Time-dependent hematological and immunological responses of olive flounder (Paralichthys olivaceus) to experimental infection with Photobacterium damselae subsp. damselae.

Photobacterium damselae subsp. damselae (Pdd) is an opportunistic pathogen in marine aquaculture, but the temporal host responses to infection remain poorly understood. In this study, olive flounder (Paralichthys olivaceus) were challenged with Pdd by intraperitoneal injection, and hematological, biochemical, cellular, humoral, and immune gene expression parameters were analyzed at 1, 2, 3, 5, 7, and 10 days post-infection (dpi). Mortality began at 2 dpi and reached 80% by 6 dpi. Hematocrit levels declined markedly and reached their lowest values at 5 dpi, coinciding with elevated plasma aspar

Virulence Genes and In Vitro Antibiotic Profile of Photobacterium damselae Strains, Isolated from Fish Reared in Greek Aquaculture Facilities.

Bacteria belonging to the species Photobacterium damselae are pathogens of cultured marine fish, causing diseases of high importance, such as Pasteurellosis. Thus, they are considered a major threat to the aquaculture sector. Despite the great importance of fish mariculture for the Greek economy, the distribution and abundance of these bacteria are not well documented in aquaculture units in Greece. Keeping this in mind, the scope of the present study was to investigate the presence, antibiotic profile, and virulence of Photobacterium bacteria originating from a representative sample of maricu

Apoptosis of sea bass (Dicentrarchus labrax L.) neutrophils and macrophages induced by experimental infection with Photobacterium damselae subsp. piscicida.

The infection of sea bass (Dicentrarchus labrax L.) by intraperitoneal (i.p.) injection of the agent of fish pasteurellosis Photobacterium damselae subsp. piscicida resulted in the apoptosis of peritoneal neutrophils and macrophages. All the eight virulent and none of the two non-virulent strains tested exhibited apoptogenic activity. A secreted bacterial protein(s) is a likely candidate as the factor(s) responsible for this activity, since no apoptosis was induced by i.p. injected UV-killed virulent strains and the virulent culture supernatants exhibited a thermo-labile apoptogenic activity i

The efficacy and safety of an oil-based vaccine against Photobacterium damsela subsp. piscicida in yellowtail (Seriola quinqueradiata): a field study.

Protection after intraperitoneal (i.p.) vaccination of yellowtail (Seriola quinqueradiata) against pasteurellosis was studied in a field trial. Yellowtail juveniles captured from the wild or artificially hatched were immunised with an oil-based vaccine against Photobacterium damsela subsp. piscicida, and the fish were observed for 15 weeks after vaccination. Outbreak of pasteurellosis was observed at all five sites (mortality in control group ranged from 7% to 77%), and significant (p<0.01) protection against pasteurellosis relative to non-vaccinated control groups was observed at all sites. Immune Response of Silver Pomfret (Pampus argenteus) CC Chemokine Ligand Gene Family to Photobacterium damselae Subsp. Damselae and Nocardia seriolae Infections.

Chemokines play a crucial role in immune responses by facilitating the migration of cells expressing corresponding chemokine receptors along concentration gradients. Photobacterium damselae subsp. Damselae (PDD) and Nocardia seriolae (NS) are known to induce substantial mortality in silver pomfret populations, yet there exists a dearth of research regarding the immune response of CCLs in PDD- or NS-infected silver pomfret. In our investigation, we identified 10 PaCCLs, which include one fish-specific CCL (PaCCL44). Phylogenetic analysis revealed considerable diversity in CCL types and copy num

Subtractive hybridization reveals a high genetic diversity in the fish pathogen Photobacterium damselae subsp. piscicida: evidence of a SXT-like element.

Photobacterium damselae subsp. piscicida is the causative agent of fish pasteurellosis, a severe disease affecting cultured marine fish worldwide. In this study, suppression subtractive hybridization was used to identify DNA fragments present in the virulent strain PC554.2, but absent in the avirulent strain EPOY 8803-II. Twenty-one genomic regions of this type (that included twenty-six distinct putative ORFs) were analysed by DNA sequencing. Twenty ORFs encoded proteins with homology to proteins in other bacteria, including four homologues involved in siderophore biosynthesis, and four homolo

Transcriptomic profiling reveals dynamic immune responses and potential immune evasion strategies of Photobacterium damselae subsp. Piscicida in spotted sea bass (Lateolabrax maculatus).

Photobacterium damselae subsp. Piscicida (Pdp) is characterized by its broad distribution and is capable of infecting numerous cultured fish species. A previous study revealed the epidemic of Pdp in Lateolabrax maculatus with high mortality. In the current study, comparative transcriptome analysis was carried out on the spleens of L. maculatus infected with Pdp at 6, 12, and 24 h post infection (hpi). The results demonstrated that compared with unchallenged individuals, Pdp infection resulted in 1631, 2085 and 2085 differentially expressed genes (DEGs) being detected at 6, 12 and 24 hpi, respe

AIP56, a novel plasmid-encoded virulence factor of Photobacterium damselae subsp. piscicida with apoptogenic activity against sea bass macrophages and neutrophils.

A strategy used by extracellular pathogens to evade phagocytosis is the utilization of exotoxins that kill host phagocytes. We have recently shown that one major pathogenicity strategy of Photobacterium damselae subsp. piscicida (Phdp), the agent of the widespread fish pasteurellosis, is the induction of extensive apoptosis of sea bass macrophages and neutrophils that results in lysis of these phagocytes by post-apoptotic secondary necrosis. Here we show that this unique process is mediated by a novel plasmid-encoded apoptosis inducing protein of 56 kDa (AIP56), an exotoxin abundantly secreted

16S rRNA gene sequence analysis of Photobacterium damselae and nested PCR method for rapid detection of the causative agent of fish pasteurellosis.

The causative agent of fish pasteurellosis, the organism formerly known as Pasteurella piscicida, has been reclassified as Photobacterium damselae subsp. piscicida on the basis of 16S rRNA gene sequence comparisons and chromosomal DNA-DNA hybridization data; thus, this organism belongs to the same species as Photobacterium damselae subsp. damselae (formerly Vibrio damselae). Since reassignment of P. damselae subsp. piscicida was based on only two strains, one objective of the present work was to confirm the taxonomic position of this fish pathogen by sequencing the 16S rRNA genes of 26 strains

Photobacterium damselae subsp. piscicida challenge reveals dietary animal protein hydrolysates modulate early immune, oxidative and barrier responses in European seabass (Dicentrarchus labrax).

Reducing dependence on marine-derived ingredients like fishmeal, while maintaining the robustness of carnivorous species, like European seabass (Dicentrarchus labrax), remains a challenge for sustainable aquaculture. Photobacterium damselae subsp. piscicida (Phdp) outbreaks are still responsible for significant economic losses, but functional ingredients capable of modulating immune responses may enhance resilience to a Phdp challenge. In this study, fish were fed four balanced diets for 89 days: a plant-based control (CTRL) and three diets including three protein hydrolysates (SHARK, FISH and

Protective efficacy and immune responses induced by a DNA vaccine encoding codon-optimized PPA1 against Photobacterium damselae subsp. piscicida in Japanese flounder.

Photobacterium damselae subsp. piscicida (Pdp) kills many cultured marine fish. As it evolves resistance to existing vaccines, new vaccines are needed. PPA1 is a major antigenic protein of Pdp. Here, DNA vaccines encoding wild-type PPA1 (pPPA1(wt)) and codon-optimized PPA1 (pPPA1(opt)) were constructed and tested against Pdp in Japanese flounder. The mRNA levels of the two antigenic genes at the vaccination site were not different, but the protein level was significantly higher in the pPPA1(opt)-vaccinated fish. In addition, after a bacterial challenge, the levels of interleukin (IL)-1β, IL-6

Characterization of Pathogenic Photobacterium damselae subsp. damselae From Whiteleg Shrimp (Litopenaeus vannamei): An Emerging Cause of Mass Mortality in the Republic of Korea.

Mass mortality events characterized by muscle whitening occurred in farmed whiteleg shrimp (Litopenaeus vannamei) in the Republic of Korea from July to September 2025. After ruling out the major crustacean pathogens listed by the World Organization for Animal Health (WOAH) through molecular screening, three dominant isolates of Photobacterium damselae subsp. damselae were recovered from the affected shrimp. The isolates were identified by 16S rRNA gene sequencing and confirmed by detection of the ureC gene. Phenotypic characterization showed that P. damselae subsp. damselae SNUABM-JN1 grew acr

Complement consumption by Photobacterium damselae subsp. piscicida in seabream, red porgy and seabass normal and immune serum. Effect of the capsule on the bactericidal effect.

A virulent strain of Photobacterium damselae subsp. piscicida (Pdp) was grown without (C form) or with (C+ form) glucose supplementation, the latter to enhance capsule formation. Both forms were resistant to killing by normal serum of seabream, red porgy and seabass. However, the C form was killed by immune serum of all three fish species while the C+ form was killed only by seabream and red porgy sera and to a lesser extent than the C form. Both C and C+ forms consumed complement in normal serum and this consumption was enhanced by precoating the bacteria in specific fish antibody. Complement

The effect of growth medium salinity of Photobacterium damselae subsp. piscicida on the immune response of hybrid bass (Morone saxatilis x M. chrysops).

Photobacterium damselae subsp. piscicida (P. damselae) was grown on various media and the effect of media salinity on certain immune responses of hybrid bass was studied. In Israel, pasteurellosis outbreaks have not been reported at water salinities below 1.38 per thousand. During vaccination experiments the salinity of the medium on which P. damselae is grown, was shown to affect stimulation of the immune system. No correlation was found between antibody response and protection. Bacterial envelopes separated by electrophoresis and subjected to western blot analysis revealed an antibody respon

Severe Wound Infection with Photobacterium damselae ssp. damselae and Vibrio harveyi, following a Laceration Injury in Marine Environment: A Case Report and Review of the Literature.

Marine microorganisms are uncommon etiologies of skin and skin structure infections, that is, wound infections. We report a case of severe wound infection, caused by the marine Photobacterium damselae (Vibrionaceae), in a 64-year-old male patient, returning from Australia. The isolate tested positive for pPHDD1, a plasmid conferring high-level virulence. Furthermore, the wound was coinfected with Vibrio harveyi, a halophile bacterium, which has never been reported from human infections before. Identification was achieved by use of Matrix-Assisted Laser Desorption-Ionization Time of Flight Mass

A siderophore biosynthesis gene cluster from the fish pathogen Photobacterium damselae subsp. piscicida is structurally and functionally related to the Yersinia high-pathogenicity island.

Photobacterium damselae subsp. piscicida, the causative agent of fish pasteurellosis, produces a siderophore which is distinct from that produced by P. damselae subsp. damselae. Using suppression subtractive hybridization, a subsp. piscicida-specific DNA region of 35 kb was identified in strain DI21, and 11 genes were defined: dahP, araC1, araC2, frpA, irp8, irp2, irp1, irp3, irp4, irp9 and irp5. The sequence of the predicted proteins encoded by these genes showed significant similarity with the proteins responsible for the synthesis and transport of the siderophore yersiniabactin, encoded wit

Characterization of Photobacterium damselae subsp. damselae isolated from diseased Asian seabass (Lates calcarifer) and the preliminary development of a formalin-killed cell vaccine.

Asian seabass (Lates calcarifer) is an economically important fish species that is widely cultivated in Thailand. However, aquaculture of Asian seabass is limited by infectious diseases. One of the most serious diseases is photobacteriosis, caused by Photobacterium damselae. Vaccination is recognized as an efficient disease prevention and pathogen control method for strengthening the aquaculture industry. To promote vaccine development, the characterization of pathogenic bacteria and their pathogenesis is required. In this study, isolates of P. damselae were obtained from commercial aquacultur
