Lutein has low stability and limited bioaccessibility in infant formulas. By integrating in vitro (static and dynamic) and in vivo digestion models, oleic acid (OA), gondoic acid (GA) and phosphatidylethanolamine (PE) were identified as agents that raise lutein bioaccessibility by 6-13% under static in vitro conditions. During digestion the additives reduced emulsion droplet size (0.3% PE most effective), inhibited lipid-droplet aggregation and enhanced micellar stability. PE additionally accelerated intestinal free fatty acid release. Dynamic digestion gave higher absolute values but identica

Energy, protein, essential fatty acids, vitamins and minerals are all necessary for normal growth and development of children. In undernutrition, there is evidence for abnormal fatty acid metabolism. High levels of oleic acid and low levels of docosahexaenoic acid in plasma phosphatidylcholine have been associated with weight-for-age Z-score shifts to the left (increased prevalence of underweight) and vice versa (reduced prevalence of underweight). An alternative hypothesis is proposed, which describes a possible physiological mechanism whereby omega3 fatty acids contribute to growth. High blo

Metabolomics has been used to study ophthalmic diseases for the past several years. However, there was rare metabolomics study was reported about primary angle-closure glaucoma (PACG). The purpose of this study was to explore a two-step metabolomics analysis method for identifying differential metabolites in the aqueous humor of patients with PACG. Aqueous humor samples were collected from 20 patients each with age-related cataracts, acute PACG, and chronic PACG. In the first step, the changed metabolites were identified using a non-targeted metabolic analysis. In the second step, metabolites

The nature of activation signals is essential in determining T cell subset differentiation; however, the features that determine T cell subset preference acquired during intrathymic development remain elusive. Here we show that naive CD4+ T cells generated in the mouse thymic microenvironment lacking Scd1, encoding the enzyme catalyzing oleic acid (OA) production, exhibit enhanced regulatory T (Treg) cell differentiation and attenuated development of experimental autoimmune encephalomyelitis. Scd1 deletion in K14+ thymic epithelia recapitulated the enhanced Treg cell differentiation phenotype

BACKGROUND: Erdosteine is a mucolytic agent with antioxidant and anti-inflammatory effects. We evaluated the protective effect of erdosteine pretreatment on oleic acid (OA)-induced acute lung injury.
MATERIALS AND METHODS: Twenty-four male Wistar Albino rats were assigned to four treatments: control (oral saline + 50 μL intravenous [i.v.] saline), OA (oral saline + 50 μL i.v. OA), erdosteine (150 mg/kg oral erdosteine + 50 μL i.v. saline), and OA + erdosteine (150 mg/kg oral erdosteine + 50 μL i.v. OA). Four hours after OA injection, lung tissues were excised for biochemical and histopatholog

The occurrence of topical fungal infections is increasing nowadays. Cutaneous fungal infections like cutaneous candidiasis are more prominent in patients associated with AIDS. Current available strategies for the treatment of cutaneous fungal infections are creams or gels which show various adverse effects on skin along with systemic absorption. These drawbacks can be overcome by using various novel drug delivery systems. So, the present investigation aims at exploring the potential of fatty acid vesicles (ufasomes) for the topical delivery of clotrimazole. Oleic acid was employed as a fatty m

UNLABELLED: Commonly, acute respiratory failure (ARF) in laboratory animals is induced through the intravenous infusion of oleic acid (OA). The methods by which OA is infused, and the methods by which droplets are generated, differ greatly among investigators. The resulting ARF, and the distribution of the underlying pulmonary pathology, are not highly reproducible. A method was developed that generated a reproducible, known spectrum of OA microdroplets. This method was applied to infuse a known volume of OA into the vena cava superior (VCS) in sheep, to induce ARF. In vitro studies were condu

Similarities between oleic acid (OA)-induced pulmonary injury and clinical adult respiratory distress syndrome (ARDS) have resulted in extensive use of this model. Using technetium 99m (Tc-99m)-labeled human serum albumin (Tc-HSA) we examined the effect of indomethacin (a prostaglandin synthetase inhibitor) and dexamethasone (a corticosteroid) alone and in combination on OA-induced pulmonary protein leak. Computer-acquired dynamic gamma camera imaging before (15 min), during, and after (60 min) OA infusion were used to generate time-activity curves for lung and heart regions. A lung:heart acti

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a chronic progressive disorder characterized by an excess accumulation of lipids in the liver. The aim of this study was to examine the role of bilirubin (BR), the catabolic heme product and a putative peroxisome proliferator-activated receptor alpha (PPARalpha) agonist, in an in vitro model of MASLD. In our study, we used human hepatoblastoma HepG2 cells exposed to oleic (OA)/palmitic acid (PA) (2:1 ratio, 24 h) with subsequent treatment with BR or fenofibrate (FF, a clinically used PPARalpha agonist) at clinically relevant c

Human leptospirosis is an acute infectious zoonosis presenting specific lipid disorders. Previous in vitro studies showed both leptospira glycolipoprotein endotoxin, and high oleic acid levels were associated with Na/K-ATPase inhibition that is amplified by the reduction of circulating albumin levels. In this study, we aimed to investigate the relationship of oleic acid/albumin (OA/A) molar ratio and clinical outcomes in Leptospirosis. Through a prospective observational cohort study employing high-performance liquid chromatography (HPLC) we sequentially determined serum concentrations of none

Marfan syndrome (MFS) is associated with progressive aortic dilatation and endothelial dysfunction that lead to early acute dissection and rupture of the aorta and sudden death. Alteration in fatty acid (FA) metabolism can stimulate nitric oxide (NO) overproduction which increases the activity of the inducible form of NO synthase (iNOS) that is involved in endothelial dysfunction. We evaluated the participation of FA in the formation of thoracic aneurysms in MFS and its relation to the iNOS. Oleic acid (OA), iNOS, citrulline, nitrates and nitrites, TGF-β1, TNF-α, monounsaturated FA and NO synt

This study was performed to establish a platform for further studies on effects of ventilatory treatment modalities on the intestines during mechanical ventilation of acute lung injury (ALI). We tested the hypotheses that oleic acid (OA) infusion causes changes in intestinal circulation, oxygenation and metabolism, and that OA is distributed to tissues outside the lung. This was performed as an experimental, prospective and controlled study in an university animal research laboratory. Thirteen juvenile anaesthetized pigs were used in the main study, where seven were given an intravenous infusi

Our purpose was to evaluate the pulmonary effects of mannitol infusion in a rat model of acute lung injury induced by oleic acid (OA) to compare the effects of mannitol to those of another diuretic, furosemide (FUR), and to assess if mannitol effects remained after correction of the volume depletion induced by this agent. Acute lung injury was induced in Wistar rats by intravenous administration of 100 mg/kg of OA. Mannitol (1 mL of a 20% solution) was infused either 15 min before or 2 h after OA infusion. FUR was infused intravenously in a dose (1 mg/kg) that induced a similar amount of diure

The effect of poly(ethylenimine) (PEI) dissolved in water on the surface pressure-area isotherm of oleic acid (OA) at the air/water interface was investigated. On a concentrated PEI solution, the isotherm of the OA monolayers exhibited a noticeable difference as a function of subphase pH. PEI caused the collapse pressure of the OA monolayer to increase up to 45 mN/m, due to a stronger acid-base-type interaction occurring between the amine group of the PEI and the carboxyl group of OA; on a pure water subphase, the collapse pressure was;28 mN/m. On the other hand, owing to a stronger OA-PEI int

OBJECTIVE: Development of a stable model of respiratory distress in pigs with oleic acid, fulfilling clinical criteria of the adult respiratory distress syndrome (ARDS).
DESIGN: Eight pigs (9.1 +/- 0.7 kg) were anesthetized with pentobarbital, paralyzed with tubocurarine and mechanically ventilated with an FIO2 of 0.6, an I:E ratio of 2:3 and a PEEP of 0.2 kPa. Oleic acid (dissolved 1:1 in 96% alcohol) was administered in a series of multiple injections of 0.1 ml until PaO2 was lower than 8 kPa.
MEASUREMENTS AND RESULTS: Careful titration of the oleic acid injections on guidance of the PaO2

Rats injected intravenously with oleic acid developed pulmonary edema leading to hypoxia and hypercarbia. These changes were accompanied by an increase in immunoreactive endothelin (ir-ET) in plasma as early as 15 min after injection. At 45 min after injection plasma levels peaked at 114 +/- 19 pg/ml plasma (n = 8) and reached basal levels again after 240 min. In contrast, much larger amounts of ir-ET were found in the bronchoalveolar lavage fluid, with a peak at 120 min (2878 +/- 258 pg/lung, n = 7) preceding the maximum hypoxia observed at 180 min. In both plasma and bronchoalveolar lavage f

This study was designed to determine whether indomethacin, a suggested treatment for adult respiratory distress syndrome (ARDS), would inhibit the efficacy of positive end-expiratory pressure (PEEP) in an animal model of ARDS. Functional residual capacity (FRC), alveolar-to-arterial oxygen difference (A-aPo2) and total lung water were measured in rabbits: 30 controls and 30 with oleic acid induced pulmonary oedema. Within each group, four treatments were administered: diluent (n = 12), PEEP (n = 6), indomethacin (n = 6), or PEEP + indomethacin (n = 6). Lung injury was induced at 30 mins and tr

This protocol outlines an acute respiratory distress model utilizing centrally administered oleic acid infusion in Yorkshire pigs. Prior to experimentation, each pig underwent general anesthesia, endotracheal intubation, and mechanical ventilation, and was equipped with bilateral jugular vein central vascular access catheters. Oleic acid was administered through a dedicated pulmonary artery catheter at a rate of 0.2 mL/kg/h. The infusion lasted for 60-120 min, inducing respiratory distress. Throughout the experiment, various parameters including heart rate, respiratory rate, arterial blood pre

An animal model experiment was conducted with nine adult sows to study the effects of long-chain fatty acids on thermogenesis when different fatty acids were replaced for 30% of the energy of a basal diet based on cereals and soybean meal. The acids were fed as commercial products containing as main constituent either palmitic acid, oleic acid, or linoleic acid, according to a latin square design in experimental periods 2 to 4. In periods 1 and 5 the sows were submitted to basal diet alone. Digestibility of palmitic acid was only 36%, whereas the unsaturated fatty acids were highly absorbed (9
