Regioselective separation of isomeric triacylglycerols by reversed-phase high-performance liquid chromatography: stationary phase and mobile phase effects.

Complete regioselective separation of five pairs of isomeric dipalmitoyl polyalkenoyl glycerols with two to six double bonds in the unsaturated acyl residues has been achieved by RP-HPLC on a single ODS column. Four ODS columns with stationary phases containing different percentages of free silanol groups have been tested. Binary mobile phases of ACN admixed with dichloromethane, tetrahydrofuran, 1-propanol, 2-propanol, ethanol, or acetone have been examined. The choice of modifier depended on the nature of the stationary phase. The more polar solvents were better suited for stationary phases

Chiral separations on polysaccharide stationary phases using polar organic mobile phases.

About 30% of a chemically diverse set of compounds were found to separate on four polysaccharide chiral stationary phases using polar organic mobile phases. No structural features appeared to correlate to successful separations. Titrations between normal and polar organic mobile phases suggested that separation mechanisms do not differ between these mobile phases. Attempts made to control retention met with varying degrees of success. Addition of hexane to alcohols had minor effects on retention although this was occasionally beneficial. Addition of water to alcohols increased retention. Addit

Phase separation of p85β modulates hepatocellular carcinoma progression through POLR1A.

PI3K complex consists of catalytic subunit p110s and regulatory subunit p85s. Emerging evidence indicates that p110-free p85 subunits play pivotal roles in diverse biological processes, including cancer progression. In this study, we demonstrate the underlying mechanism of p110-free p85β in hepatocellular carcinoma (HCC) development. PIK3R2/p85β is upregulated in HCC and correlates with poor patient survival. Nuclear p85β, but not its cytoplasmic counterpart, exhibits oncogenic activity. In the nucleus of HCC cells, p85β undergoes liquid-liquid phase separation (LLPS) and specifically accumula

α-Synuclein aggregation landscape from phase separation to neurotoxic intermediates.

The aberrant aggregation of α-synuclein (αS) into insoluble amyloid fibrils is a hallmark of Parkinson's disease. Despite recent advances in characterising the properties of mature αS fibrils, the transient and heterogeneous intermediates that underlie cellular toxicity remain largely elusive. Here, we review the mechanistic principles of αS aggregation, focussing on liquid-liquid phase separation (LLPS) as a critical intermediate step. We discuss how the structural evolution of αS within the condensed phase governs the subsequent patterns of cellular dysfunction and pathological propagation.

Liquid-Liquid Phase Separation-Enhanced Multienzyme Catalysis: Mechanisms and Applications.

Living organisms have evolved multienzyme complexes to achieve efficient and spatially ordered metabolic reactions. Recently, liquid-liquid phase separation (LLPS) has emerged as a fundamental organizational principle underlying these natural networks, providing a versatile platform for constructing artificial multienzyme catalytic systems. Owing to their reversible self-assembly and programmable nature, LLPS condensates can modulate enzyme distribution, mass transfer, and the reaction microenvironment, thereby significantly enhancing catalytic efficiency and cascade reaction throughput. Osmotic Pressure-Induced Lipid Membrane Phase Separation within Macromolecular Environments.

Lipid membrane phase separation, which is a thermodynamic physical process, has attracted attention as a dynamic function of cellular interfaces. As membrane tension influences the phase separation of cellular membranes under isothermal conditions, it is essential to clarify the physicochemical mechanism involved. Living cells contain numerous macromolecules that can generate osmotic pressure across the membrane due to the semipermeable nature of the lipid bilayer. In this research, we examined how macromolecular surroundings and the transmembrane osmotic pressure influence membrane phase sepa

Crowder Induced Phase Separation Modulates the Huntingtin Protein Aggregation Landscape.

Macromolecular crowding is a defining feature of cellular environments and plays an important role in modulating protein aggregation. Here, we examine how mixed macromolecular crowders influence the aggregation of Huntingtin exon-1 protein with a pathogenic polyglutamine length (HD39Q). Using thioflavin-T (ThT) kinetics, we show that the binary mixtures of polyethylene glycol (PEG8), Dextran (40 and 70), and Ficoll 70 produce biphasic aggregation kinetics that deviate from classical single-step behavior. Confocal imaging using labeled proteins and crowders reveals that not only do the binary c

Spontaneous Phase Separation Enables Rapid, Polymerization-Free Fabrication of Gels.

Hydrogels are cross-linked polymeric networks with wide applications in drug delivery, tissue engineering, biosensing, and environmental remediation. These hydrogels additionally host living cells, small molecules, and biological propagules, which further expand the applications of these materials. However, most, if not all, fabrication methods require covalent modifications. In this work, by deliberately selecting polymers with a known propensity to phase separate and formulating compositions far from the binodal boundary, we demonstrate the propensity of the system to transition directly int

Glycoprotein-induced phase separation drives unconventional secretion of galectin-3.

The mechanism of unconventional protein secretion remains an unresolved issue. Here, we describe an unconventional protein secretion pathway for galectin-3 that is mediated by phase separation and condensation. Using four lysosomal damage models, we observed a rapid, pronounced release of galectin-3 in large, non-exosomal particles. This secretion is driven by glycoprotein-induced galectin-3 phase separation and is independent of pyroptosis and secretory autophagy. During phase separation, the S-face of galectin-3 carbohydrate recognition domain binds glycoproteins that triggers galectin-3 N-t

DDX6 undergoes phase separation to modulate metabolic plasticity and chemoresistance.

Stress granules (SGs) and processing bodies (PBs), assembled via liquid-liquid phase separation (LLPS), are critical for spatial regulation of gene expression in the cytoplasm. However, their roles in tumorigenesis remain poorly understood. Here, we show DEAD-box helicase 6 (DDX6) as the most promising vulnerability in acute myeloid leukemia (AML) through in vitro and in vivo CRISPR screenings using a specialized library targeting RNA-binding proteins enriched in SGs and PBs. Knockout (KO) of DDX6 significantly delays leukemogenesis with minimal impact on normal hematopoiesis. Importantly, the

Tuning Targeted Liposome Avidity to Cells via Lipid Phase Separation.

The addition of both cell-targeting moieties and polyethylene glycol (PEG) to nanoparticle (NP) drug delivery systems is a standard approach to improve the biodistribution, specificity, and uptake of therapeutic cargo. The spatial presentation of these molecules affects avidity of the NP to target cells in part through an interplay between the local ligand concentration and the steric hindrance imposed by PEG molecules. Here, we show that lipid phase separation in nanoparticles can modulate liposome avidity by changing the proximity of PEG and targeting protein molecules on a nanoparticle surf

Formaldehyde Cross-Linking-Assisted Phase Separation for Protein Aptamer Selection.

With the merits of easy synthesis, strong modifiability, and high affinity, aptamers have been broadly applied for protein targeting in bioanalysis, diagnosis, and therapeutics. The selection of protein-targeted aptamers is currently largely dependent on solid-liquid separation by using different types of nano- or micro-beads. However, the use of beads inescapably introduces unwanted nonspecific binding and thus affects selection efficiency. In order to sidestep this obstacle, we herein report an integrated technique to facilitate the discovery and development of protein-targeting aptamers by

A novel hydrocyclone for use in underground DNAPL phase separation.

Halogenated organic solvents are the most commonly detected pollutants in groundwater and are particularly toxic and harmful. How to separate these dense nonaqueous phase liquid (DNAPL) pollutants efficiently from groundwater has become an important research question. Here, a novel hydrocyclone with annular overflow structure was designed, which eliminated the short-circuit flow of the traditional hydrocyclone and solved the problem of overflow entrainment caused by the enrichment of droplets near the locus of zero vertical velocities (LZVV) into turbulence. The flow field characteristics of t

Competing effect of disorder on phase separation in active systems.

We investigate the impact of random pinned disorder on a collection of self-propelled particles. To achieve this, we construct a continuum model by formulating the coupled hydrodynamic equations for slow variables: local density and momentum density of particles. The disorder in the system acts as pinning sites, effectively immobilizing the particles that come into contact with them. Our numerical results reveal that weak disorder leads to phase separation in the system at density and activity lower than the typical values for motility-induced phase separation. We construct a phase diagram usi

Triptycene-Based Lipid Mimics for Thermally Stable Membrane Phase Separation.

Membrane phase separation enables the formation of laterally compartmentalized domains with distinct local environments in lipid bilayers. Such compartmentalization offers a strategy for functional membrane materials because clustered molecules can exhibit properties that differ from those of uniformly mixed membranes. Despite this potential, the induction of thermally stable domains in fluid lipid bilayers remains a challenge owing to the high miscibility of lipid molecules. Here, we report triptycene-based lipid mimics that induce thermally stable membrane phase separation. Using the triple

Programmable DNA protonuclei reveal environmental context on protein phase separation.

Understanding protein phase separation in cellular environments remains a major challenge, as ex vivo assays often fail to capture the influence of environmental context - such as crowding, multimodal interactions, and the dynamic properties of the cytosol or nucleus. Here, we introduce programmable DNA-based protonuclei (PN) as nucleus-inspired compartments to probe phase separation of the neurodegeneration-linked protein FUS. We show that FUS partitioning and condensate formation are highly sensitive to nucleic acid sequence, spatial confinement, and viscoelastic properties of the PN core. N

Hydrodynamic Interactions Destroy Motility-Induced Phase Separation in Active Suspensions.

Motility-induced phase separation (MIPS) is a distinctive phenomenon in active matter that arises from its inherent nonequilibrium nature. Despite recent progress in understanding MIPS in dry active systems, it has been debated whether MIPS can be observed in wet systems in which fluid-mediated hydrodynamic interactions (HIs) are present. We use theory and large-scale active fast Stokesian dynamics simulations of the so-called squirmer model to show that collision-induced pusher force dipoles, which are present even for the simplest neutral squirmers (stealth swimmers), destroy MIPS when HIs a

Physiological implications of phase separation in vesicle organization and trafficking.

Phase separation has emerged as a key mechanism of cellular compartmentalization, complementing classical membrane-bound organelles. In addition to forming membraneless compartments that provide confined environments for biochemical reactions, phase separation actively organizes and regulates membrane vesicles. Protein condensates interact with vesicular organelles through defined modes of communication, influencing their assembly, trafficking, and turnover. This review highlights recent advances in understanding how phase separation contributes to vesicle biology, including synaptic vesicle c

TDRD5 facilitates piRNA biogenesis and male fertility through phase separation.

piRNAs are small regulatory RNAs with critical roles in transposon silencing and regulation of gametogenesis. Phase separation is a driving force for piRNA biogenesis machinery formation; however, the specific piRNA-related proteins that mediate phase separation and how they coordinate to drive piRNA production remain incompletely understood. Through systematic screening, we show here that TDRD5 undergoes phase separation to promote piRNA biogenesis. TDRD5 condensation is mediated by its Lotus domains and an intrinsically disordered region enriched with aromatic residues. Furthermore, TDRD1 an

Fluorogenic methodology for visualization of phase separation in chemical biology.

Phase separation is a common biological phenomenon in the liquid environment of organisms. Phase separation has been shown to be a key cause of many existing incurable diseases, such as the protein aggregates formed by phase separation of Alzheimer's Disease, Amyotrophic Lateral Sclerosis, Parkinson's disease, etc. Tracking the occurrence of phase separation in vivo is critical to many disease detection methods and solving many treatment problems. Its physicochemical properties and visual detection methods have flourished in the last few years in chemical biology, among which the fluorogenic t
