Pentobarbital in the context of possible suicides: Analysis of a Case.

Pentobarbital is a barbiturate, acting as a central nervous system depressant (CNS), being used for its anticonvulsant, sedative, hypnotic and anaesthetic properties. Barbiturates were replaced by benzodiazepines, leading to a decrease in poisoning cases with these compounds. However, pentobarbital is still used in many countries as an anaesthetic in veterinary medicine. Due to its properties, this compound is sought after by people who wish to commit suicide, acquiring it on the black market. The authors present an unusual fatal pentobarbital intoxication case, in a 37 years-old male salesper

An unusual case of relay pentobarbital toxicosis in a dog.

Sodium pentobarbital and phenytoin are common constituents of veterinary euthanasia solutions in the United States. Relay, or secondary, barbiturate toxicosis has been reported in carnivorous animals that have fed from the carcasses of euthanized livestock. This case report presents barbiturate toxicosis in a dog. A 2-year-old female spayed Australian shepherd presented comatose 2 h after ingesting an unknown substance on the beach. The material was retrieved from the stomach by gastric lavage and visually identified as fish or other animal tissue. The dog recovered with symptomatic and suppor

Potentiation of pentobarbital hypnosis by Rosa damascena in mice.

Rosa damascena has been found to act on central nervous system including brain. It inhibits the reactivity of the hypothalamous and pituitary systems in rat. In traditional medicine hypnotic effect of Rose is also suggested. In the present study hypnotic effect of ethanolic, aqueous and chloroformic extracts of R. damascena was investigated in mice. Hypnotic method was based on potentiation of pentobarbital induced sleeping time by extracts. Three doses of extracts (100, 500 and 1000 mg/kg) were injected i.p. in comparison with diazepam (3mg/kg) as positive control and saline as negative contr

Pentobarbital desensitization in a 3-month-old child.

Barbiturates, a class of medications commonly used as antiepileptics and sedatives, are known to cause adverse reaction, with the most commonly reported immune-mediated reactions being anticonvulsant hypersensitivity syndrome. Other types of allergic reactions such as immediate hypersensitivity reactions also can occur. We present a 3-month-old child with refractory generalized convulsive status epilepticus who required pentobarbital therapy in the context of phenobarbital sensitivity because of progressive generalized seizures unresponsive to other aggressive therapies. Skin tests to pentobar

Genetic determinants of sensitivity to pentobarbital in inbred mice.

RATIONALE: We postulated that genetic determinants of different responses to pentobarbital (PB) in mice would differ from response to response. OBJECTIVES: Mice from 14 standard inbred strains were tested for sensitivity to several effects of acute PB. METHODS: Strains were tested for sensitivity to PB-induced low-dose stimulation and high-dose depression of locomotor activity, reduced rearing, hypothermia, and ataxia assessed on a rotarod, using four doses of PB or saline. RESULTS: Strains differed in sensitivity to PB for all responses. Correlations among strain means indicated that strai

Human cardiac sodium channels are affected by pentobarbital.

BACKGROUND AND OBJECTIVE: To investigate the response to general anaesthetics of different sodium channel subtypes, we examined the effects of pentobarbital, a close thiopental analogue, on single sodium channels from human ventricular muscle and compared them with existing data from human brain channels. METHODS: Sodium channels from preparations of human ventricular muscle were incorporated into planar lipid bilayers in the presence of batrachotoxin, a sodium channel activator. Single channel currents were recorded in symmetrical 100 mmol L-1 and 500 mmol L-1 NaCl before and after the addit

Effect of chlorpromazine on the pharmacokinetics and pharmacodynamics of pentobarbital in rats.

The effects of chlorpromazine (4 mg/kg i.v.) on the disposition and duration of loss of the righting reflex (LRR, sleeping time) produced by intravenous pentobarbital (5 to 50 mg/kg) were studied in rats. The plasma concentration time profile following i.v. administration of pentobarbital alone was reasonably well described by a three compartment open model with Michaelis-Menten type elimination kinetics. The brain to plasma concentration ratio of pentobarbital was 1.5 and was almost constant during the experiment. Coadministration of chlorpromazine significantly reduced the systemic clearance

Effects of pyrazolopyridines and a triazolopyridazine on the pentobarbital discriminative stimulus.

Rats were trained to discriminate injections of racemic pentobarbital (5.0 mg/kg) from saline in a two-lever drug discrimination task. After stable discrimination performance was attained, stimulus generalization studies were conducted with another barbiturate (barbital), benzodiazepine derivatives (diazepam and chlordiazepoxide), pyrazolopyridine derivatives (etazolate, cartazolate, and tracazolate), and a triazolopyridazine (CL 218, 872). The pentobarbital stimulus generalized to all of these compounds, except cartazolate. In addition, the administration of the benzodiazepine receptor antago

Tolerance to ethanol and cross-tolerance to pentobarbital and barbital.

A chronic regimen of ethanol by intubation, which produced clear tolerance to ethanol-induced hypothermia, ataxia and narcosis, produced only a marginal degree of cross-tolerance to these effects of pentobarbital. The lack of appreciable cross-tolerance to pentobarbital-induced hypothermia and ataxia was also observed over a wide range of test doses. However, cross-tolerance to barbital was observed after chronic treatment with ethanol. Increased rate of drug biotransformation did not contribute significantly to the observed tolerance and cross-tolerance. The difference in the extent of cross-

The effects of pentobarbital and related compounds on frog motoneurons.

The effect of pentobarbital (PB) and related compounds on frog motoneurons was examined with sucrose gap recording from the ventral roots. PB was found to: (1) depress the action of glutamate, (2) selectively enhance the action of GABA, (3) reverse the non-competitive picrotoxin antagonism of GABA and the competitive strychnine antagonism of beta-alanine, but not the competitive bicuculline methiodide antagonism of GABA, and (4) elicit a GABAmimetic hyperpolarization. The first three actions had a threshold concentration of 10 microM, while the GABAmimetic action required a 10-fold higher conc

Pharmacologic response to pentobarbital in passively immunized mice.

Rabbits were actively immunized using a barbiturate--BGG conjugate as the immunogen. The antiserum obtained from actively immunized rabbits was administered intravenously to mice to accomplish passive immunization. The antibody binding capacity for 3H-phenobarbital was shown to be sustained in passively immunized mice for periods of up to three weeks. Serum levels of 3H-phenobarbital in passively immunized mice and control mice were compared following drug administration and found to be altered in the antibody-containing mice. There was a 4-fold higher amount of 3H-phenobarbital present in the

Determination of pentobarbital in serum by electron-capture GLC.

A GLC method was developed for pentobarbital in serum. After extraction from serum, a pentafluorobenzyl derivative was prepared and quantitated by electron-capture detection. The method has a sensitivity of 0.1 microng/ml of serum, and the amount detectable is less than 0.2 ng/injection. Hexethal was used as the internal standard. Derivatives of other barbiturates were also made. NMR and mass spectrometric analyses confirmed the proposed structure of the 1,3-bis(pentafluorobenzyl) derivative of pentobarbital. The procedure was successfully applied to measurement of serum pentobarbital levels i

Pentobarbital elimination in patients with poor renal function.

Drug oxidation is slowed in experimental uremia in animals but has been reported to be normal or accelerated in uremic patients. Eleven normal subjects and 9 uremic patients were each given 100 mg pantobarbital orally. Several blood samples were drawn over a 36-hr period starting on the morning after the dose. Plasma pentobarbital concentration was measured by GLC. The log concentration values were graphed against time elapsed after dose for each patient, and the plasma T/2 and extrapolated value of concentration at the time of drug administration (Co) were determined. The T/2 values in normal

KLF4 Exerts Sedative Effects in Pentobarbital-Treated Mice.

KLF4 is a zinc-finger transcription factor that plays an essential role in many biological processes, including neuroinflammation, neuron regeneration, cell proliferation, and apoptosis. Through effects on these processes, KLF4 has likely roles in Alzheimer's disease, Parkinson's disease, and traumatic brain injury. However, little is known about the role of KLF4 in more immediate behavioral processes that similarly depend upon broad changes in brain excitability, such as the sleep process. Here, behavioral approaches, western blot, and immunohistochemical experiments were used to explore the

Pentobarbital changes compartmental contribution to cerebral blood flow.

Barbiturates were administered to normal dogs, establishing an isoelectric electrocorticogram. Cortical cerebral blood flows (CBF) and deeper CBF's were respectively measured by krypton-85 (85Kr) and xenon-133 (133 Xe). Following barbiturate administration, the two methods of measuring CBF showed a poor coefficient of variation (r=0.12, p less than 0.05). The cortical flows decreased less than the fast compartment flows. A shifting of percentage contribution of flow to the slow compartment (60% increase, p less than 0.001) was observed after barbiturate infusion. A selective shunting of blood

Human exposures to pentobarbital-phenytoin combination veterinary drugs.

A combination of pentobarbital and phenytoin is used as a veterinary euthanasia drug. Because of its lethal effect, this study described pentobarbital-phenytoin combination veterinary drug human exposures reported to Texas poison centers during 2000-2015. Of 66 exposures, 73% involved female and 27% male patients. The distribution by patient age was 3% 0-5 years, 5% 6-19 years, 91% 20+ years, and 2% unknown. The most common routes were ocular (41%), ingestion (32%), injection (23%), and dermal (18%). The exposure reasons were unintentional (77%) and intentional (23%). The exposure site was the

Pentobarbital poisoning in Sumatran tigers (Panthera tigris sumatrae).

Three Sumatran tigers (Panthera tigris sumatrae) at the Heidelberg Zoo in Germany presented with severe neurologic signs. Physical examination and diagnostic tests did not reveal a definitive diagnosis. Two days after initial presenting signs, all of the animals appeared clinically normal. An investigation into this outbreak revealed that all animals received horse meat on the evening before the incident. A toxicologic examination was initiated and serum analysis of the affected female tiger cub and the horse meat revealed contamination with pentobarbital.
