Obesity is a modifiable risk factor for various malignancies and is mechanistically linked to hyperinsulinemia, insulin-like growth factor 1 (IGF-1) activation, adipokine imbalance, mitochondrial oxidative stress, and chronic low-grade inflammation. Tripterygium wilfordii-derived celastrol is a quinone-methide triterpenoid that possesses multiple activities targeting metabolic, inflammatory, and oncogenic signaling networks. The molecular evidence of celastrol at the obesity-cancer interface is critically discussed, particularly focusing on metabolic reprogramming, adipokine signalling, inflam

Tocoyena formosa is a medicinal species used to treat gastrointestinal disorders, which may include infections. This study analyzed the hexane (HETF) and ethanolic (EETF) extracts of T. formosa to identify their chemical composition and antimicrobial potential. Gas chromatography-mass spectrometry revealed that HETF is mainly composed of n-alkanes, especially n-nonacosane (70.02%), while EETF contains pentacyclic triterpenes, such as ursolic acid (72.08%) and oleanolic acid (16.30%). Antimicrobial activity was evaluated against multidrug-resistant bacteria and Candida species. HETF showed acti

α-Synuclein (αSyn) aggregation is associated with Parkinson's disease (PD). The region αSyn36-42 acts as the nucleation 'master controller' and αSyn1-12 as a 'secondary nucleation site'. They drive monomeric αSyn to aggregation. Small molecules targeting these motifs are promising for disease-modifying therapy. Using computational techniques, we screened thirty phytochemicals for αSyn binding. The top three compounds were experimentally validated for their binding affinity. Amongst them, celastrol showed high binding affinity. NMR analysis confirmed stable αSyn-celastrol interactions involving

The increasing global disease burden sustains interest in natural products as sources for therapeutic discovery. Among these, naturally occurring and semi-synthetic oleanane-type triterpenoids have attracted considerable attention because of their broad range of reported bioactivities and their wide distribution in nature. A literature search was conducted across PubMed, Web of Science Core Collection and Google Scholar using keywords "oleanane-type", "oleanolic acid-type" and "triterpenoids". Only studies reporting oleanane- or oleanolic acid-related triterpenoids with fully elucidated chemic

Aggregation of α-synuclein (α-SYN) into amyloid structures is closely linked to progression of Parkinson's disease (PD). Type 2 diabetes mellitus increases PD risk, sharing common pathological features like amyloid aggregation, insulin dysregulation, inflammation, oxidative stress, and mitochondrial dysfunction. Insulin resistance affects over 60% of PD cases, leading to trials of anti-diabetic drugs for potential PD benefits. The monomeric form of α-SYN tends to aggregate in a process that relies on nucleation, ultimately leading to the formation of insoluble fibrils. Recent research indicate

Triterpenoids are one of the largest groups of secondary metabolites and exhibit diverse structures, which are derived from C30 skeletons that are biosynthesized via the isoprenoid pathway by cyclization of 2,3-oxidosqualene. Triterpenoids have a wide range of biological activities, and are used in functional foods, drugs, and as industrial materials. Due to the low content levels in their native plants and limited feasibility and efficiency of chemical synthesis, heterologous biosynthesis of triterpenoids is the most promising strategy. Herein, we classified 121 triterpene alcohols/ketones ac

Rauvolfia mannii is a plant from western and eastern areas of African continent and is widely used in folk medicine for the treatment of various diseases including malaria. Herein, one previously undescribed acylated triterpene (1), together with five already published natural products (2-6) were removed from its roots. The chemical structures of these compounds were determined by spectroscopic and spectrometric means (NMR, HRESIMS, IR and UV). In addition to the isolated triterpenoids, components 5 and 6 are also newly reported from the genus Rauvolfia. Moreover, some constituents were furthe

Lipid metabolism disorders are pivotal in the development of various lipid-related diseases, such as obesity, atherosclerosis, non-alcoholic fatty liver disease, type 2 diabetes, and cancer. Celastrol, a bioactive compound extracted from the Chinese herb Tripterygium wilfordii Hook F, has recently demonstrated potent lipid-regulating abilities and promising therapeutic effects for lipid-related diseases. There is substantial evidence indicating that celastrol can ameliorate lipid metabolism disorders by regulating lipid profiles and related metabolic processes, including lipid synthesis, catab

The epithelial-mesenchymal transition (EMT) is a biological process in which epithelial cells change into mesenchymal cells with fibroblast-like characteristics. EMT plays a crucial role in the progression of fibrosis. Classical inducers associated with the maintenance of EMT, such as TGF-β1, have become targets of several anti-EMT therapeutic strategies. Natural products from the pentacyclic triterpene class have emerged as promising elements in inhibiting EMT. Oleanolic acid and ursolic acid are pentacyclic triterpenoids isolated from a variety of medicinal plants, which have antibacterial activity. Listeria monocytogenes is a Gram-positive facultative pathogen, being the causative agent of listeriosis. The present study was carried out to evaluate the in vitro effect of sub-inhibitory concentrations of both triterpene acids on the pathogenicity determinants of L. monocytogenes: their hemolytic activity and biofilm forming ability. Oleanolic and ursolic acids inhibited listeriolysin O activity without influencing toxin secretion. Biofilm formation,

Small molecule inhibitors of snake venom metalloproteinases (SVMPs) could provide a means to rapidly halt the progression of local tissue damage following viperid snake envenomations. In this study, we examine the ability of candidate compounds based on a pentacyclic triterpene skeleton to inhibit SVMPs. We leverage molecular dynamics simulations to estimate the free energies of the candidate compounds for binding to BaP1, a P-I type SVMP, and compare these results with experimental assays of proteolytic activity inhibition in a homologous enzyme (Batx-I). Both simulation and experiment sugges

Olive leaves, the main byproducts of olive cultivation, are characterized by a plethora of bioactive metabolites with significant nutritional value. Their main pentacyclic triterpenes, Oleanolic Acid (OA) and Maslinic Acid (MA), are two high added-value compounds with remarkable activities. This study aimed to develop an efficient methodology for extracting and purifying OA and MA, utilizing Supercritical Fluid Extraction (SFE) and Centrifugal Partition Chromatography (CPC)-two modern, scalable, and green techniques. A total of 21 g of olive leaves were subjected to SFE using supercritical CO2

Ursane-type pentacyclic triterpenoids exert various biological effects, including anticancer and anti-inflammatory activities. We previously reported that ursolic acid, corosolic acid, and asiatic acid interfered with the intracellular trafficking and glycosylation of intercellular adhesion molecule-1 (ICAM-1) in human lung adenocarcinoma A549 cells stimulated with the pro-inflammatory cytokine interleukin-1α. However, the structure-activity relationship of ursane-type pentacyclic triterpenoids remains unclear. In the present study, the biological activities of seven ursane-type pentacyclic tr

Betulinic acid is a naturally occurring pentacyclic triterpenoid. Betulinic acid has recently been selected by the National Cancer Institute for addition into the RAID (Rapid Access to Intervention in Development) programme. The agent exhibits potential anti-tumour activity and functions in this regard via apoptosis. The objective of the present study was to determine the pharmacokinetics of betulinic acid in CD-1 mice. Serum samples were obtained at designed times after a single 250 or 500 mg/kg intraperitoneal (IP) dose of betulinic acid. Tissue samples (skin, heart, liver, spleen, kidney, l
