Pinealectomy attenuates the effect of restraint on plus maze exploration in rats.

To investigate a possible stress modulation role of the pineal gland, male Wistar albino rats (200-250 g) were submitted to pinealectomy and divided into four groups one week after surgery: i) sham-operated unrestrained animals (N = 14); ii) pinealectomized unrestrained animals (N = 22); iii) sham-operated animals submitted to 2 h of restraint (N = 52); iv) pinealectomized animals submitted to 2 h of restraint (N = 56). Twenty-four hours later the animals were tested in the elevated plus maze for 5 min. Pinealectomized rats submitted to restraint explored the open arms of the maze to a greater

The long-term effect of pinealectomy on the crypts of the rat gastrointestinal tract.

Previously it has been found that rat small bowel crypt cell hyperplasia occurred several weeks after pinealectomy. To determine if this effect was longer-lasting (because of the possible role of the pineal in bowel malignancy) the crypt cell proliferation rate was determined in rat small bowel and colon 6 months after pinealectomy, using a stathmokinetic technique. Although the hyperproliferative effect of pinealectomy was well maintained in the small bowel crypts after 6 months, the hyperproliferative effect in the colonic crypts was much less marked. There is no obvious explanation for thes

Effect of pinealectomy on liver insulin and glucagon receptor concentrations in the rat.

The studies described here were undertaken to characterize the hepatic insulin and glucagon receptors of control (C), pinealectomized (Pn), and melatonin-treated pinealectomized (Pn + Mel) rats. Compared with C rats, an increase in plasma glucose and glucagon levels and a reduction in circulating concentrations of insulin in Pn animals were observed. Melatonin treatment of Pn rats reverses all three parameters toward the normal values. In liver membranes, insulin binding was lower in Pn than in C rats, and glucagon binding was greater in Pn than in C animals; in Pn + Mel rats both insulin and

Effects of pinealectomy and continuous light on mating behavior of male rats.

The role of pinealectomy and continuous light in the regulation of sexual activity was studied in the male rat. The parameters studied in order to evaluate the sexual behavior were initial latency, ejaculatory latency, refractory period, neuromotor activity, number of ejaculations, intromissions (false, real, and total) and percentage of ejaculating male rats. Pinealectomy as well as continuous light induced a marked facilitation of the animal's sexual behavior as shown by the shorter initial latency and refractory period, as well as by the increase in the number of intromissions (real and tot

Aspects of carbohydrate metabolism after pinealectomy in normal and pancreatectomized pigeon.

In order to examine the regulatory function in glucidic metabolism of the pigeon pineal gland the authors used the model of complete pancreatectomy associated or not with pinealectomy and treatment with Epifizan. It results that the pineal gland has a hypoglycemic action and partially compensates the lack of the endocrine pancreas.

[Characteristics of organization of the rat circadian locomotor rhythm after pinealectomy].

As compared with sham operated animals, an increase in night and daytime locomotion and a shift of the acrophase of the circadian locomotor rhythm to earlier night hours were observed in pinealectomized rats. A reorganization of the rhythm spectral characteristics took place with increase in the share of ultradian waves (14-20 h) and decrease in the number of short periodic (2-5 h) fluctuations.

Effects of pinealectomy and the treatment with melatonin on the temporal lobe epilepsy in rats.

The aim of the present work was to analyze the effects of pinealectomy in the development of the epilepsy model induced by pilocarpine in adult male rats. Group I: Wistar male adult rats were submitted to pinealectomy, and 7 days after surgery, these animals received pilocarpine (350 mg/kg, i.p.) to induce three distinct behavioral phases: status epilepticus, seizure-free, and chronic phases. This late, as well as all control groups were continuously video-recorded for 60 days, to study behavior parameters. These animals were killed and the brain sections were processed for Nissl and neo-Timm.

Pinealectomy: behavioral and neuropathological consequences in a chronic cerebral hypoperfusion model.

This experiment determined if pinealectomy (PX) affects the consequences of chronic, moderate brain ischemia. Rats were pinealectomized at 25 days of age and trained at 9 months on a tactile radial maze. They then underwent permanent occlusion of the common carotid arteries (2VO) or sham surgery, followed by maze retraining and then neurohistological assessment at 16 months. Combined PX + 2VO rats committed more working memory errors on the maze. 2VO itself caused a 10% reduction in hippocampal CA1 pyramidal cell number. PX alone caused a 21% reduction. Combined PX and 2VO caused the greatest

Effect of pinealectomy on immune parameters in rats with Staphylococcus aureus infection.

It is generally accepted that the pineal gland is a neuroendocrine organ. Several recent experiments have shown that the pineal gland has functional and anatomical connections, particularly with the immune system, and therefore the gland is now recognized as an important immunoneuroendocrine organ in both man and animals. The present study investigates the effect of pinealectomy on some immune parameters, including hematological alterations, and the response of the brain tissue against infection caused by Staphylococcus aureus. Experiments were performed on two different age groups of rats (ne

The effects of melatonin therapy on the development of scoliosis after pinealectomy in the chicken.

The mechanism underlying the development of scoliosis after pinealectomy in young chickens is unknown. However, since the main product of the pineal gland is melatonin, melatonin remains an obvious focus in studies designed to discover this mechanism. One confounding factor is that serum melatonin levels are close to zero after pinealectomy but scoliosis does not develop in all chickens that have had this procedure. Therefore, the role of melatonin in the development of scoliosis in chickens after pinealectomy remains controversial. In the current investigation, two pilot studies demonstrated

Circadian regulation of chick electroretinogram: effects of pinealectomy and exogenous melatonin.

Melatonin is an important component of the avian circadian system. This study investigates the effects of pinealectomy (Pin-X) and melatonin implantation (Mel) on electroretinogram (ERG) rhythms in chicks. Feeding rhythms were monitored to obtain a phase reference for ERG recordings. Pin-X and Mel had little or no effect on feeding rhythms. Sham-operated Pin-X and vehicle implantation had no effect on ERG rhythms in the light-dark (LD) cycle or constant darkness (DD). ERG a- and b-wave amplitudes were higher during the day than during the night. The a- and b-wave implicit times were shorter du

The production of scoliosis after pinealectomy in young chickens, rats, and hamsters.

STUDY DESIGN: This study involved weekly radiographic examination of pinealectomized rats, hamsters, and chickens to observe the development of scoliosis. OBJECTIVES: To determine whether pinealectomy produces scoliosis in animals more closely related phylogenetically to humans than to chickens, namely rats and hamsters, which are representative of mammals. SUMMARY OF BACKGROUND DATA: Pinealectomy in 3-day-old chickens has consistently resulted in the development of scoliosis with many characteristics similar to those seen in patients with adolescent idiopathic scoliosis. It has not been det

Melatonin suppresses the pinealectomy-induced elevation of collagen content in a wound.

The pineal gland hormone was shown to be involved in the regulation of repair and connective tissue accumulation in a wound. The hypothesis that melatonin injection could reverse the pinealectomy-induced elevation of the collagen content in scar tissue was verified. The effect of various melatonin doses (3, 30 and 100 micrograms/100 g body wt) on soluble, insoluble and total collagen content in the granulation tissue of a wound was investigated. The collagen level was estimated in pinealectomized rats, as well as in pinealectomized animals treated with melatonin. Ivalon sponges inserted subcut

The effect of pinealectomy and foetal pineal transplantation of collagen ageing in rats.

Recent progress in pineal research emphasized the importance of this circumventricular organ in the phenomenon of ageing as well. The present study was undertaken to elucidate the effects of pineal gland on collagen ageing by spectrofluorometrically measuring collagen-cross-link bound fluorescein. Experiments were performed on three different age groups of rats (3-4 weeks, 8-10 weeks, 15 and more weeks). Fluorescein values were found to increase causally by age in the control group. Eight weeks after sham operation of all the groups fluorescein level increments were shown to be parallel with t

Effect of pinealectomy and melatonin on vasopressin-potentiated passive avoidance in rats.

The pineal indoleamine, melatonin, and the hypothalamic neuropeptide, vasopressin, facilitate passive avoidance behaviour in rats. The similarity of the effects suggest that interaction might occur between the two substances. Therefore, the effect of intraperitoneally applied vasopressin and/or melatonin on one-trial learning passive avoidance behaviour was studied in pinealectomized rats. Intraperitoneal treatment with 500 ng vasopressin 1 hr before the retention test increased passive avoidance latency of sham-operated rats. In pinealectomized rats, an identical amount of vasopressin was ine

Effect of pinealectomy and plane of nutrition on wool growth in Merino sheep.

The effects of surgical pinealectomy and plane of nutrition on wool growth and plasma prolactin concentrations in young Merino wethers were investigated. In young pinealectomized wethers maintained at a low live weight under conditions of minimal stress, the decline of conditioned clean wool production on midside patches was slowed when compared to pineal-intact controls. This difference appeared to be due in part to the observed greater secondary wool follicle density in the pinealectomized wethers; mean fiber diameter was affected to a smaller extent, while staple length growth rate was not

Maternal pinealectomy alters the daily pattern of fetal breathing in sheep.

We have investigated the effect of pinealectomy of the pregnant ewe on the 24-h pattern of fetal breathing activity during late gestation. Fetal breathing movements were recorded during 24-h periods on 18 occasions in 5 pinealectomized ewes and on 24 occasions in 6 pineal-intact ewes between 120 and 145 days gestation. All ewes were fed once daily between 1000 and 1300 h and were kept under a light-dark cycle 12:12 h. There was no significant difference in the mean hourly incidence of fetal breathing movements between the pineal-intact (27.2 +/- 0.5 min/h) and pinealectomized (25.5 +/- 0.6 min

Effect of pinealectomy on the undernutrition-induced suppression of the reproductive axis in rats.

Food restriction is known to sensitize the reproductive axis of rats to the inhibitory action of the pineal gland and its hormone, melatonin. The present study investigated the effect of pinealectomy on the undernutrition-related reproductive suppression. Three groups of animals (control, sham pinealectomy or pinealectomy) were subjected to either 3 weeks of 50% food restriction or ad libitum feeding under a lighting regimen of 14 h light and 10 h dark. At the end of the treatment period, body weight, testes weight, accessory organ weights, serum LH and serum testosterone were determined. In t
