Legionella Persistence in Manufactured Water Systems: Pasteurization Potentially Selecting for Thermal Tolerance.

Legionella is an opportunistic waterborne pathogen of increasing public health significance. Pasteurization, otherwise known as super-heat and flush (increasing water temperature to above 70°C and flushing all outlets), has been identified as an important mechanism for the disinfection of Legionella in manufactured water systems. However, several studies have reported that this procedure was ineffective at remediating water distribution systems as Legionella was able to maintain long term persistent contamination. Up to 25% of L. pneumophila cells survived heat treatment of 70°C, but all of th

Survival of Listeria monocytogenes in milk during high-temperature, short-time pasteurization.

Milk from cows inoculated with Listeria monocytogenes was pooled for 2 to 4 days and then heated at 71.7 to 73.9 degrees C for 16.4 s or at 76.4 to 77.8 degrees C for 15.4 s in a high-temperature, short-time plate heat exchanger pasteurization unit. L. monocytogenes was isolated from milk after heat treatment in six of nine pasteurization trials done at 71.7 to 73.9 degrees C and in none of three trials done at 76.4 to 77.8 degrees C. An average of 1.5 to 9.2 L. monocytogenes cells was seen in each milk polymorphonuclear leukocyte before heat treatment in 11 of 12 pasteurization trials. Notice

Microbial safety of donor human milk before pasteurization: seasonal, maternal, and procedural determinants.

BACKGROUND: Donor human milk (DHM) is a vital alternative for preterm infants when maternal breastfeeding is unavailable, yet its microbial safety remains a critical concern. This study provides the first longitudinal evaluation of DHM contamination patterns in Iran, aiming to identify maternal, procedural, and seasonal risk factors to inform safer milk bank practices. METHODS: This repeated cross-sectional study evaluated microbial contamination in DHM over six years (2019–2025) at Valiasr Hospital’s Human Milk Bank (HMB) in Tehran. Samples were stratified into two phases following a protocol

New alternatives to holder pasteurization in processing donor milk in human milk banks.

Infectious and toxicological risks are the main potential hazards that operators of Human Milk Banks (HMBs) encounter and must eliminate. HMBs are trying to implement procedures that allow to manage and sanitize human milk without altering significantly its nutritional and biologically protective components, obtaining a product characterized by a valid balance between safety and biological quality. The history of human milk processing is linked to the origins of HMBs themselves. And although other forms of sterilization were used originally, pasteurization soon became the recognized most effec

Evaluation of Pilot-Scale Radio Frequency Heating Uniformity for Beef Sausage Pasteurization Process.

Radio frequency (RF) heating has the advantages of a much faster heating rate as well as the great potential for sterilization of food compared to traditional thermal sterilization. A new kettle was designed for sterilization experiments applying RF energy (27.12 MHz, 6 kW). In this research, beef sausages were pasteurized by RF heating alone, the dielectric properties (DPs) of which were determined, and heating uniformity and heating rate were evaluated under different conditions. The results indicate that the DPs of samples were significantly influenced (p < 0.01) by the temperature and freq

Inactivation Kinetics of Coxiella burnetii During High-Temperature Short-Time Pasteurization of Milk.

The Gram-negative, obligate intracellular bacterium Coxiella burnetii is the causative organism of the zoonosis Q fever and is known for its resistance toward various intra- and extracellular stressors. Infected ruminants such as cattle, sheep, and goats can shed the pathogen in their milk. Pasteurization of raw milk was introduced for the inactivation of C. burnetii and other milk-borne pathogens. Legal regulations for the pasteurization of milk are mostly based on recommendations of the Codex Alimentarius. As described there, C. burnetii is considered as the most heat-resistant non-spore-for

Pasteurization Modifies the Sensorial Attributes and Nutritional Profile of Orange Pulp By-Product.

After orange processing, different by-products are generated, i.e., peels, seeds and pulps. The pulp is highly perishable, being an unstable food matrix that needs a preservation process to be stored and used again in the food production chain. Pasteurization is the technique of choice before aseptically packaging and storing under refrigerated conditions. In this study, the effect of pasteurization has been evaluated on the chemical, functional and sensorial profiles. Ash content decreased (p < 0.05) after the thermal treatment. Indeed, magnesium, calcium and zinc diminished, although copper

Use of pulsed ultraviolet laser light for the cold pasteurization of bovine milk.

Because of concerns that some potentially dangerous microorganisms may survive conventional heat pasteurization of milk and because the heat needed to sterilize milk affects marketability, the ability to efficiently cold pasteurized milk may become more desirable. In this pilot study, we investigated the use of pulsed ultraviolet (PUV) laser light to nonthermally (cold) pasteurized bovine milk. Dairy bulk tank milk was treated with UV light (248 nm) emitted from a pulsed excimer laser. The samples were then analyzed for surviving bacteria by spiral plate counting and subculturing in Trypticase

Effect of pasteurization on infectivity of Cryptosporidium parvum oocysts in water and milk.

Cryptosporidium parvum is a major cause of diarrheal disease in humans and has been identified in 78 other species of mammals. The oocyst stage, excreted in feces of infected humans and animals, has been responsible for recent waterborne outbreaks of human cryptosporidiosis. High temperature and long exposure time have been shown to render oocysts (suspended in water) noninfectious, but for practical purposes, it is important to know if high-temperature--short-time conditions (71.7 degrees C for 15 s) used in commercial pasteurization are sufficient to destroy infectivity of oocysts. In this s

Pasteur at the Academy of Medicine: from hygiene to germ theory.

In the 19th century, the applications of the scientific discoveries of Louis Pasteur to medicine had difficulty in imposing themselves within the French medical community and its National Academy. Among those concerning infectious diseases, their prevention by hygiene and their etiology by microbes are described here. Louis Pasteur found it difficult to convince and had to fight, to show through the surgical practice of asepsis and examples of infections induced in animal models (anthrax), the modes of transmission of germs and their pathogenicity.

The early use of Pasteur's rabies vaccine in the United States.

Louis Pasteur's vaccine against rabies was introduced in France during 1885. A year later it became available within the United States. This article tells the story of the first use of the Pasteur vaccine in America and describes the early history of the vaccine's production and distribution across the country by Pasteur Institutes established for this purpose. Highlights of Pasteur's landmark studies on rabies are presented: research which pioneered the field of virology and the use of immunization to prevent infectious diseases.

Recent progress on sanofi pasteur's dengue vaccine candidate.

The sanofi pasteur candidate dengue tetravalent vaccine (TV) is a recombinant live attenuated vaccine. It is based on a backbone of yellow fever vaccine (YF 17D) replication genes and incorporates the envelope genes of the four dengue virus serotypes. Pre-clinical studies have demonstrated that dengue TV is genetically stable, non-hepatotropic, less neurovirulent than YF 17D and does not infect mosquitoes by the oral route. Dengue TV reactogenicity, viraemia induction and antibody responses were investigated in three Phase II trials in the USA, the Philippines and Mexico. Participants were ran

Rv1773 is a transcriptional repressor deleted from BCG-Pasteur.

Mycobacterium bovis Bacille Calmette-Guérin (BCG) is a live attenuated vaccine for the prevention of tuberculosis. Transcriptome comparison reveals dysregulated expression of two genes, Rv1774 and Rv1775, exclusively in the Pasteur strain of BCG. We show that these genes form a bicistronic operon regulated by Rv1773, a transcriptional repressor deleted during the in vitro evolution of BCG.

Histological findings in a human autogenous pasteurized bone graft.

BACKGROUND: Autogenous bone graft after pasteurization is one of the most valuable procedures for reconstruction of large bone defects following excision of malignant musculoskeletal tumors. To date, there have been no documented histological reports on pasteurized bone grafts, apart from short-term histological results. CASE REPORT: We treated a 27-year-old male with a periosteal chondrosarcoma of the tibia by wide excision and reimplantation of the large pasteurized bone. Biopsy specimens harvested from the pasteurized bone over 3 years after reimplantation were evaluated histologically. Th

[Introduction to the commemorative session of the centennial of the death of Louis Pasteur].

When elected a member of Académie de médecine in 1873, Pasteur was well known for his achievements in chemistry, fermentation and silk worm diseases. He denied spontaneity of diseases, their cause are living germs. Passionate disputations took place in Academy on subjects of septicaemia, childbirth fever, or fowl cholera. In 1881, his success was obtained by vaccination of sheep against epidemic anthrax: his method of virus action softening was available for many human infections diseases, in spite of many disputations. The same method applied to rabies gave him universal glory. When he died i

Animal Toxins: A Historical Outlook at the Institut Pasteur of Paris.

Humans have faced poisonous animals since the most ancient times. It is recognized that certain animals, like specific plants, produce toxic substances that can be lethal, but that can also have therapeutic or psychoactive effects. The use of the term "venom", which initially designated a poison, remedy, or magic drug, is now confined to animal poisons delivered by biting. Following Louis Pasteur's work on pathogenic microorganisms, it was hypothesized that venoms could be related to bacterial toxins and that the process of pathogenicity attenuation could be applied to venoms for the preventio
