Canine parvovirus-2 (CPV-2) and canine distemper virus (CDV) are two highly infectious and pathogenic pathogens that harm canids and various carnivores, and often cause co-infections in clinical settings. Although commercial attenuated live vaccines against CPV-2 and CDV have been widely used to prevent these viral infections, there are still issues of biosafety and insufficient protection against new variants, thus requiring novel vaccines. In this study, we developed a recombinant pseudovirus expression system based on the CPV-2 full-length infectious clone, and found that the recombinant CP

Molecular mimicry is hypothesized to play a key role in the pathogenesis of acute polyradiculoneuritis (APRN) in dogs, as in Guillain-Barré syndrome (GBS). GBS is linked to Campylobacter jejuni and cytomegalovirus, while Parvovirus B19 shows temporal and serological associations. In dogs, the cause of APRN is often unknown, although recent vaccination or ingestion of raw poultry contaminated with Clostridium perfringens is documented. Antiganglioside antibodies are potential serum biomarkers for APRN. Eight 2.5-month-old Sprocker littermates and a 3-month-old female Cockapoo developed severe g

Canine parvovirus type 2 (CPV-2) has high morbidity and mortality rates in canines. Nonstructural protein 1 (NS1) of CPV-2 has endonuclease activity, initiates viral DNA replication, and is highly conserved. Thus, it is a promising target for antiviral inhibitor development. We overexpressed a 41.9 kDa active recombinant endonuclease in Escherichia coli and designed a nicking assay using carboxyfluorescein and quencher-linked ssDNA as substrates. The optimal temperature and pH of the endonuclease were 37°C and pH 7, respectively. Curcumin, bisdemethoxycurcumin, demethoxycurcumin, linoleic acid

Infectious disease threats are increasingly recognized as a major contributor to mortality in wild populations of African wild dog (Lycaon pictus, AWD). Canine distemper virus (CDV) infection has been implicated as a cause of pack mortality in both captive and wild AWD populations. Ten animals were vaccinated with RecombitekTM C3, a vaccine containing a recombinant CDV, and modified live canine parvovirus (CPV) and adenovirus-2 components, at 8, 12, and 16 wk of age. Half of the pups received the vaccine IM and the other half SC. All ten pups had a positive serological response to CDV after th

A concurrent outbreak of infection by canine parvovirus 2b (CPV-2b) and Clostridium difficile producing A and/or B toxins occurred in Asian small-clawed otters (Amblonyx cinereus). The 5 clinically affected otters were 6- to 24-mo-old intact females that had severe diarrhea, dehydration, were acutely comatose, and died 1-4 d after the onset of clinical signs. Postmortem examination was performed in 3 of 7 otters. Macroscopically, the small intestine was diffusely reddened and contained red-to-brown, malodorous, watery digesta without formed feces (3 of 3). Histologic examination identified los

This randomized controlled clinical trial aimed to evaluate whether the adjunctive administration of Saccharomyces boulardii (S. boulardii) to standard treatment could accelerate clinical recovery and improve hematological indicators of systemic inflammation in dogs with naturally occurring canine parvovirus (CPV) enteritis. Additionally, the study assessed the levels of nuclear factor-κB (NF-κB) and interleukin 8 (IL-8) associated with inflammatory pathways. This study consisted of three groups, each with 10 dogs aged 3-5 months. The control group comprised apparently healthy dogs. Group 1 re

Canine parvovirus (CPV) is the main cause of viral diarrhea in dogs. CPV became a global disease in 1978 and was endemic all over the world. CPV-2 was the first strain to be identified, but with genetic mutations, new genotypes such as CPV-2a/2b/2c/new-2a/new-2b have emerged. In this study, 128 fecal samples of stray dogs suspected of CPV-2 infection were collected from January to March 2021 in Shanghai, China. All samples were screened by PCR and further analyzed by VP2 gene. The positive rate of CPV-2 was 9.4% (12/128), of which 6 CPV-2 isolates were successfully isolated. Phylogenetic tree

Canine parvovirus (CPV) is a highly contagious and worldwide cause of serious and often fatal disease in dogs, despite the widespread availability of vaccines. Which vaccine-related factors are associated with vaccination failure is largely unknown, and there are no reports from Australia. In this study - the first national population-level CPV study of its kind ever conducted - we analysed data on 594 cases of apparent CPV vaccination failure reported from an Australian national surveillance system to determine whether vaccine strain, type or administration protocol are risk factors for vacci

An optimized VP2 gene from the current prevalent CPV strain (new CPV-2a) in China was expressed in a baculovirus expression system. It was found that the VP2 proteins assembled into virus-like particles (VLPs) with antigenic properties similar to those of natural CPV and with an especially high hemagglutination (HA) titer (1:2(20)). Dogs intramuscularly or orally immunized with VLPs produced antibodies against CPV with >1:80 hemagglutination inhibition (HI) units for at least 3 months. The CPV VLPs could be considered for use as a vaccine against CPV or as a platform for research on chimeric V

BACKGROUND: Pet ferrets are typically vaccinated against distemper using a canine multivalent vaccine despite the lack of relevant safety or efficacy data. Here, we investigate the safety of a novel live canine parvovirus type 2c (CPV-2c) vaccine strain and the efficacy of the live Onderstepoort canine distemper virus (CDV) vaccine strain used in a new bivalent vaccine.
METHODS: Following an overdose of the CPV-2c component to 16 ferrets, blood, faeces and various tissues were tested to investigate viral dissemination and shedding. Serum antibody concentrations were also measured. Long-term s

BACKGROUND: Canine parvovirus (CPV) is a highly contagious virus that can lead to severe clinical complications in dogs. RNA-binding proteins (RBPs) play a crucial role in regulating translation and stabilizing RNA. Due to the failure of the immune system to respond effectively in dogs infected with CPV, this study aimed to investigate various immunological markers in CPV-infected dogs and to evaluate the impact of RBPs from these infected dogs on immune responses. Blood samples were collected from both healthy and parvovirus-infected dogs. Parvovirus infection was confirmed using immunochroma

Canine parvovirus 2 (CPV-2) and feline panleukopenia virus (FPLV) often cause acute enteric disease in their hosts. A simple, rapid, and effective method for the on-site detection of these viruses would be useful. We used a denaturation bubble-mediated strand exchange amplification (SEA) method to successfully detect CPV-2 and FPLV in fecal samples. SEA could detect as little as 3.6 pg/μL of CPV-2 and 6.6 pg/μL of FPLV genomic DNA following a 40-min incubation at an isothermal temperature of 61°C. Unlike PCR, SEA does not require complicated equipment, and positive samples produce a color chan

BACKGROUND: Canine Distemper Virus (CDV) and Canine Parvovirus (CPV) lead to infections with high mortality rates in dogs. These viruses affect unvaccinated dogs or dogs with incomplete vaccination protocols. Vaccination plays an important role in reducing death rates, preventing clinical cases and controlling the spread of virus However, the efficacy of vaccination might be affected by different factors including vaccine scheduling and the neutralization of the vaccine targets by maternal antibodies. In face of these factors, the main goals of this study are (i) to investigate the antibody re

Canine parvovirus (CPV) and Canine distemper virus (CDV) can cause fatal diseases in giant panda (Ailuropoda melanoleuca). The main capsid protein of CPV VP2 can be self-assembled to form virus-like particles (VLPs) in vitro, which is of great significance for potential vaccine development. In the present study, we remodeled the VP2 protein of a giant panda-derived CPV, where the major CDV F and N epitopes were incorporated in the N-terminal and loop2 region in two combinations to form chimeric VLPs. The reactivity ability and morphology of the recombinant proteins were confirmed by Western bl
