A comprehensive review of paratuberculosis in animals and its implications for public health.

Paratuberculosis is an infectious disease caused by Mycobacterium avium subspecies paratuberculosis (MAP). Typically, ruminant animals including cattle, buffalo, goats, and sheep are infected with MAP. Animals get infected with MAP in a number of ways, such as by eating or drinking contaminated food or water, or by nursing from an infected mother who may have contaminated teats or directly shed the organism in milk or colostrum. Animal-derived goods like meat, dairy, and tainted surface water have the potential to spread paratuberculosis through zoonotic transmission. Reports of paratuberculos

Bovine paratuberculosis: a review of the advantages and disadvantages of different diagnostic tests.

Paratuberculosis (PTB), or Johne's disease, is a chronic infectious granulomatous enteritis of ruminants, caused by Mycobacterium avium subspecies paratuberculosis (Map). It is characterized by diarrhea and progressive cachexia, which may cause the death of the animal. Calves are the most susceptible to infection. Infected animals excrete Map mainly by the feces. PTB is endemic worldwide, with high prevalence levels, strong economic impact and public health relevance because of its possible association with Crohn's disease. Although the current reference diagnostic test is identification of Ma

Comparative immunological and microbiological aspects of paratuberculosis as a model mycobacterial infection.

Paratuberculosis or Johne's disease of livestock, which is caused by Mycobacterium avium subsp. paratuberculosis (MAP), has increased in prevalence and expanded in geographic and host ranges over about 100 years. The slow and progressive spread of MAP reflects its substantial adaptation to its hosts, the technical limitations of diagnosis, the lack of practical therapeutic approaches, the lack of a vaccine that prevents transmission and the complexity and difficulty of the on-farm control strategies needed to prevent infection. More recently evidence has accumulated for an association of MAP w

The zoonotic potential of Mycobacterium avium spp. paratuberculosis: a systematic review.

BACKGROUND: The zoonotic potential of Mycobacterium avium ssp. paratuberculosis (MAP) has been debated for almost a century because of similarities between Johne's Disease (JD) in cattle and Crohn's disease (CD) in humans. Our objective was to evaluate scientific literature investigating the potential association between these two diseases (MAP and CD) and the presence of MAP in retail milk or dairy products using a qualitative systematic review. METHOD: The search strategy included 19 bibliographic databases, 8 conference proceedings, reference lists of 15 articles and contacting 28 topic-re

Intrauterine and transmammary transmission of Mycobacterium avium subsp paratuberculosis in sheep.

OBJECTIVE: To investigate intrauterine infection of foetuses with Mycobacterium avium subsp paratuberculosis and the presence of infection in mammary secretions of sheep. DESIGN: A study of 142 late-pregnant ewes and their foetuses from two heavily infected flocks. PROCEDURE: Infection of ewes was determined at necropsy by histopathology and culture of tissues and mammary secretions. Antemortem tests (clinical assessment, faecal culture and serology) were also applied. Foetuses from 59 infected ewes and 47 apparently uninfected ewes were examined by culture and histopathology. RESULTS: Five

The impact of recent tuberculin skin testing on paratuberculosis antibody responses.

There is a recognized interference between the single intradermal cervical comparative tuberculin (SICCT) test used for bovine tuberculosis surveillance and the paratuberculosis milk ELISA. Current UK guidance recommends a minimum interval of 42-60 d between a SICCT test and a paratuberculosis milk ELISA. However, recent research suggested that the post-SICCT antibody response may be greater in cows at high risk of Mycobacterium avium ssp. paratuberculosis (MAP) shedding (i.e, with an increased likelihood of shedding MAP in feces), leading to advisory guidance that high-prevalence herds could

Prevalence of bovine paratuberculosis in Chinese cattle populations: a meta-analysis.

BACKGROUND: Bovine paratuberculosis is a chronic infectious disease of ruminants primarily caused by Mycobacterium avium subsp. paratuberculosis (MAP). It is essentially a chronic granulomatous enteritis characterized by intractable diarrhea, progressive lethargy, and thickening of the intestinal mucosa with the formation of crumpled pouches. Bovine paratuberculosis not only adversely affects milk production and the quality of dairy products but also poses a significant threat to the economic development of dairy farming and human food security. Mycobacterium avium subsp. paratuberculosis and Hashimoto's thyroiditis: Is MAP the trigger?

Hashimoto's thyroiditis (HT) is an autoimmune disorder of the thyroid gland that can cause hypothyroidism. As HT is a multifactorial disorder, activation of immune responses in genetically predisposed individuals exposed to some environmental factors can contribute to it. Microorganisms, as environmental factors, including Mycobacterium avium ssp. paratuberculosis (MAP) by molecular mimicry, can be important in this autoimmune disorder. This study aimed to investigate the association between MAP and HT. This case-control study included 110 participants consisting of 60 HT patients and 50 healt

Whole-Genome association analysis of susceptibility to paratuberculosis in Holstein cattle.

The objective of this study was to identify genetic markers and genomic regions associated with susceptibility to Mycobacterium avium ssp. paratuberculosis (MAP) infection in Holstein cattle. Associated single nucleotide polymorphisms (SNPs) were identified by genotyping 521 MAP-infected Holstein cows and comparing SNP allele frequencies of these infected cows with allele frequencies estimated from specific reference populations. Reference population allele frequency estimates used Holstein sire genotype data and were weighted estimates based on sire usage within the population in question. Th

Crohn's disease in people exposed to clinical cases of bovine paratuberculosis.

Mycobacterium avium subspecies paratuberculosis (Map), the cause of ruminant paratuberculosis, has been proposed as the causative agent of Crohn's disease. The objective of this study was to determine whether exposure to clinical cases of bovine paratuberculosis was a risk factor for Crohn's disease. A questionnaire was sent to dairy farmers living on premises where the occurrence or absence of clinical cases of bovine paratuberculosis had previously been determined. The prevalence of Crohn's disease was found to be similar to that reported in other studies in the United Kingdom and showed no

Current understanding of the genetic diversity of Mycobacterium avium subsp. paratuberculosis.

Mycobacterium avium subsp. paratuberculosis (MAP) is the etiological agent of Johne's disease (or paratuberculosis). Paratuberculosis is a chronic gastroenteritis mainly affecting cattle, sheep and other ruminants. MAP is also of concern due to the heretofore unresolved issue of its possible role in Crohn's disease in humans. We present here a review of MAP (i) mobile genetic elements; (ii) repetitive elements; (iii) single nucleotide polymorphisms; and (iv) whole-genome comparisons to study the molecular epidemiology of MAP. A summary of the findings to date is presented, and the discriminato

Calibration-aware genomic prediction of paratuberculosis serostatus in indigenous Turkish goats.

Paratuberculosis, caused by Mycobacterium avium subsp. paratuberculosis (MAP), is a chronic, incurable enteritis of ruminants in which late-onset clinical signs preclude timely intervention, making early genomic stratification a key control lever. We investigated the genetic architecture underlying the host antibody response to MAP - measured as anti-MAP ELISA serostatus - using genome-wide SNP data from 474 animals representing seven indigenous Turkish goat breeds sampled across 11 provinces (237 seropositive, 237 seronegative; estimated true seroprevalence 14.1%). Fourteen architecturally di

Culture of Mycobacterium avium subsp. paratuberculosis: challenges, limitations and future prospects.

Mycobacterium avium subsp. paratuberculosis (MAP) causes paratuberculosis (Johne's disease) in ruminants and is suspected to be involved in the development of Crohn's disease and several autoimmune disorders. As such, sensitive and specific MAP detection methods are required to confirm infection in animals and identify potential sources of animal and human exposure. Despite recent developments in immunological and nucleic acid-based detection methods, culture-based detection of MAP remains the 'gold standard' against which the sensitivity and specificity of other detection methods are measured

Neglected Facts on Mycobacterium Avium Subspecies Paratuberculosis and Type 1 Diabetes.

Civilization factors are responsible for the increasing of human exposure to mycobacteria from environment, water, and food during the last few decades. Urbanization, lifestyle changes and new technologies in the animal and plant industry are involved in frequent contact of people with mycobacteria. Type 1 diabetes is a multifactorial polygenic disease; its origin is conditioned by the mutual interaction of genetic and other factors. The environmental factors and certain pathogenetic pathways are shared by some immune mediated chronic inflammatory and autoimmune diseases, which are associated

LAMP technology: Rapid identification of Brucella and Mycobacterium avium subsp. paratuberculosis.

In this study, we developed new sets of primers to detect Brucella spp. and M. avium subsp. paratuberculosis (MAP) through isothermal amplification. We selected a previously well-characterized target gene, bscp31, specific for Brucella spp. and IS900 for MAP. The limits of detection using the loop-mediated isothermal amplification (LAMP) protocols described herein were similar to those of conventional PCR targeting the same sequences. Hydroxynaphtol blue and SYBR Green(TM) allowed direct naked-eye detection with identical sensitivity as agarose gel electrophoresis. We included the LAMP-based p
