Effects of cold atmospheric plasma on patulin degradation, polyphenol oxidase inactivation and other physicochemical properties of fresh-cut apple slices during storage.

Mycotoxin elimination is one of the significant concerns in the food industry. Cold atmospheric plasma (CAP) is a novel non-thermal method with high potential in this regard. This study aimed to evaluate the dielectric-barrier discharge (DBD) cold plasma effect on fresh-cut apple slices contaminated with patulin. The contaminated slices were subjected to a 17-23 kV range of DBD for 2.5-10 min and stored for 14 days in the refrigerator. This treatment eliminated the patulin and total viable count (TVC) by 23 kV power and 2.5- and 8.5-min exposure. The polyphenol oxidase (PPO) activity also decr

Identification of mouse metabolic variations related to patulin-induced acute and subacute hepatotoxicity by ultra-high-performance liquid chromatography high-resolution mass spectrometry.

Patulin (PAT), a toxin produced by molds in fruits and related products, has caused frequent food poisoning incidents worldwide. However, its potential mechanism of hepatotoxicity remains presently unclear. Herein, we intragastrically administered the C57BL/6J mice with 0, 1, 4, and 16 mg/kg b.wt of PAT on a single occasion (acute model), and 0, 50, 200, and 800 μg/kg b.wt of PAT daily over two weeks (subacute model). Assessments of histopathology and aminotransferase activities confirmed that significant hepatic damages were induced. Metabolic profiling on the liver using ultra-high-performan

Antifungal activity of silver ion on ultrastructure and production of aflatoxin B1 and patulin by two mycotoxigenic strains, Aspergillus flavus OC1 and Penicillium vulpinum CM1.

OBJECTIVE: The antifungal activity of silver ion from silver nitrate solution was tested against two pathogenic and toxigenic fungal strains. The first was Aspergillus flavus OC1, a clinical aflatoxigenic strain that causes fungal keratitis and the second was Penicillium vulpinum CM1, a maize-pathogenic strain that is positive for patulin (PAT) producing ability. MATERIALS AND METHODS: Agar well diffusion assays on yeast sucrose (YES) agar were applied for determination of the antifungal activity of silver ions either filter- or autoclaved-sterilized. Transmission electron microscopy was used

Covalent Michael-type recognition-based dual-mode sensing platform using thiol-functionalized nitrogen-doped carbon dots and Fe-MOF nanozyme for ultrasensitive patulin detection.

Patulin is a toxic mycotoxin commonly found in apple and fruit products, requiring sensitive and reliable detection methods. Herein, a novel dual-mode sensing platform integrating turn-off fluorescence and colorimetric detection was developed for rapid patulin determination in apple juice. The strategy exploits patulin's unique electrophilic reactivity toward thiols across two independent signal channels. Thiol-functionalized nitrogen-doped carbon dots (SH-N-CDs) serve as both recognition element and signal transducer, where direct covalent Michael-type addition of patulin onto the CD surface

Green-synthesis of iron nanoparticles using wild cherry laurel (Prunus laurocerasus L.) fruit extract and their patulin and HMF adsorption capacities along with biological activities.

In this study, iron nanoparticles (YBFeNPs) were green-synthesized using the aqueous extract (YB-E) of wild cherry laurel (Prunus laurocerasus L.) fruit to evaluate their potential applications in food safety. The synthesized YBFeNPs were investigated through Fourier-transform infrared spectroscopy, X-ray diffraction, scanning electron microscopy, transmission electron microscopy, and energy-dispersive X-ray spectroscopy analyses. YBFeNPs showed higher total phenolic content, total flavonoid content, and total antioxidant capacity compared to the original extract, suggesting that plant metabol

TGF-β/Smad signaling pathway plays a crucial role in patulin-induced pro-fibrotic changes in rat kidney via modulation of slug and snail expression.

Patulin (PAT) is a mycotoxin that contaminates a variety of food and foodstuffs. Earlier in vitro and in vivo findings have indicated that kidney is one of the target organs for PAT-induced toxicity. However, no study has evaluated the chronic effects of PAT exposure at environmentally relevant doses or elucidated the detailed mechanism(s) involved. Here, using in vitro and in vivo experimental approaches, we delineated the mechanism/s involved in pro-fibrotic changes in the kidney after low-dose chronic exposure to PAT. We found that non-toxic concentrations (50 nM and 100 nM) of PAT to norma

Valorization of durian shell waste: antifungal activity of ethanol extract against Penicillium expansum and its underlying mechanisms.

Durian shells contain abundant antimicrobial active substances, representing an underutilized resource for controlling postharvest pathogens. This study evaluated the antifungal activity of durian shell ethanol extract (DSEE) against several postharvest pathogenic fungi and explored its underlying mechanism. LC-MS analysis revealed that DSEE is rich in flavonoids, phenolics, and alkaloids. In vitro experiments demonstrated that DSEE significantly inhibited the mycelial growth of Botrytis cinerea, Fusarium oxysporum, Cladosporium cladosporioides, Penicillium digitatum, P. expansum, and P. itali

Small chemical chromatin effectors alter secondary metabolite production in Aspergillus clavatus.

The filamentous fungus Aspergillus clavatus is known to produce a variety of secondary metabolites (SM) such as patulin, pseurotin A, and cytochalasin E. In fungi, the production of most SM is strongly influenced by environmental factors and nutrients. Furthermore, it has been shown that the regulation of SM gene clusters is largely based on modulation of a chromatin structure. Communication between fungi and bacteria also triggers chromatin-based induction of silent SM gene clusters. Consequently, chemical chromatin effectors known to inhibit histone deacetylases (HDACs) and DNA-methyltransfe

ROS/ER-UPR Signaling-Mediated Inhibition of Cyclo(l-Anthraniloyl-l-Pro) on Penicillium expansum and Its Application in Controlling Blue Mold.

Penicillium expansum is a major postharvest pathogen of pome fruits, causing blue mold rot and leading to patulin contamination. Cyclic(l-anthraniloyl-l-proline) (cAntP) was identified from Lacticaseibacillus paracasei ZX1231 in our previous study. Here, we evaluated the inhibitory effect of cAntP against P. expansum, including an MIC of 1.25 mg/mL, colony growth inhibition, spore germination delay, and conidiation reduction. The antifungal mechanism of cAntP was clarified to be related to the disruption of cell membrane integrity, the leakage of intracellular components, the occurrence of act

Detection of Penicillium-Toxins in Nuts Commercialized in Italy Through LC-MS/MS Analyses.

The consumption of nuts is widespread globally and constitutes a significant component of the human diet due to its nutritional value. However, the presence of mycotoxins in food products, including nuts, is a global public health concern. Mycotoxins are toxic metabolites produced by contaminating fungi such as Aspergillus spp. and Penicillium spp., which can contaminate crops during growth, harvesting, storage, or transport. The aim of this study was to conduct monitoring for the presence of mycotoxins in nuts already on the market. Specifically, secondary metabolites produced by Penicillium

[Effect of mycotoxins on the rate of fermentation in Saccharomyces cerevisiae].

The rate of fermentation of Saccharomyces cerevisiae is partially inhibited by different mycotoxins. This effect is remarkable with T2-toxin and diacetoxyscirpenol, slight with aflatoxin-B1, penicillic acid and patulin. On the contrary, the butenolide appears as a stimulator of the alcoholic fermentation.

[Toxins of Byssochlamys nivea Westling. I. Preliminary study of toxicity in sheep].

The acute intoxication in 4 sheep, due to an extract from a Byssochlamys nivea culture containing patulin is attained by administering per os with a syringe or with an esophagal tube doses of patulin varying from 15 to 50 mg per kg of bodyweight. A dose of 20 mg/kg, given with a syringe, brought on the death of a sheep within 5 hours following the administration, while a dose of 50 mg/kg given by an esophagal tube, only led to some temporary and slight problems. Intoxication brings on such symptoms such as nasal discharge, the stoppage of ruminating, a painful sensitivity in the retro-sternal

De novo biosynthesis of secondary metabolism enzymes in homogeneous cultures of Penicillium urticae.

The initiation of patulin biosynthesis in submerged batch cultures of Penicillium urticae NRRL 2159A was investigated at the enzyme level. In contrast to earlier studies, this study achieved a clear temporal separation of growing cells devoid of secondary metabolism-specific enzymes from nongrowing cells, which rapidly produce these enzymes. A spore inoculum, silicone-treated flasks, and two new media which supported a rapid, pellet-free, filamentous type of growth were used. In yeast extract-glucose-buffer medium, a marked drop in the specific growth rate (approximately equal to 0.26 h-1) coi

Physiological criteria and mycotoxin production as AIDS in identification of common asymmetric penicillia.

The taxonomy of the asymmetric (predominantly terverticillate) penicillia is based on morphological differences that leave identification difficult. The application of physiological criteria facilitated the identification of the common asymmetric penicillia investigated. Changes in the placement of some strains of these penicillia made the connection to mycotoxin-producing ability clearer. The classical criterion of conidium color was deemphasized and replaced by the following criteria: (i) growth on nitrite-sucrose agar and (ii) growth and acid (and subsequent base) production on creatine-suc

Significance of Mycotoxins to Food Safety and Human Health 1, 2.

Mycotoxins are toxic substances produced by molds, which cause disease in animals or man. Acute diseases caused by mycotoxins are called mycotoxicoses. History has recorded several human disease outbreaks and numerous animal poisonings thought to be mycotoxicoses. The outbreak of Turkey X disease in England in 1960 culminated in the discovery of aflatoxins and the realization that low levels of mold metabolites in foods and feed could cause disease in man and animals. This gave great impetus to the study of mycotoxins. Mycotoxin-producing molds are quite ubiquitous and frequently contaminate f

Species-specific inhibition of human and rat aromatase by mycotoxins: Integrated enzyme assays, docking, and structure-activity relationship analysis.

In placental tissue, human and rat aromatases convert testosterone to estradiol, which was prioritized for investigation over others due to its susceptibility to endocrine disruption by mycotoxins, yet their effects remain unclear. The objective of this study is to investigate the effects of mycotoxins on human and rat aromatase, including inhibition strength, mechanism, and structural-activity relationships (SAR). Screening was conducted to evaluate mycotoxin inhibition using placental microsomes by HPLC-MS/MS. IC50 and Ki values were determined as well as effects on estradiol secretion in hu
