Phage Therapy in Aquaculture: Mechanisms, Applications and Future Directions.

The aquaculture sector faces increasing challenges from bacterial diseases and growing restrictions on antibiotic use due to concerns regarding antimicrobial resistance (AMR). Phage therapy, the therapeutic application of bacteriophages to selectively infect and lyse pathogenic bacteria, offers a targeted strategy to combat AMR pathogens while minimizing ecological disruption. This review synthesizes current evidence on major bacterial pathogens in aquaculture, with particular emphasis on infections caused by Aeromonas, Vibrio, Edwardsiella, Flavobacterium and Streptococcus species. Effective Therapeutic Options for Melioidosis: Antibiotics versus Phage Therapy.

Melioidosis, also known as Whitmore's disease, is a potentially fatal infection caused by the Gram-negative bacteria Burkholderia pseudomallei with a mortality rate of 10-50%. The condition is a "glanders-like" illness prevalent in Southeast Asian and Northern Australian regions and can affect humans, animals, and sometimes plants. Melioidosis received the epithet "the great mimicker" owing to its vast spectrum of non-specific clinical manifestations, such as localised abscesses, septicaemia, pneumonia, septic arthritis, osteomyelitis, and encephalomyelitis, which often lead to misdiagnosis an

Vibrio cholerae infection, novel drug targets and phage therapy.

Vibrio cholerae is the causative agent of the diarrheal disease cholera. Although antibiotic therapy shortens the duration of diarrhea, excessive use has contributed to the emergence of antibiotic resistance in V. cholerae. Mobile genetic elements have been shown to be largely responsible for the shift of drug resistance genes in bacteria, including some V. cholerae strains. Quorum sensing communication systems are used for interaction among bacteria and for sensing environmental signals. Sequence analysis of the ctxB gene of toxigenic V. cholerae strains demonstrated its presence in multiple

Biocontrol of Phage Resistance in Pseudomonas Infections: Insights into Directed Breaking of Spontaneous Evolutionary Selection in Phage Therapy.

Phage therapy, long overshadowed by antibiotics in Western medicine, has a well-established history in some Eastern European countries and is now being revitalized as a promising strategy against antimicrobial resistance (AMR). This resurgence of phage therapy is driven by the urgent need for innovative countermeasures to AMR, which will cause an estimated 10 million deaths annually by 2050. However, the emergence of phage-resistant variants presents challenges similar to AMR, thus necessitating a deeper understanding of phage resistance mechanisms and control strategies. The highest priority

Characterization, genome analysis, and therapeutic evaluation of a novel Salmonella phage vB_SalS_JNS02: a candidate bacteriophage for phage therapy.

Phage therapy is gaining momentum as an alternative to antibiotics in the treatment of salmonellosis caused by Salmonella. In this study, a novel Salmonella phage, vB_SalS_JNS02, was isolated successfully from poultry farms in Shandong, China. The biological characteristics of vB_SalS_JNS02 were analysed, which revealed a short latent period of approximately 10 min and a burst size of 110 PFU/cell. Moreover, vB_SalS_JNS02 exhibited remarkable stability across a wide pH range (pH 3-12) and temperatures ranging from 30 to 80°C. Genome sequencing analysis provided valuable insights into the genet

Phage Therapy of Mycobacterium Infections: Compassionate Use of Phages in 20 Patients With Drug-Resistant Mycobacterial Disease.

BACKGROUND: Nontuberculous Mycobacterium infections, particularly Mycobacterium abscessus, are increasingly common among patients with cystic fibrosis and chronic bronchiectatic lung diseases. Treatment is challenging due to intrinsic antibiotic resistance. Bacteriophage therapy represents a potentially novel approach. Relatively few active lytic phages are available and there is great variation in phage susceptibilities among M. abscessus isolates, requiring personalized phage identification. METHODS: Mycobacterium isolates from 200 culture-positive patients with symptomatic disease were scr

Phage therapy in people with cystic fibrosis: A systematic review.

BACKGROUND: Despite advances in cystic fibrosis management, chronic infections caused by multidrug-resistant bacteria remain a major clinical challenge. Bacteriophage therapy, which uses viruses that selectively infect and lyse bacteria while sparing host cells, has emerged as a promising approach against biofilm-associated and/or multidrug-resistant pathogens, although current clinical evidence is still limited. METHODS: A systematic review was conducted in four major databases up to December 2025 for randomized controlled trials, case reports and case series reporting outcomes in cystic fib

Challenges and opportunities of phage therapy for Klebsiella pneumoniae infections.

Traditional antibiotics have been effective in many cases. However, the rise in multidrug-resistant bacteria has diminished their therapeutic efficacy, signaling the dawn of an era beyond antibiotics. The challenge of multidrug resistance in Klebsiella pneumoniae is particularly critical, with increasing global mortality and resistance rates. Therefore, the development of alternative therapies to antibiotics is urgently needed. Phages, which are natural predators of bacteria, have inherent advantages. However, comprehensive information on K. pneumoniae phages is lacking in current literature.

Exacerbations of Chronic Rhinosinusitis-Microbiology and Perspectives of Phage Therapy.

The chronically inflamed mucosa in patients with chronic rhinosinusitis (CRS) can additionally be infected by bacteria, which results in an acute exacerbation of the disease (AECRS). Currently, AECRS is universally treated with antibiotics following the guidelines for acute bacterial rhinosinusitis (ABRS), as our understanding of its microbiology is insufficient to establish specific treatment recommendations. Unfortunately, antibiotics frequently fail to control the symptoms of AECRS due to biofilm formation, disruption of the natural microbiota, and arising antibiotic resistance. These issue

Safety and efficacy of phage therapy via the intravenous route.

Increasing development of antimicrobial resistance is driving a resurgence in interest in phage therapy: the use of bacteriophages to treat bacterial infections. As the lytic action of bacteriophages is unaffected by the antibiotic resistance status of their bacterial target, it is thought that phage therapy may have considerable potential in the treatment of a wide range of topical and localized infections. As yet this interest has not extended to intravenous (IV) use, which is surprising given that the historical record shows that phages are likely to be safe and effective when delivered by

Antibodies targeting phage fiber and nozzle proteins impair Acinetobacter baumannii phage therapy by blocking infection and promoting immune clearance.

Phage therapy represents a promising strategy to tackle the growing threat of antimicrobial resistance. Increasing evidence has demonstrated that phage-specific antibodies may compromise the efficacy of phage therapy. Nevertheless, little is known about how phage protein specificity of antibodies modulates phage therapeutic outcomes. Herein, we utilized AbP20, a podovirus against Acinetobacter baumannii, to explore the effects of antibodies elicited by each structural protein on phage therapy efficacy in a mouse infection model. Intraperitoneal administration of AbP20 once a day for 7 consecut

Phage Therapy for Bone and Joint Infections: Towards Clinical Translation.

Osteoarticular infections (OAIs), including osteomyelitis, septic arthritis, prosthetic joint infections, and facture-related infections, remain a major challenge due to biofilm formation and the prevalence of multidrug-resistant (MDR) pathogens. Although OAIs are predominantly caused by Staphylococcus aureus and coagulase-negative staphylococci, the increasing incidence of MDR Gram-negative infections adds further complexity to their management. Standard approaches, combining surgery and prolonged antibiotic therapy, frequently result in recurrence and poor outcomes. Bacteriophage (phage) the

Phage therapy targeting Escherichia coli-a story with no end?

Bacteriophages (phages) or bacterial viruses have long been proposed as an alternative therapy against antibiotic-resistant bacteria such as Escherichia coli Even though poorly documented in the scientific literature, a long clinical history of phage therapy in countries such as Russia and Georgia suggests potential value in the use of phages as antibacterial agents. Escherichia coli is responsible for a wide range of diseases, intestinal (diarrhoea) and extraintestinal (UTI, septicaemia, pneumoniae, meningitis), making it an ideal target for phage therapy. This review discusses the latest res

The perspectives of the application of phage therapy in chronic bacterial prostatitis.

Chronic bacterial prostatitis (CBP) is a long-lasting and crippling disease that strongly impacts the patient's quality of life. The diagnosis of CBP is difficult and the treatment regimens are not always successful. Poor penetration of antibiotics to the prostate tissue, the drug resistance of uropathogens, the adverse events associated with antibiotic treatment, the persistence of prostatic calculi, and biofilm formation in the prostate gland are factors that contribute toward decreasing the cure rate of CBP. The phenomenon of increasing antibiotic resistance, which has also been called a cl

Molecular diversity and evolutionary dynamics of phage receptor binding proteins in Staphylococcus aureus: implications for phage therapy and resistance.

Phage-host interactions are defined by the molecular specificity of receptor-binding proteins (RBPs), which determine the efficacy of infection and the host range. The genetic and structural variability of bacterial receptor molecules is essential for designing effective phage therapeutics and mitigating resistance. This study investigated four receptor-binding proteins, tarS, tarL, tagO, and tagG, in multiple Staphylococcus aureus isolates. Single-nucleotide polymorphism (SNP) analysis was performed to assess receptor diversity and evolutionary conservation. Molecular docking was conducted be

Platelet-phage conjugates for targeted therapy of acute bacterial pneumonia.

Against the escalating global threat of antibiotic resistance, phages have emerged as a promising biological therapy capable of selectively lysing pathogenic bacteria. However, their rapid systemic clearance and limited tissue targeting remain major obstacles to clinical translation. Here, we develop platelet-phage conjugates (PPCs) as a biologically guided delivery platform to enhance targeted phage therapy for bacterial pneumonia. By harnessing the innate inflammatory tropism of platelets, phages are conjugated onto platelet carriers, enabling active homing to sites of infection. As early re
