The Small RNA Component of Arabidopsis thaliana Phloem Sap and Its Response to Iron Deficiency.

In order to discover sRNA that might function during iron deficiency stress, RNA was prepared from phloem exudates of Arabidopsis thaliana, and used for RNA-seq. Bioanalyzer results indicate that abundant RNA from phloem is small in size-less than 200 nt. Moreover, typical rRNA bands were not observed. Sequencing of eight independent phloem RNA samples indicated that tRNA-derived fragments, specifically 5' tRFs and 5' tRNA halves, are highly abundant in phloem sap, comprising about 46% of all reads. In addition, a set of miRNAs that are present in phloem sap was defined, and several miRNAs and

Magnesium deficiency in sugar beets alters sugar partitioning and phloem loading in young mature leaves.

Magnesium deficiency has been reported to affect plant growth and biomass partitioning between root and shoot. The present work aims to identify how Mg deficiency alters carbon partitioning in sugar beet (Beta vulgaris L.) plants. Fresh biomass, Mg and sugar contents were followed in diverse organs over 20 days under Mg-sufficient and Mg-deficient conditions. At the end of the treatment, the aerial biomass, but not the root biomass, of Mg-deficient plants was lower compared to control plants. A clear inverse relationship between Mg and sugar contents in leaves was found. Mg deficiency promoted

Phloem-Specific Expression of Tyrosine/Dopa Decarboxylase Genes and the Biosynthesis of Isoquinoline Alkaloids in Opium Poppy.

Tyrosine/dopa decarboxylase (TYDC) catalyzes the formation of tyramine and dopamine and represents the first steps in the biosynthesis of the large and diverse group of tetrahydroisoquinoline alkaloids. Opium poppy accumulates morphine in aerial organs and roots, whereas sanguinarine, which is derived from a distinct branch pathway, accumulates only in roots. Expression of the TYDC gene family in opium poppy was investigated in relation to the organ-specific biosynthesis of these different types of alkaloids. Members of the TYDC gene family are classified into two groups (represented by TYDC1

Characterization of Viruses in Phloem by Correlative X-Ray Microtomography (μCT)-Volume Electron Microscopy (vEM) Imaging.

Studying virus-infected phloem is of significant importance, as it not only enhances our understanding of viral pathogenesis but also leverages viruses as tools to expand knowledge about plant phloem physiology. The uneven distribution pattern of phloem-infecting viruses poses methodological challenges for such studies-requiring both large field of view (FOV) and high-resolution imaging. A comprehensive anatomical analysis of the phloem necessitates global visualization, while resolving viral structures demands local high-resolution observation. This chapter describes a method, the X-ray micro

Citrus yellow vein clearing virus infection triggers phloem remobilization of iron- and zinc-nicotianamine in citrus.

Citrus yellow vein clearing virus (CYVCV) is a worldwide and highly destructive disease of citrus, but the mechanisms involved in CYVCV-inhibited plant growth are not well understood. This study examined nutrient levels and their cellular distribution in different organs of healthy and CYVCV-affected citrus (Citrus reticulata 'Kanpei') plants. We found that CYVCV-infected plants exhibit characteristic symptoms, including a significant reduction in iron (Fe) and other elemental nutrients in the shoots. Our data suggest that CYVCV-induced chlorosis in citrus leaf veins is primarily due to iron d

Facilitating Phloem-Mediated Iron Transport Can Improve the Adaptation of Rice Seedlings to Iron Deficiency Stress.

Iron (Fe) is essential for normal plant growth and development. In rice, Fe deficiency leads to stunted growth, leaf chlorosis, reduced photosynthetic capacity, and ultimately, yield loss. Most studies have focused on investigating the mechanisms of Fe deficiency responses in rice roots; however, the effects of shoot Fe redistribution on Fe deficiency response remain poorly understood. Phloem transport plays a vital role in distributing Fe to new tissues. To investigate the effects of enhanced phloem-mediated Fe transport on rice adaptability to iron deficiency, we subjected transgenic lines w

From insect endosymbiont to phloem colonizer: comparative genomics unveils the lifestyle transition of phytopathogenic Arsenophonus strains.

UNLABELLED: Bacteria infecting the plant phloem represent a growing threat worldwide. While these organisms often resist in vitro culture, they multiply both in plant sieve elements and hemipteran vectors. Such cross-kingdom parasitic lifestyle has emerged in diverse taxa via distinct ecological routes. In the genus Arsenophonus, the phloem pathogens "Candidatus Arsenophonus phytopathogenicus" (Ap) and "Ca. Phlomobacter fragariae" (Pf) have evolved from insect endosymbionts, but the genetic mechanisms underlying this transition have not been explored. To fill this gap, we obtained the genomes

Ethylene Contributes to maize insect resistance1-Mediated Maize Defense against the Phloem Sap-Sucking Corn Leaf Aphid.

Signaling networks among multiple phytohormones fine-tune plant defense responses to insect herbivore attack. Previously, it was reported that the synergistic combination of ethylene (ET) and jasmonic acid (JA) was required for accumulation of the maize insect resistance1 (mir1) gene product, a cysteine (Cys) proteinase that is a key defensive protein against chewing insect pests in maize (Zea mays). However, this study suggests that mir1-mediated resistance to corn leaf aphid (CLA; Rhopalosiphum maidis), a phloem sap-sucking insect pest, is independent of JA but regulated by the ET-signaling

Carbohydrate-Binding Properties and Antimicrobial and Anticancer Potential of a New Lectin from the Phloem Sap of Cucurbita pepo.

A Cucurbita phloem exudate lectin (CPL) from summer squash (Cucurbita pepo) fruits was isolated and its sugar-binding properties and biological activities were studied. The lectin was purified by affinity chromatography and the hemagglutination assay method was used to determine its pH, heat stability, metal-dependency and sugar specificity. Antimicrobial and anticancer activities were also studied by disc diffusion assays and in vivo and in vitro methods. The molecular weight of CPL was 30 ± 1 KDa and it was stable at different pH (5.0 to 9.0) and temperatures (30 to 60 °C). CPL recovered its

Systemic infection of tobacco mosaic virus is limited by upward transport and phloem unloading without coat protein.

Coat protein (CP) is widely viewed as essential for systemic infection of tobacco mosaic virus (TMV), yet the mechanistic basis of this requirement remains incompletely resolved. In this study, we show that TMV can still achieve long-distance vascular transport in the absence of CP, but systemic infection becomes intermittent because the processes underlying "systemic movement" can fail at multiple, separable steps. By resolving movement into phloem loading, vascular translocation, and phloem exit, we show that CP deficiency does not completely block entry into or transport within the vasculat

Aphid Probing and Phloem Feeding Trigger Distinct Plant Immune Programs, Identifying a Broad‑Spectrum Trypsin Protease Inhibitor.

Aphid feeding begins with brief exploratory probing and transitions to prolonged phloem ingestion, yet whether these distinct phases trigger different plant immune programs remains unknown. We show that probing and phloem feeding are associated with distinct plant immune cascades. Under probing-enriched conditions, we observed selective activation of jasmonate (JA) signaling, accompanied by significant ROS bursts and vascular callose deposition, whereas phloem feeding elicits only marginal salicylic acid accumulation. Probing-enriched conditions imposes severe fitness penalties on aphids, a ph

Dual targeting of pathogen load and callose deposition improves phloem function and carbon allocation in citrus greening disease.

Huanglongbing (HLB), caused by Candidatus Liberibacter asiaticus (CLas), disrupts sugar transport in citrus due to bacterial proliferation and excessive callose deposition that blocks sieve elements. Reducing the blockage of phloem may reduce symptoms, but it is unclear whether targeting CLas or suppressing callose deposition is more critical, as both contribute to phloem dysfunction. This study evaluated whether an integrated strategy targeting both CLas and callose-mediated phloem blockage could more effectively restore phloem transport, carbohydrate allocation, and productivity in HLB-affec

High flavonoid-producing tomato reduces whitefly phloem-feeding efficiency by inducing reactive oxygen species accumulation and callose deposition.

The whitefly Bemisia tabaci (Hemiptera: Aleyrodidae) is economically one of the most threatening pests in tomato cultivation, which not only causes direct damage but also transmits many viruses. Breeding whitefly-resistant tomato varieties is a promising and environmentally friendly method to control whitefly populations in the field. Accumulating evidence from tomato and other model systems demonstrates that flavonoids contribute to plant resistance to herbivorous insects. Previously, we found that high flavonoid-producing tomato line deterred whitefly oviposition and settling behaviours, and

Differential effects of plant ontogeny and damage type on phloem and foliage monoterpenes in jack pine (Pinus banksiana).

Coniferous trees have both constitutive and inducible defences that deter or kill herbivores and pathogens. We investigated constitutive and induced monoterpene responses of jack pine (Pinus banksiana Lamb.) to a number of damage types: a fungal associate of the mountain pine beetle (Dendroctonus ponderosae Hopkins), Grosmannia clavigera (Robinson-Jeffrey & R.W. Davidson); two phytohormones, methyl jasmonate (MJ) and methyl salicylate (MS); simulated herbivory; and mechanical wounding. We only included the fungal, MJ and mechanical wounding treatments in the field experiments while all treatme

qPCR-Based Quantification As A Tool For Genetic Selection And Yield-Stable Breeding Against Restricted Phloem Pathogens In Potato.

The emerging phloem‑restricted pathogens Candidatus Arsenophonus phytopathogenicus (ARSEPH) and Candidatus Phytoplasma solani (PHYPSO), transmitted by the planthopper Pentastiridius leporinus, threaten European potato (Solanum tuberosum L.) production. This study used quantitative real‑time PCR (qPCR) to quantify ARSEPH and PHYPSO in 40 varieties grown at two field locations in southwest Germany under high natural P. leporinus pressure. Tuber assays revealed near‑complete infection (95.7-100%), dominated by co‑infections, with ARSEPH more prevalent and generally reaching higher copy numbers th

Both Jasmonic Acid- and Abscisic Acid-Mediated Signalling Pathways Regulate the Ovicidal Defence of Plants Against Phloem-Feeding Insects.

Plants perceive signals associated with herbivore eggs and in response initiate ovicidal defence. However, which phytohormone pathways regulate this defence and which defensive compounds dominate it remains largely unknown. Here, we found that the hatching rate of eggs of white-backed planthopper (WBPH) Sogatella furcifera was significantly lower on a japonica rice variety P81 than an indica rice variety NB44. When infested by WBPH, P81 plants showed higher jasmonic acid (JA) and abscisic acid (ABA) responses than did NB44 plants; moreover, P81 plants produced the ovicidal compound benzyl benz

Phloem Metabolites of Prunus Sp. Rather than Infection with Candidatus Phytoplasma Prunorum Influence Feeding Behavior of Cacopsylla pruni Nymphs.

Phytoplasmas are specialized small bacteria restricted to the phloem tissue and spread by hemipterans feeding on plant sieve tube elements. As for many other plant pathogens, it is known that phytoplasmas alter the chemistry of their hosts. Most research on phytoplasma-plant interactions focused on the induction of plant volatiles and phytohormones. Little is known about the influence of phytoplasma infections on the nutritional composition of phloem and consequences on vector behavior and development. The plum psyllid Cacopsylla pruni transmits 'Candidatus Phytoplasma prunorum', the causing a

Identification of expression profiles of sorghum genes in response to greenbug phloem-feeding using cDNA subtraction and microarray analysis.

The phloem-feeding by greenbug (Schizaphis graminum) elicits unique interactions with their host plants. To investigate the expression profiles of sorghum genes responsive to greenbug feeding, two subtractive cDNA libraries were constructed through different combinatorial subtractions in a strong greenbug resistance sorghum M627 line and a susceptible Tx7000 line with or without greenbug infestation. A total of 3,508 cDNAs were selected from the two cDNA libraries, and subsequent cDNA microarray and northern blot analyses were performed for identification of sorghum genes responsive to greenbu

Elevation-Driven Adaptive Shifts in Phloem Anatomy and Non-Structural Carbohydrates: Differential Responses of Pinus koraiensis and Acer mono.

Phloem is crucial for long-distance transport, but its coordinated phenological, anatomical, and nonstructural carbohydrate (NSC) responses to environmental changes remain unclear. Along a 750-1150 m elevational gradient on the Changbai Mountains of China, we monitored phloem growth, anatomy, and total NSC (soluble sugars and starch) in Pinus koraiensis and Acer mono during the 2024 growing season. Elevation delayed phloem formation and compressed the growing season in both species. However, the two wood types diverged in adaptive strategies. P. koraiensis employed an active structural compens
