Arthropod-borne viruses (arboviruses) are an increasingly significant threat to public health in tropical regions. In this study, we investigated the seroprevalence of various arboviruses in two species of sloth (Choloepus hoffmanni and Bradypus variegatus) in rural and peri-urban areas of Western Panama province. Between 2013 and 2018, blood samples from 60 sloths were tested for neutralizing antibodies against ten arboviruses. Significant variation in seroprevalence of different arboviruses was observed: 6.7% of sloths were seropositive for Madariaga virus, 6.7% for Venezuelan equine encepha

Epizootic hemorrhagic disease virus (EHDV), like other orbiviruses, infects and replicates in mammalian and insect vector cells. Within its ruminant hosts EHDV, like bluetongue virus (BTV), it has mainly been associated with infection of endothelial cells of capillaries as well as leukocyte subsets. Furthermore, EHDV infects and replicates within its biological vector, Culicoides biting midges and Culicoides-derived cells. A wide range of common laboratory cell lines such as BHK, BSR, and Vero cells are susceptible to infection with certain EHDV strains. Cell culture supernatants of infected c

The presence of antibodies to some alphaviruses, flaviviruses, bunyaviruses and orbiviruses was studied in the sera from humans and domestic animals of biotopes "C" and "T" in the south-east of Romania. Specific antibodies were found in humans to 3 alphaviruses (eastern equine encephalitis, western equine encephalitis and Sindbis), in a very low proportion, and to 3 flaviviruses (tickborne encephalitis, West Nile and Ntaya), at a much higher percentage. Very low percentages of antibodies to Bunyavirus and Kemerovo virus were also detected. The proportion of antibodies to flaviruses (tick-borne

Attempts to isolate viruses from 178,181 unengorged female mosquitoes collected from different ecologic areas of Sri Lanka yielded 31 isolates: 17 of Japanese encephalitis (JE) virus, nine of Getah virus, three of a Batai-related bunyavirus, and two of Arkonam virus. Culex tritaeniorhynchus and Mansonia uniformis mosquitoes were found to carry JE virus in a dry zone nonepidemic area, and Cx. pseudovishnui was found to carry it in a wet zone nonepidemic area. Japanese encephalitis virus was isolated from Cx. tritaeniorhynchus, Cx. gelidus, Cx. fuscocephala, and Cx. whitmorei during a human epid

The African horse sickness virus (AHSV) belongs to the Genus Orbivirus, family Sedoreoviridae, and nine serotypes of the virus have been described to date. The AHSV genome is composed of ten linear segments of double-stranded (ds) RNA, numbered in decreasing size order (Seg-1 to Seg-10). Genome segment 2 (Seg-2) encodes outer-capsid protein VP2, the most variable AHSV protein and the primary target for neutralizing antibodies. Consequently, Seg-2 determines the identity of the virus serotype. An African horse sickness (AHS) outbreak in an AHS-free status country requires identifying the seroty

Bluetongue virus (BTV) is a vector-transmitted orbivirus that infects ruminant livestock and wildlife, causing substantial economic losses owing to its impact on animal health and trade. Despite extensive study, the mechanisms underlying infection and immune dynamics remain unclear. Here, we develop and calibrate a mechanistic within-host model of BTV infection in experimentally infected sheep to quantify the contributions of innate and adaptive immune responses to viral control. The model integrates longitudinal data on viral RNA dynamics reverse transcriptase quantitative polymerase chain re

This review provides an overview of current and potential new diagnostic techniques against bluetongue virus (BTV), an Orbivirus transmitted by arthropods that affects ruminants. Bluetongue is a disease currently notifiable to the World Organization for Animal Health (OIE), causing great economic losses due to decreased trade associated with bluetongue outbreaks and high mortality and morbidity. BTV cross-reacts with many antigenically related viruses including viruses that causes African Horse sickness and epizootic haemorrhagic disease of deer. Therefore, reliable diagnostic approaches to de

African horse sickness virus (AHSV) is an orbivirus, a member of the Reoviridae family. Nine different serotypes have been described so far. AHSV is vectored by Culicoides spp. to equids, causing high mortality, particularly in horses, with considerable economic impacts. For development of a safe attenuated vaccine, we previously established an efficient reverse genetics (RG) system to generate Entry Competent Replication-Abortive (ECRA) virus strains, for all nine serotypes and demonstrated the vaccine potential of these strains in type I interferon receptor (IFNAR)-knockout mice. Here, we ev

Since the 1980s, a comprehensive field and laboratory investigation has been conducted throughout China, and a total of 29 virus species belonging to 7 families and 13 genera were identified through virological, morphological, and immunological methods, as well as whole-genome sequencing and molecular genetic analyses. Most of the virus isolates belong to 9 genera in the families Flaviviridae, Bunyaviridae, Togaviridae, and Reoviridae. Among them, 4 genera (Orthobunyavirus, Bunyavirus, Phlebovirus, and Nairovirus) belong to the family Bunyaviridae and 3 genera (Seadonavirus, Orbivirus, and Cyp

African horse sickness is a devastating viral disease of equids. It is transmitted by biting midges of the genus Culicoides with mortalities reaching over 90% in naïve horses. It is endemic to sub-Saharan Africa and is seasonally endemic in many parts of southern Africa. However, outbreaks in Europe and Asia have occurred that caused significant economic issues. There are attenuated vaccines available for control of the virus but concerns regarding the safety and efficacy means that alternatives are sought. One promising alternative is the use of virus-like particles in vaccine preparations, w

Brucellosis is an important bacterial disease of livestock and the most common zoonotic disease. The current vaccines are effective but unsafe, as they result in animal abortions and are pathogenic to humans. Virus-like particles are being investigated as molecular scaffolds for foreign antigen presentation to the immune system. Here, we sought to develop a new-generation vaccine by presenting selected Brucella melitensis T cell epitopes on the surface of Orbivirus core-like particles (CLPs) and transiently expressing these chimeric particles in Nicotiana benthamiana plants. We successfully de

African horse sickness (AHS) is a debilitating and often fatal viral disease affecting horses in much of Africa, caused by the dsRNA orbivirus African horse sickness virus (AHSV). Vaccination remains the single most effective weapon in combatting AHS, as there is no treatment for the disease apart from good animal husbandry. However, the only commercially available vaccine is a live-attenuated version of the virus (LAV). The threat of outbreaks of the disease outside its endemic region and the fact that the LAV is not licensed for use elsewhere in the world, have spurred attempts to develop an

This study reports the detection and genetic characterization of Tribeč virus (TRBV), a tick-borne orbivirus of zoonotic relevance. The virus was identified in Ixodes ricinus ticks collected from a goat using a newly developed RT-PCR assay. Goats grazing in the study area showed seroconversion, with low titre neutralization antibody response. The new isolate, designated 16.C/2016/Dubrava/SVK, was successfully cultured in Vero E6 cells and detected by immunofluorescence. Replication efficiency varied across rodent, human, and bovine cell lines. Whole-genome sequencing revealed that most genome

African horse sickness virus (AHSV) is an Orbivirus of the family Reoviridae that causes severe pathology in equids. Previous work in our laboratory showed the presence of AHSV-specific CD8(+) T cells in mice immunized with recombinant Modified Vaccinia Ankara (rMVA) expressing VP2 and NS1 proteins. In the present work, we selected potential CD8 T cell epitopes (MHC-class I binding peptides) for the 129 mouse strain from the VP2 and NS1 proteins of AHSV-4, using a combination of four epitope prediction algorithms (SYFPEITHI, BYMAS, NetMHC I and NetMHCpan). Orbiviruses are members of the Reoviridae family and include bluetongue virus (BTV) and epizootic hemorrhagic disease virus (EHDV). These viruses are the cause of significant regional disease outbreaks among livestock and wildlife in the United States, some of which have been characterized by significant morbidity and mortality. Competent vectors are clearly present in most regions of the globe; therefore, all segments of production livestock are at risk for serious disease outbreaks. Animals with subclinical infections also serve as reservoirs of infection and often result in significant trad

While vector-borne diseases are known to be particularly influenced by environmental factors, the impact of land-cover change on vector-borne wildlife disease patterns is poorly understood, largely due to the paucity of data on disease occurrence at extensive spatial and temporal scales. Widespread and rapid anthropogenic land-cover change, especially urbanization, has transformed the US landscape during the last century. Epizootic hemorrhagic disease virus and blue tongue virus, vectored by Culicoides biting midges, are two RNA viruses in the Orbivirus genus that cause severe hemorrhagic dise

The importance of tick-borne encephalitis (TBE) and Lyme borreliosis-the main European tick-borne diseases-is steadily growing. This fact is due to many different factors including climate changes. However, across Europe many other human pathogenic viruses, bacteria, and parasites are found in ticks. This article gives an overview of tick-borne human pathogenic viruses present in Europe as well as those considered nonpathogenic. It also deals with the various species of vector-competent ticks. Among these ticks, viruses from 6 families encompassing 8 genera and 35 species are found, whereby the

Oncolytic viruses (OVs) are promising cancer therapies whose efficacy is restricted by tumor heterogeneity and monotherapy limitations. We investigated combining the oncolytic orbivirus Epizootic Hemorrhagic Disease Virus -Tel Aviv University (EHDV-TAU) with the microtubule-targeting agent (MTA) 2-methoxyestradiol (2ME2) for the oncolysis of bladder cancer (BC) cells (T24 or SW1710) or melanoma cells (SKMEL3). Sub-lethal 2ME2 concentrations perturbed the cell cycle and enhanced multiple parameters of EHDV-TAU oncolysis of T24 and SKMEL3 cells (semi-permissive to EHDV-TAU), minimally affected t

Mosquito-borne viruses (MBVs) are a major global health threat, causing significant morbidity and mortality. MBVs belong to several distinct viral families, each with unique characteristics. The primary families include Flaviviridae (e.g., Dengue, Zika, West Nile, Yellow Fever, Japanese Encephalitis), transmitted predominantly by Aedes and Culex mosquitoes; Togaviridae, which consists of the genus Alphavirus (e.g., Chikungunya, Eastern and Western Equine Encephalitis viruses), also transmitted by Aedes and Culex; Bunyaviridae (recently reorganized), containing viruses like Rift Valley Fever an
