Insights into proton pump inhibitor-induced photosensitivity: An observational study in a tertiary photobiology service.

BACKGROUND: Proton pump inhibitors (PPIs) are extensively prescribed but may cause photosensitivity and drug-induced lupus erythematosus (DILE), which can be overlooked as the drug may have been taken for years prior to presentation. METHODS: We reviewed the clinical and investigation findings of patients diagnosed with PPI-induced photosensitivity, diagnosed through the Scottish Photobiology Service. RESULTS: We report 11 patients with median age of onset 61-years and mean duration of PPI ingestion of 5-years [DILE (n = 6), phototoxicity (n = 3) and drug-induced solar urticaria through a lu

Biophysical and photobiological basics of water-filtered infrared-A hyperthermia of superficial tumors.

Thermography-controlled, water-filtered infrared-A (wIRA) is a novel, effective and approved heating technique listed in the ESHO quality assurance guidelines for superficial hyperthermia clinical trials (2017). In order to assess the special features and the potential of wIRA-hyperthermia (wIRA-HT), detailed and updated information about its physical and photobiological background is presented. wIRA allows for (a) application of high irradiances without skin pain and acute grade 2-4 skin toxicities, (b) prolonged, therapeutically relevant exposure times using high irradiances (150-200 mW/cm2)

Chronic actinic dermatitis: a retrospective analysis of 44 cases referred to an Australian photobiology clinic.

A retrospective study was performed to analyse the clinical and photobiological features and therapeutic outcomes of 44 patients with chronic actinic dermatitis who were evaluated over an 8.3-year period. The study population comprised 37 men and seven women with a mean age of 62.7 years (range 26-85 years). The most common abnormal phototest results were decreased minimal erythema doses to both UVA and -B (73.8%), and to UVA alone (14.3%). Twenty-six patients (78.8%) had at least one allergic, photoallergic or combined allergic/photoallergic reaction. A total of 139 positive contact or photoc

PHOTOBIOLOGY OF THE SYMBIOTIC SEA ANEMONE, ANTHOPLEURA ELEGANTISSIMA: DEFENSES AGAINST PHOTODYNAMIC EFFECTS, AND SEASONAL PHOTOACCLIMATIZATION.

The sea anemone Anthopleura elegantissima, which contains photosynthetic symbionts (zooxanthellae), responds both biochemically and behaviorally to the combined environmental stresses of exposure to sunlight and photosynthetically generated hyperbaric O2. Activities of the enzymes superoxide dismutase (SOD) and catalase, which act in concert as defenses against oxygen toxicity, parallel the distribution of chlorophyll. A. elegantissima shows a finely controlled contraction behavior which shades the zooxanthellae and reduces O2 production, but which leaves the body column tissues directly expos

Photophysical and photobiological activities of a porphyrin peptide fraction derived from haemoglobin.

In a previous study, we described the preparation of a porphyrin peptide hydrolysate from haemoglobin, its isolation and its analysis by high performance liquid chromatography (HPLC) and fast atom bombardment (FAB) mass spectrometry. The purpose of the present paper is to test the photosensitizing activity of this fraction. We determined the singlet oxygen quantum yield (phi delta) in order to quantify the efficiency of the porphyrin peptide fraction. The quantum yield is about phi(1O2)=0.06. An analysis of the phototoxic effect on tumour cells in culture was performed and compared with haemat

Chronic actinic dermatitis: a clinical and photobiological study in 6 Japanese patients.

Chronic actinic dermatitis (CAD) has distinct clinical features different from polymorphous light eruption (PLE). In order to clarify the difference between CAD and PLE, not only in clinical histories and findings but also in photobiological and histopathological reactions to phototests, we investigated 6 Japanese patients with CAD and performed provocative phototests which are our standardized methods in diagnosing PLE. On provocative phototests in CAD, pruritic papules were reproduced with smaller doses of UVB, at longer hours after irradiation (48-72 h) and they lasted for more days than in

The role of PUVA in the treatment of psoriasis. Photobiology issues related to skin cancer incidence.

Photochemotherapy with methoxsalen (8-methoxypsoralen) and long wavelength ultraviolet (UV) radiation (referred to as 'PUVA' for psoralen plus UVA) is commonly used to treat psoriasis and vitiligo. These vastly different diseases respond to the therapy by different mechanisms even though the immediate effects of the therapy--the photomodification of cellular biomolecules--is the same for each. Because psoriasis is not cured by PUVA, patients receive many treatments over their lifetime and have a significantly increased risk for the development of skin cancers (primarily squamous cell carcinoma

Dynamic photobiology: A collection of experimental protocols for time-resolved structural studies on light-activated proteins.

The study of proteins that sense and respond to light reveals how nature has harnessed light for function while also mitigating its damaging effects through finely tuned molecular mechanisms. Time-resolved optical spectroscopy and time-resolved X-ray solution scattering and diffraction techniques are well-positioned to probe functional transitions triggered by light in photoactive proteins. However, integrating both fields presents technical and conceptual difficulties for many researchers. This FEBS Open Bio 'In the Limelight' special issue, Dynamic Photobiology, showcases five research proto

Photobiology, photodermatology and sunscreens: a comprehensive overview. Part 1: damage from acute and chronic solar exposure.

Sun exposure of the skin triggers several inflammatory pathways via a multitude of photochemical and photobiological effects. Furthermore, local and systemic immune suppression develops. The main clinical effects of UV exposure can be classified schematically into immediate, including sunburn, tanning, vitamin D production and exacerbation of inherited and acquired photosensitive skin disorders and long-term, including solar ageing and skin cancer. The protection against solar radiation is afforded by a healthy behavior of reasonable sun avoidance and the use of topical sunscreens as well as t

Development of different human skin colors: a review highlighting photobiological and photobiophysical aspects.

Skin color has changed during human evolution. These changes may result from adaptations to solar ultraviolet radiation (protection of sweat glands, sunburn, skin cancer, vitamin D deficiency, defence against microorganisms, etc.), and/or sexual selection. Migration to areas with high levels of UV is associated with skin darkening, while migration to areas with low levels has led to skin lightening. However, other factors may have played roles. Temperature and food have probably been secondary determinants: heat exchange with the environment is dependent on ambient temperature, and a high inta

Actinic reticuloid. A clinical photobiologic, histopathologic, and follow-u study of 16 patients.

We report a detailed clinical, histopathologic, and photobiologic study of 16 Dutch patients with actinic reticuloid. All were middle-aged or elderly men who had persistent plaques on light-exposed skin only (two patients), extension of lesions to nonexposed areas (four patients), or prolonged or persistent episodes of erythroderma (10 patients). They were sensitive to UVB, UVA, and visible light. In 10 of 13 patients tested, the dermal infiltrate contained predominantly suppressor T cells. Many also had a reversed helper-suppressor T cell ratio. Circulating lymphocytes with deeply indented nu

Violet-Blue Light Photobiological Effect on Cultured Corneal and Pigment Retinal Cells.

Artificial optical radiation, spanning from 100 nm to 1 mm, encompasses ultraviolet (UV) and infrared (IR) light. UV light is well known for its risks on the skin and eyes. Recently, there has been growing interest in light at 405 nm (violet-blue light, VBL) due to its antimicrobial properties and perceived safety for mammalian cells when administered in controlled amounts. This research delved into the impact of 405 nm VBL on corneal and retinal pigment epithelial cell cultures. ARPE-19 and corneal BCE C/D 1b cells were exposed to VBL for varying doses, according at different exposure times,

Skin type specific photobiological response to visible light is mediated by constitutional melanin.

BACKGROUND: Visible light (VL) induces varying photobiological responses between skin types, likely influenced by inherent melanization. Individual typology angle (ITA) objectively measures skin types. We hypothesize that epidermal melanin content and distribution determine VL response. OBJECTIVES: This study describes clinical and histologic responses to VL and examines the potential role of melanin in the underlying mechanistic pathways. METHODS: We grouped enrolled participants by ITA (Light = 5, Intermediate = 4, Dark = 7) per colorimetry (CR-400, Konica Minolta). Photoprotected sites we

Photobiological properties of phthalocyanine photosensitizers Photosens, Holosens and Phthalosens: A comparative in vitro analysis.

Photobiological properties of phthalocyanine photosensitizers, namely, clinically approved Photosens and new compounds Holosens and Phthalosens were analyzed on transitional cell carcinoma of the urinary bladder (T24) and human hepatic adenocarcinoma (SK-HEP-1). Photosens is a sulfated aluminum phthalocyanine with the number of sulfo groups 3.4, which is characterized by a high degree of hydrophilicity, slow cellular uptake, localization in lysosomes and the lowest photodynamic activity. Holosens is an octacholine zinc phthalocyanine, a cationic compound with significant charge. Holosens more

Phototoxicity associated with diclofenac: a photophysical, photochemical, and photobiological study on the drug and its photoproducts.

Diclofenac (1) is a photosensitizing nonsteroidal antiinflammatory drug. Its photodecomposition gives rise to chlorocarbazole 2a. This product undergoes photodehalogenation to 3a in a subsequent step. When the photobiological activities of 1, 2a, and 3a are compared by means of the photohemolysis test, it is clearly observed that chlorocarbazole 2a causes cell lysis with a markedly higher efficiency than the parent drug or the secondary photoproduct 3a. Laser flash photolysis studies suggest that photodehalogenation of 2a occurs from its excited triplet state via quenching by ground-state 2a a
