The ubiquity, high diversity and often-cryptic manifestations of fungi and oomycetes frequently necessitate molecular tools for detecting and identifying them in the environment. In applications including DNA barcoding, pathogen detection from plant samples, and genotyping for population genetics and epidemiology, rapid and dependable DNA extraction methods scalable from one to hundreds of samples are desirable. We evaluated several rapid extraction methods (NaOH, Rapid one-step extraction (ROSE), Chelex 100, proteinase K) for their ability to obtain DNA of quantity and quality suitable for th

INTRODUCTION: Globisporangium ultimum is an oomycetal pathogen causing damping-off on over 300 different plant hosts. Currently, as for many phytopathogens, its control relies in the use of chemicals with negative impact on health and ecosystems. Therefore, many biocontrol strategies are under investigation to reduce the use of fungicides.
RESULTS: In this study, the soil bacterium Pseudomonas sp. NCIMB 10586 demonstrates a strong iron-repressed in vitro antagonism against G. ultimum MUCL 38045. This antagonism does not depend on the secretion of the broad-range antibiotic mupirocin or of the

Burn spot disease, a form of shell disease in crayfish, has been linked to Fusarium avenaceum (in Astacus astacus and Pontastacus leptodactylus from Estonia and Turkey) and Fusarium solani (in a distinct form on Saaremaa Island, Estonia). Severe melanised lesions in P. leptodactylus from Belarus's Lyubanskoye Reservoir resemble the Saaremaa syndrome. This study aimed to characterise the culturable microorganisms in these lesions and to test for Fusarium species. We performed the first culture-based molecular characterisation of lesion-associated fungi and oomycetes in this host, using ITS rDNA

Nonmycorrhizal cluster root-forming species enhance the phosphorus (P) acquisition of mycorrhizal neighbours in P-impoverished megadiverse systems. However, whether mycorrhizal plants facilitate the defence of nonmycorrhizal plants against soil-borne pathogens, in return and via their symbiosis, remains unknown. We characterised growth and defence-related compounds in Banksia menziesii (nonmycorrhizal) and Eucalyptus todtiana (ectomycorrhizal, ECM) seedlings grown either in monoculture or mixture in a multifactorial glasshouse experiment involving ECM fungi and native oomycete pathogens. Roots

BACKGROUND: Oomycetes are pathogens of mammals, fish, insects and plants, and the potato late blight agent Phytophthora infestans and the oil palm and cocoa infecting pathogen Phytophthora palmivora cause economically impacting diseases on a wide range of crop plants. Increasing genomic and transcriptomic resources and recent advances in oomycete biology demand new strategies for genetic modification of oomycetes. Most oomycete transformation procedures rely on geneticin-based selection of transgenic strains.
RESULTS: We established N-acetyltransferase AAC(3)-I as a gentamicin-based selectabl

Oxathiapiprolin (OXTP), a highly effective fungicide against oomycete pathogens, represents a novel class of fungicides. In this study, a series of novel OXTP derivatives were synthesized and evaluated for their antifungal activity. Several compounds exhibited potent inhibitory effects, with B12 and B13 demonstrating particularly strong efficacy against plant diseases caused by Phytophthora species. In addition, select derivatives showed significant in vivo activity against fungal pathogens, expanding their potential application beyond oomycetes. These compounds also displayed excellent photos

Molds of the class of Oomycetes are of allergologic importance in special cases. However, probes are not commercially available for diagnostic purposes. Our case report is based on the medical history of a greenhouse worker who had atopic syndrome. He handled pure cultures of defined fungi and plants. A sensitization to pseudo mildew growing on grapevine (Plasmopara viticola) was found. Skin prick test and histamine release test results were positive when extract of P. viticola was used. Detection of IgE reactivities against pseudo mildew via binding tests, Western blot analysis, and isoelectr

This study purpose was to evaluate the in vitro inhibitory properties of Italian acacia honey extracts against pathogenic aquatic oomycete/fungal isolates that cause different diseases in crayfish, resulting in an elevated mortality rate. The antimycotic activity of acacia honey aqueous extracts was evaluated against the strain UEF88662 of Aphanomyces astaci (oomycete) and the strain SMM2 of Fusarium avenaceum (fungus). The extracts preparation was carried out with water by a cheap, not complex and organic solvent-free procedure, with low environmental impact and the higher possibility of larg

Oomycetes are water moulds in the kingdom Protista and are not considered true fungi due to the structural lack of chitin and ergosterol. Many oomycetes are pathogenic, such as Pythium spp., and many fish are prone to Saprolegnia spp. infections, particularly in stressful farming situations. A juvenile American alligator (Alligator mississippiensis) was presented for necropsy examination with white, gelatinous, raised lesions over ulcerated regions of skin on the limbs and tail. The alligator came from a hatchery with age-divided enclosures, and several of the animals within the same enclosure

Although protists occupy a vast range of habitats and are known to interact with plants among other things via disease suppression, competition or growth stimulation, their contributions to the 'phytobiome' are not well described. To contribute to a more comprehensive picture of the plant holobiont, we examined cercozoan and oomycete taxa living in association with the model plant Arabidopsis thaliana grown in two different soils. Soil, roots, leaves and wooden toothpicks were analysed before and after surface sterilization. Cercozoa were identified using 18S rRNA gene metabarcoding, whereas t

Novel fungicidal agents were designed based on the combination of two privileged scaffolds, thiohydantoin and spirocyclic butenolide, which are widely found in natural products. The synthesized compounds were characterized by 1H NMR, 13C NMR, and high-resolution electrospray ionisation mass spectrometry. The in vitro antioomycete activity evaluation showed that most of the compounds exhibited excellent inhibitory activities against different developmental stages in the life cycle of pathogenic oomycete Phytophthora capsici. Compound 5j could inhibit the mycelial growth, sporangium production,

Oxysterol-binding protein (OSBP)-related proteins (ORPs) are indispensable eukaryotic lipid transporters and promising targets for pharmaceutical and agrochemical exploitation. Among ORP family members across kingdoms, oomycete ORP1 constitutes a phylogenetically distinct subtype that evolved independently in oomycetes, which is markedly divergent from ORP homologues in animals, plants and fungi. Oxathiapiprolin, the first commercial fungicide targeting OSBP family proteins, specifically acts on oomycete ORP1 and exhibits robust inhibitory efficacy against diverse Phytophthora pathogens and do

Warmer waters are reshaping unseen interactions at the base of aquatic food webs, with potential consequences for microalgal bloom dynamics and carbon transfer to higher trophic levels. Two major groups of aquatic parasites-chytrids and oomycetes-infect the same algal host, and changes in temperature can alter the dynamics of these infections and determine their success. Using laboratory-controlled synthetic communities, we investigated the effect of temperature on chytrid and oomycete infection dynamics in the bloom-forming freshwater diatom Ulnaria ulna. Our findings reveal complex dynamics,

Oomycetes cause hundreds of destructive plant diseases, threatening agricultural production and food security. These fungus-like eukaryotes show multiple sporulation pattern including the production of sporangium, zoospore, chlamydospore and oospore, which are critical for their survival, dispersal and infection on hosts. Recently, genomic and genetic technologies have greatly promoted the study of molecular mechanism of sporulation in the genus Phytophthora and Peronophythora. In this paper, we characterize the types of asexual and sexual spores and review latest progress of these two genera.

Plasma membrane-located nucleotide transport proteins (NTTs) underpin the lifestyle of important obligate intracellular bacterial and eukaryotic pathogens by importing energy and nucleotides from infected host cells that the pathogens can no longer make for themselves. As such their presence is often seen as a hallmark of an intracellular lifestyle associated with reductive genome evolution and loss of primary biosynthetic pathways. Here, we investigate the phylogenetic distribution of NTT sequences across the domains of cellular life. Our analysis reveals an unexpectedly broad distribution of

Apple replant disease (ARD) is incited by a complex of causal agents including various fungi, oomycetes, and plant parasitic nematodes. These causal agents can differ significantly in abundance between orchard sites within a geographic region. Knowledge of the specific etiology of ARD is required in order to develop commercially viable soil management strategies to combat specific/individual components of the pathogen complex. In this study, we analyzed soil from six ARD affected orchard sites to assess the presence and composition of fungal, bacterial and oomycetes communities, as well as the

Natural products derived from Allium spp., such as garlic oil, garlic powder, and diallyl disulfide (DADS), are strong elicitors of sclerotia germination in the fungus Sclerotium cepivorum (syn. Stromatinia cepivora), the causal agent of Allium white rot. However, these compounds can also have broad antimicrobial activity against a wide range of bacteria, oomycetes, and other fungi when they are applied to soil. The objective of this study was to determine the potential impacts that DADS application has on soil microbial communities. DADS was applied to two soil types and incubated under aerob

Gene silencing mediated by small noncoding RNAs (sRNAs) is a fundamental gene regulation mechanism in eukaryotes that broadly governs cellular processes. It has been established that sRNAs are critical regulators of plant growth, development, and antiviral defence, while accumulating studies support positive roles of sRNAs in plant defence against bacteria and eukaryotic pathogens such as fungi and oomycetes. Emerging evidence suggests that plant sRNAs move between species and function as antimicrobial agents against nonviral parasites. Multiple plant pathosystems have been shown to involve a

Saprolegnia parasitica causes significant morbidity, mortality and economic loss in fish hatcheries and growing culture systems, especially during winter. The present study examined clinical and histopathological alterations in Pangasianodon hypophthalmus experimentally infected with the oomycete and evaluated the clinical efficacy of antifungal drug clotrimazole as bath treatment. Besides development of off-white cotton wool-like fungal growth on fins and skin, invasion of hyphae into underlying musculature and muscle fibre necrosis, experimental infection also induced gill lamellar hyperplas

Filamentous fungi and oomycetes have four large intracellular membrane systems consisting of tubes, lamellae, or a combination thereof. Such elongated membrane structures can extend for tens of micrometers in hyphal cells, branching and connecting into bundles and networks. These systems are represented by tubular vacuoles, elongated mitochondria, endoplasmic reticulum, and macroinvaginations of the plasma membrane. They are most developed in xylotrophic and mycorrhiza-forming basidiomycetes. This review examines three membrane systems, excluding the macroinvaginations. Various morphotypes of
