Serum pepsinogen (SP) levels were studied in 100 patients with gastroduodenal lesions, and 100 healthy volunteers. SP levels were significantly elevated in patients with duodenal ulcer (DU) and duodenitis compared to the controls. SP values above 150 ug Tyr/ml/24 hr were highly suggestive of duodenal ulcer disease. Age and sex of patients and controls did not influence SP levels. The mean values of SP in North India were found to be lower in both normal and DU subjects compared to the west.

This study was carried out to elucidate a role of pepsinogen, one of the proteases that currently introduced as a promotor of tumor invasion, on gastric cancer spread. Production of pepsinogen in cancer tissue was analysed by means of three different methods of acid proteolytic activity (APA), polyacrylamide gel electrophoresis (PAGE), and immunoperoxidase staining (IPS), respectively in the 60 resected advanced gastric cancer specimens. Mean level of APA in cancer tissue was higher than that in the control intact muscle layer of the gastric body (p less than 0.05). Through IPS and PAGE, pepsi

A hybridoma monoclonal antibody against human pepsinogen I was used to develop an enzyme-linked immunosorbent assay for pepsinogen I in serum. In the two-step competitive procedure using antimouse immunoglobulin F(ab')2 fragment coupled to alkaline phosphatase, the measurable assay range was 8-256 micrograms/l. No cross-reactivity with rat pepsinogen 1, human pepsinogen II, gastrin I, bombesin, somatostatin and peptide YY was shown. However, there was slight cross-reactivity (0.09%) with porcine pepsinogen. The coefficients of variation within and between series were 7.6% and 13.0%. This enzym

Poly(A)-containing RNA was isolated from total RNA of swine gastric mucosa by oligo(dT)-cellulose chromatography, and was translated in a wheat germ cell-free system. Analysis of the translation products by sodium dodecyl sulfate-polyacrylamide gel electrophoresis revealed that pepsinogen was a major translation product and was synthesized as two different molecular forms with apparent molecular weights of 45,000 and 43,000. The pepsinogen translated in the in vitro translation system had no autocatalytic activity. The pepsinogen mRNA was further purified by sucrose density gradient centrifuga

Hyperpepsinogenaemia has long been considered an important factor in the pathophysiology of duodenal ulcer. Moreover, H. pylori infection has been reported in virtually all duodenal ulcers. OBJECTIVE. To demonstrate the influence of H. pylori eradication on basal pepsinogen I levels in patients with duodenal ulcer. METHODS. 86 patients with endoscopically proven duodenal ulcer were prospectively studied. Different therapeutic regimens were used: Amoxycillin/clavulanate plus omeprazole or ranitidine, triple therapy, omeprazole and ranitidine. At diagnostic endoscopy and after therapy, three bio

1. The purpose of the present study was to analyse the role played by central gamma-aminobutyric acid (GABA-A) receptors in the regulation of gastric basal pepsinogen secretion in anaesthetized rats. 2. The central, but not parenteral, administration of the GABA-A receptor antagonist bicuculline or SR-95531 caused a significant and dose-dependent increase in basal pepsinogen secretion without changes in acid output. The stimulant effect exerted by bicuculline was prevented by atropine or pirenzepine, but not by vagotomy. 3. The central, but not parenteral, administration of the GABA-A receptor

BACKGROUND: Progastricsin/Pepsinogen C (PGC) is an aspartyl endoprotease predominantly expressed by gastric fundic and chief cells, with additional expression reported in select extragastric tissues. While increased PGC expression has been described in several malignancies, progressive downregulation of PGC has been observed along the spectrum of gastric inflammation to carcinoma. However, data on PGC expression in stomach adenocarcinoma (STAD) from South Asian populations remain limited. Given the distinct genetic, dietary, and environmental context of this region, we investigated the express

Serum pepsinogen (PG) testing is widely used as a non-invasive biomarker for gastric atrophy; however, its clinical interpretation varies depending on the severity and distribution of gastritis as well as on the assay methods. Autoimmune gastritis is characterized by markedly decreased PG I levels and PG I/II ratios, along with increased gastrin levels, reflecting profound corpus atrophy. In contrast, Helicobacter pylori-associated atrophy typically shows moderate reductions in PG I levels and PG I/II ratios, with variability according to the extent of antral and corpus involvement. Pepsinogen, secreted from the gastric mucosa, is the precursor of pepsin. It is categorized as pepsinogen 1 and pepsinogen 2 based on its immunogenicity. The pepsinogen content that can enter the blood circulation through the capillaries of the gastric mucosa is approximately 1% and remains stable all the time. The pepsinogen content in serum will change with the pathological changes of gastric mucosa. Therefore, the level of pepsinogen in serum can play a role in serologic biopsy to reflect the function and morphology of different regions of gastric mucosa and serve as an indicator of gastric

BACKGROUND AND AIM: Gastric cancer develops due to atrophic gastritis induced by Helicobacter pylori (H. pylori) infection. Serum levels of pepsinogen (PG) are known to be excellent markers for evaluating the degree of atrophic gastritis. We investigated whether chronic gastritis could be diagnosed by evaluating serum PG levels.
METHODS: A total of 4483 patients (average age, 49.7 years; 2879 men) were included in this study. Fasting serum samples were collected and anti-H. pylori antibody and PG levels were evaluated. We evaluated the endoscopic atrophy grade or histological extent of gastri

Studies have been made on the isozymic composition of pepsinogen (polyacrylamide gel electrophoresis) and peptidase activity in gastric mucosa of the ground squirrel at various stages of hibernation. Eight pepsinogen isoforms were found in acid-producing zone of the mucosa, pyloric mucosa lacks first three of them, its peptidase activity being two times lower than in other parts of the stomach. During hibernation, no significant changes were found in fractional composition of pepsinogen, peptidase activity being decreased twofold only in the fundulus of the stomach, remaining practically const

The human stomach mucosa contains two main group of gastric proteinases. Both pepsinogen A (PGA) and pepsinogen C (PGC) consist of molecular variants, isozymogens, which can be separated electrophoretically, PGA was found to consist of five isozymogens (Pg1-Pg2), and PGC of two isozymogens (Pg6 and Pg7). Five hundred zymograms were examined and electrophoretic mobility of pepsinogens from patients with gastric cancer was found to be higher than from other gastric diseases. The ratio of isozymogens Pg3 to Pg5 differs in to great extent in various disease. Patients with ulcer disease have this v
