Transmission of border disease virus from a persistently infected calf to seronegative heifers in early pregnancy.

BACKGROUND: This study describes the transmission of border disease virus (BDV) from a persistently infected calf to seronegative heifers in early pregnancy, resulting in persistently infected fetuses. On day 50 of pregnancy (= day 0 of the infection phase), six heifers were co-housed in a free stall with a bull calf persistently infected with BDV (pi BVD) for 60 days. The heifers underwent daily clinical examination, and blood samples were collected regularly for detection of pestiviral RNA and anti-pestivirus antibodies. After day 60 (= day 110 of pregnancy), the heifers were slaughtered, an

First detection of atypical porcine pestivirus in piglets affected by congenital tremor in Poland.

Congenital tremors are neurological disorders of newborn piglets that may lead to serious health consequences, including increased mortality. Many reports have indicated that atypical porcine pestivirus (APPV), first identified in 2015, is a possible cause of this condition. In different countries, APPV was detected in farms affected by piglets' congenital tremors. There is a lack of data regarding the presence of this virus in Poland. However, swine veterinarians report outbreaks of congenital tremor with unestablished aetiology nationwide. Therefore, this study aimed to verify the presence o

Development and Evaluation of Antigen-Specific Dual Matrix Pestivirus K ELISAs Using Longitudinal Known Infectious Status Samples.

Pestivirus K, commonly known as atypical porcine pestivirus (APPV), is the most common cause of congenital tremor (CT) in pigs. Currently, there is limited information on the infection dynamics of and immune response against APPV and no robust serologic assay to assess the effectiveness of preventative measures. To that end, known infection status samples were generated using experimental inoculation of cesarean-derived, colostrum-deprived pigs. Pigs (2 per pen) were inoculated with minimum essential medium (n = 6; negative control) or APPV (n = 16). Serum, pen-based oral fluid samples, and na

Detection and genetic analysis of a novel atypical porcine pestivirus from piglets with congenital tremor in Japan.

Atypical porcine pestivirus (APPV), which has been confirmed to be associated with congenital tremor (CT) in pigs, is a newly discovered porcine virus that has been found in the Americas, Europe and Asia; however, no report of APPV in Japan has been published. We identified an APPV in the central nervous system of Japanese piglets with CT and firstly determined and analysed the complete genome sequence. Phylogenetic analysis using the complete genome nucleotide sequence of the Japanese APPV, named Anna/2020, and those of APPVs from the NCBI database showed that APPVs were divided into three ge

Genetic characterization of atypical porcine pestivirus (APPV) in China and the successful isolation of a novel APPV strain within genotype 2.

Atypical porcine pestivirus (APPV) is one major causative agent of congenital tremor (CT) type A-II. In this study, the geographical distribution and genetic characteristics of APPV strains in China were established. To date, APPV has been detected in at least ten provinces/regions of China. Genetic variability analysis showed that NS3 genes were highly conserved among Chinese APPV strains, while NS5A-, Npro-, and Erns-encoding genes were highly variable. Phylogenetic analysis revealed that all of the reference strains could be classified into 3 genotypes (1-3) and 7 subgenotypes (1.1-1.7). Th

A review of congenital tremor type A-II in piglets.

Congenital tremor (CT) is a neurological disease that affects new-born piglets. It was described in 1922 and six different forms, designated type AI-V and type B, are described based on the causative agents, as well as specific histological findings in the central nervous system (CNS). The various forms present with identical clinical signs consisting of mild to severe tremor of the head and body, sometimes complicated with ataxia. By definition, all A-forms have hypomyelination of the CNS, whereas there are no histopathological lesions with the B-form. The cause of the A-II form was long unkn

Detection of Phocoena Pestivirus and Neutralizing Antibodies in Harbor Porpoises Found Dead at the North Sea and Baltic Sea Coasts.

In 2019, a marine pestivirus, Phocoena pestivirus (PhoPeV), family Pestiviridae, was described for the first time. To date, the impact of PhoPeV infections on harbor porpoises is still unknown. Therefore, a screening study was carried out on harbor porpoises found dead on the German North Sea and Baltic Sea coasts during 2021-2025. Analysis of organ samples collected from 84 harbor porpoises by reverse transcription (RT)-PCR showed the presence of PhoPeV RNA in samples from five animals (5.9%). Infectious virus was isolated from two of these samples. Phylogenetic analysis showed a close relati

Genome characteristics of atypical porcine pestivirus from abortion cases in Shandong Province, China.

BACKGROUND: Atypical porcine pestivirus (APPV) is a novel, highly variable porcine pestivirus. Previous reports have suggested that the virus is associated with congenital tremor (CT) type A-II in piglets, and little information is available about the correlation between the virus and sow abortion, or on coinfection with other viruses. In China, reported APPV strains were mainly isolated from South China and Central China, and data about the APPV genome from northern China are relatively scarce. METHODS: Eleven umbilical cords, one placenta, and one aborted piglet, were collected from aborted

Isolation and Full-Length Sequence Analysis of a Pestivirus from Aborted Lamb Fetuses in Italy.

Pestiviruses are distributed worldwide and are responsible for a variety of economically important diseases. They are not very host-specific, and thus sheep can be infected by well-known pestiviruses like bovine viral diarrhea virus (BVDV) and border disease virus (BDV), as well as by other recently discovered pestivirus species. The aim of this study is to describe the isolation and characterization of four pestivirus strains detected in aborted lamb fetuses from a single farm in the Brescia province (Northern Italy). A total of twelve aborted fetuses were collected and examined. After necrop

Short communication: Transmission of border disease virus to seronegative cows inseminated with infected semen.

The goal of this study was to investigate the transmissibility of border disease (BD) virus to seronegative cows via artificial insemination with cryopreserved semen from a bull persistently infected with BD virus. Five pestivirus naive cows were inseminated with BD virus-infected semen. Blood was collected for detection of pestivirus antibody by means of an ELISA on day 0 (day of insemination) and then every 7 days until day 56, at which time a serum neutralisation test (SNT) for differentiation of BD and BVD virus was carried out. Seroconversion was first noticed in two cows on day 14, in tw

Presence and shedding dynamics of Atypical Porcine Pestivirus (APPV) in semen of breeding boars: implications for virus transmission in pig production.

BACKGROUND: Atypical porcine pestivirus (APPV), genus Pestivirus, is the causative agent of congenital tremor type A-II, a neurological disease in newborn piglets that is characterized by tremors. Although the virus can spread horizontally, clinical disease is observed only after in utero infection. While venereal transmission has been proposed as a potential route, the pathogenesis and transmission dynamics of APPV remain incompletely understood. In this study, we aimed to investigate the presence of APPV in semen from Norwegian breeding boars, its temporal shedding patterns, and the potentia

Tremor tales: A cohort study of general and neurological signs in pigs with atypical porcine pestivirus-induced congenital tremor.

Congenital tremor (CT) caused by atypical porcine pestivirus (APPV) is a widespread disease in the swine industry. It is characterized by tremors in newborn piglets, but tremor description and association to other clinical signs are not well documented. This study's objectives were to characterize general and neurological clinical signs of APPV-induced CT and describe the progression and associations between the different signs. A cohort study was conducted including 37 pigs from litters with APPV-confirmed CT and 27 control pigs from healthy litters. All pigs were examined five times from bir

Epidemiological study of border disease virus infection in Southern chamois (Rupicapra pyrenaica) after an outbreak of disease in the Pyrenees (NE Spain).

In 2001 and 2002, an outbreak of a previously unreported disease, associated with a border disease virus (BDV), caused high mortality in the Southern chamois (Rupicapra pyrenaica) population in the Alt Pallars-Aran National Hunting Reserve in the Catalan Pyrenees (NE Spain). Between 2002 and 2006, sera and/or tissue samples taken from 116 healthy chamois shot during the hunting season, plus 42 from chamois affected by different diseases, were studied. A blocking enzyme-immunosorbent assay (ELISA) was used to study pestivirus seroprevalence in 114 healthy hunted and 31 diseased chamois, yieldin

No evidence of spread of Linda pestivirus in the wild boar population in Southern Germany.

Lateral-shaking inducing neuro-degenerative agent virus (LindaV) is a novel member of the highly diverse genus Pestivirus within the family Flaviviridae. LindaV was first detected in Austria in 2015 and was associated with congenital tremor in piglets. Since then, the virus or specific antibodies have been found in a few further pig farms in Austria. However, the actual spatial distribution and the existence of reservoir hosts is largely unknown. Since other pestiviruses of pigs such as classical swine fever virus or atypical porcine pestivirus can also infect wild boar, the question arises wh

Development and use of a droplet digital PCR (ddPCR) assay to achieve sensitive and fast atypical porcine pestivirus detection.

Atypical porcine pestivirus (APPV) is a recently discovered RNA virus, which mainly caused congenital tremor in piglets. Droplet digital PCR (ddPCR) is an absolute quantitative method that does not rely on the standard curve but has high sensitivity and accuracy. The present study aimed to develop a ddPCR detection assay for APPV. Furthermore, we evaluated the limit of detection, sensitivity, specificity and reproducibility of the ddPCR and real-time quantitative PCR (qPCR) and tested 135 clinical samples to calculate the detection rate of the two methods. The results showed that both methods

12 novel atypical porcine pestivirus genomes from neonatal piglets with congenital tremors: A newly emerging branch and high prevalence in China.

Atypical porcine pestivirus (APPV), a newly discovered member of the genus Pestivirus, is considered to be associated with congenital tremors (CT) in piglets. From June 2016 to January 2018, 440 serum and tissue samples from CT-affected piglets in Anhui, Guangdong and Guangxi provinces were collected to detect APPV. The results showed a high level of 63.4% preference for APPV in 27 swine farms and complicated co-infection cases between APPV and other 12 swine viruses. Meanwhile, 12 novel APPV genomes were screened and identified. Results showed that complete genomes, Npro and NS5A genes of the

HoBi-Like Pestivirus Is the Most Frequently Detected Pestivirus in Cattle Across Different Regions of Brazil.

Pestiviruses are major viral pathogens of cattle worldwide, causing substantial economic losses and posing persistent challenges to animal health and productivity. Brazil hosts the world's largest cattle population and represents a strategic setting for the emergence and maintenance of bovine pestiviruses. Although previous studies have investigated pestiviruses in Northern, Northeastern, and extreme Southern Brazil, available molecular data remain geographically fragmented across this country of continental dimensions. HoBi-like pestivirus, HoBiPeV (HoBiPeV) has been frequently detected in No
