Taxonomic position of the rickettsiae: current knowledge.

The term rickettsiae initially encompassed all intracellular bacteria. Early rickettsial taxonomy was based on a comparison of a few phenotypic characteristics and recently, molecular studies brought new bases for rickettsial taxonomy. All rickettsial species studied so far belong to the alpha and gamma groups of the Proteobacteria. Ehrlichiae complex groups Cowdria ruminantium, Anaplasma marginale and Wolbachia pipientis and the related parthenogenesis and cytoplasmic incompatibility bacteria, whereas Rochalimaea species group with Bartonella bacilliformis. Rickettsia tsutsugamushi may form a

Back to Tityus serrulatus Lutz & Mello, 1922 (Scorpiones: Buthidae): new comments about an old species.

A synopsis on the historical, geographical and ecological aspects related to the most conspicuous scorpion species of the genus Tityus known from Brazil is proposed. Tityus serrulatus Lutz & Mello, 1922 was described precisely one century ago, nevertheless many questions related to its ecological adaptations and geographical expansion remain without a precise response. This species, well known for its infamous reputation of noxious species, is also known for its capacity to reproduce asexually, by parthenogenesis. Although the individuals of a given population are considered clones, a new hypo

Inherited microorganisms, sex-specific virulence and reproductive parasitism.

Parasites show an amazing repertoire of adaptations, highlighted by complex life cycles that allow both survival in the host and transmission among hosts. However, there is one heterogeneous group of microorganisms whose adaptations are perhaps even more surprising: parthenogenesis induction, feminization of genetic males, killing of male hosts and sperm-mediated sterilization of uninfected eggs. The common feature of these microorganisms is their mode of transmission: inheritance from mother to offspring. Here, we present an introduction to hereditary symbiosis, focusing on microsporidia and

Triploidy drives vector capacity and expansion of the parthenogenetic tick Haemaphysalis longicornis.

The Haemaphysalis longicornis tick, an important arthropod vector native to East Asia, has expanded its range to Oceania and the American continents, where populations are exclusively parthenogenetic females. While the parthenogenetic population possesses a stable triploid genetic system, the absence of a triploid genome assembly has prohibited us from understanding their genetic evolution and biological traits. Here, we present a chromosome-level, haplotype-resolved triploid genome assembly of the parthenogenetic population by integrating ultra-deep long-read high-fidelity, short-read high-th

Effect of Vibration and Tilting Mechanical Loading Device on Parthenogenetic Embryos Development of Mice Immature Oocytes.

The growth and development of early mammalian embryos mainly take place in the fallopian tube, which not only provides nutrients for embryonic growth and development but also offers suitable mechanical conditions. The embryo culture system established in assisted reproductive technology mainly simulates the environment in which oocytes and embryos grow and develop in vivo. However, current in vitro embryo culture is mainly static and cannot completely mimic the mechanical environment in which embryos grow and develop in vivo. Therefore, to more accurately simulate the mechanical environment of

Effect of nucleocytoplasmic ratio on the in vitro porcine embryo development after in vitro fertilization or parthenogenetic activation.

This study was conducted to examine whether the nuclear to cytoplasmic (N/C) ratio had any influence on the timing of embryo compaction and blastocoel formation, as well as formation rate and quality of blastocyst. First, we produced embryos with increased N/C ratio by removal of approximately one-third of the cytoplasm and with decreased N/C ratio by doubling the oocyte cytoplasm with an enucleated oocyte. The initiation of compaction and cavitation in reduced cytoplasm group was significantly earlier (P < 0.05) compared with the control and doubled cytoplasm groups. The rate of blastocysts i

Developmental and reproductive biology of the ectoparasitoid, Elasmus steffani, in a substitute host, Ephestia kuehniella.

Elasmus steffani (Viggiani) (Hymenoptera: Elasmidae) is a gregarious idiobiont ectoparasitoid of Prays oleae (Bernard) (Lepidoptera: Plutellidae), an olive crop pest. In the substitute host Ephestia kuehniella (Zeller) (Lepidoptera: Pyralidae), the duration of the developmental stages was approximately 11-15 days. The preoviposition was 8.9 ± 5.0 days, and oviposition lasted 30.4 ± 10.5 days, with a reproduction capacity of 185.5 ± 62.3 eggs per female, for a mean of 5.4 ± 0.9 eggs per day. The oviposition rhythm reached its maximum when the parasitoid was 35 days of age. The lack of food nega

An obscure rider obstructing science: the conflation of parthenotes with embryos in the Dickey-Wicker amendment.

In 1996 Congress passed the Dickey-Wicker Amendment (DWA) as part of an appropriations bill; it has been renewed every year since. The DWA bans federal funding for research using embryos and parthenotes. In this paper, we call for a public discussion on parthenote research and a questioning of its inclusion in the DWA. We begin by explaining what parthenotes are and why they are useful for research on reproduction, cancer, and stem cells. We then argue that the scientific difference between embryos and parthenotes translates into ethical differences, and claim that research on parthenotes is m

Wolbachia pipientis: microbial manipulator of arthropod reproduction.

The alpha-proteobacterium Wolbachia pipientis is a very common cytoplasmic symbiont of insects, crustaceans, mites, and filarial nematodes. To enhance its transmission, W. pipientis has evolved a large scale of host manipulations: parthenogenesis induction, feminization, and male killing. W. pipientis's most common effect is a crossing incompatibility between infected males and uninfected females. Little is known about the genetics and biochemistry of these symbionts because of their fastidious requirements. The affinity of W. pipientis for the microtubules associated with the early divisions

Functional Modules in Gametogenesis.

Gametogenesis, the production of eggs and sperm, is a fundamental process in sexually reproducing animals. Following gametogenesis commitment and sexual fate decision, germ cells undergo several developmental processes to halve their genomic size and acquire sex-specific characteristics of gametes, including cellular size, motility, and cell polarity. However, it remains unclear how different gametogenesis processes are initially integrated. With the advantages of the teleost fish medaka (Oryzias latipes), in which germline stem cells continuously produce eggs and sperm in mature gonads and a

Strong cytoplasmic incompatibility and high vertical transmission rate can explain the high frequencies of Wolbachia infection in Japanese populations of Colias erate poliographus (Lepidoptera: Pieridae).

Wolbachia, belonging to Alphaproteobacteria, is ubiquitously found in arthropods and filarial nematodes, and is known to manipulate the reproduction of its hosts in various ways, such as feminization, male killing, induction of parthenogenesis or induction of cytoplasmic incompatibility. We found that the Wolbachia infection frequencies of the butterfly Colias erate poliographus were high (85.7-100%) in seven Japanese populations. Crossing experiments and rearing revealed that the Wolbachia strain exhibited strong cytoplasmic incompatibility and perfect vertical transmission in C. erate poliog

Parasitism genes and host range disparities in biotrophic nematodes: the conundrum of polyphagy versus specialisation.

This essay considers biotrophic cyst and root-knot nematodes in relation to their biology, host-parasite interactions and molecular genetics. These nematodes have to face the biological consequences of the physical constraints imposed by the soil environment in which they live while their hosts inhabit both above and below ground environments. The two groups of nematodes appear to have adopted radically different solutions to these problems with the result that one group is a host specialist and reproduces sexually while the other has an enormous host range and reproduces by mitotic parthenoge

Like mother, like daughter? Phenotypic plasticity, and heritability of size in a parthenogenetic wasp.

Parthenogenetic wasps provide an ideal natural experiment to study the heritability, plasticity, and microevolutionary dynamics of body size. Dinocampus coccinellae (Hymenoptera: Braconidae, Euphorinae) is a solitary, generalist braconid parasitoid wasp that reproduces through thelytokous parthenogenesis and parasitizes over 50 diverse species of coccinellid ladybeetles worldwide as hosts. Here, we designed an experiment with parthenogenetic lines of D. coccinellae presented with three different host ladybeetle species of varying sizes, across multiple generations to investigate heritability a

Comparison of symmetrical and asymmetrical cleavage 2-cell embryos of porcine by Smart-seq2.

Early cleavage (EC) influences the development of the pre-implantation and post-implantation embryo. Symmetric cleavage (Sym) and asymmetric cleavage (Asy) have been observed in EC, but its molecular mechanism remains unclear. This study was designed to pick out the key candidate genes and signaling pathway between Sym and Asy embryos by applying Smart-seq2 technique. In in-vitro fertilization (IVF) 2-cell embryos, Sym embryos and Asy embryos accounted for 62.55% and 37.45%, respectively. The 2-cell rate, blastocyst rate and total blastocyst cells of Sym group were significantly higher than th

Mitochondrial DNA suggests a single maternal origin for the widespread triploid parthenogenetic pest species, Paratanytarsus grimmii, but microsatellite variation shows local endemism.

Parthenogenesis is common among invasive pest species, with many parthenogenetic species also showing polyploidy. Parthenogenetic polyploid species often have multiple hybrid origins and the potential to rapidly spread over vast geographical areas. In this study, we examine patterns of mitochondrial and microsatellite variation in a widespread triploid parthenogenetic chironomid pest species, Paratanytarsus grimmii. Based on samples from five countries, including Australia, England, Germany, Japan, and Canada, we found extremely low mitochondrial diversity (<0.14%), with most individuals shari

Brevipalpus californicus, B. obovatus, B. phoenicis, and B. lewisi (Acari: Tenuipalpidae): a review of their biology, feeding injury and economic importance.

The genus Brevipalpus includes most of the economically important species of Tenuipalpidae. Many Brevipalpus species reproduce by theletokous parthenogenesis while other species reproduce by male fertilization of female eggs. Previous researchers have determined that Brevipalpus californicus (Banks), B. obovatus Donnadieu, and B. phoenicis (Geijskes) females were haploid with two chromosomes. The life cycle and developmental times for these three species are reviewed. Longevity of each Brevipalpus species is two to three times greater than corresponding longevities of various tetranychid mites

Temporal and spatial selection against parthenogenetic cells during development of fetal chimeras.

The fate of parthenogenetic cells was investigated during development of fetal and early postnatal chimeras. On day 13 of embryonic development, considerable contribution of parthenogenetic cells was observed in all tissues of chimeric embryos, although selection against parthenogenetic cells seemed to start before day 13. Between days 13 and 15 of development, parthenogenetic cells came under severe selective pressure, which was most striking in tongue. The disappearance of parthenogenetic cells from tongue coincided with the beginning of myoblast fusion in this tissue. Severe selection again

From Rigid Order to Radical Variation: Mitogenome Evolution in the Main Lineages of a Lesser-Known Animal Phylum (Gastrotricha).

Mitochondrial genomes offer valuable insights into biological and phylogenetic processes, yet the factors shaping their architecture across metazoan lineages remain poorly understood, largely due to limited taxonomic sampling. To address this gap, we analyzed mitochondrial genomes from 20 species spanning a broad taxonomic spectrum of the phylum Gastrotricha. Our findings, supported by phylogenetic analyses based on mitochondrial datasets, reveal two distinct evolutionary patterns: one lineage displays remarkable conservation in genome structure, while the other exhibits variability in gene co

Establishment of trophoblast cell line derived from buffalo (Bubalus bubalis) parthenogenetic embryo.

We have established trophoblast cell lines, from parthenogenesis-derived buffalo blastocysts. The buffalo trophoblast cells were cultured continuously over 200 days and 21 passages. These cells were observed by phase-contrast microscopy for their morphology and characterized by reverse transcriptase polymerase chain reaction and immunofluorescence against trophoblast-specific markers and cytoskeletal proteins. Trophoblast cells showed positive staining for CDX2, a marker of these cells at both blastocyst and cell line levels. Epithelial morphology of these cells was revealed by positive staini
