Development of Rapid and Economical Colorimetric Screening Method for p-Phenylenediamine in Variety of Biological Matrices and its Application to Eleven Fatal Cases of p-Phenylenediamine Poisoning.

A rapid colorimetric method for detection of p-phenylenediamine (PPD) in various biological samples is developed. The o-cresol test for acetaminophen detection has been modified to detect PPD in blood, urine, gastric contents, and liver. After precipitating protein with trichloroacetic acid solution (2 mL, 10% w/v), biological specimens were required to convert PPD metabolites to PPD by acid hydrolysis. Finally, o-cresol solution (1 mL, 1% w/v), hydrogen peroxide (200 μL, 3%v/v), and concentrated ammonium hydroxide (0.5 mL) were added in the biological samples. The presence of PPD was indicate

Cross-elicitation responses to 2-methoxymethyl-p-phenylenediamine in p-phenylenediamine-allergic individuals: Results from open use testing and diagnostic patch testing.

BACKGROUND: Allergic contact dermatitis caused by p-phenylenediamine (PPD) is a health concern for hair dye users. Because of its lower sensitization potency, the PPD derivative 2-methoxymethyl-p-phenylenediamine (ME-PPD) has been developed as an alternative hair dye for primary prevention. However, cross-elicitation responses can occur in PPD-allergic subjects. OBJECTIVES: To compare cross-elicitation responses to ME-PPD in open use and diagnostic patch testing of PPD-allergic subjects with hair dye-related allergic contact dermatitis. METHODS: Reactions to ME-PPD were investigated in 25 PP

P-Phenylenediamine Hair Dye Allergy and Its Clinical Characteristics.

BACKGROUND: P-phenylenediamine (PPD) has been identified as the most frequent contact sensitizer of hair dye and the clinical characteristics of hair dye contact allergy (HDCA) are diverse. OBJECTIVE: The purpose of this study was to identify the clinical characteristics of HDCA and to assess the relationships between HDCA, exposure time to PPD and PPD positivity. METHODS: We analyzed 105 patients with patch test-confirmed hair dye allergy who presented between July 2009 and March 2015. Clinical symptoms, signs, associated skin diseases, involved ACD area, and patterns of hair dye use were o

Prevalence of Contact Allergy to p-Phenylenediamine in the European General Population.

Population-based studies on contact allergy to p-phenylenediamine (PPD) are scarce. A cross-sectional study was performed to assess the prevalence of contact allergy to PPD and its risk factors in the general population of 5 European countries. A total of 10,425 subjects were interviewed, and a random sample (n = 2,739) was patch tested to PPD. Overall, 5,286 individuals (50.9%) reported having used hair colorants at least once in their lifetime (78% female, 20% male), and 35% had used hair colorants during the last 12 months. Hair colorant avoidance because of any skin problem during the life

Assessment of the elicitation response in subjects weakly sensitized to p-phenylenediamine.

BACKGROUND: A 30-min application of a hair dye product containing 2% p-phenylenediamine (PPD) to subjects diagnostically graded +, showed that 12 of 18 reacted; eight of 18 with a true + and four of 18 with a doubtful (?+) response, whereas six of 18 did not react at all. In vitro skin-binding experiments showed that for diagnostic patch test conditions the measured exposure level (MEL) is more than 10-fold higher than the MEL for hair dyeing conditions. OBJECTIVE: To further analyse the limited elicitation response of the diagnostically + graded subjects to a PPD hair dye product, under stan

Adsorption of Cr(VI) using synthetic poly(m-phenylenediamine).

Poly(m-phenylenediamine) (PmPD) with different oxidation state was successfully synthesized by the improved chemically oxidative polymerization. The function of oxidation state on Cr(VI) adsorption was systematically examined through adsorption experiments. Results showed that the Cr(VI) adsorptivity of all PmPD increased with decreasing the initial pH. When the oxidation state of PmPD was dropped, the equilibrium time for Cr(VI) adsorption was obviously shortened and its Cr(VI) removal and adsorption selectivity were profoundly obviously increased. Typically, PmPD with the lowest oxidation st

Allergic contact dermatitis to para-phenylenediamine in a tattoo: a case report.

It is highly popular among children and young adults to have temporary henna tattoos on their bodies in different colors and figures. Henna is a greenish natural powder obtained from the flowers and dry leaves of Lawsonia alba plant and its allergenicity is very low. Henna is also used in combination with other coloring substances such as para-phenylenediamine in order to darken the color and create a permanent tattoo effect. Para-phenylenediamine is a substance with high allergenicity potential and may cause serious allergic reactions. Here, we aimed to draw attention to the potential harms o

Identification and quantification of para-phenylenediamine in a temporary black henna tattoo.

BACKGROUND: Temporary black henna tattoos are very popular as body adornment. Although contact allergy to natural henna is unusual, the inclusion of hair dye, p-phenylenediamine (PPD), increases the risk of contact sensitization. OBJECTIVE: This study was performed to identify the presence and concentration of PPD in a black henna tattoo mixture to which our patient developed contact allergy. METHODS: The presence of PPD in a black henna tattoo mixture, various samples of commercially available henna powders, and several hair dye products was qualitatively and quantitatively detected using h

Development of novel alternative hair dyes to hazardous para-phenylenediamine.

Most of the permanent hair dye products contain p-phenylenediamine (PPD), a well-known skin sensitizer. PPD may cause cutaneous reactions and leads to allergic contact dermatitis (ACD), a condition with major medical and financial repercussions. Hair dye-induced ACD represents a growing concern both for consumers and the cosmetics industry. In this study we introduced novel side chains on the PPD molecule with the goal of overcoming the hazard potential of PPD. Our strategy relies on the replacement of the colorless PPD with new, larger and intrinsically colorled PPD derivatives to reduce derm

Clinical and experimental aspects of allergic contact dermatitis to para-phenylenediamine.

The allergenicity of para-phenylenediamine (PPD) is related to oxidative processes on and in the skin. Patients with stronger reactions to PPD are significantly more likely to have a clear history of reacting to normal consumer hair dye. Those with stronger reactions are much less likely to be still dyeing their hair. Individuals allergic to PPD have a significant frequency of simultaneous sensitivity to chemically related clothing dyes. A 24-hour test application of hair dye, which has been proposed as a self-screen, does not reliably predict all individuals allergic to PPD. Duration studies

Alternative hair-dye products for persons allergic to para-phenylenediamine.

BACKGROUND: Finding alternative hair dyes for individuals allergic to para-phenylenediamine (PPD) has been difficult. Newer permanent and demipermanent hair dyes that have replaced PPD with para-toluenediamine sulfate (PTDS) are now available. OBJECTIVE: We examined whether individuals allergic to PPD will tolerate PPD-free hair dyes containing PTDS. METHODS: A retrospective analysis of patch-test results since October 2006 was done and yielded 28 patients allergic to PPD who were also tested with a hair dye series. From January 2004 through October 2006, seven additional patients allergic t

Determination of para-phenylenediamine (PPD) in henna in the United Arab Emirates.

Henna is very popular in the United Arab Emirates (UAE); it is part of the culture and traditions. Allergy to natural henna is not usual; however the addition of para-phenylenediamine (PPD) to the natural henna increases the risk of allergic contact dermatitis. The objectives of the study were to identify the presence and concentration of PPD in henna available in UAE. Fifteen henna salons were selected randomly from three cities in UAE. Twenty five henna samples were acquired from these selected salons. The presence of PPD in henna samples was determined qualitatively and quantitatively using

A mechanistic investigation into the irreversible protein binding and antigenicity of p-phenylenediamine.

Exposure to the skin sensitizer p-phenylenediamine (PPD) is associated with allergic contact dermatitis; however, the ability of PPD to modify protein has not been fully investigated. The aims of this study were to characterize the reactions of PPD and the structurally related chemical 2,5-dimethyl-1,4-benzoquinonediamine with model nucleophiles, a synthetic peptide (DS3) containing each of the naturally occurring amino acids and His-tagged glutathione-S-transferase pi (GSTP), and to explore the effect of dimethyl substitution on PPD-specific T-cell responses using lymphocytes from allergic pa
