Bat Flies of the Family Streblidae (Diptera: Hippoboscoidea) Host Relatives of Medically and Agriculturally Important "Bat-Associated" Viruses.

Bat flies (Hippoboscoidea: Nycteribiidae and Streblidae) are obligate hematophagous ectoparasites of bats. We collected streblid bat flies from the New World (México) and the Old World (Uganda), and used metagenomics to identify their viruses. In México, we found méjal virus (Rhabdoviridae; Vesiculovirus), Amate virus (Reoviridae: Orbivirus), and two unclassified viruses of invertebrates. Méjal virus is related to emerging zoonotic encephalitis viruses and to the agriculturally important vesicular stomatitis viruses (VSV). Amate virus and its sister taxon from a bat are most closely related to

Clinical presentation of Oropouche virus infection: A systematic review and meta-analysis.

BACKGROUND: The recent surge in incidence and geographic spread of OROV infections poses an escalating threat to global public health. However, studies exploring the clinical signs of OROV infection remains exceedingly limited. METHODS: We searched for OROV studies published until June 17, 2024, in several electronic databases including MEDLINE, EMBASE, SCOPUS, and the Cochrane Library. RESULTS: In total, 15 studies involving 806 patients with OROV infection were eligible for inclusion. General symptoms with fever and headache were the most common. Gastrointestinal disturbances like nausea/v

Current View on Genetic Relationships within the Bunyamwera Serological Group.

The Bunyamwera serological group includes a number of geographically widespread viruses that are related but not identical and have serological cross-reactivity. As the first group members were obtained in the pre-sequencing era, their classifications (group attribution, species differentiation) were originally based on serological reactions. At the same time, the accuracy of the typing in each case depended on the variety of viruses that the researcher had as a comparison panel. With the advent of sequencing techniques, it has become customary to use identity thresholds (nucleotide or amino a

Collection and Processing of Adult Mosquitoes (Diptera: Culicidae) for Molecular Studies of Arbovirus Infection.

The processing of adult mosquitoes collected in the field for studies of natural infection with arboviruses of public health interest (genera: Flavivirus, Alphavirus, Orthobunyavirus) follows the assembly of homogenized mosquito pools and extraction of genetic material using commercial or manual techniques, and the detection of viral RNA by polymerase chain reaction (nested RT-PCR) [11]. This paper describes the procedures for capturing, taxonomic determination, obtaining genetic material, and molecular techniques for detecting alphaviruses.

Oropouche virus infection and pathogenesis are restricted by MAVS, IRF-3, IRF-7, and type I interferon signaling pathways in nonmyeloid cells.

UNLABELLED: Oropouche virus (OROV) is a member of the Orthobunyavirus genus in the Bunyaviridae family and a prominent cause of insect-transmitted viral disease in Central and South America. Despite its clinical relevance, little is known about OROV pathogenesis. To define the host defense pathways that control OROV infection and disease, we evaluated OROV pathogenesis and immune responses in primary cells and mice that were deficient in the RIG-I-like receptor signaling pathway (MDA5, RIG-I, or MAVS), downstream regulatory transcription factors (IRF-3 or IRF-7), beta interferon (IFN-β), or th

Rhabdomyolysis and Hepatitis Associated with Oropouche Fever: Brazil.

This study presents a rare case of rhabdomyolysis caused by Orthobunyavirus oropoucheense in a 21-year-old male patient in the Amazon region. The diagnosis was confirmed by the reverse transcription polymerase chain reaction test. Neutropenia, thrombocytopenia, and hepatitis were also identified by laboratory tests, demonstrating the clinical relevance as they are alterations with potential severity. The patient's good recovery is noteworthy, with only simple analgesia and intravenous hydration. The case highlights the need for better clarification regarding the pathophysiology of this distinc

Dicer-2 mutations in Aedes aegypti cells lead to a diminished antiviral function against Rift Valley fever virus and Bunyamwera virus infection.

Mosquitoes are known to transmit different arthropod-borne viruses belonging to various virus families. The exogenous small interfering RNA pathway plays an important role in the mosquito defence against such virus infections, with Dicer-2 (Dcr2) as one of the key proteins that initiates the cleavage of viral dsRNAs into 21 nt long virus-derived small interfering RNAs. Previous data identified the importance of various motifs in Dcr2 for its small interfering RNA (siRNA)-mediated antiviral activity. However, all these data focus on positive-strand RNA viruses, although negative-strand RNA viru

Redescription of immature stages of Culicoides paraensis (Goeldi) (Diptera: Ceratopogonidae), vector of the Oropouche virus.

Oropouche fever is an emerging zoonotic disease caused by Oropouche virus (OROV). It has two distinct transmission cycles, with the anthropophilic biting midge Culicoides paraensis (Goeldi) (Diptera: Ceratopogonidae) being the primary vector in the urban cycle. Species identification of Culicoides typically has been carried out on the basis of morphological characters, but molecular tools applied to taxonomy can provide rapid and efficient methods to the identification of vector species. The aim of this work was to obtain the first DNA barcode for C. paraensis collected in Argentina and redesc

First isolation of Bunyamwera virus (Bunyaviridae family) from horses with neurological disease and an abortion in Argentina.

Bunyamwera virus (BUNV) is the prototype virus for both the Orthobunyavirus genus and the Bunyaviridae family. Different strains of BUNV have been associated with clinical diseases in domestic animals, mainly ruminants. During 2013, in Argentina's Santa Fe Province, three new isolates of BUNV were recovered from the brain and spleen of two horses with encephalitis, and from the brain of an aborted equine fetus. This isolation of BUNV from domestic animals provided the first association of BUNV infection with disease of the central nervous system and abortion in equines in Argentina.

Powassan Virus Encephalitis in an Immunocompromised Patient: A Diagnostic Challenge With Case Report and Literature Review.

Powassan virus (POWV) is a tick-borne flavivirus transmitted to humans by Ixodes ticks. In this report, we describe an immunocompromised patient who presented with progressive neurologic symptoms and was diagnosed with POWV encephalitis. Additionally, the patient tested positive for Jamestown Canyon virus (JCV), a mosquito-borne orthobunyavirus, creating a diagnostic dilemma. This case underscores the importance of considering vector-borne illnesses in immunocompromised individuals who present with neurologic symptoms, particularly during peak arboviral transmission seasons in the United State

Diversity and Genetic Reassortment of Keystone Virus in Mosquito Populations in Florida.

Keystone orthobunyavirus (KEYV), a member of the genus Orthobunyavirus, was first isolated in 1964 from mosquitoes in Keystone, Florida. Although data on human infections are limited, the virus has been linked to a fever/rash syndrome and, possibly, encephalitis, with early studies suggesting that 20% of persons in the Tampa, Florida, region had antibodies to KEYV. To assess the distribution and diversity of KEYV in other regions of Florida, we collected > 6,000 mosquitoes from 43 sampling sites in St. Johns County between June 2019 and April 2020. Mosquitoes were separated into pools by speci

Development of a Diagnostic IgM Antibody Capture ELISA for Detection of Anti-Cache Valley Virus Human IgM.

Cache Valley virus (CVV), a mosquito-borne orthobunyavirus, causes epizootics in ruminants characterized by congenital malformations and fetal death in North America. Only seven human infections have been identified; limited information exists on its potential as a human teratogen. Diagnosis of CVV infections relies on the plaque reduction neutralization test (PRNT), which requires live virus, is time-consuming, and cannot differentiate between recent and past infections. To improve diagnostics for CVV, we developed an IgM antibody capture ELISA (MAC-ELISA) for detection of anti-CVV human IgM

Ecology and public health burden of Keystone virus in Florida.

Keystone virus (KEYV) is an under-studied orthobunyavirus that is transmitted via both horizontal and vertical cycles involving various mosquito species and vertebrate hosts. Historical evidence indicates that KEYV causes sub-clinical infections in humans, and some case studies draw links between this virus and encephalitis. Given KEYV's potential to cause human infections, it is plausible that it causes an under-appreciated proportion of both generic viral infections and unidentified viral encephalitis cases. This review summarizes current knowledge of KEYV and its disease dynamics in order t

Virome of Giant Panda-Infesting Ticks Reveals Novel Bunyaviruses and Other Viruses That Are Genetically Close to Those from Giant Pandas.

Tick infestations have been reported as one of the factors threatening the health of giant pandas, but studies of viral pathogens carried by ticks feeding on the blood of giant pandas are limited. To assess whether blood-sucking ticks of giant pandas can carry viral pathogens and if so, whether the viruses in ticks are associated with those previously detected in giant panda hosts, we determined the viromes of ticks detached from giant pandas in a field stocking area in Sichuan Province, southwest China. Using viral metagenomics we identified 32 viral species in ticks, half of which (including

Schmallenberg virus-two years of experiences.

In autumn 2011, a novel species of the genus Orthobunyavirus of the Simbu serogroup was discovered close to the German/Dutch border and named Schmallenberg virus (SBV). Since then, SBV has caused a large epidemic in European livestock. Like other viruses of the Simbu serogroup, SBV is transmitted by insect vectors. Adult ruminants may show a mild transient disease, while an infection during a critical period of pregnancy can lead to severe congenital malformation, premature birth or stillbirth. The current knowledge about the virus, its diagnosis, the spread of the epidemic, the impact and the

Characterization of five unclassified orthobunyaviruses (Bunyaviridae) from Africa and the Americas.

The Bunyaviridae family is made up of a diverse range of viruses, some of which cause disease and are a cause for concern in human and veterinary health. Here, we report the genomic and antigenic characterization of five previously uncharacterized bunyaviruses. Based on their ultrastructure, antigenic relationships and phylogenomic relationships, the five viruses are classified as members of the Orthobunyavirus genus. Three are viruses in the California encephalitis virus serogroup and are related to Trivittatus virus; the two others are most similar to the Mermet virus in the Simbu serogroup,

Epidemiological investigation of Shuni virus infections in hospitals in two provinces of South Africa (2019-2021) using molecular and serological surveillance.

Shuni Virus (SHUV) is a reemerging zoonotic orthobunyavirus in the Peribunyaviridae family associated with neurological infections and birth defects in humans and animals in Africa and has emerged in the Middle East in the past 10 years. Limited epidemiological data exist in humans, partially due to a lack of clinical awareness and availability of diagnostic assays to determine the incidence of clinical cases and seroprevalence in the population. The goal of this study was to establish serologic and molecular diagnostic assays for SHUV for hospital-based surveillance and to investigate the cli

Identification and Molecular Characterization of Shamonda Virus in an Aborted Goat Fetus in South Africa.

Viruses in the Orthobunyavirus genus, Peribunyaviridae family, are associated with encephalitis, birth defects and fatalities in animals, and some are zoonotic. Molecular diagnostic investigations of animals with neurological signs previously identified Shuni virus (SHUV) as the most significant orthobunyavirus in South Africa (SA). To determine if other orthobunyaviruses occur in SA, we screened clinical specimens from animals with neurological signs, abortions, and acute deaths from across SA in 2021 using a small (S) segment Simbu serogroup specific TaqMan real-time reverse transcription po

Emerging orthobunyaviruses associated with CNS disease.

The Orthobunyavirus genus comprises a wide range of arthropod-borne viruses which are prevalent worldwide and commonly associated with central nervous system (CNS) disease in humans and other vertebrates. Several orthobunyaviruses have recently emerged and increasingly more will likely do so in the future. Despite this large number, an overview of these viruses is currently lacking, making it challenging to determine importance from a One Health perspective. Causality is a key feature of determining importance, yet classical tools are unfit to evaluate the causality of orthobunyaviral CNS dise
