Toxicological interactions of pesticide mixtures: an update.

Pesticides can interact with each other in various ways according to the compound itself and its chemical family, the dose and the targeted organs, leading to various effects. The term interaction means situations where some or all individual components of a mixture influence each other's toxicity and the joint effects may deviate from the additive predictions. The various mixture effects can be greatly determined by toxicokinetic and toxicodynamic factors involving metabolic pathways and cellular or molecular targets of individual pesticides, respectively. However, the complexity of toxicolog

Novel insecticide resistance mutations associated with variable PBO synergy in Anopheles gambiae s.l. from the Democratic Republic of Congo.

Over-reliance on pyrethroid insecticides in insecticide-treated nets (ITNs) has imposed significant selection pressure for the evolution of insecticide resistance among major malaria vector species. In the Democratic Republic of Congo (DRC), the country with the second highest malaria burden globally, pyrethroid resistance is pervasive, but there is a paucity of information regarding the molecular mechanisms driving resistance. A clear understanding of the specificity of resistance mechanisms to individual insecticides and the likelihood of selecting for cross-resistance mechanisms is crucial

Joint acute toxicity of the herbicide butachlor and three insecticides to the terrestrial earthworm, Eisenia fetida.

The herbicide butachlor and three insecticides phoxim, chlorpyrifos, and lambda-cyhalotrhin are widely used pesticides with different modes of action. As most previous laboratory bioassays for these pesticides have been conducted solely based on acute tests with a single compound, only limited information is available on the possible combined toxicity of these common chemicals to soil organisms. In this study, we evaluated their mixture toxicity on the terrestrial earthworm, Eisenia fetida, with binary, ternary, and quaternary mixtures. Two different types of bioassays were employed in our wor

[Pyrethroid resistance mechanisms in the body lice Pediculus humanus humanus L.: detoxification enzyme systems].

Synergists (piperonyl butoxide, MGK-264 (N-octyl bicycloheptene dicarboximide), DEF (S, S, S, tributyl phosphorotrithioate), and diethylmaleate) were used to investigate the metabolic resistance of body lice to permethrin. Instances of the synergistic activity ofpermethrin when combined with enzyme system inhibitors have been identified. The synergic ratios were 3.3 to 52.0 for monooxygenase inhibitors; 5.2 to 7.4 for esterases, and about 4 for glutathione-S-transferases. Since the use of the synergists did not lead to full inhibition of resistance, it was concluded that the lice had also a kd

Pesticide mixtures show potential toxicity to aquatic life in U.S. streams, water years 2013-2017.

During water years (WY) 2013-2017, the U.S. Geological Survey, National Water-Quality Assessment (NAWQA) Project, sampled the National Water Quality Network - Rivers and Streams (NWQN) year-round and reported on 221 pesticides at 72 sites across the United States in agricultural, developed, and mixed land use watersheds. The Pesticide Toxicity Index (PTI) was used to estimate the potential chronic and acute toxicity to three taxonomic groups - fish, cladocerans, and benthic invertebrates. For invertebrates (either cladocerans, benthic invertebrates, or both), the maximum PTI score exceeded the

Pesticides in Honeybee Products-Determination of Pesticides in Bee Pollen, Propolis, and Royal Jelly from Polish Apiary.

The bioaccumulation of pesticides in honeybee products (HBPs) should be studied for a number of reasons. The presence of pesticides in HBPs can provide new data on the risk related to the use of pesticides and their role in bee colony losses. Moreover, the degree of contamination of HBPs can lower their quality, weaken their beneficial properties, and, in consequence, may endanger human health. The aim of this study was to quantify a broad range of pesticide residues in three different HBPs-bee pollen, propolis, and royal jelly. Samples were collected in the years 2017-2019 from the apiary in

The quest for metabolic biomarkers of agrochemicals exposure via in vitro studies and suspect screening.

Numerous agrochemicals, including pesticides and herbicides, are applied in modern agriculture, resulting in concerns for the ecosystem and human safety as humans are easily exposed to these compounds. Many agrochemicals, and their transformation products or metabolites, have shown toxicity in in vitro and in vivo studies. However, given the rapid development of novel agrochemicals, for many there is no information about their effects nor about metabolic transformations when ingested by humans. Tracing biomarkers may be the best method for assessing the impacts of agrochemicals. A combination

Pesticide-Virus Interactions in Honey Bees: Challenges and Opportunities for Understanding Drivers of Bee Declines.

Honey bees are key agricultural pollinators, but beekeepers continually suffer high annual colony losses owing to a number of environmental stressors, including inadequate nutrition, pressures from parasites and pathogens, and exposure to a wide variety of pesticides. In this review, we examine how two such stressors, pesticides and viruses, may interact in additive or synergistic ways to affect honey bee health. Despite what appears to be a straightforward comparison, there is a dearth of studies examining this issue likely owing to the complexity of such interactions. Such complexities inclu

Caffeine and related methylxanthines: possible naturally occurring pesticides.

Natural and synthetic methylxanthines inhibit insect feeding and are pesticidal at concentrations known to occur in plants. These effects are due primarily to inhibition of phosphodiesterase activity and to an increase in intracellular cyclic adenosine monophosphate. At lower concentrations, methylxanthines are potent synergists of other pesticides known to activate adenylate cyclase in insects. These data suggest that methylxanthines may function as natural insecticides and that phosphodiesterase inhibitors, alone or in combination with other compounds, may be useful in insect control.

In Vitro Evaluation of the Combined Toxicity of Pirimiphos-methyl and Piperonyl Butoxide.

Pirimiphos-methyl (PMM) and piperonyl butoxide (PBO) are commonly used in crops to combat pests. This study examined their cytotoxicity and genotoxicity in various cell lines (RAW 264-7, H9C2, HepG2, HEK 293) across different exposure times. HepG2 cells showed heightened sensitivity to the combination (MIX) at 100 µg/mL after 48 and 72 hours, with no genotoxic effects observed in both HepG2 and mouse whole blood cells. HEK 293 cells experienced cytotoxicity at higher concentrations of PMM, PBO, and MIX (50 and 100 µg/mL) after 48 and 72 hours. H9C2 cells exhibited the most pronounced cytotoxic

Differential effects of dual and synergist-based insecticide-treated bed nets on pyrethroid resistance and L995F/S knockdown resistance mutation dynamics in Anopheles gambiae s.l. populations in south-western Burkina Faso.

BACKGROUND: The introduction of next-generation insecticide-treated nets (ITNs) in Burkina Faso aims to mitigate pyrethroid resistance in malaria vectors. This study evaluated the impact of different ITN types on phenotypic resistance and kdr mutation frequencies in Anopheles gambiae sensus lacto (s.l.) populations across three health districts over 3 years. METHODS: Annual mosquito collections were conducted in Banfora (where pyrethroid-chlorfenapyr nets had been distributed), Gaoua (pyrethroid-only ITNs) and Orodara (pyrethroid-piperonyl butoxide [PBO] ITNs). Two populations were analysed:

Impacts of warming and acidification on pesticide toxicity in continental aquatic environments: A scientometric and systematic map.

Carbon dioxide emissions are altering aquatic ecosystems by causing water acidification and temperature increases, and these environments are also facing pesticide contamination. We present a scientometric and systematic map of these impacts in continental aquatic environments, aiming to provide an overview of research investigating the effects of temperature and acidification on pesticide toxicity. Our findings reveal a significant increase in research output on this topic, especially over the past seven years, with the United States leading due to high pesticide use and rigorous environmenta

Methylenedioxyphenyl insecticide synergists as potential human health hazards.

The effects of methylenedioxyphenyl insecticide synergists on human health arise, not from their intrinsic toxicity which is relatively low, but from their dual effect on the oxidative detoxication or intoxication processes of the body. They are able to inhibit these reactions by two mechanisms. They are substrates for mixed function oxidation and, thus, compete with other xenobiotics for available enzyme, and an intermediate in their metabolism is able to bind with cytochrome P-450 to form an inactive complex which absorbs maximally at 455 nm. In addition, they are able to activate xenobiotic
