Photoperiodic responses in Djungarian hamsters (Phodopus sungorus): importance of light history for pineal and serum melatonin profiles.

Male Djungarian hamsters with previous light experience of long photoperiods of 16 h of light per day (16L:8D) or short photoperiods (8L:16D) were transferred either to the opposite photoperiods or to intermediate photoperiods (14L:10D). It was demonstrated that the same intermediate photoperiod could exert inhibitory or stimulatory effects on coat color, body weight, and the reproductive system, dependent on the previous light history. The response was graduated in accordance with the degree of change in day length. Despite opposite responses to the same photoperiod, diurnal patterns of melat

Influence of prenatal photoperiods on postnatal reproductive responses to daily infusions of melatonin in the Siberian hamster (Phodopus sungorus).

Prepubertal reproductive development in juvenile male Siberian hamsters can be strongly influenced by photoperiod information received during gestation. Information transmitted from the mother hamster to her fetuses appears to modify the photoperiodic mechanism of the developing hamsters so that they may respond differently to certain intermediate day lengths [i.e. 14 h of light and 10 h of darkness/day (14L)] experienced after birth depending on whether gestation occurred in longer or shorter day lengths. In adult and juvenile hamsters, the duration of the nocturnal elevation of pineal and se

Trans-pineal microdialysis in the Djungarian hamster (Phodopus sungorus): a tool to study seasonal changes of circadian clock activities.

The Djungarian hamster is a highly seasonal small mammal. The rhythmic secretion of melatonin by the pineal gland is under control of the circadian clock, conveying the photoperiodic message to the organism. Trans-pineal microdialysis permits the in vivo study of this well-defined and precise clock output by measuring melatonin release directly in the pineal gland. The aim of this study was to adapt this method to the Djungarian hamster in order to monitor clock properties during photoperiodic changes. Male adult Djungarian hamsters were kept in a long photoperiod (LD 16:8) and melatonin relea

Expression of daily torpor in the Djungarian hamster (Phodopus sungorus) is correlated with the testosterone-driven vasopressin content in the lateral septum.

In the Djungarian hamster (Phodopus sungorus), torpor bouts appear after exposure to short photoperiods (SP) for 11 weeks. Castration accelerates the occurrence of daily torpor when performed between the 1st week before and the 4th week after transfer to SP. In animals castrated 8 weeks before transfer to SP, daily torpor was observed nearly at the same time as in the control group. Vasopressin-immunoreactivity (VP-ir) in the lateral septum (LS) of the Djungarian hamster was examined. Dense VP-ir was seen in sexually active and in sexually inactive hamsters implanted with testosterone (T) caps

Nuclear receptor sites for vitamin D-soltriol in midbrain and hindbrain of Siberian hamster (Phodopus sungorus) assessed by autoradiography.

Autoradiograms were prepared from midbrains and hindbrains of male and female Siberian hamsters (Phodopus sungorus), kept under short-day or long-day illumination, after injection of tritium-labeled 1,25-dihydroxycholecalciferol (vitamin D, soltriol). Concentration and retention of radioactivity was noted in nuclei of certain neurons, glial cells, and ependymal cells, and in choroid epithelium. Labeled neurons of varying intensity were found throughout the brainstem in distinct populations at characteristic topographical sites, which include cranial nerve motor nuclei, the nucleus (n.) reticul

Thyroid medullary carcinoma of the Djungarian hamster Phodopus sungorus. Ultrastructural evidence for the production of normal and atypical intracellular granules.

We have carried out an electron microscopic and immunocytochemical study of thyroid medullary carcinoma arising spontaneously in the Djungarian hamster, Phodopus sungorus. At the ultrastructural level the cytoplasm of tumor cells contained numerous round to slightly elongated, dense-cored secretory granules. The number of secretory granules differed from cell to cell in the tumor, being scanty in some cells but more or less abundant in most. Electron microscopic-immunocytochemistry demonstrated that all dense-cored secretory granules in all tumor cells exhibited calcitonin immunoreactivity. In

Molecular characterization of circannual interval timing in the preoptic area and anteroventral periventricular nucleus in the Djungarian hamster (Phodopus sungorus).

Most temperate animals exhibit seasonal rhythms in reproductive physiology and behaviour. Gonadotropin-releasing hormone (GnRH) cells in the preoptic area (POA) and kisspeptin (Kiss) cells in the anteroventral periventricular (AvPv) nucleus are critical for timing photoperiod-induced changes in seasonal reproduction. In response to prolonged exposure to photoperiod cues, many rodents exhibit endogenously generated programmed changes in physiology that reflect a circannual timing mechanism. Here we used transcriptome sequencing coupled with measurements of surface body temperature and testes ma

Daily torpor alters multiple gene expression in the suprachiasmatic nucleus and pineal gland of the Djungarian hamster (Phodopus sungorus).

Circadian rhythms are still expressed in animals that display daily torpor, implying a temperature compensation of the pacemaker. Nevertheless, it remains unclear how the clock works in hypothermic states and whether torpor itself, as a temperature pulse, affects the circadian system. To reveal changes in the clockwork during torpor, we compared clock gene and neuropeptide expression by in situ hybridization in the suprachiasmatic nucleus (SCN) and pineal gland of normothermic and torpid Djungarian hamsters (Phodopus sungorus). Animals from light-dark (LD) 8ratio16 were sacrificed at 8 time po

Diurnal changes in the content of indoleamines, catecholamines, and methoxyindoles in the pineal gland of the Djungarian hamster (Phodopus sungorus): effect of photoperiod.

Previous studies in Syrian hamster have shown that the correlations between the daily fluctuations in the contents of pineal indoleamines and methoxyindoles are influenced by the photoperiod, and that dopamine may play a role in the regulation of pineal function. The present study investigated the 24 hour changes in the content of 5-hydroxytryptophan (5-HTP), serotonin (5-HT), 5-hydroxyindoleacetic acid (5-HIAA), N-acetylserotonin (NAS), melatonin, 5-methoxytryptophol (5-MT), noradrenaline (NA), dopamine (DA), and 3,4-dihydroxyphenylacetic acid (DOPAC) in the pineal gland of female Djungarian

Increased oxidation of pineal serotonin as a possible explanation for reduced melatonin synthesis in the aging Djungarian hamster (Phodopus sungorus).

Groups of young adult (1.7 months) and old adult (9.7 months) female Djungarian hamsters were exposed to short-day photo-periods (SD; 8L:16D) or long-day photoperiods (LD; 16L:8D) for 6 weeks. Pineal and serum melatonin rhythms were dampened in old SD animals when compared with young SD animals. The melatonin precursor serotonin showed diurnal variations with opposite phases to melatonin synthesis. However, pineal serotonin levels were even lower in old SD animals than in young SD animals, contrary to the expected accumulation of the precursor. The synthesis of a metabolite of serotonin, 5-hyd

Substance P-like immunoreactivity in the hypothalamic suprachiasmatic nucleus of Phodopus sungorus--relation to daytime, photoperiod, sex and age.

The immunohistochemical distribution of substance P (SP) in the hypothalamic suprachiasmatic nucleus (SCN) was studied in adult male and female Djungarian hamsters (Phodopus sungorus) held under either long or short photoperiods. Intact animals were killed by perfusion with a fixative at the middle of the light or dark periods, respectively. The tissue was processed by routine immunohistochemical methods. Perikarya exhibiting SP-like immunoreactivity (LI) were found in the SCN of animals of all groups. These cell bodies predominantly were restricted to a distinct portion of the nucleus extendi

Influence of short photoperiods on reproductive organs and estrous cycles of normal and pinealectomized female Djungarian hamsters, Phodopus sungorus.

This study was designed to analyze morphological changes in the reproductive organs and endocrine changes in the serum and pituitaries of female Djungarian hamsters during regression, the period of involution and early spontaneous recrudescence in response to short photoperiods. In addition, the involvement of the pineal gland on the inhibitory response to short photoperiods was investigated. In the first study, 63 adult female hamsters were divided into five groups, which were either pinealectomized at the time of transfer or 11 wk after transfer into short photoperiods, or exposed to differe

Alteration of the bed nucleus of the stria terminalis (BNST) in young female Djungarian hamsters (Phodopus campbelli) exposed to adult males.

Central components of the vomeronasal system appear to mediate extrahypothalamic control of gonadotropin release during male-induced acceleration of puberty onset in female rodents. In order to describe the bed nucleus of the stria terminalis (BNST) and to determine whether this component of the vomeronasal system is altered following early puberty onset, the anterior BNST of female Djungarian hamsters (Phodopus campbelli) was subdivided into three regions: medial, lateral, and ventral. The cross-sectional area, soma size, and neuronal density of each subdivision was compared among female hams

Pineal and pituitary involvement in the photoperiodic regulation of body weight, coat color and testicular size of the Djungarian hamster, Phodopus sungorus.

Hypophysectomy of 40-day-old male hamsters kept under long photoperiods resulted in a change in coat color from the brownish summer coat to the whitish winter coat, within 5 wk of the surgery, cessation of growth, and, most frequently, an additional loss in body weight. The testes of the hypophysectomized animals were involuted within 14 days after surgery. Continuous substitution of prolactin by implantation of two anterior pituitaries under the kidney capsule was able to reverse partly or totally the inhibitory effects of hypophysectomy on coat color and body weight, although there was no in

Sustained response of pineal melatonin synthesis to a single one-minute light pulse during night in Djungarian hamsters (Phodopus sungorus).

Previous experiments have demonstrated that short light pulses during the night suppress pineal melatonin formation almost completely for the remainder of the night. Here, the effects of a single 1 min light pulse during the night on melatonin synthesis during the following night was investigated in Djungarian hamsters (Phodopus sungorus). It is shown that the melatonin pattern during the consecutive night is suppressed to a greater extent than that caused by acute light exposure, indicating a very effective light memory of the melatonin-generating neuronal network, possibly a key to understan

1,25(OH)2 vitamin D3 binding sites in male sex organs of the Siberian hamster (Phodopus sungorus). An autoradiographic study.

Using autoradiography, binding sites for 1,25(OH)2 vitamin D3 are found in certain genital organs of male Siberian hamsters (Phodopus sungorus), in particular in basal epithelial cells and fibroblasts of the lamina propria of prostate glands. Scattered labeled cells are also present in the epithelium of coagulation and urethral glands. In contrast to the findings in mice, under the conditions of the experiment, 1,25(OH)2 vitamin D3 binding sites are not recognizable in other accessory sex glands and gonads. Effects of age, photoperiod and follicle-stimulating hormone on lactate production by cultured Sertoli cells from prepubertal Siberian hamsters (Phodopus sungorus).

To define a functional difference in Sertoli cells of animals exposed to different photoperiodic conditions, we isolated Sertoli cells from the testes of juvenile Siberian hamsters and cultured them in serum-free medium. In all age groups studied, Sertoli cells isolated from hamsters with delayed and normal puberty responded to follicle-stimulating hormone (FSH) with an increase in lactate production. The increase in lactate production induced by 1000 ng FSH ml-1 was significantly greater in Sertoli cells isolated from hamsters with delayed puberty than in those with normal puberty. These resu

Influence of photoperiod on pineal melatonin synthesis, fur color, body weight, and reproductive function in the female Djungarian hamster, Phodopus sungorus.

In order to investigate female Djungarian hamsters' reactions to changes of the photoperiod, the following two experiments were performed. Experiment I: Age-matched female hamsters were exposed to either short (8L:16D) or long days (16L:8D) for 38 weeks. Initially, the short-day group showed a decline in body weight, associated with changes in gonadal function and fur color. This was not maintained by the short-day group which returned, on the most part, to long-day levels, thus becoming insensitive to this regressive lighting regimen.
