The vector competence of Phlebotomus perniciosus for Leishmania infantum zymodemes of Tunisia.

Experimental infections of Phlebotomus (L.) perniciosus from a colony established in Madrid (Spain) carried out with the Leishmania (L.) infantum zymodemes MON-1, MON-24, and MON-80 isolated in Tunisia are reported here. Laboratory-reared female sand flies were experimentally fed via membrane feeding device on a suspension of L. infantum promastigotes in defibrinated rabbit blood (107/ml). Engorged females were dissected at progressive time points postfeeding to observe the intravectorial cycle of different L. infantum zymodemes. Development in the sand fly midgut of L. infantum parasites to t

Analysis of salivary transcripts and antigens of the sand fly Phlebotomus arabicus.

BACKGROUND: Sand fly saliva plays an important role in blood feeding and Leishmania transmission as it was shown to increase parasite virulence. On the other hand, immunity to salivary components impedes the establishment of infection. Therefore, it is most desirable to gain a deeper insight into the composition of saliva in sand fly species which serve as vectors of various forms of leishmaniases. In the present work, we focused on Phlebotomus (Adlerius) arabicus, which was recently shown to transmit Leishmania tropica, the causative agent of cutaneous leishmaniasis in Israel. RESULTS: A cDN

Phlebotomus (Paraphlebotomus) alexandri: a probable vector of Leishmania infantum in Iran.

The incidence of human visceral leishmaniasis (VL) caused by Leishmania infantum, which is endemic in several parts of Iran, has recently increased in the Nourabad-Mamassani district of Fars province, in the south of the country. Between 2003 and 2005, 12,688 sandflies were caught in this focus, using CDC miniature light traps, sticky traps and aspirators, in an attempt to identify the main vector of L. infantum. When 120 of the parous, female Phlebotomus (Paraphlebotomus) alexandri caught were individually checked for Leishmania infection, in an assay based on PCR and kinetoplast minicircle p

The development of Leishmania infantum in Phlebotomus langeroni nitzulescu (Diptera: Psychodidae).

The extrinsic development of Leishmania infantum was observed in Phlebotomus langeroni the potential vector of visceral leishmaniasis in El-Agamy, Egypt. Flies were infected with L. infantum isolated from the same area, using membrane feeding technique. Flies were examined for infection at 1-10 days post-infection. The initial establishment of the parasites was in the posterior midgut region beginning at sixth day post-infection. Parasites were first observed in the esophagus at day 8, and in the posterior armature region of the pharynx at day 9. The pattern of development of L. infantum in P.

Phlebotomus guggisbergi (Diptera: Psychodidae), a vector of Leishmania tropica in Kenya.

Sand flies were collected in light traps and on oiled papers at four active case sites of human cutaneous leishmaniasis due to Leishmania tropica at Muruku Sublocation, Laikipia District, Kenya. Nearly 5,200 females of five species, including Phlebotomus guggisbergi, were dissected and examined for flagellates. Of 3,867 P. guggisbergi females collected at a multiple case site, 168 (4.3%) harbored mature infections (to include metacyclic promastigotes) of flagellates morphologically identical to Leishmania, while all other flies were negative. Of the infected flies, 164 were collected in a cave

Anthropophagy and aggregation behaviour of the sandfly Phlebotomus argentipes in Sri Lanka.

The visceral leishmaniasis (VL) vector Phlebotomus argentipes Annandale & Brunetti is widely distributed throughout the Indian sub-continent and S.E. Asia. The absence of VL in areas such as Sri Lanka has been attributed to the zoophilic nature of P.argentipes, since they were not recorded biting man. Field studies on P.argentipes were undertaken in the central highlands of Sri Lanka, near Kandy, in May 1988. Male sandflies outnumbered females on cows by 19:1, and were regularly spaced at all densities. This behaviour is considered analagous to swarming in other Nematocera. However, all-night

Impact of Vgsc-1014 mutations on the feeding pattern of Phlebotomus argentipes.

Knockdown resistance alleles (kdr alleles) within the para voltage-gated sodium channel gene (Vgsc) are a common mechanism of DDT and pyrethroid resistance in insect vectors. In the primary Asian visceral leishmaniasis vector, Phlebotomus argentipes, two kdr alleles in codon 1014 of the Vgsc are associated with insecticide resistance, potentially presenting challenges to vector control efforts in the Indian subcontinent. Here, we screened Vgsc-1014 alleles and blood meal origin in P. argentipes females collected between September 2013 and August 2015 in Bangladesh (Mymensingh), to understand h

Phlebotomus perniciosus response to volatile organic compounds of dogs and humans.

The olfactory response of insect vectors such as phlebotomine sand flies is a key facet for investigating their interactions with vertebrate hosts and associated vector-borne pathogens. Such studies are mainly performed by assessing the electrophysiological response and the olfactory behaviour of these arthropods towards volatile organic compounds (VOCs) produced by hosts. Nonetheless, few studies are available for species of the subgenera Lutzomyia and Nyssomyia in South America, leaving a void for Old World sand fly species of the genus Phlebotomus. In this study, we evaluated the olfactory

Malformations of the genitalia in male Phlebotomus papatasi (Scopoli) (Diptera: Psychodidae).

Phlebotomus papatasi (Scopoli, 1786) (Diptera: Psychodidae) is a major vector of Leishmania major (Kinetoplastida: Trypanosomatidae), a causative agent of zoonotic cutaneous leishmaniasis. Morphological characters of sand fly genitalia are key indicators for species identification. Various anomalies affecting male genitalia have been previously described. We take advantage of a large sand flies survey conducted in 32 stations in Central and Southern Morocco to systematically quantify the prevalence and spatial distribution of malformations affecting the genitalia of P. papatasi. Among 597 exam

Phlebotomus perfiliewi transcaucasicus, a vector of Leishmania infantum in northwestern Iran.

Visceral leishmaniasis is caused by Leishmania infantum, which is transmitted to humans by bites of phlebotomine sand flies and is one of the most important public health problems in Iran. To identify the vector(s), an investigation was carried out in Germi district, an important focus of the disease in Ardebil province in northwestern Iran, during July-September 2004 and 2005. Using sticky papers, CDC light traps and aspirators, 3,560 sand flies were collected and identified to species. Host bloodmeal preference and Leishmania infections in female specimens were evaluated using enzyme-linked

The structure of the female sand fly (Phlebotomus papatasi) alimentary canal.

In the sand fly vector, Leishmania parasites are confined to the alimentary canal. During much of their development, promastigotes are attached to the wall of the gut via their flagella. In this context, the surface of the different regions of the sand fly alimentary tract lumen warrants scientific attention. In this paper, the various regions are described, for the first time using scanning electron microscopy. The cibarium and the pharynx, which function as pumping organs, are lined with cuticle. Parts of the cibarium and the pharynx bear different types of cuticular spines and appendages. T

Knockdown resistance mutations in Phlebotomus argentipes sand flies in Bihar, India.

BACKGROUND: Vector control based on indoor residual spraying (IRS) is one of the main components of the visceral leishmaniasis (VL) elimination programme in India. Dichlorodiphenyltrichloroethane (DDT) was used for IRS until 2015 and was later replaced by the synthetic pyrethroid alpha-cypermethrin. Both classes of insecticides share the same target site, the voltage-gated sodium channel (Vgsc). As high levels of resistance to DDT have been documented in the local sand fly vector, Phlebotomus argentipes, it is possible that mutations in the Vgsc gene could provide resistance to alpha-cypermeth

Behavioural responses of Phlebotomus duboscqi to plant-derived volatile organic compounds.

Phlebotomine sand flies are vectors of Leishmania parasites that cause leishmaniases. Both sexes of sand flies feed on plants primarily for sugars, although the chemical cues that mediate attraction to host plants remain largely unknown. Previously, using coupled gas chromatography-mass spectrometry, the authors identified several volatile organic compounds (VOCs) common to preferred host plants for selected Afrotropical sand flies from the Fabaceae family. Of the identified volatiles, the significance of the monoterpenes linalool oxide, ocimene and p-cymene and the benzenoid m-cresol, p-creso

Human immune response to salivary proteins of wild-caught Phlebotomus papatasi.

Phlebotomine sand flies are the known vectors of Leishmania parasites. New approaches in vaccination against Leishmania have investigated the possibility of integrating Phlebotomus papatasi salivary proteins to enhance the immune response and protect against the transmission of the infection. The aim of the present study was to screen human immune responses to wild sand fly saliva and evaluate immunogenic salivary proteins. Blood samples were collected from donors in control and sand fly infested areas. Antibodies specific for sand fly antigens in donor plasma were probed using immunoblotting.
