In the presence of polyunsaturated fatty acids, including cis-4,7,10,13,16,19-docosahexaenoic acid (DHA), 2,7-dichlorodibenzo-p-dioxin (DCDD) was treated with manganese peroxidase (MnP) from white rot basidiomycete Phanerochaete sordida YK-624. After incubation with MnP, DCDD could not be extracted from the reaction mixture with n-hexane and was trapped in the water layer. DCDD was released by alkalification of the water layer. DCDD was also trapped after treatment with lipoxidase, which produces hydroperoxides from unsaturated lipids. The amounts of thiobarbituric acid-reactive substances pro

Alcohol oxidases (AOxs) catalyze the aerobic oxidation of alcohols to the corresponding carbonyl products (aldehydes or ketones), producing only H2O2 as the byproduct. The majority of known AOxs, however, have a strong preference for small, primary alcohols, limiting their broad applicability, e.g., in the food industry. To broaden the product scope of AOxs, we performed structure-guided enzyme engineering of a methanol oxidase from Phanerochaete chrysosporium (PcAOx). The substrate preference was extended from methanol to a broad range of benzylic alcohols by modifying the substrate binding p

The environmental persistence and toxicity of neonicotinoids, such as imidacloprid (IMI) and thiamethoxam (THX), pose substantial threats to aquatic ecosystems. This study evaluated the performance of constructed wetlands (CWs) amended with a novel Fungus-Fe/Mn biochar substrate (synthesized by immobilizing the white-rot fungus Phanerochaete chrysosporium onto Fe/Mn-modified biochar) for enhanced removal of IMI and THX from simulated agricultural wastewater. CWs integrated with Fungus-Fe/Mn biochar demonstrated significantly improved removal efficiencies of 73.3 % for IMI and 66.7 % for THX, s

Benzotriazole ultraviolet stabilizers (BUVSs) are widely used as additives in a variety of materials to protect against UV-induced degradation. Their widespread distribution in environmental and biological samples has raised concerns regarding their potential ecological and health risks. 2-(2'-Hydroxyphenyl)benzotriazole (UV-P), a structurally simple BUVS, was selected as a model compound to evaluate its metabolism by the white-rot fungus Phanerochaete sordida in potato dextrose broth cultures. UV-P gradually decreased, with approximately 10% and 40% removed in 2 and 4 weeks, respectively. Two

Heme-containing enzymes, critical across life's domains and promising for industrial use, face stability challenges. Despite the demand for robust industrial biocatalysts, the mechanisms underlying the thermal stability of heme enzymes remain poorly understood. Addressing this, our research utilizes a 'keystone cofactor heme-interaction approach' to enhance ligand binding and improve the stability of lignin peroxidase (LiP). We engineered mutants of the white-rot fungus PcLiP (Phanerochaete chrysosporium) to increase thermal stability by 8.66 °C and extend half-life by 29 times without losing

Biodegradation of soil humic acids by Streptomyces viridosporus ATCC 39115 growing in a mineral salts--glucose medium was demonstrated. This biodegradation accompanies bacterial growth and is, therefore, presumed to be a primary metabolic activity, but humic acids were not used as the sole source of carbon. This bacterial activity was enhanced when cells were shaken and within a pH range of 6.5-8.5. In further experiments, the relative abilities of S. viridosporus to mineralized [14C]melanoidin, used as synthetic humic acid, were also established. In contrast to the white rot fungus Phanerocha

In order to improve the adsorption performance of microorganisms, we synthesized a novel material - phanerochaete chrysosporium cells covered with a layer of ZnS nanoparticles (ZnS-cells). The preparation of the ZnS-cells is based on the Sonochemical method to synthesize the ZnS nanoparticle layer on the surface of the microbial cells. The ZnS-cells were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), energy dispersive X-ray spectroscopy (EDS) and Fourier transform infrared spectroscopy (FTIR). Characterization results showe

The 1H NMR spectroscopy was used to study lignin peroxidase (LiP) and manganese peroxidase (MnP) containing deuterated histidines. LiP and MnP were obtained from a histidine auxotroph of the fungus Phanerochaete chrysosporium grown in the presence of deuterated histidines. The derivatives with deuterated histidines have allowed a firm assignment of the protons of the distal and proximal histidines. We have also found that the LiP from this strain exhibits different orientations of the 2-vinyl group compared to the LiP from the strain previously studied. Mobility of the group has also been dete

White-rot and brown-rot fungi (WRF and BRF, respectively) decompose lignocellulose, the main structural component of plant biomass, through distinct mechanisms. This study examines the transcriptomic responses of three WRF species (Pleurotus ostreatus, Phanerochaete chrysosporium, and Heterobasidion irregulare) and two BRF species (Fomitopsis schrenkii and Rhodonia placenta) grown on poplar wood (W) and glucose (G) as sole carbon sources. RNA-seq analysis revealed upregulation of carbohydrate-active enzymes (CAZymes) linked to lignocellulose degradation. WRF displayed a broader enzymatic reper

Wastewaters produced by the recycled paper and cardboard sector (RPCE) pose significant environmental risks due to high levels of chemical oxygen demand (COD) and a lack of essential nutrients required for effective biological degradation. In response, this investigation introduces a novel and cost-effective valorization approach: employing nutrient-laden dairy wastewater (DWW) as an eco-friendly source of nitrogen and phosphorus to enhance the bioremediation performance of a fungal consortium comprising Bjerkandera adusta and Phanerochaete chrysosporium. A primary aim of the study was to opti

Heterogous expression of lignin peroxidase (LiP) from Phanerochaete chrysosporium was performed in by E. coli prokaryotic expression system, and pure LiP was prepared by washing, refolding, and purification. The enzyme activity was measured by the resveratrol oxidation method. The effects of different chemicals on LiP activity were explored by adding different kinds of metal ions, acids/phenols, and surfactants. The optimal pH and temperature are 4.2 and 40 °C. The single-factor screening experiment showed that adding 1 mM Mn2+, 0.1 mM DL-lactic acid, and 2% PEG-4000 had the best promotion eff

Subunit vaccines are considered the safest immunization platforms but typically require adjuvants to elicit robust immune responses. Although nanoparticles have emerged as promising adjuvants, biogenic silver nanoparticles (AgNPs) produced by mycosynthesis remain largely unexplored. Herein, nine different AgNPs were mycosynthesized using local strains of Phanerochaete chrysosporium, Penicillium expansum and Punctularia atropurpurascens grown in three different culture media (PDB, MEB, MGYP). Mycosynthesized AgNPs were all shown to be spherical, negatively charged and containing fungal proteins

Over the past few years biodegradation of trichloroethylene (TCE) using different microorganisms has been investigated by several researchers. In this review article, an attempt has been made to present a critical summary of the recent results related to two major processes--reductive dechlorination and aerobic co-metabolism used for TCE biodegradation. It has been shown that mainly Clostridium sp. DC-1, KYT-1, Dehalobacter, Dehalococcoides, Desulfuromonas, Desulfitobacterium, Propionibacterium sp. HK-1, and Sulfurospirillum bacterial communities are responsible for the reductive dechlorinatio

The search for novel microorganisms able to degrade olive mill wastewaters (OMW) and withstand the toxic effects of the initially high phenolic concentrations is of great scientific and industrial interest. In this work, the possibility of reducing the phenolic content of OMW using new isolates of fungal strains (Coriolopsis gallica, Bjerkandera adusta, Trametes versicolor, Trichoderma citrinoviride, Phanerochaete chrysosporium, Gloeophyllum trabeum, Trametes trogii, and Fusarium solani) was investigated. In vitro, all fungal isolates tested caused an outstanding decolorization of OMW. However

The present study focused on the bioaccumulation and cytotoxicities of Cd2+, CdSe quantum dots (QDs) and CdSe/ZnS QDs in Escherichia coli (E. coli, represents prokaryotic system) and Phanerochaete chrysosporium (P. chrysosporium, represents eukaryotic system), respectively. Two types of QDs were characterized by transmission electron microscopy (TEM) and dynamic light scattering. The inductively coupled plasma optical emission spectrometer results showed that the bioaccumulation amounts of CdSe QDs by E. coli and P. chrysosporium were larger than those of CdSe/ZnS QDs due to the smaller partic

The fibrolytic enzymes and the hindgut fungi in donkey cecum-colon ecosystem play an important role in dietary fiber digestion. A better understanding of the fibrolytic enzyme profiles and the fungal community along donkey caecum and colon is key for optimizing hindgut function. In the present study, the fibrolytic enzyme activities within donkey caecum and colon were firstly measured by spectrophotometry. Activities of carboxymethyl cellulase, avicelase, xylanase, and acetyl esterase were greater in donkey dorsal colon than in caecum, indicating that the colon microorganisms may be more effic
