[Peptide hormones LH, FSH, TSH, prolactin, beta-HCG and PTH in patients with urogenital tumors].

Serum levels of the peptide hormones, LH, FSH, TSH, prolactin, beta-HCG and PTH are observed to significantly modulate in patients with urogenital tumors, beta-HCG appears elevated in patients with chorioncarcinoma, teratoma and embryonal carcinoma and is shown to increase serum prolactin. TSH and prolactin are measured in pathological range in patients with renal carcinomas. Following nephrectomy serum TSH persists in high levels only in patients with advanced diseases, while PTH is more frequently suppressed in these patients. Serum LH and prolactin show a more distinct change of values in i

Targeted expression of constitutively active receptors for parathyroid hormone and parathyroid hormone-related peptide delays endochondral bone formation and rescues mice that lack parathyroid hormone-related peptide.

Mice in which the genes encoding the parathyroid hormone (PTH)-related peptide (PTHrP) or the PTH/PTHrP receptor have been ablated by homologous recombination show skeletal dysplasia due to accelerated endochondral bone formation, and die at birth or in utero, respectively. Skeletal abnormalities due to decelerated chondrocyte maturation are observed in transgenic mice where PTHrP expression is targeted to the growth plate, and in patients with Jansen metaphyseal chondrodysplasia, a rare genetic disorder caused by constitutively active PTH/PTHrP receptors. These and other findings thus indicat

A helical peptide antagonist of the human growth hormone receptor.

The binding of human growth hormone (hGH) to the human growth hormone receptor (hGHR) is a key endocrinological process that controls critical aspects of cell growth, proliferation, and differentiation. Mechanistically, this sequential, asymmetric binding event involves the interaction between a single hGH molecule and distinct sites (sites 1 and 2) on the extracellular domain of a preformed hGHR homodimer. Our group recently identified S1H, a rationally designed peptide sequence mimetic of the hGH site 1-binding helix (residues 36-51) that disrupts the hGH-hGHR interaction and inhibits hGH-me

Salmon cardiac natriuretic peptide is a volume-regulating hormone.

The present study tested the hypothesis that salmon cardiac peptide (sCP), a new member of the family of natriuretic peptides, has an important role in the regulation of fluid balance and cardiovascular function. Intra-arterial administration of sCP increased urine output in salmon. It had a diuretic effect in rat as well, but the potency was lower. sCP increased the sodium excretion in proportion to the increased urine flow. Blood pressure was not affected by sCP in either species. Acute volume expansion elevated the plasma level of sCP in salmon, and an acute transfer of salmon from fresh to

Contribution of parathyroid hormone-related peptide to the evaluation of hypercalcemia.

Hypercalcemia of malignancy is due either to local osteolysis at the site of bone metastases or to production by the malignancy of parathyroid hormone-related peptide, which shares some of the effects of parathyroid hormone. We used a radioimmunoassay (antiserum specific to the amino-terminus) to measure serum parathyroid hormone-related peptide levels in controls (n = 61), chronic renal failure patients (n = 10), patients with primary hyperparathyroidism (n = 19), cancer patients with (n = 35) or without (n = 57) hypercalcemia and/or bone metastases (n = 53 and n = 39, respectively), and pati

[Parathyroid hormone related peptide (PTHrP) and bone metabolism].

The discovery of PTHrP was the result of research on the mechanisms by which some cancers cause hypercalcemia (humoral hypercalcemia of malignancy) without necessarily metastasizing to bone. PTHrP is also present in various normal adult and fetal tissues. Its concentration is normally very low (picomolar) in blood, but it is more abundant in milk (nanomolar concentration). PTHrP seems able to exert autocrine/paracrine as well as endocrine effects on bone metabolism. A major role for PTHrP in regulation of fetal bone metabolism has been demonstrated in mice. Homologous recombination has been us

Thyrotrophin releasing hormone and related peptides in canine tissues.

Thyrotrophin releasing hormone (TRH)-immunoreactive peptides have been quantified in canine serum, hypothalamus, liver, pancreas, adrenal, thyroid, prostate, testis, epididymis and semen by TRH radioimmunoassay, SP-Sephadex C-25 cation exchange chromatography, Sephadex G-10 exclusion chromatography and high pressure liquid chromatography. The total concentration of TRH and TRH-like peptides was highest in the hypothalamus, followed by liver, adrenal, pancreas, thyroid, prostate, epididymis, testis and serum. All of the TRH immunoreactivity (TRH-IR) within extracts of the hypothalamus was due t

Quinacrine accumulates in certain peptide hormone-producing cells.

Quinacrine is a fluorescent anti-malarial acridine derivative which binds selectively to a population of nerves, presumably peptidergic, and to certain peptide hormone-producing cells. Among these cells are glycopeptide hormone-producing cells in the adenohypophysis, the calcitonin cells in the thyroid, the insulin, glucagon and PP cells in the pancreatic islets, and the gastrin cells in the pyloric antrum. Available evidence suggests that the fluorophore accumulates in the secretory granules. The half-life of the fluorescence varies from one cell type to another, from 6 h in the gastrin cells

Calcemic fraction-A: biosynthetic peptide precursor of parathyroid hormone.

Calcemic fraction-A (CF-A) is a biologically active, hypercalcemic and bone resorptive peptide, which was detected in, and isolated from, bovine parathyroid glands [Hamilton et al. (1971) Endocrinology 89, 1440-1447]. It has been further purified, and its relationship to parathyroid hormone clarified. The peptide is present in fresh glands at a concentration of about 3 mug/g (parathyroid hormone, 100 mug/g). It contains 109 amino acids (hormone, 84), each of which is present in equal or greater molar ratio than in the hormone. Its molecular weight, calculated from amino-acid composition, is 12

Peptide hormone sensors using human hormone receptor-carrying nanovesicles and graphene FETs.

Hormones within very low levels regulate and control the activity of specific cells and organs of the human body. Hormone imbalance can cause many diseases. Therefore, hormone detection tools have been developed, particularly over the last decade. Peptide hormones have a short half-life, so it is important to detect them within a short time. In this study, we report two types of peptide hormone sensors using human hormone receptor-carrying nanovesicles and graphene field-effect transistors (FETs). Parathyroid hormone (PTH) and glucagon (GCG) are peptide hormones present in human blood that act

Parathyroid hormone/parathyroid hormone-related peptide type 1 receptor in human bone.

The parathyroid hormone/parathyroid hormone-related peptide (PTH/PTHrP) receptor (denoted as PTH-1R) is a key signaling factor through which calcium-regulating hormones PTH and PTHrP exert their effects on bone. There are contradictory reports regarding the capability of osteoclasts to express PTH-1R. To address this issue in humans, bone biopsy specimen samples from 9 normal controls and 16 patients with moderate to severe secondary renal hyperparathyroid bone disease (2 degrees HPT) with elevated PTH levels were studied to determine whether osteoclasts in the bone microenvironment express PT

C-peptide in diabetes: A player in a dual hormone disorder?

C-peptide, a byproduct of insulin synthesis believed to be biologically inert, is emerging as a multifunctional molecule. C-peptide serves an anti-inflammatory and anti-atherogenic role in type 1 diabetes mellitus (T1DM) and early T2DM. C-peptide protects endothelial cells by activating AMP-activated protein kinase α, thus suppressing the activity of NAD(P)H oxidase activity and reducing reactive oxygen species (ROS) generation. It also prevents apoptosis by regulating hyperglycemia-induced p53 upregulation and mitochondrial adaptor p66shc overactivation, as well as reducing caspase-3 activity

Parathyroid hormone and its related peptides in bone metabolism.

Parathyroid hormone (PTH) is an 84-amino-acid peptide hormone that is secreted by the parathyroid gland. It has different administration modes in bone tissue through which it promotes bone formation (intermittent administration) and bone resorption (continuous administration) and has great potential for application in sbone defect repair. PTH regulates bone metabolism by binding to PTH1R. PTH plays an osteogenic role by acting directly on mesenchymal stem cells, cells with an osteoblastic lineage, osteocytes, and T cells. It also participates as an osteoclast by indirectly acting on osteoclast

Hepcidin: a novel peptide hormone regulating iron metabolism.

BACKGROUND: Hepcidin is a low-molecular weight hepatic peptide regulating iron homeostasis. Hepcidin inhibits the cellular efflux of iron by binding to, and inducing the internalization and degradation of, ferroportin, the exclusive iron exporter in iron-transporting cells. It has been recently recognized as a main hormone behind anemia of chronic disease. METHOD: A comprehensive literature search was conducted from the websites of Pubmed Central, the US National Library of Medicine's digital archive of life sciences literature (http://www.pubmedcentral.nih.gov/) and the National Library of M

Parathyroid hormone and related peptides for the treatment of postmenopausal osteoporosis.

Osteopenia, a skeletal condition characterised by bone loss, affects over 10% of the North American population, with a worldwide incidence of 200 m. In the absence of treatment, osteopenia usually progresses to osteoporosis, characterised by more severe bone loss and, ultimately, by fractures. In the US alone osteoporosis affects more than 25 m people, and causes more than 1.3 m fractures a year. This skeletal deterioration is accelerated subsequent to menopause and thus can affect women at a relatively young age. Treatment of postmenopausal osteoporosis primarily involves the use of hormones

Physiological and pathological roles of parathyroid hormone-related peptide.

Parathyroid hormone-related peptide was originally identified as a tumor-produced factor that mediated malignancy-associated hypercalcemia by binding to the common parathyroid hormone/parathyroid hormone-related peptide receptor to stimulate osteoclastic bone resorption and renal tubular resorption of calcium. Its role as a humoral factor in hypercalcemia of malignancy is well established, and recent work has demonstrated its importance as a tumor-produced factor in the pathogenesis of bone metastasis. Besides these cancer-related functions, work in the past decade has clearly established that

Opioid peptides, adrenocorticotrophic hormone, and idiopathic (orthostatic) edema.

The effect of dextroamphetamine sulfate (Dexedrine) on plasma opioid peptides, hormones, and other metabolites was studied in eight female subjects with idiopathic (orthostatic) edema and five healthy females. All subjects were given 20 mg of dextroamphetamine sulfate, a drug widely used in the treatment of this disorder, and blood samples were collected before and 30, 60, and 90 minutes after treatment. Patients with idiopathic (orthostatic) edema had significantly lower plasma sodium levels but higher blood urea nitrogen, aldosterone, and renin levels. D-amphetamine decreased aldosterone and

Atrial natriuretic peptide: a hormone with implications for clinical practice.

The heart functions as an endocrine organ, releasing atrial natriuretic peptide (ANP), a hormone, in response to sodium and fluid overload. Specifically, ANP is released by cardiac myocytes in response to atrial distension. As a hormone, ANP has far-reaching multiorgan effects. The body systems affected include the cardiovascular, renal, neural, gastrointestinal, and endocrine systems. However, the main effects of this hormone are on the renin-angiotensin-aldosterone system. ANP acts to oppose this system by causing vasorelaxation, blocking the secretion and sodium-retaining effects of aldoste

Thyroid hormones and pericardial effusion may influence plasma levels of atrial natriuretic peptide (ANP) in humans.

Fluid and electrolyte homeostasis is impaired in patients suffering from hypothyroidism and myxedema because myxedema induces retention of salt and water. We have measured plasma levels of human atrial natriuretic peptide (hANP) in 8 female patients who had been totally thyroidectomized because of thyroid carcinoma. Estimations of the hormone were done 4 weeks after diagnostic withdrawal (searching for iodine retaining metastases) and after 2 weeks and 4 weeks of reinitiation of thyroid suppressive therapy by L-thyroxine. hANP levels, although within the normal range (10-80 ng/l) throughout th
