Neonatal methyl parathion exposure affects the growth and functions of the male reproductive system in the adult rat.

Methyl parathion (MP) is a well-known organophosphorus pesticide, to which humans are exposed in fruit and vegetables as residues of 0-2 mg/kg, children being at higher risk of exposure. The present study was planned to investigate the effects on the adult male reproductive functions of MP following neonatal exposure. New born male Wistar rat pups were treated orally with either 0 or 0.5 mg/kg MP from postnatal day (PND) 3 to PND 28 and sacrificed on PND 98 for the purpose of examination of the reproductive system. Methyl parathion lowered the body weights from days 10 to 24 (p < 0.01), the we

Down-regulation of muscarinic receptors and the m3 subtype in white-footed mice by dietary exposure to parathion.

The effect of ad libitum dietary exposure (as occurs in the field) to parathion for 14 d was investigated on the muscarinic acetylcholine receptor (mAChR) in brains and submaxillary glands of adults of a field species, the white-footed mouse Peromyscus leucopus. Immunoprecipitation using subtype selective antibodies revealed that the relative ratios of the m1-m5 mAChR subtypes in Peromyscus brain were similar to those in rat brain. There was little variability in acetylcholinesterase (AChE) activity in control mice brains but large variability in 39 exposed mice, resulting from differences in

Altered capillary filtration coefficient in parathion- and paraoxon-induced edema in isolated and perfused rabbit lungs.

Changes in pulmonary endothelium permeability and in microvascular hemodynamics induced by parathion (Pth) and paraoxon (Pox), its active metabolite, were investigated in isolated, perfused rabbit lungs. Blood-free perfusate was recirculated through isolated and ventilated lungs in an isogravimetric state and in zone III conditions. The arterial/venous/double occlusion technique was used to divide the total vascular resistance (Rt) into four components: arterial, precapillary, postcapillary, and venous. The capillary filtration coefficient (Kfc) was evaluated by measuring the amount of fluid f

Quantitative analysis of methyl parathion pesticides in a polydimethylsiloxane microfluidic channel using confocal surface-enhanced Raman spectroscopy.

A fast and ultra-sensitive trace analysis of methyl parathion pesticides in a polydimethylsiloxane (PDMS) microfluidic channel was investigated using confocal surface-enhanced Raman spectroscopy (SERS). A three-dimensional PDMS-based passive micromixer was fabricated for this purpose. This PDMS micromixer showed a high mixing efficiency because a strong chaotic advection was developed by the simultaneous vertical and transverse dispersion of the confluent streams. The confocal SERS signal was measured after methyl parathion pesticides were effectively adsorbed onto silver nanoparticles while f

Toward a better understanding of pesticide dermal absorption: diffusion model analysis of parathion absorption in vitro and in vivo.

Human skin absorption of radiolabeled parathion was studied in vitro at specific doses (mass loadings) of 0.4, 4.0, 41, or 117 microg/cm(2), with and without occlusion. The compound was applied in small volumes of acetone solution to split-thickness skin. Permeation of radiolabel into the receptor solutions was monitored for 76 h, after which the tissue was dissected and analyzed for residual radioactivity. For the 3 lower doses, cumulative permeation after 76 h was approximately dose-proportional, ranging from 28.5-30.5% of applied dose (unoccluded) to 45.5-55.7% (occluded). Total absorption,

Evaluation of residual levels of benomyl, methyl parathion, diuron, and vamidothion in pineapple pulp and bagasse (Smooth cayenne).

The objective of this research was to study the residual levels of benomyl, methyl parathion, diuron, and vamidothion in pineapple bagasse and pulp. Benomyl (benlate), methyl parathion (Folidol 600), diuron (Krovar), and Vamidothion (Kilval 300) were applied pre-harvest to pineapples (smooth cayenne). After harvesting, the fruits were washed (100 ppm sodium hypochlorite) and the pulp was separated from the sub-products (peel, core, tops, and tails). The pulp was not submitted to any heat treatment. The sub-products and the juice expressed from them, were submitted to a blanching process (95 de

Chronic toxicity of ethyl parathion and isobutoxyethanol ester of 2,4-dichlorophenoxyacetic acid to estuarine juvenile and adult crabs.

The chronic toxicity of ethyl parathion and isobutoxyethanol ester of 2,4-dichlorophenoxyacetic acid was determined for the crabs Uca uruguayensis and Chasmagnathus granulata collected in Argentina in 1989. Parathion toxicity was more time-dependent than 2,4-D. Four-week LC50 values for parathion were size and species independent, as expected under chronic exposure conditions. As shown in previous studies, the opposite was noted when crabs were exposed to acute exposure. Parathion was much more toxic than 2,4-D, in all cases. Also, parathion exerted a very much lower chronic lethal effect than

Superoxide dismutase, catalase and acetylcholinesterase: biomarkers for the joint effects of cadmium, zinc and methyl parathion contamination in water.

Heavy metals are known to reduce the activities of antioxidant enzymes (e.g. superoxide dismutase, catalase), while organophosphorous insecticides are known to inhibit the activity of the enzyme acetylcholinesterase. In this study, the activities of these three enzymes in zebrafish (Danio rerio) tissues were assessed to evaluate the consequences heavy metal and organophosphate contamination in aquatic systems. When the fish were contacted with water containing a single pollutant, superoxide dismutase activity was affected by the presence of Cd but not by methyl parathion or Zn. However, catala

Changes in nocturnal pineal indoleamine metabolism in rats treated with parathion are prevented by beta-adrenergic antagonist administration.

Parathion, an organophosphorous insecticide, was previously shown to enhance the nighttime rise in pineal N-acetyltransferase (NAT) activity and serum melatonin levels. The purpose of the present study was to test whether parathion acts on the pineal gland by means of a beta-adrenergic receptor mechanism. Whereas parathion (total dose 6.5 mg/kg body wt over 6 days) by itself significantly augmented nocturnal pineal NAT activity and serum melatonin levels in otherwise untreated rats, the insecticide was ineffective in reference to this enzyme when it was given in conjunction with the beta-adren

Interaction of parathion and malathion with hepatic cytochrome P-450 from rats treated with phenobarbital and carbon disulfide.

Alterations in the activity of drug-metabolizing enzymes and the concentration of cytochrome p-450 from liver microsomes of rats pretreated with phenobarbital for enzyme induction and later given a single oral dose (0.4 ml/kg) of carbon disulfide (CS2) have been examined. In vitro incubations of these microsomes with parathion and malathion showed no significant change in the amount of cytochrome p-450 from that present in the corresponding controls. This inhibition of oxidative desulfuration of these insecticides by microsomes from CS2-treated animals could be attributed to the prior binding

Hierarchical MOF-derived NiCoS/rGO composite for high-performance electrochemical detection of methyl parathion in fruits and vegetables.

The rapid and reliable detection of organophosphorus pesticide residues in food is essential for ensuring food safety and public health. In this study, a hierarchical metal-organic framework-derived nickel‑cobalt sulfide/reduced graphene oxide composite was fabricated as a high-performance electrochemical sensing platform for methyl parathion (MPN). Bimetallic nickel cobalt framework precursors with tunable metal ratios were synthesized by a solvothermal method and subsequently converted into nickel cobalt sulfide (NiCoS) nanoparticles uniformly anchored on reduced graphene oxide (rGO) sheets

Multiscale Simulations Reveal Distal Allosteric Reprogramming of Pocket Dynamics and Dual Hydrolytic Pathways in Engineered Methyl Parathion Hydrolase.

Methyl parathion hydrolase (MPH) is an important organophosphorus (OP) detoxifying enzyme, yet achieving a balance between catalytic activity and thermostability remains a major challenge. Notably, two engineered MPH variants, MPHase-m5a and MPHase-m5b, achieve this balance through predominantly distal substitutions, yet the underlying mechanisms remain unclear. Understanding how such remote mutations simultaneously enhance activity and stability is essential for elucidating MPH function and guiding the design of related detoxifying enzymes. Here, using multiscale simulations, we find that dis

Synthesis and characterization of g-C3N5/CuS/AgNPs nanocomposite as a Z-scheme photocatalyst for efficient methyl parathion degradation.

Water pollutants constitute a significant environmental concern today. Organophosphorus compounds, notably parathion, represent a critical category of water pollutants. Their extensive usage poses substantial risks to human health and the environment, necessitating their removal from water sources. This study focuses on the photocatalytic degradation of parathion, utilizing a g-C3N5/CuS/AgNPs nanocomposite synthesized through a combination of hydrothermal and ultrasonic methods. The nanocomposite was characterized using microscopic and spectroscopic techniques and demonstrated the capacity to

A Cu2+-triggered turn-on fluorescence non-enzymatic probe based on covalent organic framework for the detection of methyl parathion.

BACKGROUND: Methyl parathion, a potent organophosphorus insecticide, is extensively employed in agriculture and animal husbandry, leading to significant environmental contamination with pesticide residues, posing a grave threat to human health. This compound irreversibly inhibits acetylcholinesterase (AChE) in the human nervous system, resulting in the accumulation of acetylcholine (ACh), which is detrimental. Various enzyme activity-based assays have been explored due to its pathogenic mechanism, yet these methods are fraught with limitations. Thus, the development of a highly selective, sens

Water-Regulated Mechanisms for Degradation of Pesticides Paraoxon and Parathion by Phosphotriesterase: Insight from QM/MM and MD Simulations.

The enzymatic degradation of pesticides paraoxon (PON) and parathion (PIN) by phosphotriesterase (PTE) has been investigated by QM/MM calculations and MD simulations. In the PTE-PON complex, Znα and Znβ in the active site are five- and six-coordinated, respectively, while both zinc ions are six coordinated with the Znα -bound water molecule (WT1) for the PTE-PIN system. The hydrolytic reactions for PON and PIN are respectively driven by the nucleophilic attack of the bridging-OH- and the Znα -bound water molecule on the phosphorus center of substrate, and the two-step hydrolytic process is pre

Development of surface imprinted core-shell nanoparticles and their application in a solid-phase dispersion extraction matrix for methyl parathion.

Applying molecular imprinting techniques to the surface of functionalized SiO2 allows the preparation of molecularly imprinted polymers (MIPs) with accessible, high affinity and surface exposed binding sites. This paper demonstrates a new strategy for producing such hybrid organic-inorganic surface imprinted silica nanoparticles for specific recognition of methyl parathion. The technique provides surface grafting imprinting in chloroform using amino modified silica nanoparticles as supports, acrylamide as the functional monomer, γ-methacryloxypropyl trimethoxy silane as the grafting agent, and
