A Rosella-PLIN2 knock-in mouse reveals lipophagy and immunometabolic interplay in atherosclerosis.

Cytosolic lipid droplets (LDs) regulate lipid homeostasis, with abnormal LD dynamics linked to metabolic diseases like atherosclerosis. In macrophage foam cells, LDs undergo autophagic degradation via lipophagy, but the extent of this process in vascular smooth muscle cell (VSMC) foam cells remains unclear. To track lipophagy in real time, we developed a Rosella-PLIN2 (perilipin 2) biosensor by tagging PLIN2 with the fluorescent pH-biosensor Rosella. We show that proatherogenic lipoproteins and autophagy activators stimulate lipophagy in human macrophages. Targeting LDs with an LC3 fusion prot

The role of TP53INP2 as an adaptor protein in the regulation of lipophagy in mature adipocytes.

AIM: Lipid droplet (LD) accumulation is caused by an imbalance between energy intake and expenditure, leading to adipose tissue dysfunction, obesity, and other metabolic disorders. Lipophagy is a selective autophagy process responsible for LD degradation. However, no specific adaptor proteins have been identified as regulators of lipophagy. This study investigates the role of TP53INP2 as a potential adaptor protein in the regulation of the lipophagy process. METHODS: Murine 3T3L1 cells were subjected to starvation treatment to stimulate autophagy, followed by gene and protein expression analy

Identification of shared pathophysiological molecules of major psychiatric disorders: A comprehensive analysis of serum immune complex antigens before and after electroconvulsive therapy.

Recent studies indicate common inflammatory findings have been identified in peripheral blood in patients with major psychiatric disorders, including schizophrenia (SCZ), bipolar disorder (BD), and major depressive disorder (MDD). Electroconvulsive therapy (ECT) frequently improves both severe symptoms and inflammatory markers in these conditions. However, the shared inflammatory mechanisms underlying these disorders, and thus, reliable biomarkers remain unclear. We hypothesized that the activation of immune complexes (ICs) contributes to inflammatory pathogenesis of these disorders. Using imm

Antiobesity activity of a sphingosine 1-phosphate analogue FTY720 observed in adipocytes and obese mouse model.

Higher levels of body fat are associated with an increased risk for development numerous adverse health conditions. FTY720 is an immune modulator and a synthetic analogue of sphingosine 1-phosphate (S1P), activated S1P receptors and is effective in experimental models of transplantation and autoimmunity. Whereas immune modulation by FTY720 has been extensively studied, other actions of FTY720 are not well understood. Here we describe a novel role of FTY720 in the prevention of obesity, involving the regulation of adipogenesis and lipolysis in vivo and in vitro. Male C57B/6J mice were fed a sta

Myocardial Fibrosis and Steatosis in Patients with Aortic Stenosis: Roles of Myostatin and Ceramides.

Aortic stenosis (AS) involves progressive valve obstruction and a remodeling response of the left ventriculum (LV) with systolic and diastolic dysfunction. The roles of interstitial fibrosis and myocardial steatosis in LV dysfunction in AS have not been completely characterized. We enrolled 31 patients (19 women and 12 men) with severe AS undergoing elective aortic valve replacement. The subjects were clinically evaluated, and transthoracic echocardiography was performed pre-surgery. LV septal biopsies were obtained to assess fibrosis and apoptosis and fat deposition in myocytes (perilipin 5 (

Long-term exercise increases the DNA binding activity of peroxisome proliferator-activated receptor gamma in rat adipose tissue.

The aim of the present study was to examine the effect of 8 weeks of voluntary wheel running on the gene expression, at the protein level, of 2 enzymes involved in lipogenesis (fatty acid synthase [FAS] and diacylglycerol acyl transferase 1), 2 proteins involved in lipolysis (hormone-sensitive lipase [HSL] and perilipin), and 3 transcription factors mediating the induction of genes involved in lipid metabolism (the alpha, gamma, and delta members of the peroxisome proliferator-activated receptor, or PPAR, family) in rat liver, gastrocnemius muscle, epididymal fat, and subcutaneous fat. Aggressive multiple myeloma with lymph node involvement, loss of CD138, and adipophilin-positive cytoplasmic vacuolization: a case report.

We describe a rare and aggressive case of multiple myeloma (MM) characterized by extensive lymph node involvement, loss of CD138 expression, and adipophilin (ADP)-positive cytoplasmic vacuolization, highlighting the role of lipid metabolism in disease aggressiveness. An 83-year-old woman presented with painless cervical lymphadenopathy and widespread osteolytic lesions. Bone marrow examination confirmed MM, while lymph node biopsy showed diffuse infiltration of atypical lymphoid cells with numerous tingible body macrophages, initially mimicking a high-grade lymphoma. Immunophenotyping showed C

Adipocytes in Aortic Stenosis: Association With Clinical and Morphological Indices.

Recently, great attention has been given to understanding the new pathogenetic mechanisms underlying aortic stenosis (AS). The study aims to understand the role of mature adipocytes in AS and their association with histologic, clinical, and echocardiographic data, an area previously overlooked in AS research. We enrolled 25 patients (15 women and 10 men) with severe AS undergoing elective aortic valve replacement. Each patient underwent clinical and transthoracic echocardiographic evaluations before surgery. We obtained AS valves and left ventricular (LV) septal biopsies to assess the presence

Thyroid hormone promotes lipid droplet remodeling and lipophagy through KAT2B/PCAF-dependent histone acetylation in preimplantation embryos.

Thyroid hormones are central regulators of metabolic homeostasis and developmental programming. The active hormone triiodothyronine (T3) modulates transcription through nuclear receptors that recruit epigenetic cofactors to remodel chromatin and regulate metabolic gene networks. Although thyroid hormone signaling is known to influence lipid metabolism, whether it coordinates lipid droplet turnover with autophagy-related pathways during early embryonic development remains largely unknown. Here, transcriptomic profiling revealed distinct metabolic signatures between in vivo and in vitro embryos,

Endomyocardial proteomic signature corresponding to the response of patients with dilated cardiomyopathy to immunoadsorption therapy.

UNLABELLED: Dilated cardiomyopathy (DCM) is a disease of the myocardium with reduced left ventricular ejection fraction (LVEF). Cardiac autoantibodies (AAbs) play a causal role in the development and progression of DCM. Removal of AAbs using immunoadsorption (IA/IgG) has been shown as a therapeutic option to improve cardiac function. However, the response to therapy differs significantly among patients. The reasons for this variability are not completely understood. Hitherto, no potential biomarker is available to predict improvement of cardiac function after therapy accurately. This shotgun p

Osteocytes secrete adiponectin and display adipocyte-like phenotype under control of PPARG nuclear receptor.

Osteocytes and adipocytes represent cells with disparate functions. Osteocytes regulate bone metabolism (remodeling) and bone homeostasis, while adipocytes regulate energy metabolism and energy storage. Here, we demonstrate that osteocyte phenotype consists of adipocytic features which are under control of peroxisome proliferator-activated receptor gamma (PPARG), a master regulator of adipocyte differentiation and function. Using a mouse model with osteocyte-specific deletion of PPARG (OTγKO) and osteocyte cellular model of MLO-Y4 cells edited with CRISPR/Cas9 for PPARG deficiency, we are demo

Time course of histomorphological changes in adipose tissue upon acute lipoatrophy.

BACKGROUND AND AIMS: Crown-like structures (CLS) are characteristic histopathology features of inflamed adipose tissues in obese mice and humans. In previous work, we suggested that these cells derived from macrophages primarily involved in the reabsorption of dead adipocytes. Here, we used a well-characterized transgenic mouse model in which the death of adipocytes in adult mice is inducible and highly synchronized. In this "FAT ATTAC" model, apoptosis is induced through forced dimerization of a caspase-8 fusion protein. METHODS AND RESULTS: 0, 0.5, 1, 2, 3 and 10 days post induction of adip

Lipases and lipid droplet-associated protein expression in subcutaneous white adipose tissue of cachectic patients with cancer.

BACKGROUND: Cancer cachexia is a multifactorial metabolic syndrome characterized by marked loss of adipose tissue and skeletal muscle. Fat loss from adipose tissue in cancer cachexia is partly the result of increased lipolysis. Despite the growing amount of studies focused on elucidating the mechanisms through which lipolysis-related proteins regulate the lipolytic process, there are scarce data concerning that profile in the adipose tissue of cancer cachectic patients. Considering its fundamental importance, it was our main purpose to characterize the expression of the lipolysis-related prote

Cytosolic lipid droplets: from mechanisms of fat storage to disease.

The lipid droplet (LD) is a phylogenetically conserved organelle. In eukaryotes, it is born from the endoplasmic reticulum, but unlike its parent organelle, LDs are the only known cytosolic organelles that are micellar in structure. LDs are implicated in numerous physiological and pathophysiological functions. Many aspects of the LD has captured the attention of diverse scientists alike and has recently led to an explosion in information on the LD biogenesis, expansion and fusion, identification of LD proteomes and diseases associated with LD biology. This review will provide a brief history o

The perilipin homologue, lipid storage droplet 2, regulates sleep homeostasis and prevents learning impairments following sleep loss.

Extended periods of waking result in physiological impairments in humans, rats, and flies. Sleep homeostasis, the increase in sleep observed following sleep loss, is believed to counter the negative effects of prolonged waking by restoring vital biological processes that are degraded during sleep deprivation. Sleep homeostasis, as with other behaviors, is influenced by both genes and environment. We report here that during periods of starvation, flies remain spontaneously awake but, in contrast to sleep deprivation, do not accrue any of the negative consequences of prolonged waking. Specifical

Iron regulates lipid droplet formation in hepatocytes via heme oxygenase-1 mediated ferroptosis.

BACKGROUND: Iron overload has been implicated in the disruption of hepatic lipid metabolism, potentially contributing to non-alcoholic fatty liver disease and other hepatic disorders. However, the underlying mechanisms connecting iron overload to lipid metabolism dysregulation remain elusive. This study aimed to investigate the effect of iron overload on lipid droplet formation, and to explore the regulatory mechanism of iron overload on lipid metabolism through the lens of ferroptosis. METHODS: Iron overload and ferroptosis models were established by treating AML12 mouse hepatocytes with fer
