A facile solvothermal method for the synthesis of multifunctional magnetic CuFeMnO4 nanospheres affinity probe (NSAP) with controllable morphology and size was developed for the first time. The CuFeMnO4 nanospheres combine the brilliant features of Cu2+, Fe3+, and Mn2+ ions, so their multifunction performances are embodied by strong coordination to carboxyl and amine groups of peptides (Cu2+ and Fe3+), special affinity to phosphate groups of phosphopeptides (Fe3+ and Mn2+), and high magnetic responsiveness in a magnetic field. Their potential as an affinity probe was evaluated for highly effec

Gout is a painful form of inflammatory arthritis characterized by the deposition of monosodium urate (MSU) crystals in the joints. The aim of this study was to investigate the effect of peptide P140 on the inflammatory responses in crystal-induced mouse models of gout and cell models including MSU-treated human cells. Injection of MSU crystals into the knee joint of mice induced neutrophil influx and inflammatory hypernociception. Injection of MSU crystals subcutaneously into the hind paw induced edema and increased pro-inflammatory cytokines levels. Treatment with P140 effectively reduced hyp

A novel multifunctional composite comprising mesocrystalline SnO2 nanorods (NRs) vertically aligned on reduced graphene oxide (rGO) sheets was synthesized and developed for sequential capture of endogenous peptides and phosphopeptides. With the hydrophobicity of rGO and high affinity of SnO2 nanorods, sequential enrichment of endogenous peptides and phosphopeptides could be easily achieved through a modulation of elution buffer. With this multifunctional nanomaterial, 36 peptides were observed from diluted bovine serum albumin (BSA) tryptic digest and 4 phosphopeptides could be selectively cap

In this work, we synthesized α-Fe(2)O(3)@SnO(2) core-shell structure nanotubes by a facile two-step hydrothermal method for selective enrichment of phosphopeptides. The core-shell structure nanotubes were found to have outer diameters of 90-110 nm, thicknesses of 30 nm, and lengths of 260-280 nm. SnO(2) nanoparticles with an average crystallite size of around 5 nm form a layer of about 10 nm on both walls of the α-Fe(2)O(3) nanotubes. To demonstrate their ability for selective enrichment of phosphopeptides, we applied α-Fe(2)O(3)@SnO(2) nanotubes to the isolation and enrichment of the phosphop

Dental caries is a common disease resulting from tooth demineralization caused by bacterial plaque. Probiotics have shown great potential against caries by regulating the balance of oral flora. However, obstacles such as poor colonization and lysozyme sensitivity in oral cavity hinder their further application. In this study, an efficient layer-by-layer surface coating of tannic acid (TA)-Ca2+ and casein phosphopeptide (CPP) was applied to the probiotic Bifidobacterium infantis (BI) with potential anti-caries activity. Multi-functionalized probiotics thus prepared (called BI@TA-Ca2+@CPP) exhib

Spermatozoa are functionally inert when they emerge from the testes. Functional competence is conferred upon these cells during a post-testicular phase of sperm maturation in the epididymis. Remarkably, this functional transformation of epididymal spermatozoa occurs in the absence of nuclear gene transcription or protein translation. To understand the cellular mechanisms underpinning epididymal maturation, we have performed a label-free, MS-based, comparative quantification of peptides from caput, corpus and caudal epididymal spermatozoa. In total, 68 phosphopeptide changes could be detected d

A number of germ-line mutations in the BRCA1 gene confer susceptibility to breast and ovarian cancer. However, it remains difficult to determine whether many single amino-acid (missense) changes in the BRCA1 protein that are frequently detected in the clinical setting are pathologic or not. Here, we used a combination of functional, crystallographic, biophysical, molecular and evolutionary techniques, and classical genetic segregation analysis to demonstrate that the BRCA1 missense variant M1775K is pathogenic. Functional assays in yeast and mammalian cells showed that the BRCA1 BRCT domains c

There is a growing need for effective methods in the management of early stage carious lesions. Therefore, the aim of this study was to evaluate the effect of combined casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) and fluoride on white spot lesions (WSLs) compared to fluoride-only interventions. This meta-analysis was performed according to PRISMA guidelines and registered in PROSPERO (CRD42021286245). The Medline, Embase, and Cochrane Central databases were searched until October 17, 2022. Eligible studies were randomized controlled trials. Outcome variables included laser fluor

Introduction Radiotherapy, one of the most important modalities of treating head and neck cancer, causes xerostomia, mucositis, and structural and chemical changes in dental hard tissues. These factors favour the commencement of radiation-induced caries. Aim The study aimed to evaluate the changes in enamel microhardness and fracture resistance after irradiation and the effect of casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) on the same. Materials and methods Fifty extracted human premolar teeth were debrided and cleansed using an ultrasonic scaler. In order to test for fracture

BACKGROUND: Concerns regarding enamel demineralization due to the acidic nature of probiotic tablets have led to investigations into potential solutions. This study aimed to investigate the remineralization potential of probiotic tablets containing Casein phosphopeptide-amorphous calcium phosphate (CPP-ACP).
METHODS: Forty-eight enamel blocks extracted from human teeth were prepared and randomly assigned to four groups (n = 12 per group). Group 1 was treated with a 20 mg CPP-ACP-containing probiotic lozenge dissolved in 5 mL of Phosphate Buffered Saline (PBS) and applied to the enamel surface

Casein is one of the most studied proteins with activity against dental caries. In particular, casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) has shown promising remineralizing properties. In vivo evidence on the anticaries potential of CPP-ACP added to foodstuffs is elusive, nonetheless. Hence, this systematic review aimed at determining whether the use of CPP-ACP added to foodstuffs has a remineralizing or inhibitory action on dental demineralization either in vivo or in situ. The review protocol followed the PRISMA-P criteria and was registered in PROSPERO. PubMed, Scopus, and

INTRODUCTION: Phosphopeptides represent interesting compounds to study and elucidate cellular protein phosphorylation/dephosphorylation processes underlying various signal transduction pathways. However, studies of phosphopeptide action in cells are severely constrained by the negatively charged phosphate moiety of the phosphopeptide resulting in poor transport through the cell membrane. The following study describes the synthesis and radiopharmacological evaluation of two (18)F-labeled phosphopeptide-cell-penetrating peptide dimers. Fe3O4@ZrO2 microspheres with well-defined core-shell structure were prepared and applied for the highly selective enrichment of phosphopeptides from tryptic digest product of proteins. To successfully coat iron oxide microspheres with uniform zirconia shell, magnetic Fe3O4 microspheres were first synthesized via a solvothermal reaction, followed by being coated with a thin layer of carbon by polymerization and carbonization of glucose through hydrothermal reaction. Finally, with the use of the Fe3O4@C microspheres as templates, zirconium isopropoxide was prehydrolyzed and absorbed onto the mic

This study aimed to determine the skin protective effect of egg yolk phosvitin phosphopeptides (PPPs). Phosvitin was separated from the egg yolk, and PPPs were produced using high-temperature and mild-pressure (HTMP) pretreatment and enzyme-sterilization hydrolysis combinations. The elastase and melanogenesis inhibitory activities and anti-inflammatory effects of egg yolk PPPs were determined. All PPPs significantly inhibited elastase activity, but the PPPs prepared with HTMP pretreatment and trypsin-sterilization (HTMP-T-S) combination suppressed the tyrosinase activity the most. PPPs (3 mg/m
