM) and adjacent nucleus reticularis magnocellularis (NRMC) project to the spinal cord where they are likely to modulate nociceptive transmission. Previous studies have suggested that these cells are physiologically and anatomically heterogeneous. In the present investigation, we examined whether subclasses of serotonergic RM and NRMC cells can be delineated based on their response to a visceral stimulus, and whether any such subclasses are morphologically distinct. Most RM and NRMC serotonergic cells tested (81 of 116) responded to ret

Antinociceptive effects of stimulation of discrete sites in the rat hypothalamus: evidence for the participation of the lateral hypothalamus area in descending pain suppression mechanisms.

The sites in the rat hypothalamus where microinjection of morphine (5 micrograms/0.5 microliters) or electrical stimulation depresses the tail withdrawal reflex to noxious heating of the skin were examined. Among other hypothalamic sites found to be sensitive to morphine or to an electrical stimulus, the posterior part of the lateral hypothalamic area (LHA) was the only portion of the hypothalamus that was strongly sensitive to both manipulations. A 15-sec period of 35 microA sine-wave stimulation of the LHA significantly increased the latency of the tail reflex for periods up to 30 min. The e

Motor Cortex Stimulation Reversed Hypernociception, Increased Serotonin in Raphe Neurons, and Caused Inhibition of Spinal Astrocytes in a Parkinson's Disease Rat Model.

Persistent pain is a prevalent symptom of Parkinson's disease (PD), which is related to the loss of monoamines and neuroinflammation. Motor cortex stimulation (MCS) inhibits persistent pain by activating the descending analgesic pathways; however, its effectiveness in the control of PD-induced pain remains unclear. Here, we evaluated the analgesic efficacy of MCS together with serotonergic and spinal glial modulation in an experimental PD (ePD) rat model. Wistar rats with unilateral striatal 6-OHDA and MCS were assessed for behavioral immobility and nociceptive responses. The immunoreactivity

Functional connectome of brainstem nuclei involved in autonomic, limbic, pain and sensory processing in living humans from 7 Tesla resting state fMRI.

Despite remarkable advances in mapping the functional connectivity of the cortex, the functional connectivity of subcortical regions is understudied in living humans. This is the case for brainstem nuclei that control vital processes, such as autonomic, limbic, nociceptive and sensory functions. This is because of the lack of precise brainstem nuclei localization, of adequate sensitivity and resolution in the deepest brain regions, as well as of optimized processing for the brainstem. To close the gap between the cortex and the brainstem, on 20 healthy subjects, we computed a correlation-based
