Enzymic acylation of mucus glycoprotein with palmitic acid in rat submandibular salivary gland.

The enzymic activity which catalyses transfer of palmitic acid from palmitoyl coenzyme A to mucus glycoprotein was found in Triton X-100 extracts of the microsomal fraction of rat submandibular and sublingual salivary glands. The acyltransferase activity of this fraction was 1.3-1.4 times greater in submandibular gland than in sublingual gland. Further subcellular fractionation of submandibular gland showed that the enzyme activity was associated with a Golgi-rich membrane fraction. Optimum enzyme activity for fatty acylation of mucus glycoprotein was at pH 7.4 using 0.5 per cent Triton X-100,

Syringic acid improves cognitive impairment in palmitic acid-induced obesity model: an in vitro and in vivo study.

OBJECTIVE: Obesity-mediated cognitive impairment is increasingly known as high levels of saturated fatty acid intake and accumulation in the body. Saturated fatty acids, mainly palmitic acid (PA) is recognized as the leading cause of oxidative stress, metabolic dysregulation, and neuronal degeneration in obesity. METHOD: The current study investigated the neuroprotective effects of syringic acid (SA) against PA-induced neuronal injury in N2a neuronal cells, as well as in a high-saturated-fat-induced obese mouse (DIO) model. RESULT: The study demonstrated that PA significantly reduced cell vi

Docosahexaenoic Acid Protects Schwann Cells Against Palmitic Acid-Induced Lipotoxicity by Modulating Autophagy, ER Stress, and Lipid Handling.

Elevated saturated fatty acids, such as palmitic acid (PA), induce lipotoxicity in peripheral nerve cells, a pathological feature of metabolic disorders such as type 2 diabetes and obesity that are frequently associated with neuropathic pain (NP). PA overload elicits a maladaptive stress response characterized by endoplasmic reticulum (ER) stress, disrupted intracellular calcium homeostasis, and impaired autophagic flux, ultimately promoting cell death. Although omega-3 polyunsaturated fatty acids such as docosahexaenoic acid (DHA) protect against PA-induced lipotoxicity (PA-LTx), the mechanis

The Different Mechanisms of Lipid Accumulation in Hepatocytes Induced by Oleic Acid/Palmitic Acid and High-Fat Diet.

Non-alcoholic fatty liver disease (NAFLD) is the primary chronic liver disease worldwide, mainly manifested by hepatic steatosis. Hepatic lipids may be derived from dietary intake, plasma free fatty acid (FFA) uptake, or hepatic de novo lipogenesis (DNL). Currently, cellular and animal models of hepatocellular steatosis are widely used to study the pathogenesis of NAFLD and to investigate therapeutic agents. However, whether there are differences between the in vivo and in vitro models of the mechanisms that cause lipid accumulation has not been reported. We used OA/PA-induced NCTC 1469 cells

Myo-inositol alters the effects of glucose, leptin and insulin on placental palmitic acid and oleic acid metabolism.

Well-regulated placental palmitic acid (PA) and oleic acid (OA) metabolism is vital for optimal placental function and fetal development, but dysregulation occurs with gestational diabetes (GDM). We hypothesized that such dysregulation might arise from increased maternofetal glucose, leptin or insulin concentrations present in GDM, and that dysregulated PA and OA lipid metabolism could be moderated by myo-inositol, a natural polyol and potential GDM intervention. Placental explants from 21 women were incubated with stable isotope-labelled 13 C-PA or 13 C-OA for 48 h. Explants were treated with

Myristic acid potentiates palmitic acid-induced lipotoxicity and steatohepatitis associated with lipodystrophy by sustaning de novo ceramide synthesis.

Palmitic acid (PA) induces hepatocyte apoptosis and fuels de novo ceramide synthesis in the endoplasmic reticulum (ER). Myristic acid (MA), a free fatty acid highly abundant in copra/palmist oils, is a predictor of nonalcoholic steatohepatitis (NASH) and stimulates ceramide synthesis. Here we investigated the synergism between MA and PA in ceramide synthesis, ER stress, lipotoxicity and NASH. Unlike PA, MA is not lipotoxic but potentiated PA-mediated lipoapoptosis, ER stress, caspase-3 activation and cytochrome c release in primary mouse hepatocytes (PMH). Moreover, MA kinetically sustained PA

Dietary palmitic acid to oleic acid ratio modulates energy metabolism and biological rhythms in young healthy Japanese males.

The present study investigated the potential role of the composition of dietary fatty acids in the regulation of biological rhythms, such as the sleep architecture, core body temperature and leukocyte clock gene expression, in subjects fed meals rich in palmitic acid (PA) or oleic acid (OA). Eleven males participated in two sessions of indirect calorimetry in a whole-room metabolic chamber. In each session, subjects consumed three meals rich in PA (44·3 % of total fat as PA and 42·3 % as OA) or OA (11·7 % of total fat as PA and 59·3 % as OA) in the metabolic chamber. The ratio of PA to OA in p

Levels of palmitic acid ester of hydroxystearic acid (PAHSA) are reduced in the breast milk of obese mothers.

To achieve optimal development of a newborn, breastfeeding is extensively recommended, but little is known about the role of non-nutritive bioactive milk components. We aimed to characterize the fatty acid esters of hydroxy fatty acids (FAHFAs), namely palmitic acid hydroxystearic acids (PAHSAs)-endogenous lipids with anti-inflammatory and anti-diabetic properties, in human breast milk. Breast milk samples from 30 lean (BMI=19-23) and 23 obese (BMI>30) women were collected 72h postpartum. Adipose tissue and milk samples were harvested from C57BL/6J mice. FAHFA lipid profiles were measured usin

Palmitic acid reprograms neutrophils to compromise vascular integrity and promote breast cancer lung metastasis.

Pre-metastatic niche formation in the lung creates a permissive microenvironment for breast cancer metastasis, characterized by metabolic reprogramming of resident cells, recruitment of suppressive neutrophils, and vascular remodeling. However, the role of lipids in regulating neutrophil-endothelial interactions, particularly in facilitating tumor cell extravasation, remains largely undefined. Here, we showed that triple-negative breast cancer established a palmitic acid-enriched lung microenvironment that drove tumor cell extravasation and colonization. Pulmonary endothelial cells were a majo

Antitumor efficacy and reduced systemic toxicity of palmitic acid-modified pirarubicin nanoparticles in mice.

PURPOSE: Pirarubicin (THP) is a potent anthracycline anticancer agent; however, its clinical use is limited by nonselective tissue distribution and systemic toxicity. This study aimed to evaluate whether palmitic acid-modified THP nanoparticles (Pal-THP nanoparticles) could improve the in vivo therapeutic profile of THP. METHODS: Pal-THP nanoparticles stabilized by albumin as a formulation stabilizer were prepared by antisolvent precipitation. Antitumor efficacy, biodistribution, and drug forms (intact Pal-THP and the free THP released from Pal-THP) were evaluated in colon 26 tumor-bearing mi

Zebrafish model of palmitic acid induced MAFLD recapitulates pathways conserved in mice and humans.

Metabolic Dysfunction-Associated Fatty Liver Disease (MAFLD) is a major global health issue, affecting millions, yet its underlying molecular mechanisms remain poorly understood. Here, we propose a novel diet-induced zebrafish model to investigate pathophysiology of MAFLD. To validate the model, we performed comprehensive histological analysis and molecular assessments, including RNA-sequencing, to characterize the disease progression. These approaches enabled us to examine the molecular alterations underlying MAFLD and identify key genes and pathways involved in its development. Our results d

Selection and Characterization of Palmitic Acid Responsive Patients with an OXPHOS Complex I Defect.

Mitochondrial disorders are genetically and clinically heterogeneous, mainly affecting high energy-demanding organs due to impaired oxidative phosphorylation (OXPHOS). Currently, effective treatments for OXPHOS defects, with complex I deficiency being the most prevalent, are not available. Yet, clinical practice has shown that some complex I deficient patients benefit from a high-fat or ketogenic diet, but it is unclear how these therapeutic diets influence mitochondrial function and more importantly, which complex I patients could benefit from such treatment. Dietary studies in a complex I de

Cytoprotective effects of melatonin on astroglial cells subjected to palmitic acid treatment in vitro.

Melatonin, an endogenously produced neurohormone secreted mainly by the pineal gland, has a variety of physiological functions and neuroprotective effects. Saturated fatty acids (SFAs) have been known to induce neurotoxicity and oxidative stress in central nervous system injuries and neurodegenerative pathologies. However, the effect of melatonin on SFAs-induced cytotoxicity in astroglial cells, if any, has remained to be explored. This study reports that in primary cultured astroglial cells, melatonin significantly attenuated palmitic acid (PA)-induced cytotoxicity in a concentration- and tim

Palmitic acid opens a novel cyclosporin A-insensitive pore in the inner mitochondrial membrane.

An assortment of agents can induce mitochondria to undergo a permeability transition, which results in the inner mitochondrial membrane becoming nonselectively permeable to small (<1500 Da) solutes. This mitochondrial permeability transition (MPT) is characterized by a strict dependence on matrix Ca2+ and sensitivity to cyclosporin A (CsA). However, it is becoming increasingly clear that other experimental conditions can elicit increases in mitochondrial permeability that are distinct from this classic MPT. For example, butylated hydroxytoluene (BHT; Sokolove, P. M., and Haley, L. M. (1996) J.

Targeting LDHA Palmitoylation Sensitizes GBM to Chemoradiotherapy via Suppressing Palmitic Acid Induced Glycolytic Reprogramming.

Glioblastoma (GBM) is a lethal malignancy characterized by therapeutic resistance and recurrence, which are driven by dynamic cellular metabolic adaptation within the tumor microenvironment. However, the precise molecular metabolic mechanisms remain poorly understood. Here, we found that palmitic acid (PA) accumulates in hypoxic pseudopalisading regions of GBM and drives the formation of mesenchymal (MES) GBM and the hypoxia myeloid-derived macrophages (MDM) subpopulation, establishing an interactive tumor-promoting niche that fuels GBM recurrence. Maechanistically, PA induces ZDHHC12-mediated

Palmitic acid activates c-Myc via dual palmitoylation-dependent pathways to promote colon cancer.

c-Myc is broadly hyperactivated in colon cancer, yet the mechanisms sustaining its transcriptional activation remain elusive. Here we identify palmitic acid (PA) as a metabolite cue that activates c-Myc via dual palmitoylation-dependent pathways operating across tumor initiation and progression. In colitis models, PA-rich diets exacerbate inflammation and enrich MYC target programs without increasing Myc mRNA. Mechanistically, the palmitoyltransferase ZDHHC9, upregulated by IL-1β, directly palmitoylates c-Myc at C171, enhancing c-Myc/MAX dimerization and transcriptional activity; genetic or ph

Epithelial-Mesenchymal Transition Shapes the Lipotoxic Response of Colon Cancer Cells to Palmitic Acid.

Saturated fatty acids such as palmitic acid (PA) can induce lipotoxic stress, whereas monounsaturated fatty acids like oleic acid (OA) often promote adaptive responses through lipid droplets (LDs) formation. Here, we reveal that epithelial-mesenchymal transition (EMT) profoundly influences the lipotoxic response of colorectal cancer cells. Using the epithelial-like HCT15 and mesenchymal-like HCT116 cell lines, we combined proteomic, metabolic, and imaging analyses to elucidate how EMT status determines lipid storage capacity and resistance to PA-induced toxicity. A basal proteomic profiling hi

Palmitic acid in cancer research: mechanistic insights from in vitro and in vivo studies.

Palmitic acid (PA), a long-chain saturated fatty acid abundantly present in dietary lipids and endogenous metabolic pathways, has traditionally been associated with metabolic dysregulation. However, emerging experimental evidence has revealed a contrasting role for PA in cancer biology. This review critically summarizes recent in vitro and in vivo studies investigating the anticancer potential of PA across multiple malignancies, including gastric, breast, colorectal, prostate, and endometrial cancers. Accumulating data demonstrate that PA suppresses tumor cell proliferation, induces apoptosis

Bilirubin Ameliorates Oleic and Palmitic Acid Accumulation in an In Vitro Model of MASLD.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a chronic progressive disorder characterized by an excess accumulation of lipids in the liver. The aim of this study was to examine the role of bilirubin (BR), the catabolic heme product and a putative peroxisome proliferator-activated receptor alpha (PPARalpha) agonist, in an in vitro model of MASLD. In our study, we used human hepatoblastoma HepG2 cells exposed to oleic (OA)/palmitic acid (PA) (2:1 ratio, 24 h) with subsequent treatment with BR or fenofibrate (FF, a clinically used PPARalpha agonist) at clinically relevant c

Salidroside alleviates palmitic acid-induced hepatocyte injury by regulating the LILRB2-mediated autophagy pathway.

Non-alcoholic steatohepatitis (NASH) is a progressive form of non-alcoholic fatty liver disease (NAFLD), characterized by hepatic steatosis, inflammation, and fibrosis, which may advance to cirrhosis and hepatocellular carcinoma. Treatment options remain limited, underscoring the need to elucidate its mechanisms and develop effective therapeutics. Salidroside (Sal), a primary active compound of Rhodiola rosea, has shown potential in alleviating NASH, yet its underlying mechanisms are not fully understood. This study investigates whether Sal mitigates palmitic acid (PA)-induced hepatocyte injur
