The oomycete Phytophthora infestans is one of the most destructive phytopathogens globally. It has a proven ability to adapt to changing environments rapidly; however, molecular mechanisms responsible for host invasion and adaptation to new environmental conditions still need to be explored. The study aims to understand the epigenetic mechanisms exploited by P. infestans in response to nitrosative stress conditions created by the (micro)environment and the host plant. To characterize reactive nitrogen species (RNS)-dependent acetylation profiles in avirulent/virulent (avr/vr) P. infestans, a t

Phytophthora palmivora is a destructive oomycete plant pathogen with a wide host range. So far, little is known about the factors governing its infection structure development and pathogenicity. From the culture filtrate of a P. palmivora strain isolated from papaya, we identified a secreted glycoprotein of 15 kDa, designated as Ppal15kDa, using liquid chromatography tandem mass spectrometry. Two gene variants, Ppal15kDaA and Ppal15kDaB were amplified from a P. palmivora papaya isolate. Transient expression of both variants in Nicotiana benthamiana by agroinfiltration enhanced P. palmivora inf

The oomycete Phytophthora infestans is the causal agent of late blight, the most devastating disease of potato. The importance of vesicle fusion processes and callose deposition for defense of potato against Phytophthora infestans was analyzed. Transgenic plants were generated, which express RNA interference constructs targeted against plasma membrane-localized SYNTAXIN-RELATED 1 (StSYR1) and SOLUBLE N-ETHYLMALEIMIDE-SENSITIVE FACTOR ADAPTOR PROTEIN 33 (StSNAP33), the potato homologs of Arabidopsis AtSYP121 and AtSNAP33, respectively. Phenotypically, transgenic plants grew normally, but showed

Fifty-two Phytophthora isolates from necrotic roots of olives were characterized. Colony morphologies on carrot-agar medium led us to separate them into two groups: A (36 isolates) and B (16 isolates). The optimum growth temperature for Group A was about 21°C, with slow growth at 30°C. In contrast, Group B isolates had an optimum temperature for growth of 26°C, and grew rapidly at 30°C. Growth rates, sporangial and oogonial characteristics of the Group A isolates conformed to P. megasperma "BHR-type" sensu stricto. This designation was supported by a sequence analysis of their ITS rDNA regions

An oligopeptide elicitor from Phytophthora megasperma f.sp. glycinea (Pep-13) that induces phytoalexin accumulation in cultured parsley cells was radioiodinated and chemically cross-linked to its binding site in microsomal and plasma membrane preparations with each of three homobifunctional reagents. Analysis by SDS/PAGE and autoradiography of solubilized membrane proteins demonstrated labeling of a 91-kDa protein, regardless of which reagent was used. Cross-linking of this protein was prevented by addition of excess unlabeled Pep-13. The competitor concentration found to half-maximally reduce

Breeding for Phytophthora fruit rot (PFR) resistance caused by the devastating pathogen Phytophthora capsici remains challenging in watermelon mainly due to phenotyping constraints. To overcome these bottlenecks, we developed and validated genomic selection models using an interspecific recombinant inbred line (RIL, F11) population derived from USVL531-MDR (resistant, Citrullus mucosospermus) × USVL677-PMS (susceptible, Citrullus lanatus) and a segregating F2 population (USVL531-MDR × 'Calhoun Grey'). Mature fruits from both populations were artificially inoculated under controlled conditions

The oomycete Phytophthora capsici causes Phytophthora blight, a major constraint on global pepper production. Our previous observations indicated that pretreating plants with thiamethoxam (TMX) and imidacloprid (IMI) could reduce the incidence of pepper blight, but the underlying mechanisms remained unclear. Here, we investigated how TMX and IMI induced resistance in pepper (Capsicum frutescens) against P. capsici. Both in vitro and in vivo assays demonstrated that TMX and IMI suppressed disease, not by directly impairing pathogen virulence but by inducing systemic resistance in susceptible (C

In this study, we determined the sensitivity of 148 Phytophthora litchii isolates to cyazofamid, yielding a mean EC50 value of 0.0091 ± 0.0028 μg/mL. Through fungicide adaptation, resistant mutants (RMs) carrying the F220L substitution in PlCyt b were derived from wild-type isolates. Notably, these RMs exhibited a lower fitness compared with the parental isolates. Molecular docking analysis further revealed that the F220L change contributed to a decrease in the binding energy between cyazofamid and PlCyt b. The total phenol and flavonoid contents in the litchi pericarp treated with cyazofamid

A cascade formed by phosphorylation events of mitogen-activated protein kinases (MAPKs) takes part in plant stress responses. However, the roles of these MAPKs in resistance of potato (Solanum tuberosum) against Phytophthora pathogens is not well studied. Our previous work showed that a Phytophthora infestans RXLR effector targets and stabilizes the negative regulator of MAPK kinase 1 of potato (StMKK1). Because in Arabidopsis thaliana the AtMPK4 is the downstream phosphorylation target of AtMKK1, we performed a phylogenetic analysis and found that potato StMPK4/6/7 are closely related and are

The mRNA differential display technique was used to identify genes from Habanero pepper (Capsicum chinense Jacq.) seedlings whose expression is modified systemically by infection with the oomycete Phytophthora capsici L. Experiments with different oligonucleotide primer combinations revealed that no single gene was synthesised de novo. Instead, the quantitative accumulation of multiple transcripts was found. From these transcripts, levels of a nitrate reductase (Capsicum chinense nitrate reductase, CcNR), which has a high percentage of identity with other Solanaceae NRs, showed a consistent in

Many filamentous pathogens employ environmental signals to regulate sporulation, maximizing the likelihood that their spores survive to colonize a new host. In the oomycete Phytophthora, little is known of the transcriptional program that underlies sporangia formation and how it is influenced by environmental inputs such as light, humidity, and nutrients. By studying time courses of the potato blight pathogen P. infestans on artificial media and tomato leaves, we identified over 4000 genes up-regulated during sporulation. These are associated with signal transduction, cell structure, vesicle t

Monomeric compounds from Humulus scandens that effectively inhibit Phytophthora nicotianae were isolated, and their antimicrobial effects were analyzed. Methanol extracts were isolated using a combination of activity tracking and chemical separation methods. Compound structures were identified using NMR and other techniques. Antimicrobial activity against P. nicotianae was assessed via the mycelial growth rate method with mycelial morphology further observed using optical microscopy and scanning electron microscopy. Five compounds were isolated from the ethyl acetate (EtOAc) layer of H. scande

Twenty-three novel flavonol derivatives bearing a 1,2,3,4-tetrahydroquinoline (THQ) scaffold were designed and synthesized, and their antifungal activities against eight plant pathogenic fungi were evaluated. Several compounds exhibited 100% inhibition against Sclerotinia sclerotiorum and Phytophthora capsici (Pc). Among them, Q8 showed the most potent activity against Pc with an EC50 value of 4.1 μg mL-1, superior to azoxystrobin (69.0 μg mL-1) and fluopyram (123.2 μg mL-1). In vivo tests on chili fruits demonstrated that Q8 at 200 μg mL-1 provided superior curative and protective effects (73

Phytophthora infestans, the causal agent of late blight in potato and tomato, is a historically devastating pathogen that continues to pose a major threat to global food security. Although conventional fungicides remain a cornerstone of crop protection, growing concerns over their environmental impact and long-term sustainability have driven the search for safer and more selective alternatives. In this context, next-generation fungicides with high specificity and minimal ecological footprint are urgently needed. This study introduces CP32, a small cyclic peptide, identified via yeast two-hybri

Plasmid-mediated DNA transformation is a foundational molecular technique and the basis for most CRISPR-Cas9 gene editing systems. While plasmid transformations are well established for many agricultural Phytophthora pathogens, development of this technique in forest Phytophthoras is lacking. Given our long-term research objective to develop CRISPR-Cas9 gene editing in a forest pathogenic Phytophthora species, we sought to establish the functionality of polyethylene glycol (PEG)-mediated plasmid transformation in five species: P. cactorum, P. cinnamomi, P. cryptogea, P. ramorum, and P. syringa

INTRODUCTION: Phytophthora infestans, the causative agent of late blight disease, has gained notoriety for its destructive potential, leading to substantial losses in potato yields. Although conventional systemic fungicides have been shown to be effective in controlling plant pathogens, growing environmental concerns have prompted the need for more integrated disease management approaches. Hence, in this study, the effectiveness of wild Origanum elongatum extracts as biopesticides was explored in controlling P. infestans and potentially mitigating its devastating impact in planta.

A study of the in vitro sensitivity of 12 isolates of Phytophthora infestans to metalaxyl, azoxystrobin, dimethomorph, cymoxanil, zoxamide and mancozeb, was conducted. The isolates derived from infected potato leaves collected at eight different localities in Serbia during 2005-2007. The widest range of EC(50) values for mycelial growth of the isolates was recorded for metalaxyl. They varied from 0.3 to 3.9 μg mL(-1) and were higher than those expected in a susceptible population of P. infestans. The EC(50) values of the isolates were 0.16-0.30 μg mL(-1) for dimethomorph, 0.27-0.57 μg mL(-1) f

Phytophthora root rot, caused by Phytophthora sojae, is a devastating soilborne disease of soybean (Glycine max). However, the epigenetic regulation of soybean responses to P. sojae remains incompletely understood. Here, using genetic, molecular and biochemical approaches, we characterized the functions of LIKE HETEROCHROMATIN PROTEIN 1 (GmLHP1) and its downstream regulatory network. We demonstrated that GmLHP1, as a reader of H3K27me3, negatively regulates soybean resistance to P. sojae. GmLHP1 binds to H3K27me3 peptides in vitro and colocalizes with H3K27me3 marks genome-wide in vivo. The in

AIMS: Previously, we selected a bacterial strain (GSE09) antagonistic to Phytophthora capsici on pepper, which produced a volatile compound (2,4-di-tert-butylphenol), inhibiting the pathogen. In this study, we identified strain GSE09 and characterized some of the biological traits of this strain in relation to its antagonistic properties against P. capsici. In addition, we examined bacterial colonization on the root surface or in rhizosphere soil and the effect of various concentrations of the volatile compound and strain GSE09 on pathogen development and radicle infection as well as radicle g

A series of ten geranylated benzaldehydes (11-20) was designed, synthesized via Suzuki cross-coupling for the C-C series and SN2 substitution for the C-O derivatives, and structurally characterized. The antifungal activity of all compounds against the phytopathogens Botrytis cinerea and Phytophthora cinnamomi, two important plant pathogens associated with substantial agricultural and economic losses worldwide, was evaluated in vitro using a mycelial growth inhibition assay. Dose-response treatment of the data indicates that (E)-2-((3,7-dimethylocta-2,6-dien-1-yl)oxy)-5-hydroxybenzaldehyde (19)
