The Phospholipid N-Methyltransferase and Phosphatidylcholine Synthase Pathways and the ChoXWV Choline Uptake System Involved in Phosphatidylcholine Synthesis Are Widely Conserved in Most, but Not All Brucella Species.

The brucellae are facultative intracellular bacteria with a cell envelope rich in phosphatidylcholine (PC). PC is abundant in eukaryotes but rare in prokaryotes, and it has been proposed that Brucella uses PC to mimic eukaryotic-like features and avoid innate immune responses in the host. Two PC synthesis pathways are known in prokaryotes: the PmtA-catalyzed trimethylation of phosphatidylethanolamine and the direct linkage of choline to CDP-diacylglycerol catalyzed by the PC synthase Pcs. Previous studies have reported that B. abortus and B. melitensis possess non-functional PmtAs and that PC

Three phosphatidylcholines as a potential early-diagnostic signature for hyperglycemia in preschool-aged children: a cross-sectional pilot study.

OBJECTIVE: Although lipid metabolism dysregulation commonly accompanies elevated blood glucose, lipidomic studies in hyperglycemic children remain limited. We hypothesized that distinct lipid metabolic signatures and derived lipid ratios are associated with pediatric hyperglycemia and may aid in its identification. METHODS: Based on the Infant Development and Health Cohort, 59 preschool-aged children (3-4 years) with fasting plasma glucose (FPG) ≥ 5.6 mmol/L were randomly selected from 210 participants, alongside 60 matched healthy controls (FPG < 5.6 mmol/L). Semiquantitative lipidomics was

Amphotericin B and monoacyl-phosphatidylcholine form a stable amorphous complex.

Amphotericin B (AmB) is a "life-saving" medicine for the treatment of invasive fungal infections and visceral leishmaniasis. To date, all marketed AmB formulations require parenteral administration, which causes high rates of acute infusion-related side effects and dose-dependent nephrotoxicity. The development of an oral AmB formulation will entail numerous advantages including increased patient compliance, eliminated infusion-related toxicities and reduced nephrotoxicity. Unfortunately, the gastrointestinal absorption of AmB is negligible due to its extremely low solubility in both aqueous a

Reduced phosphatidylcholine synthesis suppresses the embryonic lethality of seipin deficiency.

Seipin plays a vital role in lipid droplet homeostasis, and its deficiency causes congenital generalized lipodystrophy type II in humans. It is not known whether the physiological defects are all caused by cellular lipid droplet defects. Loss-of-function mutation of seip-1, the Caenorhabditis elegans seipin ortholog, causes embryonic lethality and lipid droplet abnormality. We uncover nhr-114 and spin-4 as two suppressors of seip-1 embryonic lethality. Mechanistically, nhr-114 and spin-4 act in the "B12-one-carbon cycle-phosphatidylcholine (PC)" axis, and reducing PC synthesis suppresses the e

The studies on the membrane activity of triester of phosphatidylcholine in artificial membrane systems.

Due to the increasing number of infections together with the appearance of bacteria exhibiting multi-drug resistance, new antibiotics are being sought. In this context the interest of the cationic lipoids increases because of their amphiphilic structure and positive charge that can stimulates the antibacterial action of these compounds. Thus, in this work we have performed the studies on the effect of one selected triesters of phosphatidylcholine, namely 1,2-dipalmitoyl-sn-glycero-3-ethylphosphocholine (EDPPC), on the model lipid membranes. The investigations included the analysis of the impac

Suitability of phosphatidylcholine-based formulations for liquid filling in hard capsules.

The current study aimed to evaluate whether phosphatidylcholine-based formulations have suitable properties for liquid filling in hard capsules. Liquid or semisolid formulations were obtained with Miglyol 812 (medium chain triglycerides) or Softisan 378 (semisolid hard fat), respectively, in combination with three different phospholipids (Phospholipon 90 G, Lipoid S 75 or Lipoid S LPC 80) and ethanol. Suitability for the filling process was evaluated by rheological characterization and stringing tests. Liquid and semisolid mixtures with Lipoid S 75 or Phospholipon 90 G (also with addition of L

Loss of Phosphoethanolamine N-Methyltransferases Abolishes Phosphatidylcholine Synthesis and Is Lethal.

Plants use several pathways to synthesize phosphatidylcholine (PC), the major phospholipid of eukaryotic cells. PC has important structural and signaling roles. One pathway plants use for synthesis is the phospho-base methylation pathway, which forms the head-group phosphocholine through the triple methylation of phosphoethanolamine (PEA) catalyzed by phosphoethanolamine N-methyltransferases (PEAMTs). Our understanding of that pathway and its physiological importance remains limited. We recently reported that disruption of Arabidopsis thaliana PEAMT1/NMT1 and PEAMT3/NMT3 induces severe PC defi

Determination of Phosphatidylcholine Hydroperoxide (PCOOH) as a Marker of Membrane Lipid Peroxidation.

Increasing evidence for phosphatidylcholine hydroperoxide (PCOOH) as a marker of oxidative food deterioration and oxidative diseases has revealed the need for a pure PCOOH standard and a reliable quantification method. Recently, we synthesized the PCOOH isomers 1-palmitoyl-2-(9-hydroperoxyoctadecadienoyl)-sn-glycero-3-phosphocholine, (16:0/9-HpODE PC) and 1-palmitoyl-2-(13-hydroperoxyoctadecadienoyl)-sn-glycero-3-phosphocholine (16:0/13-HpODE PC). Using these standards along with liquid chromatography-tandem mass spectrometry, a reliable quantification method was developed. This mini-review de

Instead the fact that chemical lipolysis through phosphatidylcholin injections really works, this procedure eliminating limited fat deposits remains confidential in France. Inconstant results, necessity to repeat injections, unclear legacy may explain that this very basic procedure remains unsuccessful. We have proceeded to lipolysis injections for five years on a very limited number of patients: in our hands, it may be efficient on puffy cheeks, double chin, superficial cellulitis, liposuction and lipofilling sequellaes. Eyelid bags may also be considered but not recommended. On the other han

Prevention of hepatocarcinogenesis with phosphatidylcholine and menaquinone-4: in vitro and in vivo experiments.

BACKGROUND/AIMS: We examined whether phosphatidylcholine inhibited growth of hepatic cancer, as previously shown for menaquinone-4 (vitamin K2). METHODS: Growth inhibitions by phosphatidylcholine and/or menaquinone-4 and apoptosis induction by phosphatidylcholine were evaluated in vitro using human hepatic cancer cell lines (Hep-3B, Hep-G2, HuH-7, and Alexander). Effects of these agents were then investigated in male Sprague-Dawley rats against hepatocarcinogenesis induced by diethylnitrosamine plus phenobarbital. All rats were killed to examine livers to evaluate inhibitory potential macrosc

Brucella abortus synthesizes phosphatidylcholine from choline provided by the host.

The Brucella cell envelope is characterized by the presence of phosphatidylcholine (PC), a common phospholipid in eukaryotes that is rare in prokaryotes. Studies on the composition of Brucella abortus 2308 phospholipids revealed that the synthesis of PC depends on the presence of choline in the culture medium, suggesting that the methylation biosynthetic pathway is not functional. Phospholipid composition of pmtA and pcs mutants indicated that in Brucella, PC synthesis occurs exclusively via the phosphatidylcholine synthase pathway. Transformation of Escherichia coli with an expression vector

Improvement of antischistosomal activity of praziquantel by incorporation into phosphatidylcholine-containing liposomes.

Praziquantel (PZQ) is effective against all known species of Schistosomes that infect humans. The failure of mass treatment of schistosomiasis has been attributed to the fact that therapy is not sufficiently long-lasting. This effect may be due to the low bioavailability of PZQ that has a low hydrosolubility and fast metabolism. Liposomes have been used to prolong drug levels, reduce the side effects, direct drugs to specific sites and increase bioavailability after administration. The aim of this work was to study the effect of phosphatidylcholine (PC)-containing liposomes to vehiculate PZQ t

Dietary phosphatidylcholine alleviates fatty liver induced by orotic acid.

OBJECTIVE: We compared the effect of dietary phosphatidylcholine (PC) with that of triacylglycerol (TG), both with the same fatty acid profiles, on fatty infiltration in orotic acid (OA)-induced fatty liver of Sprague-Dawley rats. METHODS: Rats were fed an OA-supplemented diets containing TG (TG+OA group) or PC (20% of dietary lipid, PC+OA group) for 10 d. Rats fed the TG diet without OA supplementation served as the basal group. RESULTS: Administering OA significantly increased the weights and TG accumulation in livers of the TG+OA group compared with the basal group. These changes were att

Phosphatidylcholine prevents postoperative peritoneal adhesions: an experimental study in the rat.

Phosphatidylcholine (PC) is the main constituent of the surface-active material coating peritoneal mesothelium. It may prevent postoperative adhesion formation through production of a lubricant film on mesothelial defects. We therefore examined the effect of its soluble form on surgically induced intraabdominal adhesions in rats. The adhesions were induced at laparotomy by any of four different operative models. PC was administered intraperitoneally (20 mg/rat) or intravenously (20 mg/rat or 50 mg/rat) at the end of the operation and on the second and third postoperative day. It was found that

A cytosolic phospholipase in human neutrophils that hydrolyzes arachidonoyl-containing phosphatidylcholine.

In stimulated neutrophils the production of eicosinoids and the lipid mediator, platelet-activating factor, is thought to be initiated by the activation of a phospholipase A2 which cleaves arachidonic acid from choline-containing glycerophospholipids. Accordingly, studies were undertaken in human neutrophils to characterize phospholipase enzymes that can hydrolyze 1-acyl- and 1-alkyl-linked arachidonoyl-containing phosphatidylcholine (PC). Cellular homogenates were incubated with sonicated dispersions of the arachidonoyl-labeled phospholipid substrates and the hydrolysis of radiolabeled arachi

Lipidomic Sample Preparation to Detect Phosphatidylcholine Alterations in Skeletal Muscle.

Altered phospholipid compositions in skeletal muscle occur in muscular dystrophy and other pathological conditions; however, origins and relationships to the disease course are unclear. Liquid chromatography-mass spectrometry (LC-MS) technologies allow the determination of phospholipid compositions and alteration patterns in disease states. Here, we describe a basic protocol to prepare methanolic extracts from skeletal muscle, which may be analyzed by lipidomic LC-MS in order to detect compositional alterations in phosphatidylcholine, a major phospholipid species of skeletal muscle, as well as

Association of SOGPI in mediating the effect of Phosphatidylcholine on polycystic Ovary Syndrome.

BACKGROUND: Polycystic ovary syndrome (PCOS) is a prevalent endocrine disorder in women of reproductive age, marked by hormonal imbalances and disruptions in glucose and lipid metabolism. Emerging research has indicated a correlation between lipids and PCOS, yet the specific lipid profiles or associated genes identified in various studies vary, and observational data alone cannot establish causation. Therefore, our study seeks to establish a causal association between lipidome and PCOS. METHODS: Data from genome-wide association studies, liposomes, metabolites, and PCOS-related information we
