Passive immunity is critical for protection of neonatal piglets against porcine epidemic diarrhea virus (PEDV). Here, we investigated the immunogenicity of an orf virus (ORFV) vector expressing the full-length spike (S) protein of PEDV (ORFV-PEDV-S) in pregnant gilts and its ability to confer passive immunity and protection in piglets. Three doses of ORFV-PEDV-S were given to two groups of PEDV-negative pregnant gilts, with the last dose being administered two weeks prior to farrowing. One of the two groups immunized with the ORFV-PEDV-S recombinant virus was also exposed to live PEDV orally o

Orf is a highly contagious viral disease affecting goats and sheep. It is caused by Orf virus (ORFV) and has caused severe economic losses to the global goat industry, including in China. In this study, an indirect ELISA method for recombinant proteins based on truncated dominant antigenic epitopes of B2L and F1L genes of ORFV was established. A series of conditions and its performance were comprehensively evaluated. The optimized ELISA reaction conditions were: the optimal coating amount of antigen was 0.25 μg/mL, 5% skim milk powder was closed for 1 h, the optimal dilution of serum was 1:200

The emerging worldwide distributed porcine circovirus type 3 (PCV3) infection poses a serious threat to swine herds. An important means of preventing and controlling PCV3 infection is the development of the vaccine, while, the inability to cultivate in vitro has become the biggest obstacle. Orf virus (ORFV), the prototypic member of the Parapoxviridae, has been proven to be a novel valid vaccine vector for preparing various candidate vaccines. Here, recombinant ORFV expressing capsid protein (Cap) of PCV3 was obtained and proved its favorable immunogenicity inducing antibody against Cap in BAL

BACKGROUND: The success of new sequencing technologies and informatic methods for identifying genes has made establishing gene product function a critical rate limiting step in progressing the molecular sciences. We present a method to functionally mine genomes for useful activities in vivo, using an unusual property of a member of the poxvirus family to demonstrate this screening approach.
RESULTS: The genome of Parapoxvirus ovis (Orf virus) was sequenced, annotated, and then used to PCR-amplify its open-reading-frames. Employing a cloning-independent protocol, a viral expression-library was

Orf virus (ORFV), the prototype species of the Parapoxvirus genus within the Poxviridae family (subfamily Chordopoxvirinae), is a global pathogen infecting sheep, goats, and other ruminants, with zoonotic potential for humans. In this study, an outbreak of ORFV infection occurred in a sheep flock in Changling County, Jilin province, China, causing papules, pustules, and crusting lesions on the lips and eyelids. Typical parapoxvirus particles were observed using electron microscopy, and a wild ORFV strain was isolated, characterized, and designated as ORFV-CL24. To clarify the epidemiological a

BACKGROUND: Inactivated Orf virus (iORFV) has been used as a preventative as well as a therapeutic immunomodulator in veterinary medicine in different species. iORFV elicits strong effects on cytokine secretion in mice and human immune cells leading to an auto-regulated loop of initial up-regulation of inflammatory and Th1-related cytokines followed by Th2-related cytokines that attenuate immunopathology. The therapeutic potential of iORFV has been recognized in several models for difficult-to-treat disease areas such as chronic viral diseases, liver fibrosis or various forms of cancer.

Orf is a contagious zoonotic disease caused by Orf virus (ORFV), posing a threat to both animal and human health. The ORFV113 gene, located in the terminal variable region of the ORFV genome, has been demonstrated as a significant virulence gene, but its function remains largely unknown. In the study, we first amplified the truncated version of the ORFV113 gene (ORFV113t) by removing its transmembrane domain at the 5' end. We then constructed the pET-32a-ORFV113t recombinant plasmid and expressed the truncated ORFV113 recombinant protein in Escherichia coli (E.coli). The purified ORFV113t fusi

Contagious ecthyma is a skin disease, caused by Orf virus, creating great economic threats to livestock farming worldwide. Zoonotic potential of this disease has gained recent attention owing to the re-emergence of disease in several parts of the world. Increased public health concern emphasizes the need for a predictive understanding of the geographic distributional potential of Orf virus. Here, we mapped the current distribution using occurrence records, and estimated the ecological niche in both geographical and environmental spaces. Twenty modeling experiments, resulting from two- and thre

Orf virus, a member of the Parapoxvirus genus within the Poxvirus family, is the causative agent of Contagious Ecthyma, a zoonotic infection primarily affecting goats and sheep. The immune response against Orf virus is short-lived, enabling the virus to repeatedly infect animals, regardless of their vaccination status. Several virulence factors, including the OH1 tyrosine phosphatase, are responsible for the modulation of the host immune response. Here, we report the direct interaction between the viral OH1 and the human transcription factor STAT1 identified as a physiological OH1 substrate. I

Merino sheep exhibit a high follicular density, enhancing wool yield but predisposing them to inherited cutaneous disorders, such as breech wrinkle, which is characterized by the appearance of multiple skin folds in the breech area. This study provides the first clinicopathological description of the so-called body wrinkle, a congenital skin condition in Merino lambs where the folds affect the whole-body skin. Four affected lambs displayed generalized alopecia with excessive skin folds. Hematological analysis showed neutrophilia, lymphopenia, and eosinopenia. Histological examination revealed

Recombinant Orf virus (rORFV) vectors have proven safe and immunogenic in human clinical trials but the cellular correlates of antibody induction by this parapoxviral platform remain ill-defined. To characterize antigen-specific B cell responses at the cellular level, we performed adoptive transfer experiments with monoclonal B cells specific for the glycoproteins of either vesicular stomatitis virus or lymphocytic choriomeningitis virus. Immunizations of mice with rORFV delivering these glycoproteins stimulated antigen-specific B cells to engage in germinal center (GC) reactions and to differ

Contagious ecthyma (Orf), an acute and highly contagious zoonosis, is prevalent worldwide. Orf is caused by Orf virus (ORFV), which mainly infects sheep/goats and humans. Therefore, effective and safe vaccination strategies for Orf prevention are needed. Although immunization with single-type Orf vaccines has been tested, heterologous prime-boost strategies still need to be studied. In the present study, ORFV B2L and F1L were selected as immunogens, based on which DNA, subunit and adenovirus vaccine candidates were generated. Of note, heterologous immunization strategies using DNA prime-protei

This study was performed to differentiate vaccine-type strains from wild-type strains and determine the genotype of varicella-zoster virus (VZV) in 51 Korean children. A sequencing analysis of ORF 62 identified two cases of herpes zoster caused by the vaccine-type virus, without a previous history of varicella, 22 months and 5 months after VZV vaccination. The wild-type strain was identified in the remaining children. A genotype analysis of ORF 22 amino acids revealed genotype J in all children except one. Genotype E was identified in an infant with varicella imported from Egypt.

We describe a burn patient who developed skin lesions on her skin-graft harvest and skin-graft recipient (burn) sites. Orf virus infection was confirmed by a combination of diagnostic assays, including molecular tests, immunohistochemical analysis, pathologic analysis, and electron microscopy. DNA sequence analysis grouped this orf virus isolate among isolates from India. Although no definitive source of infection was determined from this case, this is the first reported case of orf virus infection in a skin graft harvest. Skin graft recipients with exposures to animals may be at risk for this

Orf virus (ORFV), a member of the genus Parapoxvirus within the family Poxviridae, causes contagious ecthyma in sheep and goats and is zoonotic. In this study, a scab sample collected from an affected goat in Binzhou, Shandong Province, China, was subjected to virus isolation and whole-genome characterization. ORFV was isolated in HeLa cells, and its identity was confirmed by F1L-targeted PCR with Sanger sequencing and indirect immunofluorescence assays using anti-F1L and anti-B2L polyclonal antibodies. Reference-independent de novo assembly of Illumina reads generated a 133,972-bp near-comple

Orf, resulting from infection with the Orf virus (ORFV), is a zoonotic disease that threatens the health of both animals and people. Reliable serological detection methods are critical for managing and preventing Orf outbreaks. The ORFV114 protein, highly conserved across ORFV strains, serves as a promising antigen for diagnostic applications. In this study, we developed a novel indirect ELISA for ORFV serodiagnosis using a recombinant ORFV114 protein. The antigen was produced in Escherichia coli via the pET-32a-ORFV114 plasmid, then purified and refolded for use in the assay. Receiver operati
