Thyroid hormone (triiodothyronine, T3) promotes neurodevelopment but under strict control because unconstrained exposure to T3 impairs brain and sensory functions. Thyroid hormone-inactivating type 3 deiodinase, encoded by Dio3, critically limits T3 signaling and controls diverse neural functions. Accordingly, understanding the cellular basis of T3 action requires identification of Dio3-expressing cell types but this is difficult because of low level, transient expression within the complexity of the nervous system. Here, we derived a knock-in Dio3Cre driver that sensitively labels Dio3-expres

Salt loading (SL) in rats for 7 days raises mean arterial pressure (MAP) and induces astrogliosis in the paraventricular nucleus of the hypothalamus (PVN). However, the exact role of the PVN in elevating MAP and sympathetic nerve activity (SNA), as well as its effects on astrocytes in forebrain osmosensitive areas, remain unclear. Male Holtzman and Wistar rats (300-320 g) received only 2% NaCl solution (SL) to drink for 7 days, while controls drank water (WT). Daily fluid and food intake, body weight, urinary volume, and MAP were recorded. On day 7, blood and cerebrospinal fluid (CSF) were col

Thirst is usually characterized as an unpleasant sensation provoking drinking of water. The purpose of the present review is to draw attention to the importance of thirst in overall regulation of body fluid homeostasis in health and pathology. Intensity of thirst is determined by signals generated in multiple groups of osmosensitive neurons engaged in dipsogenic and antidipsogenic activities, which are located in the brain cortex, the insula, the amygdala, the median preoptic area, the hypothalamic nuclei and the organum vasculosum laminae terminalis. Water ingestion is also influenced by sign

The circumventricular organs (CVOs) are small structures lining the cavities of brain ventricular system. They are associated with the semitransparent regions of the blood-brain barrier (BBB). Hence it is thought that CVOs mediate biochemical signaling and cell exchange between the brain and systemic blood. Their classification is still controversial and development not fully understood largely due to an absence of tissue-specific molecular markers. In a search for molecular determinants of CVOs we studied the green fluorescent protein (GFP) expression pattern in several zebrafish enhancer tra

The inflation of an intravascular balloon positioned at the superior vena cava and right atrial junction (SVC-RAJ) reduces sodium or water intake induced by various experimental procedures (e.g. sodium depletion; hypovolaemia). In the present study we investigated if the stretch induced by a balloon at this site inhibits a rapid onset salt appetite, and if this procedure modifies the pattern of immunohistochemical labelling for Fos protein (Fos-ir) in the brain. Male Sprague-Dawley rats with SVC-RAJ balloons received a combined treatment of furosemide (Furo; 10 mg (kg bw)(-1)) plus a low dose

Igf2 and H19 are reciprocally imprinted genes that are closely linked and coexpressed in tissues of mesodermal and endodermal origin. Here we report that coexpression of these genes is also found in specific fetal tissues of neuroectodermal origin, that is in the ventral midline region of both the hindbrain and spinal cord. For cells of neuroectodermal origin, complete absence of Igf2 and H19 transcription was previously described. Analysis of allele-specific expression of both Igf2 and H19 in the ventral midline region of the hindbrain shows that H19 is expressed monoallelically, with the pat

To study GnRH neuron projections to the systemic circulation during sexual maturation and in photoperiod-induced delayed puberty, we injected fluorogold (i.p.) into long- or short-day-reared male Djungarian hamsters. Five days after fluorogold treatment, at 15, 25, or 40 days of age, most GnRH neurons were found to access the systemic vasculature from terminal fields in the median eminence and organum vasculosum of the lamina terminalis. More than 80% of GnRH cell bodies in the medial preoptic area and diagonal band of Broca contained fluorogold, the locus for the majority of the GnRH neuron s

Central nervous system neurons which have access to the general circulation were identified by injecting the retrograde tracer Fluoro-Gold peripherally. Fluoro-Gold does not penetrate the blood-brain barrier but is taken up by nerve terminals which project to areas supplied by fenestrated capillaries or to the periphery. Fluoro-Gold-accumulating neurons were present in the following regions or cell groups of the central nervous system: diagonal band of Broca; medial preoptic area; organum vasculosum of the lamina terminalis; subfornical organ; anterior periventricular area; paraventricular nuc

Colchicine was injected into the region of the organum vasculosum lamina terminalis/anterior region of the third cerebral ventricle (OVLT/AV3V) to produce a temporary disruption of the nervous connections of this area and the rest of the anterior hypothalamus, but to maintain the vascular connections intact. Rats were kept in metabolism cages throughout the experiment and food and water intake plus urine and electrolyte excretion and body weight were measured each day. Food intake, body weight gain and urine and sodium excretion were reduced for several days after the injection of colchicine a

Chronic intermittent hypoxia (CIH) is a key feature of obstructive sleep apnea (OSA) and leads to physiological changes that can cause cardiovascular and neurological issues. This review explores the role of nitric oxide (NO) and inflammation in the development of CIH-induced health problems, specifically focusing on hypertension and cognitive dysfunction. We synthesize current evidence regarding how CIH modulates inflammatory processes and NO signaling in different brain regions, especially autonomic control centers crucial for cardiovascular regulation. We also discuss the activation of proi

Llamas are induced non-reflex ovulators, which ovulate in response to the hormonal stimulus of the male protein beta-nerve growth factor (β-NGF) that is present in the seminal plasma; this response is dependent on the preovulatory gonadotrophin-releasing hormone (GnRH) release from the hypothalamus. GnRH neurones are vital for reproduction, as these provide the input that controls the release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the pituitary gland. However, in spontaneous ovulators, the activity of GnRH cells is regulated by kisspeptin neurones that relay th

The brainstem plays an important role in controlling sodium and water homeostasis. It is a major regulatory site for autonomic and motor functions. Moreover, it integrates cerebrospinal fluid (CSF) signals with neuronal and hormonal signals. Evidence suggests that the CSF-contacting nucleus (CSF-CN) transmits and integrates CSF signals, but, the definitive role of CSF-CN in sodium homeostasis is poorly understood. In this study, we used c-Fos as a marker of neuronal activity and causing colocalization of Nax channel and 5-HT. This proved that CSF-CN played a role in sensing the increase of CSF

Circumventricular organs (CVOs) are specialized ventricular structures around the third and fourth ventricles of the brain. In humans, these structures are present during the fetal period and some become vestigial after birth. Some of these organs, such as the pineal gland (PG), subcommissural organ (SCO), and organum vasculosum of the lamina terminalis, might be the sites of origin of periventricular tumors, notably pineal parenchymal tumors, papillary tumor of the pineal region and chordoid glioma. In contrast to the situation in humans, CVOs are present in the adult rat and can be dissected

1. Accumulating evidence in both humans and animals indicates that acute increases in plasma osmolality elevate sympathetic nerve activity (SNA). In addition, plasma hyperosmolality (or hypernatraemia) can produce sustained increases in SNA and arterial blood pressure (ABP) through stimulation of forebrain osmoreceptors. 2. Although an abundance of information exists regarding the osmoregulatory circuits for thirst and secretion of antidiuretic hormone, much less is known about those pathways and synaptic mechanisms linking osmotic perturbations and SNA. To date, the available evidence suggest

Thirst motivates animals to seek fluid and drink it. It is regulated by the central nervous system and arises from neural and chemical signals from the periphery interacting in the brain to stimulate a drive to drink. Our research has focussed on the lamina terminalis and the manner in which osmotic and hormonal stimuli from the circulation are detected by neurons in this region and how that information is integrated with other neural signals to generate thirst. Our studies of osmoregulatory drinking in the sheep and rat have produced evidence that osmoreceptors for thirst exist in the dorsal

Oestrogen plays a key role in the regulation of the endocrine and behavioural events associated with the oestrous cycle. It is important, therefore, to know the location of neurones receptive to this steroid and to know whether their distribution varies with the oestrous cycle. We have undertaken experiments to identify the location of oestrogen receptors (ER) within the preoptico-hypothalamic continuum of ovariectomized ewes submitted to a variety of different hormone replacement regimes which mimic the different stages of the oestrous cycle. We used a monoclonal antibody to ER and detected r

The indirect peroxidase-antiperoxidase immunohistochemical technique was used to investigate the possible presence of vasoactive intestinal peptide (VIP) in the circumventricular organs of the rat. Considerable numbers of VIP-immunoreactive fibers were seen in the pineal gland. A moderate amount of VIP-immunoreactive fibers was present in the median eminence, the posterior lobe of the pituitary and the area postrema, but only few fibers were found in the organum vasculosum laminae terminalis. No immunoreactivity was observed in the subfornical organ or the subcommissural organ. The circumventr

The location of neurons that concentrate progestin in the brains of female cynomolgus monkeys was mapped by autoradiography using the specific synthetic progestin receptor ligand 3H-ORG 2058. Three females were ovariectomized and treated with estrogen (20 micrograms estradiol benzoate daily for 7 days), and one of them was also pretreated with progesterone. Each received an i.v. injection of 1 mCi 3H-ORG 2058 and was killed 1 hour later. Thaw-mount autoradiograms revealed intense accumulation of radioactivity in the nuclei of many neurons in the mediobasal hypothalamus, particularly in the ven

Atrial natriuretic factor (ANF) is widely distributed in the preoptic area and the hypothalamus, it is present there both in cell bodies and nerve terminals. Effect of experimental alterations in the salt and water balance was examined on preoptic-hypothalamic ANF levels measured in ten microdissected nuclei. Immunohistochemical analysis was also performed to confirm radioimmunological results. Following interventions were performed in adult male rats: adrenalectomy (5 days), daily 0.9% NaCl, aldosterone (5 micrograms/100 g) and dexamethasone (2 micrograms/ml drinking water) treatments in both

PURPOSE: The circumventricular organs (CVOs) occupy seven midline locations around the ventricles. They contain specialized ependymal cells called tanycytes and have an incomplete blood-brain barrier (BBB). We hypothesized that appearances of the lesser known CVOs on contrast-enhanced MRI might lead to confusion in image interpretation whereby they might be mistaken for pathology-related abnormal contrast enhancement. We therefore assessed the normal appearances and prevalence of contrast enhancement of the CVOs on routine clinical brain MRI and reviewed the functional anatomy of the CVOs.
