Calcium regulates the mode of exocytosis induced by hypotonic shock in isolated neuronal presynaptic endings.

A decrease in the osmolarity of incubation medium is accompanied by calcium influx in neuronal presynaptic endings. We studied the influence of Ca2+ on exocytosis induced by hypotonic shock using the hydrophilic fluorescent dye acridine orange and the hydrophobic fluorescent dye FM2-10. It was shown using acridine orange that lowering of osmolarity to 230 mOsm/l induces exocytosis both in calcium-containing and calcium-free medium. By contrast, we were able to demonstrate calcium-dependence of exocytosis using styryl dye FM2-10. Lowering of osmolarity leads to increase of [3H]D-aspartate and [

Mechanisms of Long-Term Nanoplastic Release in Soil and Their Interaction with Soil Colloids.

The quantification of nanoplastic retention in soils and release mechanisms mediated by soil colloids remains challenging. This study used palladium-labeled nanoplastics (Pd-NPs) in column experiments to investigate their retention and potential long-term release, focusing on interactions with soil colloids. Pd-NPs were primarily immobilized through low energy barriers related to charge heterogeneity or nanoscale roughness, but also influenced by blocking, ripening, charge reversal, and cation bridging. These interactions limited particle release under steady-state conditions or flow interrupt

Measurement of dissolved reactive phosphorus in water with polyquaternary ammonium salt as a binding agent in diffusive gradients in thin-films technique.

Diffusive gradients in thin-films (DGT) sampler with a polyquaternary ammonium salt (PQAS) aqueous solution as a binding phase and a dialysis membrane as a diffusive phase (PQAS DGT) was developed for the measurement of dissolved reactive phosphorus (DRP) in water. The performance of PQAS DGT was not dependent upon pH 3-10 and ionic strength from 1 × 10(-4) to 1 mol L(-1). The effective binding capacity of PQAS DGT containing 2.0 mL of 0.050 mol L(-1) PQAS solution was estimated as 9.9 μg cm(-2). The measurement of DRP in a synthetic solution by PQAS DGT over a 48 h deployment period demonstra

Preparation and Characterisation of a Cyclodextrin-Complexed Mānuka Honey Microemulsion for Eyelid Application.

Honey has been widely purported as a natural remedy due to its antimicrobial and anti-inflammatory effects. In recent years, several studies have suggested that the considerably high methylglyoxal (MGO) concentration in Mānuka honey (MH) makes it particularly effective to manage bacterial overload, such as that observed in blepharitis. However, the poor solubility, high viscosity, and osmolarity of aqueous honey solutions, especially at the high MGO concentrations studied in the literature, render the formulation of an acceptable dosage form for topical application to the eyelids challenging.

Anabolic steroid excess promotes hydroelectrolytic and autonomic imbalance in adult male rats: Is it enough to alter blood pressure?

AIM: The present study investigated the effects of anabolic steroid (AS) excess on blood pressure regulation. METHODS: Male Wistar rats were treated with nandrolone decanoate (AS) or vehicle (CTL) for 8 or 10 weeks. Saline (1.8%) and water intake were measured in metabolic cages. Urinary volume, osmolarity, Na+ and K+ concentrations, and plasma osmolarity were measured. The autonomic balance was estimated by heart rate variability at baseline or after icv injection of losartan. Cardiac function was assessed by echocardiography and ex vivo recordings. Myocardial collagen deposition was evaluat

Osmotic flow transients during acetylcholine stimulation in the perfused rat submandibular gland.

Osmotic stress was applied to the perfused rat submandibular gland during steady-state fluid secretion. Alterations of perfusate osmolarity, by addition or withdrawal of sucrose or NaCl, caused transient changes in secretory rate during continuous stimulation with 1 microM acetylcholine (ACh). Hyposmotic perfusates transiently increased, and hyperosmotic perfusates transiently reduced, the secretory rate. The transients were attributed to changes in osmotic flow resulting from changes in the instantaneous transepithelial osmotic gradient. The time course of the change in interstitial osmolarit

Inward flux of lactate⁻ through monocarboxylate transporters contributes to regulatory volume increase in mouse muscle fibres.

Mouse and rat skeletal muscles are capable of a regulatory volume increase (RVI) after they shrink (volume loss resultant from exposure to solutions of increased osmolarity) and that this RVI occurs mainly by a Na-K-Cl-Cotransporter (NKCC)-dependent mechanism. With high-intensity exercise, increased extracellular osmolarity is accompanied by large increases in extracellular [lactate⁻]. We hypothesized that large increases in [lactate⁻] and osmolarity augment the NKCC-dependent RVI response observed with a NaCl (or sucrose)-induced increase in osmolarity alone; a response that is dependent on l

[POSSIBILITY OF CORRECTION OF METABOLIC DISORDERS WITH REAMBERIN IN ACUTE PERIOD OF TRAUMATIC INJURY].

56 patients at the age of 18-60 years with severe trauma were examined. Influence of the polyelectrolytic (Reamberin)solution on an acid-base state, osmolarity and electrolytic composition of plasma in the acute posttraumatic period was evaluated. It was found that patients, who was treated by isotonic sodium chloride solution and Ringer's solution, had metabolic acidosis and hyperchloremia. In contrast, in the reamberin group 82% of patients had lower concentrations of chloride and had nothing acid-base disturbances on the second day after trauma. Reamberin didn't influence on plasma osmolari

Plastic movement in variably saturated porous media: The role of ionic strength and surface roughness.

We investigated colloid deposition mechanisms in saturated and unsaturated porous medias, focusing on roles of ionic strength (IS, 1-100 mM) and collector surface roughness. Through a customized column setup, experiments were conducted with smooth and rough (HF-etched) glass beads. Rough collectors featured fractal, multi-scale surface craters. Following colloid application, we evaluated released colloid via flushing with 0.4 mM NaHCO₃ solutions. Finally, column dissection was performed to quantify moderately and tightly attached colloids within the primary minimum. Increasing IS reduced mobil

Osmolality (mosmol/kg H2O) versus osmolarity (mosmol/L): applied physiology to improve patient safety.

Severe osmotic pressure imbalances may be associated with substantial morbidity and mortality. As a consequence, osmolality should be a routine parameter in clinical medicine, but actual clinical practice often looks different, and confusion or inaccurate interpretations are common. This paper therefore reviews the physiological background of osmolality to achieve a better understanding and to prevent complications in hospitalized patients. The use of osmolality (mosmol/kg H2O) instead of osmolarity (mosmol/L) is recommended for clinicians to avoid potentially misleading results when comparing

Combined Effects of Pressure and Ionic Strength on Protein-Protein Interactions: An Empirical Approach.

Proteins are exposed to hydrostatic pressure (HP) in a variety of ecosystems as well as in processing steps such as freeze-thaw, cell disruption, sterilization, and homogenization, yet pressure effects on protein-protein interactions (PPIs) remain underexplored. With the goal of contributing toward the expanded use of HP as a fundamental control parameter in protein research, processing, and engineering, small-angle X-ray scattering was used to examine the effects of HP and ionic strength on ovalbumin, a model protein. Based on an extensive data set, we develop an empirical method for scaling

Effect of ionic strength on the structural properties and emulsion characteristics of myofibrillar proteins from hairtail (Trichiurus haumela).

Myofibrillar proteins (MPs) are susceptibly affected by ionic strength. The effect of ionic strength on the structure and emulsifying properties of MPs from hairtail (Trichiurus haumela) is still unclear. Therefore, the effect of ionic strength on the structural properties and emulsification of myofibrillar proteins from hairtail was analyzed. The increase in ionic strength led to the increase in endogenous fluorescence intensity of MPs. The α-helix content in MPs first increased and then decreased from ionic strength of 0 to 1.0 mol/L and β-sheet content exhibited the oppositive trend, indica

Serum osmolality and hyperosmolar states.

Serum osmolality is the sum of the osmolalities of every single dissolved particle in the blood such as sodium and associated anions, potassium, glucose, and urea. Under normal conditions, serum sodium concentration is the major determinant of serum osmolality. Effective blood osmolality, so-called blood tonicity, is created by the endogenous (e.g., sodium and glucose) and exogenous (e.g., mannitol) solutes that are capable of creating an osmotic gradient across the membranes. In case of change in effective blood osmolality, water shifts from the compartment with low osmolality into the compar

Interpenetrating polymeric network (IPNs) in ophthalmic drug delivery: Breaking the barriers.

To maintain the therapeutic drug concentration for a prolonged period of time in aqueous and vitreous humor is primary challenge for ophthalmic drug delivery. Majority of the locally administered drug into the eye is lost as to natural reflexes like blinking and lacrimation resulting in the short span of drug residence. Consequently, less than 5% of the applied drug penetrate through the cornea and reaches the intraocular tissues. The major targets for optimal ophthalmic drug delivery are increasing drug residence time in cul-de-sac of the eye, prolonging intraocular exposure, modulating drug
