Development and degeneration of retina in rds mutant mice: ultraimmunohistochemical localization of opsin.

In normal retina the developing photoreceptor cells first show presence of opsin over the distal ends of the ciliary protrusions. In a fully differentiated cell intense activity is seen over the rod outer-segment discs; some activity is also seen over the Golgi zone and near the distal ends of the inner segments but the other parts of the receptor cell appear negative. In the pigment epithelium opsin is seen only over phagosomes containing rod outer segment debris. In the homozygous rds mutant retina, developing receptor cells show opsin activity over the ciliary protrusions as in the normal.

Attenuation of cocaine seeking in rats via enhancement of infralimbic cortical activity using stable step-function opsins.

RATIONALE: The infralimbic cortex (IL) and its downstream projection target the nucleus accumbens shell (NAshell) mediate the active suppression of cocaine-seeking behavior. Although an optogenetic approach would be beneficial for stimulating the IL and its efferents to study their role during reinstatement of cocaine seeking, the use of channelrhodopsin introduces significant difficulties, as optimal stimulation parameters are not known. OBJECTIVES: The present experiments utilized a stable step-function opsin (SSFO) to potentiate endogenous activity in the IL and in IL terminals in the NAsh

Ocular and extraocular expression of opsins reveals evolutionary trends underlying visual and non-visual functions in bivalves (Pteriomorphia).

Opsin proteins are central to photoreception and have diversified extensively to support both visual and non-visual functions in animals. While the molecular conservation is striking, the evolutionary pathways that shaped opsin diversity, particularly in relation to visual and extraocular roles, remain poorly understood. To address this gap, we examined opsin expression in Pteriomorphia, a diverse group of mostly sessile marine bivalves with multiple photoreceptor systems and ecologies. We generated transcriptomes for 12 species, including eyed and eyeless adults, and analysed published RNA-se

Disease mechanisms revealed in the P23H opsin knock-in mouse model of retinitis pigmentosa.

The molecular mechanisms driving disease progression in autosomal dominant retinitis pigmentosa (adRP) due to a proline-to-histidine substitution (P23H) in opsin are not completely understood. Thus, we undertook to unlock features of the P23H/+ knock-in mouse model. Bisretinoids, the source of short-wavelength fundus autofluorescence (SW-AF), were elevated in albino P23H rhodopsin transgenic rats and in black and albino P23H/+ mice relative to wild type. Conversely, bisretinoid levels were lower in albino P23H/+ relative to black P23H/+ mice. Fluorescence microscopy revealed an inherent and ab

Seeing in the Deep: Evolution of the Opsin Gene Expression in Bermin Crater Lake Cichlids.

Cichlid visual systems can evolve rapidly during adaptive radiations. The Bermin crater lake species flock in Cameroon, comprising 13 (nine valid and four undescribed) Coptodon species, offers an ideal model to investigate visual adaptation to the deep-water light environments. Here, we examine visual opsin genes sequences and expression using 109 retina transcriptomes, focusing on interspecific variation with habitat depth, as well as on seasonal changes in the migratory species between depths. All species possess a multichromatic system with at least five cone opsins. While opsin coding sequ

Physiological studies of the interaction between opsin and chromophore in rod and cone visual pigments.

The visual pigment in vertebrate photoreceptors is a G protein-coupled receptor that consists of a protein, opsin, covalently attached to a chromophore, 11-cis-retinal. Activation of the visual pigment by light triggers a transduction cascade that produces experimentally measurable electrical responses in photoreceptors. The interactions between opsin and chromophore can be investigated with electrophysiologial recordings in intact amphibian and mouse rod and cone photoreceptor cells. Here we describe methods for substituting the native chromophore with various chromophore analogs to investiga

Vertebrate ancient (VA) opsin and extraretinal photoreception in the Atlantic salmon (Salmo salar).

A member of a new photopigment family first isolated from teleost fish, vertebrate ancient (VA) opsin, has recently been shown to form a functional photopigment and to be expressed within a subset of horizontal and amacrine cells of the inner retina. These sites of expression (and structural features) of VA opsin suggest that this photopigment might mediate non-image-forming light-detection tasks. We attempted to gain support for this hypothesis by examining the expression of VA opsin within the central nervous system (CNS) (pineal and deep brain) of the Atlantic salmon Salmo salar. In additio

Characterization of acquired resistance in lesion-mimic transgenic potato expressing bacterio-opsin.

The lesion-mimic mutants of certain plants display necrotic lesions resembling those of the hypersensitive response and activate local and systemic defense responses in the absence of pathogens. We have engineered a lesion-mimic phenotype in transgenic Russet Burbank potato plants through constitutive expression of a bacterio-opsin (bO) proton pump derived from Halobacterium halobium. Transgenic potato plants exhibiting a lesion-mimic phenotype had increased levels of salicylic acid and overexpressed several pathogenesis-related messenger RNAs, all hallmarks of systemic acquired resistance (SA

Shared nuclear protein binding sites in the upstream region of the rat opsin gene.

DNase I protection and gel retardation assays have identified two sequences 5' to the rat opsin gene that interact with nuclear proteins from retina but not from a number of neuronal and non-neuronal tissues. These sites, Ret 2 and Ret 3, are over 1200 base pairs apart but seem to interact with the same protein(s). Synthetic oligonucleotides corresponding to each site were able to inhibit complex formation in a gel retardation assay using an oligonucleotide corresponding to the other site. The proteins binding to the Ret 2 and Ret 3 sites co-eluted in both ion exchange and gel filtration chrom

The fate of immunoreactive opsin following phagocytosis by pigment epithelium in human and monkey retinas.

Polyclonal and monoclonal antibodies to human rhodopsin were used to identify and localize this principal glycoprotein of the photoreceptor outer segment discs on thin sections of human and monkey retinal pigment epithelium (RPE) and on immunoblots of RPE subcellular fractions following gel electrophoresis. Antiopsin was visualized with protein A-gold labeling by electron microscopy or peroxidase-linked second antibody on immunoblots. In immunocytochemical studies using polyclonal antibodies, the rod outer segments (ROS) were heavily labeled whereas cone outer segments labeling was variable an

Ultra-low power deep sustained optogenetic excitation of human ventricular cardiomyocytes with red-shifted opsins: a computational study.

The main challenge in cardiac optogenetics is to have low-power, high-fidelity deep excitation of cells with minimal invasiveness and heating. We present a detailed computational study of optogenetic excitation of human ventricular cardiomyocytes (HVCMs) with new ChRmine, bReaChES and CsChrimson red-shifted opsins to overcome the challenge. Action potentials (APs) in ChRmine-expressing HVCMs can be triggered at 6 µW mm-2 (10 ms pulse) and 0.7 µW mm-2 (100 ms pulse) at 585 nm, which is two orders of magnitude lower than ChR2(H134R). This enables safe sustained excitation of deeply situated card

Proteomic analysis of opsins and thyroid hormone-induced retinal development using isotope-coded affinity tags (ICAT) and mass spectrometry.

PURPOSE: Analyses that reveal the relative abundance of proteins are informative in elucidating mechanisms of retinal development and disease progression. However, popular high-throughput proteomic methods do not reliably detect opsin protein abundance, which serve as markers of photoreceptor differentiation. We utilized thyroid-hormone (TH) treatment of rainbow trout (Oncorhynchus mykiss) as a model of cone apoptosis and cone regeneration. We used this model to investigate if emerging proteomic technology allows effective analysis of retinal development and opsin protein abundance. We also so

Opsin 5 mediates violet light-accelerated wound healing in mammalian corneas.

The cornea is the transparent tissue at the ocular surface that generates most of the refractive power of the eye. Due to its exposed location, the cornea is uniquely in danger of injury. Rapid and efficient healing is required for high-acuity vision. Here, we show that epithelial corneal wounding induces Opn5, an opsin G-protein-coupled receptor (GPCR) sensitive to ultraviolet/violet light, at the cornea's surface in both mice and primates. The expression of corneal Opn5 is coincident with the direct light sensitivity of multiple pathways, including circadian rhythms, damage-response genes, a

The co-chaperone and reductase ERdj5 facilitates rod opsin biogenesis and quality control.

Mutations in rhodopsin, the light-sensitive protein of rod cells, are the most common cause of autosomal dominant retinitis pigmentosa (ADRP). Many rod opsin mutations, such as P23H, lead to misfolding of rod opsin with detrimental effects on photoreceptor function and viability. Misfolded P23H rod opsin and other mutations in the intradiscal domain are characterized by the formation of an incorrect disulphide bond between C185 and C187, as opposed to the correct and highly conserved C110-C187 disulphide bond. Therefore, we tested the hypothesis that incorrect disulphide bond formation might b

Cone photoreceptor mosaic disruption associated with Cys203Arg mutation in the M-cone opsin.

Missense mutations in the cone opsins have been identified as a relatively common cause of red/green color vision defects, with the most frequent mutation being the substitution of arginine for cysteine at position 203 (C203R). When the corresponding cysteine is mutated in rhodopsin, it disrupts proper folding of the pigment, causing severe, early onset retinitis pigmentosa. While the C203R mutation has been associated with loss of cone function in color vision deficiency, it is not known what happens to cones expressing this mutant opsin. Here, we used high-resolution retinal imaging to exami

NinaB is essential for Drosophila vision but induces retinal degeneration in opsin-deficient photoreceptors.

In animals, visual pigments are essential for photoreceptor function and survival. These G-protein-coupled receptors consist of a protein moiety (opsin) and a covalently bound 11-cis-retinylidene chromophore. The chromophore is derived from dietary carotenoids by oxidative cleavage and trans-to-cis isomerization of double bonds. In vertebrates, the necessary chemical transformations are catalyzed by two distinct but structurally related enzymes, the carotenoid oxygenase beta-carotenoid-15,15'-monooxygenase and the retinoid isomerase RPE65 (retinal pigment epithelium protein of 65 kDa). Opsin photoisomerases in the chick retina and pineal gland: characterization, localization, and circadian regulation.

PURPOSE: The chick retina and pineal gland exhibit circadian oscillations in biochemical and physiological processes in vivo and in vitro, which entrain to 24-hour light-dark cycles. However, the phototransduction mechanisms responsible for entrainment are largely unknown. The present study characterizes two candidate opsinlike genes that may play a role in entrainment of the clocks in these tissues. METHODS: Bioinformatics, cladistic techniques, and in situ hybridization and Northern blot analyses were conducted to characterize, localize, and determine the circadian expression of the candida

Opsin synthesis in the C57BL/6-mivit/mivit mouse model of retinal degeneration.

The capacity of photoreceptor cells to synthesize opsin was evaluated in a newly-described mouse model of retinal degeneration, the C57BL/6-mivit/mivit. The mivit/mivit mouse loses photoreceptor cells at a rate of about one row per month beginning at 8 weeks, ROS are severely disrupted at 4 months, RPE is unevenly pigmented. Retinas of affected and control mice ages 4, 6, 8, 12, 16, 20, 24, 28, 32 and 52 weeks were incubated for 2 hours in medium containing [3H] leucine. Homogenates of retina samples were subjected to SDS-PAGE using disc gels. The gels were sliced and counted by scintillation.

The regulation of skin pigmentation in response to environmental light by pineal Type II opsins and skin melanophore melatonin receptors.

Coupling skin colour with the light/dark cycle helps regulate body temperature in ectotherms. In X. laevis, nocturnal release of melatonin from the pineal complex induces pigment aggregation and skin lightening. This nocturnal blanching is initiated by a sensor (type II opsin) that triggers melatonin release when light intensity falls below a minimum threshold, and an effector (melatonin receptor) in the skin which induces pigment aggregation. The sensor/s and effector/s belong to two families of G-protein coupled receptors that originated from a common ancestor, but diverged with subsequent e
